2. Inhibitory effect of Bcl-2 short hairpin RNA on growth of BEL-7402 and Caco2 cells

Objective: To investigate the effects of Bcl-2 shRNA on the growth of liver carcinoma cell line BEL-7402 and colorectal carcinoma cell line Caco2. Methods: Bcl-2 shRNA was cloned into Pgenesil-1 plasmid labeled with fluorescent protein and the product was transfected into BEL-7402 and Caco2 cells with Lipofectamine 2000. The study also included shRNA as negative control, Pgenesil-1, Lipofectamine and blank control groups. Transfected cells were visualized by inverted fluorescent microscope and then assayed by flow cytometry. The expression levels of Bcl-2 protein were assayed by Western-blot and cell proliferation was measured by MTT method. Results: There was no difference in transfection rate among cells in Bcl-2 shRNA, shRNA and Pgenesil-1 vector groups. Western-blot assay showed that the expression levels of Bcl-2 protein in BEL-7402 and Caco2 cells were significantly decreased in Bcl-2 shRNA group compared with those in other 4 groups (P<0.05). Furthermore, Bcl-2 shRNA had a stronger inhibitory effect on Caco2 cells than on BEL-7402 cells (P<0.05). MTT assay showed that Bcl-2 shRNA significantly inhibited the growth of BEL-7402 and Caco2 cells (at 72 and 96 h after treatment, respectively) compared with the other 4 groups (P<0.05). Conclusion: Bcl-2 shRNA can specifically inhibit the growth of BEL-7402 and Caco2 cells.
3.The hospital emergency management for a large number of earthquake patients in 2008 Sichuan Wenchuan Earthquake, China

Objective A large number of injured earthquake patients were accepted by the hospital whilethe professional surgeons were relatively lack. This article introduced the hospital emergency management in 2008Sichuan Wenchuan Earthquake, China. Method Within 3 days, Central Hospital of Mianyang accepted andtreated over 1000 patients after Wenchuan Earthquake jolted on 12 May 2008,and within 2 weeks, the number ofpatients reached 1500. The hospital carded out emergency management plan: (1) emergency comprehensive treat-ment district was established, which was divided into traumatic surgery district, general surgery district, and gen-eral medical district. Traumatic surgery district is responsible for treating traumatic patients, and most doctors andnurses were in this district. The district also had preview, contamination, operation, isolation, monitoring sec-tions, and the tents were numbered and labeled. General surgery district and general medical district were responsi-ble for patients not from earthquake, and only few doctors and nurses were in the two districts. According to the in-jury degree, all the wounded were classified into acute and severe, moderate and minor injuries, and wore red,yellow and blue label on the wrists, respectively. The name, gender, age and diagnosis of each patient and thename of doctor were written on the label. The infectious patients and non-infectious patients were separated.Results Near 200 operations and near 300 operations were performed at one night and at one day, respectively.Within one week, only 1 patient had the lung infection, and one patient with gangrenous emphysema was effective-ly treated. In-hospital cross infection and epidemic of infection disease didn't happen. Conclusions Emergencymanagement model and mechanism, which referred to the model of the battlefield ambulance, played an importantrole in treating a large number of injured patients.
4.Comparative study between the features of FFA and OCT in diabetic macular edema

AlM: To assess the correlation between the features of optical coherencetomography ( OCT ) and fundus fluorescein angiography ( FFA) in diabetic macular edema ( DEM) .
METHODS: Totally 70 patients (135 eyes) with diabetic retinopathy ( DR) were evaluated by central vision, best corrected visual acuity ( BCVA ) , intraocular pressure, indirect ophthalmoscopy, slit lamp microscope combined+ 90D front mirror mydriatic fundus examination, mydriatic fundus color photography, OCT, FFA, the correlation between FFA and OCT were analyzed.
RESULTS: ln mild macular oedema cases, abnormalities in FFA was 56 eyes, abnormalities in OCT was 68 eyes (P=0. 0009);FFA showed 12 normal eyes, 10 eyes in OCT were characterized by diffused macular oedema; FFA was performed with cystoid macular oedema, OCT was 46. 7% with cystoid type .
CONCLUSlON: DME is diagnosed by Combination FFA with OCT, OCT is an indispensable tool when following up DME, and it has advantage in early application.
5.Gastrointestinal manifestations and acute abdominal pain of childhood systemic lupus erythematous

Objective To analyze the clinical characteristics of childhood systemic lupus erythematosus (SLE) patients with severe gastrointestinal manifestations,especially cases with acute and severe abdominal pain,so as to improve the recognition of severe and acute gastrointestinal manifestations of SLE during pediatric diagnosis and treatment.Methods Medical records of 119 patients with SLE under the age 16 years old in Peking Union Medical College Hospital from Jan.2010 to Mar.2013 were reviewed and gastrointestinal manifestations were retrospectively analyzed.All patients were diagnosed as SLE according to 1997 American College of Rheumatology (ACR) revised classification criteria.Results Gastrointestinal involvement was recorded in 24 children (20.2%).The median (range) age at the time of initial gastrointestinal manifestations was (13.5 ± 2.0) years (6-15 years).The ratio of female to male was 1.0 ∶ 2.4.And in 4 cases,gastrointestinal manifestations occurred as the initial symptoms.Abdominal pain was the most frequent symptom,present in 13 patients (54.2%),11 cases (45.8%) had nausea and vomiting,4 cases (16.6%) had abdominal distension,and 3 cases (12.5%) had diarrhea.Abnormal liver function was found by lab test in 8 cases,without obvious symptoms or the proofs of any virus infection.Acute and severe abdominal pain was found in 9 cases,of whom 6 patients were diagnosed as intestinal pseudo-obstruction (2 cases with bilateral ureterohydronephrosis),1 case had acute peritonitis with surgery,1 case was diagnosed as protein losing enteropathy and 1 case had acute pancreatitis.Liver impairment also occurred in SLE.All cases got alleviated with the treatment of steroids and immunosuppressive drugs.Significant difference was found in the occurrence of hydroureter and hydronephrosis and albumin concentrations between SLE digestive and non-digestive system involvement children.However,there was no remarkable difference in the erythrocyte sedimentation rate,or the involvement of kidney and blood system and central nervous system between the 2 groups.Conclusions SLE is a common autoimmune disease in children involving multiple systems.The severity of gastrointestinal involvement is underestimated.Gastrointestinal manifestations can be initial symptoms of SLE.Intestinal pseudo-obstruction,protein losing enteropathy,and acute pancreatitis are uncommon but severe gastrointestinal manifestations of SLE patients should be fully recognized.It is vital for pediatric physicians to make early diagnosis and begin timely treatment on SLE with Corticosteroid and cyclophosphamide.
6.Therapeutic efficacy of first-line CHOP or CHOP-like chemotherapy on patients with advanced staged angioimmunoblastic T-cell lymphoma

Objective To evaluate the efficacy of first-line CHOP or CHOP-like regimen on patients with advanced staged angioimmunoblastic T-cell lymphoma (AITL).Methods Between Aug 2006 and Sep 2014,twenty-nine AITL patients who were newly diagnosed without prior treatment were included in study.The clinical features,efficacy and survival were analyzed retrospectively.Results Median age of these patients was 59 years old.All patients had stage Ⅲ/Ⅳ disease.17 (58.6 %) cases presented with B symptoms.26 (89.7 %) cases had an international prognostic index (IPI) score ≥2,and 20 (69.0 %) cases had elevated LDH,9 (31.0 %) cases had ≥2 extranodal involvements.The median follow-up time was 20 months.Overall response rate was 69.0 % (20/29).Five (17.2 %) patients achieved complete remission (CR+CRu),15 (51.7 %) patients achieved partial remission,and 3 (10.3 %) patients had stable disease (SD),6 (20.7 %) patients had progressive diseases(PD).Median progression-free survival (PFS) was 6 months.1-and 2-year PFS rates were 39.0 % and 20.0 %.1-,2-and 5-year overall survival (OS) rates for all patients were 76.8 %,53.4 % and 17.1%,respectively.PFS was significantly better in chemotherapy-sensitive patients (P < 0.001).The responses to chemotherapy had a tendency of affecting the OS,but it failed to reach statistical significance (P > 0.05).Conclusions The CHOP or CHOP-like regimen maybe induce unfavorable efficacy in AITL patients.Further therapeutic options are required to improve the outcome.
7.Effect of human telomerase reverse transcriptase gene antisense oligodeoxynucleotide on telomerase activity in leukemia cells.

To explore the effect of human telomerase reverse transcriptase (hTERT) gene antisense oligodeoxynucleotide (ASODN) on telomerase activity in primary acute myeloid leukemia (AML) cells and chronic myeloid leukemia (CML) cells, polymerase chain reaction enzyme-linked immunosorbent assay (PCR-ELISA) was used to determine telomerase activity. The expression levels of hTERT protein were assayed by flow cytometry using immunofluorescence labeling. Immunofluorescence assay showed that the expression levels of hTERT protein in AML and CML cells decreased with time after hTERT ASODN treatment. There was no difference in hTERT protein levels between control and sense oligodeoxynucleotide (SODN)-treated cells. Telomerase activity decreased when AML and CML cells were treated with ASODN for 48 h. Telomerase activity of AML and CML cells was significantly inhibited when the cells treated with ASODN for 72 h. There was no difference in telomerase activity levels between control and hTERT SODN-treated cells. It was concluded that hTERT ASODN could inhibit hTERT protein expression level, thus decreasing the telomerase activity in primary AML and CML cells.

9.Effect of Inhibition of Telomerase on Cisplatin-Induced Apoptosis in HL-60 Cells

To explore the effect of inhibition of telomerase activity on cisplatin-induced apoptosis in HL-60 cells, polymerase chain reaction-enzyme-linked immunosorbent assay (PCR-ELISA) was used to determine telomerase activity in HL-60 cells untreated or treated with human telomerase reverse transcriptase (hTERT) gene antisense oligodeoxynucleotide (ASODN); apoptosis was detected by morphological observation and cell cycle analysis by flow cytomertry. Telomerase activity in HL-60 cells treated with hTERT ASODN was shown to decrease with time of treatment. The combination of hTERT ASODN and cisplatin could induce apoptosis of HL-60 cells. Morphological characteristics of apoptosis, such as cell shrinkage, chromatin condensation, nuclear segmentation and formation of apoptotic bodies, were observed. The percentage of apoptotic HL-60 cells treated with cisplatin for 48 or 72 hours after 24 hours of exposure to ASODN was significantly increased than that of groups with sense oligodeoxynucleotide (SODN) plus cisplatin or cisplatin alone (P < 0.01), respectively. However, there was no difference in percentage of apoptotic HL-60 cells between hTERT ASODN plus DNR or Ara-c, SODN plus DNR or Ara-c, and DNR or Ara-c alone, respectively. The results here demonstrated that inhibition of telomerase activity by hTERT ASODN could enhance the cisplatin-induced apoptosis of HL-60 cells.
10.Effect of Cognitive Therapy and Acupuncture on Insomnia

ObjectiveTo observe the effect of cognitive therapy and acupuncture on insomnia. Methods72 insomnia sufferers were divided into two groups averagely. 36 sufferers were treated with cognitive therapy and acupuncture (treatment group), and other 36 sufferers were treated with acupuncture (control group) only.ResultsA month after treatment, ,15 sufferers were cured, 18 sufferers were obviously improved, 2 sufferers were improved and one did not show any improvement in the treatment group.Otherwise, 6 sufferers were cured, 12 sufferers were obviously improved, 12 sufferers were improved and 6 did not show any improvement in the control group. (P<0.05)ConclusionCognitive therapy and acupuncture used together shows more efficacy on insomnia compared with acupuncture alone.
