1.Professional Harmfulness of Personnel in Supply Division and Its Countermeasures

OBJECTIVE To reinforce the personnel′s protection in supply division, to reduce the harmfulness of profession. METHODS The reasons of education, standard operation, dedicated supplies for protection, and building up wholesome institution were carried out. RESULTS The personnel′s consciousness of protection was reinforced and the arrangement of protection was carried out. Incidence wasn′t happened. CONCLUSIONS Better precaution and professional competency of personnel are the bases of protection for high quality work in supply division.

4.Application of combined pancreaticoduodenectomy in operation for upper abdominal malignant tumor

Objective To probe the surgical approach and effect on upper abdominal malignant tumor infiltrating pancrea. Methods Thirty patients with upper abdominal malignant tumor infiltrating pancreas or tissue around pancrea were treated by radical resection on primary tumor combined pancreaticoduodenectomy. Results Three patients died in 1 month after operation due to multiple organ failure. The patients with gallbladder cancer and metastatic lymph nodes fixed behind caput pancreatis were followed up averaged 35 months. The patients with gastric antrum carcinoma infiltrating caput pancreatis were followed up averaged 31 months. The patients with recurrent gastric cancer infiltrating caput pancreatis were followed up averaged 13 months. The patients with pancreatic and duodenal invasion by cancer of hepatic flexure of colon were followed up averaged 41 months. The patients with hilar cholangiocarcinoma and metastatic lymph nodes fixed behind caput pancreatis were followed up averaged 11 months. Conclusion The patients with upper abdominal malignant tumor infiltrating pancreas should be operated by radical resection on primary tumor combined pancreaticoduodenectomy, which can increase the rate of tumor resection, and be expected to prolong the survival period.
5.Wedge-shaped autograft reconstructing deformity of severe varus knee in primary total knee arthroplasty

[Abstrct] Objective The deformity of severe varus knee (varus angulation > 15°) can be treated by many ways in primary total knee arthroplasty , but the efficacy and cost are different. The topic of this study provides a new surgery: autograft (wedge grafts) implant and substitute medial tibial bone defects. Methods From Jan. 2006 to Jan. 2014 , a retrospective study was conducted in the First Affiliated Hospital of BaoTou Mendical College. Out of 687 patients, 721 primary knees were operated. Bone defects in proximal tibia (varus angulation >15° ) were encounted in 16 patients in 21 knees in the pattern of non-inclusive slope bone defects in the mean varus degree of 27°. Their IBM was 31 kg/m2. The average age of patients was (63 ± 5) years. The score of clinical knee society score (KSS) before operation was 18 on average. Among of them, the pain score were 12 points, the average range of motion (ROM) were 12 points. the average funtional score were 20 points. During operation,the slope bone defect of the medial tibial plateau was tailored into wedge-shaped ladder bone defect by osteotomy,and then the defect was filled with the resected tibial plateau autograft in which the thickness and shape were matched completely. The high-intensity cortical part of the autograft was placed in the rim to sustain the tibial prosthesis. Three months after operation ,patients started to walk without the help of crutch. Results The average followup period was 5.3 y , We observed that an average postoperative scores of clinical KSS from 42 points to 88 points with the score of functional KSS from 57 points to 92 points. Fusion was observed by trabeculae crossing from the graft to the tibial metaphysis on plain films at least 6 mouths without collapse, stress fracture, untight and bone ununion. Confirmation of vascularization of the graft was observed in bone scans of nuclide. showing increased uptake in the operated area. Inflammatory makers were normal. Conclusion Autograft (wedge grafts) reconstruction of medial tibial bone defects, is of high rate of healing in implant bones, good restoration of the function and low cost.

7.Application of Thermal Shift Assays in Purification of Recombinant TBEV Serine Protease

Thermal Shift Assays was applied for high-throughput screening optimized buffer system used in the gel filtration chromatography for purification of recombinant TBEV serine protease. The optimization included kinds of salt and pH. It was optimized new buffer remarkably improved the quality of the purified TBEV serine protease by increasing its monomer from 75% to 99%. This result may improve future crystal studies of this protein.
8.The effect and mechanism of indole-3-carbinol on alleviating the radiation-induced bone marrow hematopoietic cell injury in mice

Objective To investigate the protective effect of indole-3-carbinol (I3C) on radiation-induced mouse bone marrow hematopoietic cell injury and the involved mechanisms. Methods (1) The bone marrow nuclear cells (BMNCs) from CD45.1 subtype of C57BL/6J mice were collected by a density gradient centrifugation method. The BMNCs were pretreated with a series doses of I3C (0 mol/L, 10-8 mol/L-10-3 mol/L) and then exposed with radiation of 137Csγ-ray (doses of irradiation were 0 Gy, 1 Gy and 4 Gy). After 18-hour culturing, the bioluminescence method was used to detect the cell viability. (2) These cells were divided into control group and 10-6 mol/L I3C group. Both groups were received the irradiation (0 Gy, 1 Gy and 4 Gy) and inoculated into the methylcellulose semi-solid culture medium to incubate 7 days, the colony forming unit-granulocyte monocytes (CFU-GM) were observed. (3) Twenty-four CD45.2 subtype mice used as the receptor were exposed with 8 Gy radiation. The CD45.1 BMNCs were divided into control group, 4 Gy irradiation group, 4 Gy irradiation and 10-6 mol/L I3C group. Donor cells were harvested from C57BL/6J (CD45.1) mice after they received various treatments, and were then mixed with competitive BMNCs from C57BL/6J (CD45.2) mice. The mixed cells were transplanted into recipient mice (8 mice/group). Flow cytometry was used to analyze the proportion of donor cells in peripheral blood of receptor. (4) The cells were divided into control group, 10-6 mol/L I3C group, 1 Gy irradiation group, 1 Gy irradiation with 10-6 mol/L I3C group. After 24-hour culturing, Western blot assay was used to detect the expression levels of nuclear factor erythroid 2-related factor 2 (Nrf2) and hemeoxygenase-1 (HO-1). Results (1) I3C showed a significant cytotoxic effect on the BMNCs when its concentration was above 10-4 mol/L. 10-7-10-6 mol/L I3C could reduce the radiation injury of BMNCs under the same dose of irradiation. Therefore, 10-6 mol/L I3C was chosen for subsequent experiments. (2) The CFU-GM was significantly higher in 10-6 mol/L I3C group than that of control group (P<0.05). (3) Results of flow cytometry showed that the proportion of donor cells in receptor was significantly higher in 4 Gy irradiation group than that of control group, which decreased the engraftment capability of irradiated HSCs (P<0.05), although the engraftment capability of irradiated HSCs improved after 10-6 mol/L I3C treatment. (4) I3C significantly enhanced the increased protein expression of Nrf2 and HO-1 caused by radiation (P<0.05). Conclusion I3C has a protective effect on hematopoietic cells following radiation-induced injury, which may be related with activating the Nrf2/HO-1 signal pathway.
9.Protective effects of 3,3′-diindolylmethane on radiation damage of hematopoietic system in mice

Objective To investigate the protective effect of 3,3′-diindolylmethane (DIM) on radiation-induced injury in mouse hematopoietic system. Methods Thirty C57BL/6 mice were randomly divided into control group, 2 Gy irradiation group and 2 Gy irradiation+DIM group (n=10 for each group). Mice of control group received sham irradiation, and the other two groups accepted 2 Gy 137Cs γ-ray total body irradiation. Mice in 2 Gy irradiation +DIM group were intraperitoneally injected 75 mg/kg DIM 30 min before irradiation. Mice of other two groups were treated with reference solution. After 7 d and 15 d of 2 Gy irradiation, the peripheral blood samples were collected to count the number of bone marrow nuclear cells (BMNCs). The level of reactive oxygen species(ROS) was measured by DCFH-DA. The levels of colony forming units-granulocyte-macrophage (CFU-GM) were also detected. Results The numbers of white blood cell (WBC), platelet count (PLT), BMNCs and CFU-GM were significantly decreased and the ROS level of bone marrow cells increased significantly in the irradiated group than those of control group (P<0.05). Compared to 2 Gy irradiation group, the numbers of WBC, PLT, BMNCs and CFU-GM were significantly increased in 2 Gy irradiation +DIM group, and the level of ROS was decreased significantly (P<0.05). Conclusion DIM has a protective effect on hematopoietic cells following radiation-induced injury, which may be related with the decreased ROS level.
10.Effect of Music Electroacupuncture on Perifocal Ferric Ions and Neuronal Apoptosis Percentage and the Brain Water Content in Rats with Acute Cerebral Hemorrhage

Objective To investigate the effect of music electroacupuncture on perifocal ferric ions and neuronal apoptosis and the brain water content in rats with acute cerebral hemorrhage and explore the mechanism of its action on acute cerebral hemorrhage.Methods One hundred and twenty-eight healthy adult male Wister rats were randomized to normal, model, electroacupuncture and music electroacupuncture groups. In each group, four time points of 6 hrs, 24 hrs, 3 days and 7 days were set up, eight rats each time point. A rat model of acute cerebral hemorrhage was made using collagenase. In the electroacupuncture and music electroacupuncture groups, point Baihui was connected to the anode and point Taiyang to the cathode, 2 voltage was used as a selection parameter and electroacupuncture lasted 30 min. Intracerebral perifocal ferric ions and neuronal apoptosis and the brain water content were measured at four time points of 6 hrs, 24 hrs, 3 days and 7 days after model making.Results Intracerebral ferric ion content was higher in the model, electroacupuncture and music electroacupuncture groups of rats than in the normal group at 6 hrsafter model making (P<0.05), lower in the electroacupuncture and music electroacupuncture groups than in the model group at 24 hrs, 3 and 7 days after model making (P<0.05) and lower in the music electroacupuncture group than in the electroacupuncture group at 3 and 7 days after model making (P<0.05). Intracerebral neuronal apoptosis was higher in the model, electroacupuncture and music electroacupuncture groups of rats than in the normal group at 6 hrs after model making (P<0.05) and lower in the music electroacupuncture group than in the electro- acupuncture group at 7 days after model making (P<0.05). The brain water content was higher in the model, electro- cupuncture and music electroacupuncture groups of rats than in the normal group at 6 and 24 hrs after model making (P<0.05), lower in the electroacupuncture and music electroacupuncture groups than in the model group at 3 days after model making (P<0.05) and lower in the music electroacupuncture group than in the electroacupuncture group at 7 days after model making (P<0.05).Conclusions Music electroacupuncture has a benign regulating effect on intracerebral perifocal ferric ions, inhibits neuronal apoptosis around hematoma in the early stage and reduces cerebral edema in rats with acute cerebral hemorrhage. Its therapeutic effect is superior to that of electroacupuncture.
