1.Content Comparison of Polydatin and Emodin between Polygonum Cuspidatum Pieces and Dispensing Gran-ules

Objective:To compare the content of polydatin and emodin between polygonum cuspidatum pieces and dispensing gran-ules to provide theoretical basis for the improvement of preparation process of the granules. Methods:An HPLC method with linear gra-dient elution was used to determine the content of polydatin and emodin in the pieces and dispensing granules, and the difference in the two contents was compared. Results: The content of polydatin and emodin in polygonum cuspidatum pieces met the requirement de-scribed in China pharmacopoeia, which was lower than that in the dispensing granules, and the contents of the two components in the commercial dispensing granules were not equivalent to the labeled amount, which was 1/15 and 1/27 of that in the pieces. Conclu-sion:The transport rate of the traditional preparation process of dispensing granules is with poor efficiency, which should be improved.
2.EFFECTS OF MALOTILATE ON ACUTE LIVER INJURY INDUCED BY CARBON TETRACHLORIDE AND D-GALACTOSA-MINE IN MICE

A study was made; of the effect of malotilate on the acute liver injury induced by carbon tetrachlorid,e ( CC14 ) and d-galactosamine in mice. Malotilate ( 50~150mg/kg ig?3 ) significantly inhibited the elevation of serum glu tamic pyruvic transaminase ( SGPT ) in CC14- intoxicated mice.At the dose of 100mg/kg ig?3, malotilate remarkably increased the content of hepatic glycogea in CCl4-injected mice. The contents of serum protein, liver protein, and cytochrom P-450 in liver hemogenate were increased by malotilate ( 100mg/kg ig?3) in CC14-intoxicated mice. The drug also reduced the accumulation of liver triglycerides induced by CCl4 in mice.In addition to, malotilate(50mg /kg, ip?5) could act against the increase of SGPT and the decrease of liver protein content induced with d-galactosamine in mice. These results suggest that malotilate may be a new therapeutic agent for liver injury.

8.Application and Progress of Helper Phage in Phage Display

Phage display is a widely used gene engineering high technology.Through the display of exogenous peptides or proteins fused specific coat protein on the surface of phages,it is possible to construct proteins or peptides libraries and screen interesting proteins,peptides and antibodies successfully.Most commonly used phage display technology is phagemid/helper phage system,in which helper phages are essential for the replication and assembly of phagemid particles.In this review,in combination with the newest research dynamic status,we summarize phagemid/helper phage double-genome system.We mainly emphasized the features and mutation mechanisms of different helper phages.We also made some prospects for the future directions,in the meanwhile,we also expect that our experience can provide some help for further maturity of the technology.
9. Genetic polymorphism analysis of cytochrome CYP3A4, CYP2C9, CYP2C19, CYP2D6 in Chinese Han and Mongolian population

OBJECTIVE: To study the genotype distribution of cytochrome CYP3A4, CYP2C9, CYP2C19, CYP2D6 in Han and Mongolian Chinese population and the allele and genotype frequency are compared between the Han and Mongolian. METHODS: CYP3A4, CYP2C9, CYP2C19, CYP2D6 of Han and Mongolian Chinese population were genotyped by PCR-RFLP. RESULTS: The allele frequencies of CYP3A4 * 5 in Han and Mongolian are 0, the allele frequencies of CYP3A4 * 18 in Han and Mongolian are 0.1838, 0.2025. The allele frequencies of CYP2C9 * 2 in Han and Mongolian are 0.0110, 0.0253; the allele frequencies of CYP2C9 * 13 in Han and Mongolian are 0, 0.003 2.The allele frequencies of CYP2C19 * 2 in Han and Mongolian are 0.386 0, 0.4146, the allele frequencies of CYP2C19 * 3 are 0.0515, 0.0443. The allele frequencies of CYP2D6 * 10 in Han and Mongolian are 0.5735, 0.4652. CONCLUSION: The study shows that there are no significant ethnic differences in the distribution of CYP3A4 * 18, CYP2C19 * 2, CYP2C19 * 3, CYP2D6 * 10 genotypes in Han and Mongolian. The CYP3A4 * 5 genotype is not found in this study and only one CYP2C9 * 1/* 13 genotype is found in Mongolian. For CYP2C9 * 2, the Mongolian is significantly lower than the Han, (P=0.023).
10.Tertiary peritonitis treated with integrated traditional Chinese and western medicine and its APACHE Ⅲscoring

Objective To investigate the clinical features , treatment regimen , and prognosis evaluation of tertiary peritonitis (TP). Methods Seventy-eight cases with TP were randomly enrolled into 2 groups, including the simple western medicine-treated group (32 cases) and the integrated traditional Chinese and western medicine-treated group (46 cases). The prognoses were evaluated according to the acute physiology and chronic health evaluationⅢ (APACHEⅢ, APⅢ) scoring. Results The mortality rate was 71.9% (23 of 32) in patients received the simple western medicine and was 32.6%(15 of 46) in patients received the integrated traditional Chinese and western medicine with significant difference between these two groups (P < 0.01). There was a significant correlation between AP Ⅲscore and actual mortality (r=0.73,P<0.01), and predicted mortality (r=0.76, P<0.01). Conclusions The therapeutic effect is acceptable and satisfactory for the TP patients received the integrated traditional Chinese and western medicine. The AP Ⅲ scoring system can be used to predict the prognosis of TP patients.
