1.Expression of HSP27 and GPC-3 in primary hepatic cancer and its clinical implication

Objective To investigate the clinical significance of heat shock protein 27(HSP-27) and glypican-3(GPC-3) in diag-nosing primary hepatic cancer (PHC) and the clinicopathologic characteristics by detecting their expression in PHC .Methods Im-munohistochemistry was used to detect the expression of HSP-27 and GPC-3 in 32 cases of PHC tissue ,30 cases of live cirrhosis tissue ,25 cases of benign liver occupying tissue and 31 cases of hepatitis tissue .The relationship between their expression and the clinicopathologic features of PHC was analyzed .Results HSP-27 and GPC-3 were highly expressed in the PHC tissues ,the differ-ence had statistical significance compared with their expression levels in the liver cirrhosis tissues ,liver benign neoplasm tissues and hepatitis tissues(P<0 .05) .The expression of HSP-27 in the PHC tissues was correlated with the tumor pathological grade and the TNM stage ,while the expression of GPC-3 was not correlated with the tumor pathological grade and the TNM stage .Conclusion The high expression of HSP-27 and GPC-3 in the PHC tissue is closely related with the tumorigenesis ,progress ,metastasis of PHC ,which may become new pathological diagnosis markers of PHC .
2.Survey of Phytomedicine's Development in Germany and Its Meaning for Traditional Chinese Medicine

OBJECTIVE:To outline the development of phytomedicine in Germany to offer a reference for traditional Chinese medicine.METHODS:From the aspects of research,production and market to make an exposition of phytomedicine's develop?ment in Germany.RESULTS:There existed a gap between Germany and China in technology and standards in production of botanicals.CONCLUSION:We should use the experience of Germany in developing botanicals to draw up international quality standard of traditional Chinese medicine.
3.IDENTIFICATION OF HUMAN PAPILLOMAVIRUSES DNA IN LESIONS FROM WOMEN WITH CONDYLOMA ACUMINATA BY MOLECULAR HYBRIDIZATION

HPV DNA were detected in 60 biopsies from women with condyloma acuminata (vulva 47,Vagina 12,cervix 1)by dot blot hybridiztion;physical states of 8 HPV 11 DNA were analyzed by techniques of restriction endonuclease and Southern blot hybridization. HPV 11 was found in 19biopsies of vulva (13 ). vagina (5)and cervix (1),and HPV 16 and 18 in lvaginal and 1 vulvar biopsy respectively. All of HPV 11 DNA were episomal in cells . Our results provided the data for further studying the relationship between HPV and genital cancer in our country.
4.THE UPPER AIRWAY CT SCAN OF OBSTRUCTIVE SLEEP APNEA PATIENTS

Compared with CT scan results of OSAS patients and normal adults to explore the value of CT diagnosis of upper airway stricture in patients with OSAS, 35 patients with OSAS and 35 normal adults were included in the study. 22, 19, 12, 8 cases had obstruction at palate, uvula, lingua and epiglottis region respectively, the awaked CT scan results of 61 obstructive sites were compared with those of normal adults. Obvious difference was found between patients and normal adults in area, left right dimension and coronal dimension. difference was also found in thickness of lateral pharyngeal tissue of palate, uvula and lingua region and retropharyngeal tissue of palate and uvula. The normal extent was determined by unilateral 95% confidence interval in normal adult group. And 43 sites had abnormal CT scan results, the positive ratio was 72 13%. The results sugget that there are obvious differences between the patients with OSAS and normal adults, the CT scan measure can show the upper airway obstructive site of patients with OSAS.
5.Effect of Acupuncture plus Medication on HAMD, SDS and H-P-O Axis in Perimenopausal Depression Patients

Objective By observing the clinical efficacy of acupuncture plusBu Shen Jie Yu Qing Xin decoction (herbal decoction for tonifying kidney, anti-depression and clearing heart fire) in treating perimenopausal depression, to discuss about the effect of acupuncture plus medication on Hamilton Depression Scale (HAMD), Self-rating Depression Scale (SDS) and hypothalamic-pituitary-ovarian (H-P-O) axis in perimenopausal depression patients.Method By following a completely randomized design, sixty perimenopausal depression patients were divided into a Chinese medication group (Bu Shen Jie Yu Qing Xin decoction) with 30 cases, and an acupuncture-medication group (acupuncture plusBu Shen Jie Yu Qing Xin decoction) with 30 cases, to receive 12-week treatment in total. The HAMD and SDS were estimated prior to the treatment and in the 4th, 8th and 12th weeks; the serum follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2), testosterone (T), prolactin (PRL), and progesterone (P) contents were detected before and after the treatment.Result Acupuncture plus medication and Chinese medication alone were both effective in treating perimenopausal depression, and the therapeutic efficacy of acupuncture plus medication was superior to that of Chinese medication alone (P<0.05). In the acupuncture-medication group, the HAMD score dropped significantly in the 4th, 8th and 12th weeks compared to that before the treatment, and the score dropped significantly in the 8th and 12th weeks compared to that before the treatment in the Chinese medication group (P<0.01). The HAMD score in the acupuncture-medication group was significantly lower than that in the Chinese medication group in the 4th, 8th and 12th weeks (P<0.01). After the treatment, the items including anxiety, insight, sleep, and despair were significantly changed in the Chinese medication group (P<0.05); 7 items were significantly changed in the acupuncture-medication group (P<0.05). The scores of anxiety, insight, sleep, diurnal-nocturnal, obstruction, and body weight in the acupuncture-medication group were significantly superior to those in the Chinese medication group (P<0.05). The SDS score dropped significantly in the 4th, 8th and 12th weeks in the acupuncture-medication group, and in the 8th and 12th weeks in the Chinese medication group (P<0.01,P<0.05); in the 4th and 8th weeks, the SDS score in the acupuncture-medication group was significantly lower than that in the Chinese medication group (P<0.05), while the between-group difference was statistically insignificant in the 12th week (P>0.05). After 12-week treatment, the contents of FSH and LH declined significantly in both groups (P<0.01, P<0.05), and the contents of E2 and T increased significantly in the acupuncture-medication group (P<0.01,P<0.05). The decrease of FSH and LH, and the increase of E2 were more significant in the acupuncture-medication group (P<0.05,P<0.01).Conclusion Acupuncture plus medication can improve the symptoms of perimenopausal depression, and it takes a faster action than Chinese medication alone. Acupuncture plus medication can mitigate 7 items in HAMD, while Chinese medication can improve anxiety, insight, sleep and despair. Acupuncture plus medication can produce a more significant efficacy in improving anxiety, insight, sleep, diurnal-nocturnal, obstruction, and body weight compared to Chinese medication. For perimenopausal depression, acupuncture plus medication can correct the endocrine disorder and improve depression state by down-regulating the levels of FSH and LH, and up-regulating the contents of E2 and T to improve the function of H-P-O axis.
6.Pathological changes of soft tissues and transforming growth factor beta 1 expression in radiation masseter injury models

BACKGROUND: Long-term radioactive ray accumulation leads to surrounding tissues fibrosis accompanied by limitation of mouth-opening, which seriously affects life quality of tumor patients. OBJECTIVE: To observe pathological change of soft tissues in rats with radiation masseter injury and the level of transforming growth factor β1 (TGF-β1) mRNA expression in vivo.METHODS: A total of 30 Wistar rats were randomly divided into control group (n=10) and radiated group (n=20). Radiated group were radiated with linear accelerator at a dose of 40 Gy. The pathological changes of vessels were observed under light microscope and electron microscope and expression of TGF-β1 was detected by RT-PCR. RESULTS AND CONCLUSION: The microvessel density of radiated group was obviously decreased than that of the control group (P < 0.01), but the mRNA expression of was higher than that in the control group (P < 0.01). It suggests that radiated injury can induce fibrosis to repair the radiated injury.
7.Treatment of the Pipkin fractures

Objective To investigate the treatment of Pipkin fractures and curative effect. Methods From January 2003 to November 2009, we treated 19 cases of posterior dislocation of the hip with fracture of the femoral head,with type Ⅰ 8 cases, type Ⅱ6 cases, type Ⅲ 1 case, type Ⅳ4 cases. Seventeen patients were treated according to the illness with internal fixation treatment. Results Among type Ⅰ 8 cases, 2 cases were good with conservative treatment, 3 were excellent and 3 were good with surgical treatment;Among type Ⅱ6 cases, 3 were excellent, 2 were good, 1 was fair; 1 patient of type Ⅲ was fair; In 4 cases of type Ⅳ, 1 case was excellent, 2 cases were good, 1 case was fair. Conclusions Surgical treatment as soon as quickly is preferred for posterior dislocation of the hip with fracture of the femoral head( Pipkin fractures) , but the surgical time and methods should be chosen according to patients' detailed illness. It is important reserving the hip bone for preventing traumatic arthritis, and surgical skills to protect blood supply should not be ignored.
8.Flexible spectral imaging color enhancement for determining the demarcation of early gastric cancer and precancerous lesions

Objective To evaluate the flexible spectral imaging color enhancement (FICE) system for determine the margin of early gastric cancer and precancerous lesions.Methods From February 2008 to October 2011,a total of 51 patients with early gastric cancer or high-grade intraepithelial neoplasm who received ESD were enrolled and randomly divided into experimental group to determine lesion margin by FICE (n =26) and control group to identify lesion margin by indigo carmine (n =25).Histological complete resection rate (the lateral and vertical margins were free of cancer),operation time,complications,post-ESD ulcer-healing rates and local recurrence were assessed.Results The histological complete resection rate,acute minor bleeding rate and post-ESD abdominal pain rate in experimental group were 2.3% (24/26),88.5% (23/26),15.4% (4/26) and 73.1% (19/26),which were not significantly different from those of control group,i.e.,92.0% (23/25),84.0% (21/25),12.0% (3/25)and 72.0% (18/25) (P>0.05).The mean operation time of in experimental group was shorter than that of control group (P < 0.05).No massive bleeding,delayed bleeding or perforation occurred in either group.Follow-up showed no local residue or recurrence.Conclusion FICE is safe and effective to determine the tumor demarcation of early gastric cancer and high-grade intraepithelial neoplasm,and needs less operation time.
9.Effect of quercetin and psoralen on proliferation in MCF-7 cells

Aim To investigate the effects of quercetin and psoralen on the proliferation of human breast carcinoma cells in vitro.Methods Effects of quercetin and psoralen on the cell proliferation was tested in ER-positive MCF-7 cells by flow cytometer and Western blot.And the estrogen-like effect of psoralen and its relation with the estrogen-receptor were evaluated by pure estrogen receptor antagonist ICI182,780 as a tool.Results ① Psoralen(10 ?mol?L-1) and quercetin(10 ?mol?L-1) stimulated proliferation of MCF-7 cells compared with vehicle control,and the cell cycle was impulsed from G1 to S,DNA synthesizing was enhanced.It was also found the above function on boosting MCF-7 cell proliferation could be inhibited by adding estrogen receptor antagonism ICI182,780.② Psoralen(10 ?mol?L-1) and quercetin(10 ?mol?L-1) up-regulated ER? protein levels without altering ER? levels.The above up-regulation on ER? protein could be inhibited by adding estrogen receptor antagonism ICI182,780.Conclusions Psoralen and quercetin have the estrogen-like activities through the estrogen response pathway.And they exert estrogenic activity to MCF-7 cell proliferation through interaction with ER? expression.
10.Preparation and study of monoclonal antibody to matrix metalloproteinase-2

AIM: To obtain a McAb that can inhibit the function of matrix metalloproteinase-2 (MMP-2), we expressed the fibronectin-like domain of MMP-2 (MFD) in vitro and prepared a McAb against MMP-2. METHODS: The purified MFD protein was used to immunize BALB/C mouse three times. Then the spleen of mouse was taken out and hybridized with hybridoma cells SP2/0. The positive cell clones were screened with ELISA method. The subtype and tissue specificity of the McAb were identified and its effect on endothelial cell migration and tube-formation was analyzed. RESULTS: After the spleen cells of the mouse and hybridoma cells SP2/0 were hybridized, a piece of cells that continuously secreted McAb against MMP-2 was obtained and named SZ-117. The titers of this McAb in culture supernatants and ascites were 2?10~-3 and 2?10~-5 , respectively. The heavy chain of the McAb belongs to IgG1 subclass. The McAb identified native MMP-2. MMP-2 existed in the stromal tissue of stomach, cholecystis, spleen, ovarian, prostate, salping and lymph node. It inhibited the invasion behavior of endothelial cells Eahy926 and pancreatic carcinoma cells 1990 and inhibited the tube-formation of Eahy926 cells. CONCLUSION: A useful tool for testing MMP-2 is obtained and it will be helpful to look for a kind of new anti-tumor material.
