1. Establishment of fingerprint of Ginkgo biloba leaves and study on its quality evaluation

To compare the quality Ginkgo biloba L. leaves collected form various habitats, different tree ages, and different harvesting times and to confirm the source of G. biloba leave used in Xin Mai Capsule. To establish the HPLC fingerprint of G. biloba leaves to determine the fingerprint and content of total flavonol glucoside and terpene lactones in G. biloba leaves collected from various habitats, different tree ages, and different harvesting time, and then to analyze the results. The contents of total flavonol glucoside and terpene lactones and fingerprint similarity in G. biloba leaves from various regions, different tree ages, and collecting times were different. The fingerprint control method has provided the scientific basis for the reasonable collection and quality evaluation of G. biloba leaves. The G. biloba leaves in June to August, 2-3 year-old from Pizhou, Jiangsu province are better, which could be considered as the source of G. biloba leaves used in Xin Mai Capsule.
2. Variation analysis on triterpene compounds in hawthorn from different regions

Objective: To establish an HPLC method for the content determination of ursolic acid and oleanolic acid in hawthorn (Crataegi Fructus) from different regions, whose differences were compared. Methods: Waters Symmetry C18 column (4.6 mm× 250 mm, 5 μm) was used with the mobile phase of acetonitrile-methanol-0.5% ammonium acetate (69:12:19) at a flow rate of 1 mL/min. The detection wavelength was set at 210 nm and the column temperature was 30°C. To determine the content of hawthorn from 10 different regions, variance analysis and cluster analysis were carried out for the results. Results: The linear ranges of ursolic acid and oleanolic acid were 0.078-0.780 mg/mL(r = 0.9998, n = 6) and 0.0169-0.169 mg/mL (r = 0.9996, n = 6), respectively. The average recoveries of the two components were 102.6% and 100.2%, in accordance with the determination requirement in Chinese Pharmacopoeia 2010. Conclusion: This method is simple, quick, accurate, and has better reproducibility for the determination of triterpene compounds in hawthorn. And hawthorn from Chengde and Jiangsu provinces is better. It could be used as the origin of hawthorn in the prescription of Xin mai Capsule.
3.Quantitative analysis of coronary artery ostia anatomy using three-dimensional trans-esophageal echocardiography

Objective To quantitatively analyze the coronary artery ostia by three-dimensional trans-esophageal echocardiography (3D-TEE).Methods The full-volume images of aortic root and coronary artery ostia were acquired by 3D-TEE in 95 adult patients.The Philips QLab 3DQ measurement technology was employed to determine three mutually perpendicular planes:(1) The transverse plane cross the bottom of three coronary artery sinus.(2) The sagittal plane perpendicular to sino-tubular junction.(3) The coronal plane perpendicular to the aforementioned two planes .The following relevant parameters were measured and recorded:(1) Length, width, height and area of bilateral coronary artery ostia .(2) The angle between coronary arterial outflow tract and aortic root in sagittal plane .(3) The spatial distribution of coronary artery ostia, aortic root and coronary artert sinus .Results The shape of left coronary artery ostia were more regular (round or oval) than right coronary artery ostia ( teardrop-shape or oval ).Calcification was more frequent in right coronary artery ostia (81/95, 85.26%) than that in left coronary artery ostia. There were statistical differences between left and right coronary artery in the parameters of ostial wide , area and height (t =3.85, 3.86, -4.49, all P<0.01).Most left coronary artery ostia were located inside the sinus (76/95, 80.00%), mainly in the upper third segment (69/95, 72.63%); while more than half of the right coronary artery ostia were found outside the sinus ( 53/95, 55.79%).The difference was statistically significant( χ2 =25.91, P<0.01).Conclusion The quantitative analysis of aortic root and coronary artery ostia based on the full-volume images originated from real-time 3D-TEE is feasible, which is helpful for further clinical research .
4.Methodology study for the anatomic structure of aortic root using three-dimensional transesophageal echocardiography

Objective To establish a quantitative analysis method for the anatomic structure of aortic root by real-time three-dimensional transesophageal echocardiography,and to explore the reliability and repeatability of this method.Methods Ninety five adult patients underwent RT-3DTEE were included in this study.Full-volume 3D images of aortic root were acquired and analyzed offline with QLab-3DQ (iE33, Philips).The three mutually perpendicular plane were determined.Results 1 )Aortic root:At the end of diastole,the diameter and area of coronary artery sinus bottoms level was 1 .84 cm and 3.45 cm2 ;the diameter and area of top commissural level was 2.97 cm and 7.77 cm 2 ;the diameter and area of sino-tubular junction level was 2.72 cm and 5.44 cm 2 .2)Aortic sinus:right coronary sinus was widest,left coronary sinus was the most stenosis(P <0.05).The repeatability of each parameters was well (P >0.50).The measurement of each parameter,which analysis among different angles showed that 0° region were more accurate than others.Conclusions 3D quantitative analysis to aortic root can be established by full-volume RT-3DTEE images and has better reliability and repeatability.
5.The mechanical characterization study of mitral annulus at different sites in normal adults using ultrasonic dual pulse-wave Doppler

Objective To define the mechanical features of mitral annulus at various sites,and to investigate the specific mechanics characterization at different mitral annulus sites in evaluation consequences of left ventricular function by dual pulse-wave Doppler (DPW) technology.Methods The DPW spectrums were obtained at lateral and aboral interval,anterior and inferior and posterior mitral annular from 112 normal adults.The peak systolic velocity (Sm),peak early diastolic velocity (Em),peak late diastolic velocity (Am),the beginning time of the peak and the time to peak were measured,and E/A,Em/Am,E/Em,left ventricular ejection fraction (LVEF) and left ventricular fractional shortening (LVFS) were computed.Results 1)Sm,Em,Am and Em/Am measured in the free wall annulus were significantly greater than measured in the interval annulus of mitral annular sites.However,E/Em was opposited (P < 0.05).Sm of the posterior wall mitral annulus accelerated frist and experienced shortest duration in all the mitral annular sites (P < 0.01).There were no significant differences of Em time parameters among different mitral annulus sites.Am of the beginning time and peak time in the free wall annulus were significantly longer than that in the interval annulus of anterior mitral annular sites.However,the acceleration time was opposited(P <0.05).2) Sm was correlated with LVEF and LVFS (r =0.243 and r =0.227,P <0.01) only at the posterior mitral annular site,Em/Am of anterior and posterior wall mitral annulus had the highest correlations with mitral orifice flow E/A(r =0.545 and 0.545 respectively,P < 0.01).Conclusions There are significant differences among the mechanics patterns at different mitral annulus sites in normal adults.The mechanics characterization at different mitral annulus sites have different conclusions of left ventricular function.
6.Value of Deoxyribonucleic Acid Content Determination on Evaluation of Biological Characteristic of Nephroblastoma in Children

Objective To explore the value of deoxyribonucleic acid(DNA) content on clinical pathological characteristics of nephroblastoma in children.Methods Twenty-four cases of pediatric nephroblastoma from Jan.1993 to Dec.2007 were proved by pathology.The distribution of the 24 cases according to National Wilms Tumor Study group(NWTSG) criteria of USA was stage Ⅰ(12 cases),stage Ⅱ(7 cases),stage Ⅲ(5 cases).The pathology was highly differentiated in 10 cases,medium differentiated in 8 cases and low differentiated in 6 cases according to the level of differentiation.There were 9 cases of lymphatic metastasis and 15 cases of non-lymphatic metastasis in accordance with lymphatic metastasis.The DNA contents of 24 cases were determined by flow cytometry.Chi-square test was used to analyze the relationship among DNA ploidy and clinical stage,level of differentiation,lymphatic metastasis of children with nephroblastoma.Results Aneuploidy mainly occurred in clinical stage Ⅱ or stage Ⅲ(57.14%,100.0%),there was significant difference compared with that of stageⅠ(25.0%)(P
7.Effect of Traditional Chinese Massage on Upper Limbs Spasticity in Stroke Patients Accepted Botulinum Toxin Type A Injection

Objective To explore the effect of traditional Chinese massage on upper limbs spasticity in stroke patients who accepted Botulinum toxin type A (BTX-A) injection. Methods 74 stroke patients with upper limbs spasticity were divided into treatment group (n=36)and control group (n=38). The control group received BTX-A injection, and the treatment group received massage after injection. Both groups received rehabilitation training after injection and were assessed with modified Ashworth scale, Fugl-Meyer assessment and modified Barthel index before and 2 weeks, 4 weeks, 8 weeks and 12 weeks after treatment. Results All the scores of assessments improved in both groups after treatment (P<0.001), and there was no significant difference between them (P>0.05). Conclusion Addition of traditional Chinese massage doesn't work more on spasticity of upper limbs after stroke when accepted BTX-A injection.
8.Qualitative and quantitative detection of Poria cocos by near infrared reflectance spectroscopy.

OBJECTIVEThe present study is concerning qualitative and quantitative detection of Poria cocos quality based on FT-near infrared (FT-NIR) spectroscopy combined with chemometrics.
METHODThe Poria cocos polysaccharides contents were determined by UV. Transmission mode was used in the collection of NIR spectral samples. The pretreatment method was first derivation and vector normalization. Then principal component analysis (PCA) was used to build classification model and partial least square (PLS) to build the calibration model.
RESULTThe results showed that conventional criteria such as the R, root mean square error of calibration (RMSEC), and the root mean square error of prediction (RMSEP) are 0.944 0, 0.072 1 and 0.076 2, respectively. The misclassified sample is 0 using the qualitative model built by PCA.
CONCLUSIONThe prediction models based on NIR have a better performance with high precision, good stability and adaptability and can be used to predict the polysaccharose content of Poria cocos rapidly, which can provide a fast approach to discriminate the different parts of Poria cocos.

10.Effect of wound to growth of larva of host to Ophiocordyceps sinensis during artificial breeding.

To clear the effect of the wound to the growth of the larva of the host to the Ophiocordyceps sinensis, the wounds of same severity at the same position were made artificially to the larva and which were artificial fed at the same environment and condition. The results indicated that, over the winter, the survival rate of the wounded of the infection larva was lower than that of the healthy larva, but the weight had no significant difference between the wounded and the healthy larva. The survival rate of the wounded of the no infection larva was lower than that of the healthy larva, but except with black skin, the wounded larva with offwhite and dusty red had no influence on the variety of the weight. In summery, wound had no advantage to the survival rate, but had no influence to the weight. The result had provided theoretical basis to the reforming of the system of the artificial culture O. sinensis.
