3.Transport of melanocytes by chitosan membrane: an animal experiment

Objective To study the feasibility of using chitosan membrane to carry and transport melanocytes, in order to refine the technique for melanocyte transplantation with chitosan membrane. Methods Melanocytes were inoculated onto chitosan membrane and cultured for a period of time, then, electron microscopy,MTT assay and NaOH assay were carried out to estimate the adherence, growth and melanogenesis of the melanocytes. Skin wound surface was prepared in 12 nude mice, which were equally divided into 3 groups, test group inoculated with melanocytes on chitosan membrane, negative control group I treated with chitosan membrane without melanocytes, and negative control group II directly dressed immediately after the preparation of wound surface. On day 10 and 20 after the transplantation, confocal laser microscopy and immunohistochemistry were performed to observe the migration of melanocytes into the skin wound surface. Results Scanning electron microscopy and inverted microscopy showed that melanocytes were evenly distributed on and adhered well to the underlying chitosan membrane. As the growth curve of melanocytes demonstrated, chitosan membrane could support the normal growth of melanocytes, and no significant difference was observed in the synthesized melanin content between melanocytes cultured on the chitosan membrane and those in culture disks (0.087 ± 0.027 vs. 0.101 ± 0.036, t = 0.79, P > 0.05). Melanocytes were seen at the transplantation sites by confocal laser microscopy, and biopsy specimens from the transplantation sites stained positive for antimelan-A monoclonal antibody. Conclusions Melanocytes can adhere to and grow on the chitosan membrane,which can facilitate the migration of melanocytes to the transplantation sites in animals with the maintenance of biological activity of melanocytes.
5.Evaluation of clinical efficacy of three different placental preparations in the treatment of childhood vitiligo

ObjectiveTo evaluate the clinical efficacy of three different placental preparations in the treatment of childhood vitiligo.MethodsEighty-two children with vitiligo were divided into 3 groups to be topically treated with melagenine,human placental tissue fluid and human placental lipopolysaccharide respectively.All the patients were additionally treated with infrared rays for 20 minutes twice daily.The drugs were topically applied with an interval of 5 minutes during the irradiation.ResultsThe response rate was 75.65%,35.00% and 44.44% respectively in the melagenine-,human placental tissue fluid- and human placental lipopolysaccharide-treated patients,and significant differences were observed between the three groups of patients(all P < 0.05).In particular,melagenine was superior to the other two preparations for the treatment of large areas of vitiligo on the scalp.ConclusionThe 3 placental preparations combined with infrared irradiation show favorable efficacy for the treatment of childhood vitiligo.
6.Risk factors of early renal damage in children with Henoch Schonlein purpura

Objective To analyze the risk factors of early renal damage in children with Henoch Schonlein purpura(HSP).Methods The clinical data of 196 children with HSP admitted to our hospital from April 2012 to January 2016 were analyzed retrospectively.They were divided into the renal damage group and non-renal damage group within 90 d after confirmed diagnosis.The related clinical data such as serum immunoglobulin and urinary microalbumin were compared between the two groups,and the risk factors of early renal damage in children with HSP were screened.Results There were significant differences between the two groups on age,joint symptoms,recurrent purpura,persistent rash,gastrointestinal bleeding and abdominal pain(with χ2 or t of 11.345,16.223,11.275,43.211,12.592,17.771,P<0.05).The white blood cell count,platelet count,immunoglobulin A(IgA) level and urinary albumin level also showed significant differences between the two groups(t=33.750,60.442,9.451,8.458,P<0.05).The multivariate regression analysis showed that the independent risk factors for early renal damage in children with HSP included age(OR=2.703),recurrent purpura(OR=2.721),persistent skin rash(OR=1.782),gastrointestinal bleeding(OR=11.472),abdominal pain(OR=2.046),IgA level(OR=1.221) and urine microalbumin(OR=3.214).Conclusion Age,recurrent purpura,persistent skin rash,gastrointestinal bleeding,abdominal pain,IgA level and urine microalbumin are closely related to early renal damage in children with HSP.
7.Life Quality and Life Satisfaction among the Elderly Population in Ji'nan

Objective:To study quality of life and life satisfaction among the elderly population dwelling in Ji'nan. Method:1386 elderly people were interviewed with“the Short-Form 36 Health Survey Scale”by the investigators who were trained in advance.Results:The prevalence rate of chronic diseases was 65.1%. The main influencing factors of life satisfaction were mental health condition, family harmony, marriage condition, chronic diseases, and income. The main influencing factors of life quality are life satisfaction, chronic diseases, mental health condition, activities of daily life, family harmony, and income.Conclusion:Life satisfaction is closely associated with life quality of elderly people. The physical, psychological, social and economic conditions have significant effect on life quality of elderly people.
8.Effects of mutation in Cosmc non-coding region on the transcription level of Cosmc mRNA in Tn antigen positive tumor cells

Objective To explore the mutation in corel β3-galactosyl-transferase specific molecular chaperone(Cosmc、no-coding region and it's effects on the transcription level of Cosmc in Tn antigen positive tumor cells.Methods The Tn antigen positive(Tn+)and negative(Tn-)cells were separated from tumor tissues by immune magnetic bead,then the genomic DNA(gDNA),total RNA were prepared by Qiagen AllPrep DNA/RNA mini kit. In these cells.the transcription levels of T-synthase and Cosmc mRNA were tested by RT-PCR.the DNA of Cosmc non-coding region was amplified by PCR,the mutation in Cosmc non-coding region were further detected by sequencing.Results There are no mutation appearing in Tn-cells,one or more mosaic sequence allele appearing in portion of patient's Tn-cells.Almost of the Tn+cells which separated from tumor tissues and Jurkat T cell exists mutation.but the mutation style and mutation point were not saine in different tumor.Thtee patient's Tn+cells have loss of hetemzygosity(LOH),four patient's Tn+cells and Jurkat T cell have point mutation.Although no difference of transcription level of T-synthase mRNA in Tn+ and Tn-cells.but the transcription level of Cosmc mRNA in Tn+ cell was much lower than that in Tn-cell.The ratio of T-synthase/Cosmc mRNA in Tn+ tumor cells was hiigher than that in Tn-cell.Conclusion The tumor Tn antigen arise from mutation in Cosmc non-coding region maybe result from transcription level decreased of Cosmc mRNA.

10.The effect of anti-CD4-epirubicin to T lymphoma in vitro and in vivo

Objective:To detect the specificity killing effect of anti-CD4-epirubicin immunoconjugate on T origin lymphoma cells.Methods:The immunoconjugate was made by linking anti-CD4 monoclonal antibody with epirubicin using Dextran T10 as linkage. The affinity and the killing effect of the immunoconjugate in vitro and in vivo were detected by complement independent cytotoxicity, immuno-histochemistry, CFU-GM and ~ 125 I-anti-CD4 radio-immuno-imaging in nude mice.Results:Anti-CD4 immunoconjugate showed a strong affinity and specific killing effect to T origin lymphoma CEM cells.Conclusion:Anti-CD4-epirubicin and ~ 125 I-anti-CD4 could be used for diagnosis and treatment of T origin lymphoma.
