Objective The TCS gene without N-and C-terminal sequences was cloned into pET-29b in order to get the recombinant trichosanthin(rTCS) at the relatively high level in E.coli, and the rTCS immunogenicity was compared with natural TCS(nTCS). Methods TCS gene without the N-and C-terminal coding sequences was amplified by RT-PCR,and then inserted into pET-29b.The soluble rTCS was induced with IPTG and purified through metal chelate affinity chromatography, the purity and content were showed by the thin-layer scan analysis and bradford assay. After the rabbits were immunized with purified rTCS and nTCS,respectively, the titer and specificity of polyclonal antibody were detected by indirect ELISA assay and Western blotting. Results 1.The vector pET-29b-TCS was constructed successfully after it was confirmed with the same restriction enzymes.2.The rTCS attained the maximum in the presence of 1 mmol/L IPTG for 4?h at 30℃.3.The soluble rTCS was expressed at relatively high level(36% of total protein in E.coli) and the yield of rTCS with a purity of 95% was 1.2?g in 1 L bacterium.4.The polyclonal antisera titer of nTCS was significantly higher than that of rTCS(1∶40?960 and 1∶5?120 respectively).5.The specific reaction between rTCS and anti-nTCS antibody was detected by Western blotting. Conclusion The high level expressing vector pET-29b-TCS,carrying TCS gene withoug the N-and C-terminal coding sequences, was constructed successfully. The immunogenicity of rTCS was remarkably lower than that of nTCS,which indicates that glycosylation of nTCS has a relationship with the antigenicity.

Cytotoxic substances and ionizing radiation can easily induce DNA damage, and double strand breaks (DSBs) are the main form of DNA damage. DNA damage can activate intracellular DNA damage responses and further induce related biological effects, such as DNA damage repair and cell cycle arrest. Homologous recombination (HR) is the primary DSB repair mechanism in eukaryotes. Abnormal expression of Rad51C, which is a key factor in the HR pathway, may result in DNA repair disorder, genomic instability, and eventually lead to tumor formation. In recent studies, researchers considered Rad51C as a potential target for cancer treatment. We reviewed the research progress on Rad51C in DNA damage repair and radiotherapy.

Objective To evaluate the effect of vacuum suction on shaping sliming and breast anaplasty, to expand indications of vacuum suction, and to improve its result. Methods Based on injection of swelling solution more than routine dosage, vacuum suction was used in totally 825 cases, including local fat deposit in face or other sites of body, macromastia in female, hypermastia in male, lump after breast anaplasty by autograft of fat granule, as well as infection and induration after breast anaplasty by injection of polyacrylamide. Results Satisfactory rate of subjects to vacuum suction was up to 96 %, and clinical efficacy rate of the operation was 100 %. As to sliming cases, there were significant decreases in their measurements of figures or dimensions. In case of the lump from breast shaping by fat granule autograft, as well as the infection and induration caused by injection of polyacrylamide during breast anaplasty, vacuum suction could resolve all of them wonderfully. Conclusions When used properly, vacuum suction may play an excellent role in shaping sliming and breast anaplasty, and it can also resolve those complications brought up in breast shaping by fat granules autograftor and by polyacrylamide injection. Vacuum suction decreases the pain and cicatrisation caused by invasion surgery significantly.

OBJECTIVE To establish a rapid method of detecting class D carbapenemase gene in Acinetobacter baumannii by multiplex PCR.METHODS Design primers according to the conservative region of blaOXA-51 and blaOXA-23,the class D carbapenemase gene were detected by Multiplex PCR.RESULTS 24 of 30 A.baumannii isolates produced two bands of blaOXA-51 and blaOXA-23;other 6 isolates showed a band of blaOXA-23.CONCLUSIONS The multiplex PCR could detect rapidly the class D carbapenemase gene.

OBJECTIVE To explore the features, developing trend and factors of hospital infection in patients with iatrogenic injury.METHODS Retrospective surveys of hospital infections in 750 cases with iatrogenic injury were carried out in hospital between 2000 and 2007.RESULTS The incidence of hospital infection in patients with iatrogenic injury was 87.39%.Among them, hospital wound infection was the highest(87.39%),the second one was cavity infection(8.98%).The leading causative microorganisms were Gram-negative bacteria.CONCLUSIONS It is imperative to make effort to decvease the morbidity of hospital infections. We hope that the iatrogenic injury can be brought into the category of public health as soon as possible to enhance the medical care service and secure safty and health of the patients.

Objective To investigate the prevention and management of anastomotic leakage in patients with middle and lower rectal cancer after neoadjuvant therapy. Methods In this study,50 cases of middle or lower rectal carcinoma underwent anus preserving radical resection and anastomosis after a 4 cycles of chemotherapy with FOLFOX and 46 Gy(2 Gy×23)radiotherapy.Anastomotic leakage, the prevention and management of this complication were analyzed retrospectively. Results All these 50 patients received low anterior resection after chemoradiotherapy, among the 19 cases receiving prophylactic ileac stoma simultaneously, there was no anastomotic leakage, whereas anastomotic leakage developed in 4 out of the 31 patients who did not receive prophylactic ileac stoma, including concomitant rectovaginal fistula in 2 cases. All these fistulae were cured conservatively. Conclusion Concomitant treatment of FOLFOX regimen and RT in neoadjuvant setting of rectal cancer could increase the rate of anal interior sphincter reservation, prophylactic ileac stoma helps to reduce the rate of anastomotic leakage after a anus-preserving low anterior resection of the cancer.

Objective To clone, express and purify the extracellular carbohydrate recognition do-main(CRD) of Dectin-1 in mouse peritoneal macrophages and to further investigate its ability to recognize and bind to β-glucans. Methods The Dectin-1 CRD gone was amplified by RT-PCR from RNA of mouse peritoneal macrophages and cloned into prokaryotic expression vector pET28a (+), the constructed pET-CRD recombinant plasmid was transformed into Escherichia coli BL21 (DE3) and the fusion protein was in-duced to express. After affinity purification and renaturation, the fusion protein was incubated with Candida albicans yeast and its ability to recognize and bind to β-glucans in the cell wall of fungi. Results The fu-sion protein could recognize β-glucans in the fungal cell wall. Conclusion The recombinant expression plasmid pET28a-CRD was successfully constructed and the fusion protein was induced. The fusion protein is able to recognize and bind to β-glucans in the fungal cell wall, thus laying a good foundation for fungal de-tection and the exploration of the biological role of β-glucans.

Journal of Leukemia & Lymphoma 2010;19(1):4-7
This critical review was summarized more systematically about the JAK2V617F mutation of related research progress in myeloproliferative disorders (MPD) research fields and the identification of JAK2V617F mutation represents an important advance in our understanding of MPD was agreed. The authors focused on several sensitive problems of post the JAK2 mutation era, and expressed their opinions. The Guideline of the MPD diagnostic criteria recommended by WHO in 2008 was accepted. The authors recommend the MPD, rather than myeloproliferative neoplasm (MPN). The treatment for the MPD (not including the CML) is recommended. Before the effective targeting of JAK2V617F specific inhibitors for the treatment of the MPD, short-term of use hydroxyurea (HU) was suggested to suppress excessive proliferation of bone marrow of MPD and a long course of treatment application of inteferon-α(IFN-α), and low-dose of aspirin in a timely manner were recommended to prevent thrombosis and other complications.
