2.An Immunohistochemical Study on Expression of Proliferating Cell Nuclear Antigen, Transglutaminase, and Glutathione-S -Transferase pi on DMBA induced Oral Carcinogenesis in Hamsters.

The purpose of this study was to evaluate the relationship between proliferation and cell death during oral carcinogenesis. Syrian golden hamsters which were 3 month old and 90-120 gm-weight were used in this study. The 9, 10-dimethyl-1, 2-benzanthracene(DMBA) in a 0.5% solution in mineral oil was painted on the buccal pouch mucosa 3 times per week in the experimental group during 20 weeks. Control group was painted with mineral oil. In each control and experimental group of 6,8, 10, 12, 14, 16, 18,20 weeks, specimen were sectioned for immunohistochemical study with anti-Proliferating Cell Nuclear Antigen(PCNA), anti-transglutaminase transglutaminase and GST-pi were obtained by counting the positive cells to those antibodies. The following results were obtained. 1. Histopathologically, finding of epithelial dysplasia of the 6 and 8 weeks experimental group and carcinoma in situ in the 12 weeks and squamous cell carcinoma in those of the 14 weeks were seen. 2. PCNA positive cells were mainly mild expressed in the basal cell layer of normal oral mucosa, increased moderately, after 6 weeks. In suprabasal cell layer, control group is negative but retained moderately between 6 weeks and 14 weeks, and decreased after 16 weeks. In spinous cell layer, restricted only between 12 weeks and 16 weeks, other period is mild or negative. 3. PCNA index of experimental group revealed the increased peak in 6 weeks and 20 weeks than control group, and retained between 12 weeks and 18 weeks. All experimental group expressed higher PCNA index than control group(p.<0.05). 4. Tranglutaminase expression was localized in outer and suprabasal layers on control group, but after 6 weeks, expression site moved spinous & suprabasal cell layers, and after 8 weeks, expression is spreaded to basal cell layer, and this patters retained to 20 weeks. Transglutaminase expression of experimental group was higher than control group after 8 weeks. 5. The positive staining of detoxifying agent, G1utathione S-Transferase(GST)pi of experimental group was radually increased from 6 weeks. After 10 weeks, all layer of experimental group was seen positive reaction. The strong positive staining in center of tumor and weak positive staining in periphery of tumor were seen at the stage of squamous cell carcimoma in 14 weeks. According to the results, we should suggest that the more increased proliferation of tumor cell, the more increased expression of PCNA, transglutaminase and GST-pi as a detoxifying agent during carcinogenesis by induced DMBA were seen.

5.Evidence for neutrophil activation in induced sputum from patients with TDI - induced occupational asthma.

BACKGROUND: Toluene diisocyanate (TDI) is the most prevalent agent to cause occupational asthma (OA) in Korea. The pathogenic mechanism of TDI-induced OA is still unclear. Involvement of both immunological and non-immunologicaI mechanisms have been suggested. OBJECTIVE: To evaluate a possible role of neutrophil in the development of TDI-asthma. OBJECT AND METHOD: Myeloperoxidase (MPO) as a neutrophil activation marker in both serum and induced sputum, and IL-8 in induced sputum were measured. Induced sputa and sera were collected from 15 TDI-induced OA patients (classified to group I) during TDI- bronchoprovocation test and were compared with those from 11 asthmatic subjects with negative TDI-bronchoprovocation test (group II). MPO levels were measured by radioimmunoassay, IL-8 levels, by enzyme linked immunosorbent assay and albumin levels, by nephelometry. Sputum MPO and IL-8 levels were presented as a ratio to albumin. RESULT: Serum MPO level tended to decrease during the TDI-bronchoprovocation test in two groups, but no statistical significance was reached (p>0.05). However, the ratios of MPO (the ratio of MPO level measured at 30 min to MPO level at baseline, and the ratio MPO level measured at 360 min to MPO baseline) in group I were significantly lower than group II (p=0.004, p=0.03 respectively). The IL-8/albumin and MPO/albumin levels in induced sputum from group I were significantly increased after the TDI-bronchprovocation test in comparison to the baseline value which was obtained before the bronchoprovocation test (p=0.0l, p=0.02 respectively). There was a significant correlation between the percent increase of IL-8/albumin and the MPO/albumin in induced sputum (r=0.89, p<0.05). CONCLUSION: These findings suggest a possible involvement of neutrophil in the development of bronchoconstiction after the TDI exposure, and IL-8 might contribute to neutrophil recruitment to airway mucosa. Further investigation will be needed to investigate mechanism of neutrophil activation in the pathogenesis af TDI-induced OA.

6.Evaluation of the effect of abutment preparation angles on the repeatability and reproducibility using a blue light model scanner

. The purpose of the study is to evaluate the repeatability and reproducibility of the abutment angle using a blue light scanner. MATERIALS AND METHODS. 0°, 6°, and 10° wax cast abutment dies were fabricated. Each of the silicone impression was produced using the replicable silicone. Each study die was constructed from the prepared replicable stone used for scans. 3-dimensional data was obtained after scanning the prepared study dies for the repeatability by using the blue light scanner. The prepared 3-dimensional data could have the best fit alignment using 3-dimensional software. For reproducibility, each abutment was used as the first reference study die, and then it was scanned five times per each. 3-dimensional software was used to perform the best fit alignment. The data obtained were analyzed using a nonparametric Kruskal-Wallis H test (α=.05), post hoc Mann-Whitney U test, and Bonferroni correction (α=.017). RESULTS. The repeatability of 0°, 6°, and 10°abutments was 3.9, 4.4 and 4.7 μm, respectively. Among them, the 0° abutment had the best value while the 10° abutment showed the worst value. There was a statistically significant difference (P<.05). The reproducibility of 0°, 6°, and 10° abutments was 6.1, 5.5, and 5.3 μm, respectively. While the 10° abutment showed the best value, the 0° abutment showed the worst value. However, there was no statistically significant difference (P>.05).CONCLUSION. In repeatability, the 0° abutment showed a positive result. In reproducibility, the 10° abutment achieved a positive result.

8.Revival of Hop Japanese pollinosis in asthmatic subjects in Kyungki area.

BACKGROUND: Hop Japanese (Hop J) pollens are abundant in the air of Korea during the autumn season. Their significances as a source of allergenic sensitization have been underestimated in this country. MATERIAL AND METHOD: In other to observe clinical features of Hop J-sensitive asthmatic patients in this country, skin prick test with Hop J pollen was performed. The serum specific IgE antibodies to Hop J pollen antigen were detected by enzyme linked immunosorbent assay (ELISA) in positive responders (>2+ of A/H ratio) on skin prick test. To confirm the respiratory sensitization, bronchoprovocation test was performed in 17 asthmatic patients sensitive to this pollen. RESULT: Ten asthmatic subjects showed a significant bronchoconstriction following the inhalation of Hop J pollen extract (6 early and 4 dual astmatic responses) and all of them had high serum specific IgE bindings, with minimal bindings in negative responders. They have suffered from seasonal aggravation of asthmatic symptoms with or without rhinitis, and/or conjunctivitis symptoms. The skin reactivity to Hop J had more than 5+ of A/H ratio on skin prick test in nine positive responders, whlie negative responders showed from 1+ to 3+ response. Moreover, four (40%) asthmatic subjects showed a positive response to only the Hop J pollen on skin prick test and an isolated positive asthmatic response to the Hop J bronchoprovocation test. CONCLUSION: We believe that the Hop J pollen should be considered as an allergen during the Autumn season, and thus included in skin test batteries in this area. Some labelled having intrinsic asthma or rhinitis might be sensitized to this pollen or other unknown allergens.
