Chinese Journal of Experimental Ophthalmology 2013;(4):316-321
Background It is widely appreciated that many animal cells rely on the mechanism of regulatory volume decrease (RVD) after swell under the hypotonic environment, which involved in some processes of cellular physiology. But the RVD of lens epithelial cells(LECs) still is being further researched. Objective Present study was to clarify the possible mechanisms and influencing factors in the RVD of LECs.Methods Human LECs line (HLE B-3)were cultured and passaged in DMEM/F12 containing 10% fetal bovine serum(FBS),and before volume measurement, cells were stuck to the base of a perfusion chamber,Ringer solution osmolality was decreased from 15%Hypo to 45% Hypo, and the cells stimulated by 45% Hypo Ringer solution were used as the control group. Some experiments were performed in the presence of high extracellular K+ concentration, chloride or potassium channel inhibitor, experiments were also carried out in the nominal absence of Ca2+,Cl-or HCO-3 to test the effect of a decrease in intracellular concentration of these ions on the cell volume response. The volume changes of living cells were measured by lag-time microphotograph acquisition and analysis system (IPP6.0).Results Time course of cell volume change after hypotonic shock in HLE B-3 cells was observed. The cell swelling was followed by a gradual volume recovery, indicating the presence of RVD was influenced by the hypotonic stress.Under the stimulation of 45 %Hypo Ringer solution, the rates of RVD were (59.1 ±7.8)%.RVD was correlated positively to the maximum cell volume (r =0.99,P<0.05)in S shape, and RVD changes were sensitive to alter maximum cell volume in the range of 115%-135%.RVD reduced to (16.5 ± 1.6) %,(14.7 ± 2.3) %,respectively after acted by potassium channel inhibitor,TEA(10 mmol/L)and BaCl2(5 mmol/L)as well as chloride channel inhibitor,NPPB(100 μmol/L)and DIDS(100 μmol/L),with significant differences in comparison with the control group(all P<0.01).RVD decreased by(5.8±1.6)% and(2.7±0.8)% in high concentration of K+ in extracellular fluid and the absence of Cl-(P<0.01).RVD was significantly inhibited under the absence of Ca2+.When the 45% Ringer solution was pH6.8,the process of RVD delayed. The rate of RVD in the first ten minutes was (0.86±0.24)%/min,showing a significant decline in comparison with (3.24±0.84) % / min of pH 7.4 (P <0.05).Conclusions HLE B-3 have RVD ability under the hypotonic stress stimulation.A certain intracellular Ca2+ concentration is the premise of RVD activation, and Cl efflux and K+ efflux are the key mechanism of RVD of HLE B-3.Acidic environment of hypotonic solution delays the occurrence of RVD.

Objective To evaluate the safety, feasibility and curative effect of mini-incision ex-ploration of common bile duct. Methods In this study, 290 patients underwent min-incision explora-tion of common bile duct and 120 patients underwent open-incision exploration of common bile duct for bile duct stones and/or gallstones from 2005 to 2007. The iatrogenic bile duct injury, postoperative complication, residual stone, stone recurrence, therapeutic effect and clinical data were evaluated by randomized contrast analysis. Results Time of operation, bleeding, volume of drain pipe, time of re-covery of intestinal peristalsis and average duration in hospital were significantly lower in the group of min-incision exploration(MCE) than in the group of open-incision exploration(OCE). The iatrogentic bile duct injury occurred in 5 cases(1.72%), residual stone in 10 cases(3.45%), stone recurrence in 15 cases(5.18%) in the group of MCE, and in 2 cases(1.67%), 4 cases(3. 33%) and 6 cases respec-tively in the group of OCE. There was no marked difference between the two groups. Howevert post-operative complications occurred in 17 cases(6.8%) and 16(13.3%) in the group of MCE and OCE,respectively. There was remarkable difference between the 2 groups(P<0. 05). Conclusion Mini-in-cision exploration of common bile duct is a feasible and safe method resulting in fewer complications of iatrogentic bile duct injury, stone recurrence and residual stone.

Iron is abnormally accumulated in the substantia nigra pars compacta (SNc) in Parkinson’s disease (PD) patients. The disorder of iron metabolism and the neurotoxicity induced by excessive iron are proved in PD patients, and these indicate that iron is closely correlative with PD. This article presented an overview of research advances of three aspects including the distribution and metabolism of iron in the brain, the neurotoxicity of iron, and the relationship between the disorder of iron metabolism in central nervous system and PD.

Objective Cardiac muscle cells play a critical role in maintaining normal function of the heart.Cardiotrophin-1(CT-1),a potent cardiac survival factor,is capable of inhibiting apoptosis or promoting survival in cardiomyocytes. To elucidate the mechanism of CT-1 promoting cardiac myocyte survival in cultured neonatal rat cardiomyocytes. To explore the potential signaling pathway that might be responsible for this effect. Methods We examined the cardiac myocyte survival effect of CT-1 in cultured neonatal rat cardiomyocytes. The cardiomyocytes were stained [3-(4,5-dimethyl-thiaziazol-2-yl)-2-5-diphenyltetrazolium bromide,MTT] and the counted. Results The survival rate of cardiac myocytes was increased by CT-1 in a dose-dependent manner(10-10~10-7 mol/L) and in a time-dependent manner(1~4 d,10-8 mol/L) in cultured neonatal rat cardiomyocytes.Pretreatment of PD098059(5?10-5mol/L),a MAPK blocker, decreased significantly survival rate of cardiac myocytes by promoted CT-1.The phorbol 12-myristate 13-acetate(PMA)(10-5mol/L),a PKC activator, increased significantly this effect of CT-1,but inhibited significantly by MAPK blocker PD098059.Conclusion CT-1 is a potent factor of promoting cardiac myocyte survival, and increase significantly survival rate of cardiac myocytes in a dose-dependentand a time-dependent manner in cultured neonatal rat cardiomyocytes. The MAPK signaling pathway mediates CT-1 induced cardiac myocyte survival.PKC signaling molecule may be a upstream signaling transduction pathway which cascades of MAPK in CT-1 induced cardiac myocyte survival.

Objective To determine the advantages of laparoscopic cholecystectomy (LC) by establishing pneumoperitoneum under direct vision. Methods A 1cm incision was made just below the umbilicus; lifting and cutting out of the peritoneum at the line alba abdominis with direct vision; then a 10mm trocar was inserted into the pneumoperitoneum cavity. Results There were 107 patients underwent LC.Of them, 93 patients suffered from chronic cholecystitis with gallstone, 6 from acute cholecystitis with gallstone, and 8 from cystopolyps. Among them, 16 patients had previous abdominal operations. Two patients with atrophic cholecystitis converted to open cholecystectomy(OC) owing to the unclear bile duct anatomy. The average operation time was 45min. Postoperative complications included pulmonary infection in 3 patients, bile leakage in 1( due to the titanic clip falling off),but no bile duct injury or other severe complications occurred;and no mortality in this series. Conclusions Establishing pneumoperitoneum under direct vision has following advantages:rapid and safe, and favorable to avoid the severe trocar-related complications.

Objective To assess the predictive value of serum uric acid levels to the incident nonalcoholic fatty liver disease(NAFLD)in a cohort of healthy population. Methods A prospective cohort study of NAFLD incidence was conducted in Xinjiang province, from 2012 to 2014. A cohort study was performed on 2 207 subjects with no evidence of fatty liver disease by liver ultrasound and with no major risk factors for liver disease at baseline. All participants were interviewed to carry out the epidemiological questionnaire survey, physical examination, abdominal ultrasonography, as well as blood biochemistry measurements. The hazard ratios of NAFLD were compared among groups with different uric acid levels. All subjects were classified according to serum uric acid (within normal range grouped by quartile: Q1-Q4 group; above the normal range: Q5 group). Results NAFLD was newly diagnosed in 13.72% subjects within the 3-year period. The incidence of NAFLD was increased with elevated serum uric acid quartiles(P<0.01). In multivariate logistic regression, after adjustment for gender, age, race, metabolic syndrome and its components, OR for incident fatty liver in Q2 to Q5 of serum uric acid concentration as compared to Q1 were 2.509, 3.172, 3.392, and 4.041 respectively. Conclusion Elevated serum uric acid concentrations may predict NAFLD in the subjects for a regular health checkup.
