2.Contribution of history-taking, physical examination and laboratory investigation to diagnoses for medical outpatients

Objective To quantitatively evaluate relative contribution of medical history,physical examination and laboratory investigation to diagnoses for medical outpatients.Methods In total,145 medical visitors to the outpatient department of Peking Union Medical College Hospital (PUMCH) during October 10 to 16,2008 were recruited and followed-up for 12 months.Results Nineteen of 145 visitors (13.1%) were lost during the period of follow-up and diagnoses were established for 86 of them (68.3%)finally with medical history and for 20 (15.9%) with physical examination or laboratory investigation,respectively.Confidence index of internists in their correct diagnosis increased to 7.3 with medical history and to 7.9 and 8.7 with physical examination and laboratory investigation in average,respectively.Conclusions Most visitors to internal medicine department could be diagnosed correctly with medical history only.On the basis of physical diagnosis,selection of adequate laboratory investigation for them is critical to improvement of clinical diagnosis.
3.EFFECT OF TASPINE ON WOUND HEALING AND FIBROBLAST PROLIFERATION

Objective To study the effect and mechanism of taspine on wound healing and fibroblast proliferation. Methods The effect of taspine on skin wound was observed in vivo. The different concentration of taspine hydrochloride was added to L929 fibroblast cultivated in vitro, and lactate dehydrogenase was detected and MTT method was applied to observe effect of taspine on fibroblast proliferation. Results The local application of taspine 3 mg/Ml and 1.5 mg/mL accelerated the healing of skin wounded. In vitro, 0.01~0.5 μg/mL of taspine hydrochloride showed no effect on the change of lactate dehydrogenase activity and fibroblast proliferation. Conclusion Taspine is a kind of active alkaloid from leontice robustum which can enhance wound healing, its mechanism on wound healing is not by means of accelerating the proliferation of fibroblast, other mechanisms are necessary for being further studied.

5.Protective Effect of Total Flavone of Camellia Against Cerebral Ischemic Injury

Objective To observe the protective effect of total flavone of Camellia (TFC)against cerebral ischemic injury.Methods Decapitation method and close hypoxia method were used to observe the effect of TFC on anoxic tolerance of mice and step down test was used to observe the effect of TFC on learning and memory; after ischemia, the content of malondialdehyde (MDA)and nitric oxide (NO)and the activity of lactate dehydrogenase (LDH)were detected. Rat models with acute incompletely cerebral ischemia were established by means of ligating right common carotid arteries and effect of TFC on cerebral water volume, permeability of cerebral vessels and cerebral histopathological changes were also observed.Results TFC prolonged the grasping time after decapitation and the survival time after anoxia in mice, and improve the learning and memory during the step down test. TFC decreased MDA and NO contents, counteracted the de creases of LDH activities in the mice cerebral cortex, reduced the water volume and permeability of cerebral vessels in ischemic rats and improve the cerebral hitstopathological changes. Conclusion TFC has protective effects against cerebral ischemic injury and the mechanism may be related to the inhibition of free radicals and NO production.
6.Effects of Danqi Granules combined with Xianling Gubao Capsules on bone metabolism and bone mineral density in patients with postmenopausal osteoporotic vertebral compression fractures after surgery

Objective To observe the effects of Danqi Granules combined with Xianling Gubao Capsules on bone metabolism and bone mineral density (BMD) in patients with postmenopausal osteoporotic vertebral compression fractures (OVCF) after surgery. Methods 90 cases of postmenopausal women with OVCF were randomly divided into the control group and the observation group, 45 cases in each group. The two groups were treated by vertebroplasty (PVP), and after the surgery, the control group was treated with Xianling Gubao Capsules while the observation group was additionally given Danqi Granules. The serum markers of bone metabolism [(25 (OH) D, PINP, β-CTX] and BMD of L2-4 and left femoral neck were detected before and after treatment. Results The level of serum 25 (OH) D in the observation group after treatment was significantly higher while the levels of PINP and β-CTX were significantly lower than those in the control group (P<0.05), and the BMD of L2-4 and left femoral neck of the observation group were significantly higher than those of the control group (P<0.05). There was no significant difference in the incidence of adverse reactions between the two groups. Conclusion Danqi Granules combined with Xianling Gubao Capsules can significantly accelerate fracture recovery, and improve bone mineral density in the treatment of postmenopausal women with OVCF.
7.Effect of liver detoxification soup combined with compound glycyrrhizin on hepatitis related factors in patients with chronic hepatitis B

Objective To investigate the effect of liver detoxification soup combined with compound glycyrrhizin on hepatitis-related factors in patients with chronic hepatitis B.MethodsFrom April 2014 to August 2015, 86 patients with chronic hepatitis B were enrolled in this study.Random number table was divided into observation group and control group (43 cases).The control group was given routine glycyrrhizin and other conventional (ALT), serum total bilirubin (TBil), serum albumin transferase (ALT) and serum total bilirubin (TBil) were measured before and after treatment.(HA), laminin (LN), type Ⅳ collagen (Ⅳ-C) and type Ⅲ procollagen (PCⅢ) were used to analyze the effect of hepatic fibrosis, And observe adverse reactions.ResultsThe effective rate of the observation group was 93.02% higher than that of the control group (76.74%, P<0.05).After treatment, the patients were treated with chest discomfort, emotional depression, epigastric fullness, fatigue, ALT (35.28±1.35) U/L, HA (165.25±1.44) mg/L, and the treatment group (P<0.05).After treatment, the levels of TBil (30.34±1.59)μmol/L, Alb (48.32±1.33)g/L, and the level of serum was significantly higher than that of the control group (P<0.01).The incidence of adverse reactions was 11.63% and 6.98% in the two groups.ConclusionTraditional Chinese medicine hepatectomy decoction combined with compound glycyrrhizin can effectively treat chronic hepatitis B and improve the level of serum hepatitis-related factors, and the adverse reaction is light, it is worthy of clinical application.
8.Effect of total flavone of ginkgo biloba on apoptosis induced by cerebral ischemia in rats

BACKGROUND: Total flavone of ginkgo biloba(TFG) can affect on free radical, but the effect on apoptosis induced by cerebral ischemia is unclear.OBJECTIVE: To study the inhibitory effect of TFG on apoptosis induced by cerebral ischemia.DESIGN: Completely randomized controlled experimental study based on experimental animals.SETTING: Department of pharmacology in a university.MATERIALS: Totally 24 SD rats in half genders with clean grade and body mass of(250 ± 50) g, were divided into 4 groups at random: sham-operation group, model group, TFG 40 rmg/kg group and TFG 80 mg/kg group (Certificate No. 01).METHODS: This study was completed in the Department of Pharmacology,Anhui Medical University during October 2001 and January 2002. Incomplete cerebral ischemia was made by ligating bilateral common carotid arteries(CCA) in rats. The cerebral injury was evaluated by brain edema. The apoptosis was determined by terminal deoxy-nucleotidyl transforase-mediated dUTP-digoxigenin nick end-labeling(TUNEL) and transmission electron microscopy (TEM) method. The DNA fragmentation analysis was measured with the diphenylamine reagent method.MAIN OUTCOME MEASURES: Major factor: Effect of TFG on ultrastructral alteration of apoptotic cerebral cortex cells; Secondary factor: Effect of TFG on DNA fragmentation induced by cerebral ischemia.RESULTS: Ligating of bilateral CCA markedly induced apoptotic cell in cerebral cortex. TFG 80 mg/kg significantly inhibited brain edema( P < 0.05 )and decreased the numbers of apoptotic cells in cortex( P < 0.01 ) and improved ultrastructral alteration of apoptotic cells; TFG 40, 80 rmg/kg also inhibited the increase of DNA fragmentation induced by cerebral ischemia (P <0.05, P <0.01).CONCLUSION: TFG has inhibitory effect on ischemia-induced apoptosis of cerebral cortex and improve the ultrastructual changes of apoptosis. Moreover,TFG can relieve the occurrence of edema of ischemic brain tissue and inhibit the increase of DNA section induced by cerebral ischemia.
9.Apoptotic effect of RIZ1 expression on human myeloid leukemic cell lines

Background and purpose:Few reports demonstrate the relation between the expression of RIZ1 which has been found to be a tumor suppressor gene recently and leukemia. This study investigated the effect of RIZ1 expression on the apoptosis of human myeloid leukemic cell lines. Methods:The expression of RIZ1 mRNA was observed in human myeloid leukemic cell lines AML193, KG-1, KG-1a, K562 and Ery-1 by reverse transcription polymerase chain reaction assay. RIZ1 was forced to express in the expression-lacking cell lines by transfecting pRIZ1 RH containing full length of RIZ1 cDNA to the cell lines. As controls, the cell lines were transfected with pcDNA3.1. The apoptotic cells were determined by using the annexin V/propidium iodide stain 24 h after transfection.Results:Among the 5 cell lines, no expression of RIZ1 mRNA had been detected in AML193 and low expression in K562. The forced expression could be found in both cell lines 24 h after transfection of pRIZ1 RH, accompanied by obviously increased apoptotic rates which were (22.7? 0.7)% in AML193 and (28.6?1.2)% in K562 compared to controls (11.7%?1.6% and 9.0%?0.8%, respectively) (P

Objective:To investigate the expression of cyclooxygenase-2 in primary hepatocellular carcinoma, in cancer surrounding tissues and normal liver tissue and its relationship with clinical pathological features.Methods:The expression of COX-2 was detected in 30 cases of hepatocellular carcinoma, 20 cases of cancer surrounding tissue and 10 normal liver tissue by flow cytometry (FCM) and immunohistochemistry (SP). The clinical data were analyzed retrospectively.Results:(1)The expression of COX-2 in the HCC tissue was significantly higher than in cancer surrounding tissues and normal liver tissue (P0.05).Conclusion:The hyperexpression of COX-2 in tissue can reflex the biological behavior of HCC,and have very important role in the development of HCC.The specificness of COX-2 protein expression make it to be new target of tumor diagnosis and treatment.These results provide a theoretical basis for the chemoprevention of hepatoma.
