Objective:To construct eukaryotic expression vectors pEGFP-N2/MafA and pEGFP-C1/MafA,encoding the mouse musculus v-maf musculoaponeurotic fibrosarcoma oncogene family,protein A(MafA)gene,for the further research of its expression in HepG-2 cells.Methods:pRNA of the mouse MafA gene was distilled by RT-PCR from the mouse,and inserted into Hind Ⅲand SalⅠrestriction sites by PCR,then cloned into pEGFP-N2 and pEGFP-C1 vectors to obtain plasmids pEGFP-N2/MafA and pEGFP-C1/MafA.Human liver cancer cells(HepG-2) were transfected with formed plasmids by means of lipidosome.The MafA-EGFP fused protein was viewed directly with fluorensce microscope,and expression of MafA and insulin Ⅱ was detected by RT-PCR.Results:The mouse MafA gene was amplified through RT-PCR and successfully cloned into transfer vectors.The favorite gene sequence could be expressed and was completely consistent with that reported in genebank,but the insulin Ⅱ gene expression was not detected.Conclusion:The recombinant plasmids pEGFP-N2/MafA and pEGFP-C1/MafA were successfully constructed.But the insulin Ⅱgene was not expressed.

Aim To explore the effects of Nimodipine(NDP) treatment on the expression of neural cell adhesion molecule(NCAM) after the ischemic brain injury in rats.Methods One hundred male Wistar rats(180~220g) were randomly divided into four groups:sham operation control group(SOCG,n=10),cerebral ischemia control group(CICG,1,3,7 d,n=30),high and low dose Nimodipine treatment group(H-NTG,L-NTG,1,3,7 d,n=10).Their middle cerebral arteries of CICG and NTG were blocked by filament forming brain injured,the NTG groups were treated with NDP(0.5 mg?kg-1 and 1mg?kg-1),respectively.and then,their cerebral blood flow was reperfused.Their blood vessel shape of cerebri surface was observed and the expression of NCAM was determined,after neurologic impairment(NIP) was scored and regional cerebral blood flow(RCBF) was surveyed,respectively.Results 3 d,7 d PO2 of H-NTG were higher than those of CICG(P

Objective To discuss the treatment value of Jinxiangdan tablets on the pre-thrombus state of patient with coronary heart disease and angina pectoris. Methods 166 cases of clinical diagnosed coronary heart disease and angina pectoris were randomly divided into the treatment group and the control group. The changes of Fg, vWF, P-selectin, D-dimer, t-PA, ET-1 of two groups before and after the treatment were observed. Results Before the treatment, Fg, vWF, P-selectin, D-dimer and ET-1 elevate, while t-PA reduces. After the treatment of Jinxiangdan, Fg, vWF, P-selectin, D-dimer and ET-1 were decreased, t-pA were increased. Conclusion Jinxiangdan tablets can improve the hemoglutination fibrinloytic system and secretion function of endothelial cell, reduce the activity of platelet, to prevent the cardiovascular events.

Objective To evaluate the pharmacokineties of ropivacaine for single sciatic nerve block in dogs.Methods Twelve healthy adult mongrel dogs weighing 14-17 kg were randomized to receive 0.5% ropivaeaine 10 mg/kg or 20 mg/kg for single sciatic nerve block(n=6 each).ECG,BP and HR were monitored and recorded during anesthesia.Blood samples were obtained from femoral artery before ropivacaine injection (baseline)and at 10,20,30,40,60,90,120,150,180,240,360 and 720 min after ropivacaine injeetion for determination of plasma ropivaeaine concentration(by reversc-phasc high performance liquid chromatography).Arterial blood samples were also taken when adverse reactions occurred.The pharmacokinetic parameters were eMeulated with DAS 1.0 software package.Results The concentration-time curve of ropivacaine for single sciatic nenre block was fitted to two-compartment open model in both groups.The peak plasma concentration of ropivaeaine was significantly lower in 10 mg/kg group than in 20 mg/kg group.Two dogs developed convulsion in 20 mg/kg group.The plasma ropivaeaine concentration was 12.56 and 13.67 mg/L respectively during convulsion.Conclusion Pharmaeokinetic profile of ropivaeaine for single sciatic nerve block is best described by two-compartment model.Bopivaeaine 20 mg/kg for sciatic nerve block can hardly be tolerated by dogs.

OBJECTIVE:To evaluate the permeation enhancing effect of Flos Caryophylli volatile oil versus azone on diclofenac sodium.METHODS:Percutaneous absorption experiment was performed on the apparatus of isolated skin,and the penetrated amounts of diclofenac sodium at different time were measured,then the steady-state flux and enhancing rate were calculated,and the deposit effect of diclofenac sodium was investigated.RESULTS:Flos Caryophylli volatile oil and azone all had promotion effect on percutaneous absorption of diclofenac sodium,especially when they used in combination.Used either in combination or alone,they all had remarkable enhancement property on deposit effect of diclofenac sodium(P

Objective To investigate the effect and function analysis by sulfentanyl in patients with tongue cancer postoperative analgesia.Methods A total of 74 patients with radical resection of tongue cancer in department of anesthesiology with general anesthesia from our hospital were collected, patients or their families signed consent, according to postoperative self-controlled intravenous analgesia drugs divided into experimental group and control group with 37 cases in each group.Patients in control group were treated by fentanyl self-controlled intravenous analgesia;patients in experimental group were treated by sulfentanyl self-controlled intravenous analgesia, determination of the analgesic effect, sedative effect and the changed of vital signs postoperative 2, 4, 8, 12, 24, 48 h,at the same time recorded complications.Results After operation, at different time points analgesic effect and sedative effect were changed in the two groups, the VAS scored of the experimental group was lower than the control group postoperative 2, 4, 8, 12, 24, 48 h time points, the Ramsay Sedation scored of the experimental group was lower than the control group postoperative 2 h, 4 h, 8 h, 12 h time points ( P <0.05 ); the changed of vital signs and adverse reactions between the two groups had no difference within 48 hours ( P >0.05 ) . Conclusion Sulfentanyl in the treatment of patients with radical resection of tongue cancer the analgesic effect and sedative effect are better than fentanyl, has no effect on the vital signs, and the role is security and exact.
