1.Control of Postpartum Bleeding by Rectal Misoprostols: A Report of 3 Cases.

Postpartum hemorrhage is an important cause of maternal mortality and morbidity. Especially uterine atony is the most common cause of postpartum hemorrhage. Conventional method to control postpartum uterine atonic bleeding is based on the use of oxytocin and ergot preparations. Prostaglandin F2alpha analogue such as carboprost can be used to promote contraction when these agents fail to produce uterine contraction. Prostaglandin E1 analogue, misoprostol has uterotonic effect by oral or vaginal administration. They are used to induce labor and first or mid trimester abortion. In postpartum uterine atonic bleeding, misoprostols cannot be used via oral or vaginal route. Recently we have experienced that postpartum uterine atonic bleedings unresponsive to conventional methods were controlled by rectal misoprostols. So we report these cases with a brief review of literatures.

2.Independent or combined effects of endothelin-1 and prostaglandin F2alpha on cardiomyocytes.

AIMTo elucidate the independent or combined effects of endothelin-1 (ET-1) and prostaglandin F2alpha (PGF2alpha) on cardiomyocytes and investigate the relationship between hypertrophy and apoptosis of cardiomyocytes.
METHODSCultured neonatal rat cardiomyocytes were stained with FITC-conjugated phalloidin and eosin to detect the cardiomyocyte hypertrophy evidenced by increased sarcomeric structure and cell size. Cardiomyocytes were stained with Hoechst 33258 to detect apoptotic nuclei showing features of condensation and fragmentation.
RESULTSCardiomyocyte hypertrophy induced by ET-1 or PGF2, shown a dose dependent effect. The area of cardiomyocytes treated by 10 nmol/L or 100 nmol/L of ET-1 for 24 h increased 68% or 84% as compared with control, respectively. The area of cardiomyocytes exposed to 10 nmol/L or 100 nmol/L of PGF2alpha for 24 h increased 28% or 106% as compared with control, respectively. The ET-1 and PGF2alpha had a synergic effect on cardiomyocyte hypertrophy, but not superimposed effect. The area of cardiomyocytes increased 80%, 122%, 96%, and 199% in 10 nmol/L ET-1 plus 10 nmol/L PGF2, 10 nmol/L ET-1 plus 100 nmol/L PGF2alpha, 100 nmol/L ET-1 plus 10 nmol/L PGF2alpha, and 100 nmol/L ET-1 plus 100 nmol/L PGF2alpha group, respectively. There were no changes in apoptotic rate of cardiomyocytes treated by ET-1 or PGF2alpha alone for 48 h. The apoptotic rate of cardiomyocytes also didn't increase in ET-1 plus PGF2alpha treatment for 24 h groups, but significantly increased in ET-1 plus PGF2alpha treatment for 48 h groups. ET-1 or PGF2alpha could induce an increase in apoptotic rate of hypertrophic cardiomyocytes. There was a positive relationship between hypertrophic extent and apoptotic rate in cardiomyocytes.
CONCLUSIONThe cardiomyocytes treated by ET-1 or PGF2alpha alone only show hypertrophy, but treatment of ET-1 plus PGF2alpha for 48 h induces apoptosis of cardiomyocytes.

3.Factors affecting the success of resynchronization protocols with or without progesterone supplementation in dairy cows.

The objective of this study was to investigate factors that influence the success of resynchronization protocols for bovines with and without progesterone supplementation. Cow synchronized and not found pregnant were randomly assigned to two resynchronization protocols: ovsynch without progesterone (P4) supplementation (n = 66) or with exogenous P4 administered from Days 0 to 7 (n = 67). Progesterone levels were measured on Days 0 and 7 of these protocols as well as 4 and 5 days post-insemination. Progesterone supplementation raised the P4 levels on Day 7 (p < 0.05), but had no overall effect on resynchronization rates (RRs) or pregnancy per artificial insemination (P/AI). However, cows with Body Condition Score (BCS) > 3.5 had increased P/AI values while cows with BCS < 2.75 had decreased P/AI rates after P4 supplementation. Primiparous cows had higher P4 values on Day 7 than pluriparous animals (p = 0.04) and tended to have higher RRs (p = 0.06). Results of this study indicate that progesterone supplementation in resynchronization protocols has minimal effects on outcomes. Parity had an effect on the levels of circulating progesterone at initiation of the protocol, which in turn influenced the RR.

4.Observation on therapeutic effect of acupoint application on dysmenorrhea of excess syndrome and effect on prostaglandins.

OBJECTIVETo observe therapeutic effect of acupoint application on dysmenorrhea of excess syndrome and its effect on prostaglandins.
METHODSThe patients with primary dysmenorrhea of excess syndrome were randomly divided into an application group and a medication group. The application group of 31 cases were treated with application of Chinese medicine composed of Zhinanxing (Rhizoma Arisaematis), Sanleng (Rhizoma Sparganii), Ezhu (Rhizoma Zedoariae), etc. at Zhongji (CV 3), Guanyuan (CV 4), Qihai (CV 6); and the medication group of 30 cases were treated with oral administration of Tianqi Tongjing Capsules. The scores for the pain degrees and the duration of dysmenorrhea were observed and prostaglandin F2alpha (PGF2alpha) and prostaglandin E2 (PGE2) contents in peripheral blood were determined before and after treatment.
RESULTSThe total effective rate of 93.5% in the application group was significantly better than 73.3% in the medication group (P < 0.05); after treatment, the scores of the symptoms were significantly decreased in the two groups (both P < 0.01), with more obvious improvement in the application group than the medication group (P < 0.01). The PGE2 content was significantly increased, and the PGF2alpha content and PGF2alpha/PGE2 were significantly decreased in the application group (all P < 0.01).
CONCLUSIONAcupoint application has a better therapeutic effect on dysmenorrhea of excess syndrome and has benign regulative action on synthesis of prostaglandins in the patients with primary dysmenorrhea.

5.Calcium metabolism in cows receiving an intramuscular injection of 1,25-dihydroxyvitamin D3 combined with prostaglandin F2alpha closely before parturition.

To determine the effect of exogenous 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] combined with induced parturition on calcium (Ca) metabolism, cows received a single intramuscular injection of 1,25(OH)2D3 and prostaglandin F2alpha (PGF2alpha) closely before calving. Ten late-pregnant, multiparous Holstein cows were assigned to 1,25(OH)2D3 group (five treated with both 1,25(OH)2D3 and PGF2alpha) and control group (five treated with PGF2alpha). 1,25(OH)2D3 group showed an increase in plasma Ca concentration around parturition, whereas control group revealed a decrease in plasma Ca level. Plasma Ca concentration in 1,25(OH)2D3 group were significantly higher than that in control group during .0.5 to 3 days after parturition.

6.Monitor on influence of quality standard improvement upon Guizhi Fuling capsules efficacy.

In 2012, the preparation process and quality standard for Guizhi Fuling capsule were improved. To compare the effects and differences of capsules before (2011) and after(2012-2014) the improvement, evaluation models for intrinsic dysmenorrhea, pelvic inflammation and hysteromyoma were applied in rats. Models were induced by oxytocin, liqiud bacteria mixture and estrogen loading, respectively. The capsules (12 batchs/year, 48 bathcs in all), sampled randomly in 2011-2014, the effects were assessed using the three models. In anti-dysmenorrhea models, remarked reduction of writhing frequency, ET-1 and PGF2α content in uterus could be detected, as well as extension of writhing latency. In pelvic inflammation rats, depression of TNF-α and raise of IL-2 were induced by earh batch of capsules. In hysteromyoma model, uterine weight and smooth muscle proliferation, including E2 and P level in plasma, were lowered obviously by all batchs of capsules. Secondly, Guizhi Fuling capsules produced in 2012-2014 revealed better effectiveness than the ones manufactured in 2011. Moreover, pharmacodynamics indexes of the samples made in 2011 differed significantly between groups, which could not be observed in the ones ot 2012-2014. After tne preparation process and quality standard improvement, the effectiveness and homogeneity of Guizhi Fuling capsules were enhanced.
