2.Preparation and immunogenicity identification of artificial antigen for luteoloside.

Lonicerae Japonicae Flos was one of the most widely used traditional Chinese medicine for its special biological activities. The content of luteoloside, one of its major compounds, was an important standard for the quantity control of Lonicerae Japonicae Flos. The major method used for the detection of luteoloside was instrumental analysis. Compared with the ELISA method, instrumental analysis was time-consuming, complex pretreatment and low-throughout. Thus, it was significantly important to develop an enzyme-linked immunosorbent assay (ELISA) for luteoloside analysis. Here, the conjugates of luteoloside-bovine (LG-BSA) and luteoloside-ovalbumin (LG-OVA) were produced as the immunogen and coating antigen by the carbodiimide ( CDI) method, respectively. The conjugation ratio of carrier protein and the hapten in the conjugate were determined by UV-Vis spectrophotometry (UV). LG-BSA conjugate was used to immunize Bal b/c mice to produce antiserum. The titer and specificity of antiserum were detected by ELISA. The conjugation ratio of hapten and carries protein were 3. 7: 1 (LG-BSA) and 1. 0: 1 (LG-OVA). The antiserum titer was higher than 2 000 with the linear range of 18.4-4 852.4 μg x L(-1), R2 = 0.988 4 and IC50 = 298.7 μg x L(-1). The result showed that the conjugate antigen LG-BSA was synthesized successfully and the mice can produce specific antiserum injected with artificial antigen.

3.Synthesis and biodistribution of a bi-functional agent 99Tcm-Gd-DTPA-DG in tumor bearing nude mice

Objective To evaluate the stability and biodistribution of a novel SPECT-MRI bi-functional agem 99Tcm-Gd-DTPA-DG in tumor-bearing nude mice. Methods DTPA-DG was synthesized and then conjugated with Gd2O3 to generate Gd-DTPA-DG. The tumor-bearing nude mice were scanned by MRI to evaluate the tumor targeting ability of Gd-DTPA-DG. The orthogonal experiment was applied to optimize pH value of reaction medium and reaction temperature. The radiolabeling efficiency was measured by thin layer chromatography. The distribution of 99Tcm-Gd-DTPA-DG in nude mice was evaluated by scintigrapy in vivo. The % ID/g was measured at different time after intravenous injection of 99Tcm-Gd-DTPA-DG. Results The tumor was significantly enhanced by Gd-DTPA-DG with MRI. The radiochemical purity of 99Tcm-Gd-DTPADG was about 98.5% and remained 96.2% at room temperature for 6 h. The tumor was well visualized by 99TcmGd-DTPA-DG SPECT at 2 h after injection. The tumor uptake was (1.48 ±0.12) %ID/g, and the rumor to muscle radioactivity ratio was 2.91. Conclusions MRI contrast of Gd-DTPA-DG may enhance tumor detection. 99Tcm-labeled Gd-DTPA-DG may be useful for tumor imaging and might have a potential role as a SPECT-MRI bi-functional agent.
4.Treatment of cicatricial stricture subsequent to esophageal chemical burns with transverse colon replacing esophagus in children.

OBJECTIVETo study the validity of transplanting transverse colon to replace esophagus in treating cicatricial stricture resulting from severe esophageal chemical burns in children.
METHODSA retrospective study was carried out on the clinical data of 46 patients with severe chemical esophageal burns who were treated from November 1972 to September 2008. The transverse colon with the ascending branch of the left colic artery was brought through a retrosternal tunnel to replace strictured esophagus. Thirty-two patients underwent colon-esophageal anastomosis and 14 patients underwent colon-pharyngeal anastomosis.
RESULTSAll patients survived after surgery, but complications occurred in 7 cases, including leakage of anastomosis in cervical region in 4 cases, stenosis of anastomosis in 2 cases, and dyspnea in 1 case, and they were cured after due treatment. Follow-up study (1 - 26 years) in 39 patients revealed that there was no difference in growth, development and diet between the patients and the normal children of the same age.
CONCLUSIONSEsophageal reconstruction with transverse colon together with the ascending branch of the left colic artery through a retrosternal tunnel is a valuable method for treating cicatricial stricture of the esophagus secondary to severe chemical burns of the esophagus in children.

5.Comparison of plaque-forming characteristics and morphogenetic changes of Newcastle disease virus isolated from chickens and geese on host cells.

The plaque-forming characteristics of Newcastle disease viruses of chickens and geese source were compared on various cells. The result showed that there were obvious differences of plaque formation between F48E9 and NA-1 on Vero cells, chicken embryo fibroblast cells (CEF) and goose embryo fibroblast cells (GEF). The plaque-forming ability of NA-1 was higher than F48E9 on GEF, but lower than F48E9 on CEF. On Vero cells, the plaque-forming ability of NA-1 was slightly stronger than F48E9. It demonstrated that the plaque-forming characteristics were consistent with host tropism of virus. The morphogenesis of F48E9 and NA-1 on Vero cells was observed with transmission electron microscope. There were different replication processes between F48E9 and NA-1 on cells at different stages. NA-1 had stronger adaptability to host than F48E9 according to budding processes and envelope integrity.

6.Effects of different concentration extract from shenghua decoction on contractile activity of the uterine smooth muscle isolated from normal, estrogen-treated and postpartum mice.

OBJECTIVETo investigate the effects of different concentration extract from Shenghua decoction on contractile activity of the uterine smooth muscle isolated from normal, estrogen-treated and postpartum mice.
METHODMedlab/4 s vital signal recorder was used to measure the effects of extract from Shenghua decoction (3-12 mg x mL(-1)) on contractile amplitude and frequency of the isolated uterus from normal, estrogen-treated and postpartum mice.
RESULTShenghua decoction extract (3-12 mg x mL(-1)) significantly decreased the contractile activity of the mouse isolated uterus in normal non-pregnancy and postpartum, but significantly increased that of the mouse isolated uterus treated with estrogen, and didn't show significant concentration-response relationship.
CONCLUSIONThe effects of Shenghua decoction extract on contractile activity of mouse-isolated uterus treated with estrogen cannot represent the pharmacological effects on that of in normal non-pregnancy and postpartum uterus.

7.A prospective assessment of the diagnostic value of MAIPA test in idiopathic thrombocytopenic purpura.

OBJECTIVETo evaluate the clinical significance of MAIPA test in diagnosis of idiopathic thrombocytopenic purpura (ITP) and in the differential diagnosis of antoimmune thrombocytopenias from nonimmune thrombocytopenias.
METHODSA total of 321 thrombocytopenic patients (118 males, 203 females) from 14 centers were studied. A modified monoclonal antibody immobilization of platelet antigen (MAIPA) method was used to detect the platelet glycoprotein-specific autoantibodies (anti-GP IIb/IIIa, anti-GPIb/IX) to double-blindly evaluate its sensitivity and specificity for the diagnosis of ITP and to investigate the impact of the antibodies on platelet count.
RESULTSThe results showed that for the diagnosis of ITP, anti-GPIIb/IIIa, anti-GPIb/IX and both of them had the sensitivity of 39.75%, 32.64% and 55.23%; the specificity of 97.56%, 93.94% and 92.68%; the positive predictive value of 97.94%, 93.98% and 95.65%; the negative predictive value of 35.71%, 32.35% and 41.53%; and the total efficiency of 54.51%, 48.29% and 64.80%, respectively. The positivity of the autoantibodies in immune thrombocytopenias was incredibly higher than that in nonimmune thrombocytopenias. The platelet counts in the immune thrombocytopenias with autoantibody positivities were significantly lower than those without the autoantibodies. The platelet counts were negatively correlated with the concentration of the autoantibodies. The levels of anti-GPIIb/IIIa or anti-GPIb/IX or both of them dropped or disappeared in patients being responsive to steroid therapy.
CONCLUSIONMAIPA assay is proved to be of great value for the diagnosis of ITP and for differential diagnosis of immune thrombocytopenias from nonimmune thrombocytopenias.

8.Effect of stromal cell-derived factor-1 and its receptor CXCR4 on liver metastasis of human colon cancer.

OBJECTIVETo investigate the effect of chemokine stromal cell-derived factor-1 (SDF-1) and its receptor CXCR4 on liver metastasis of human colon cancer.
METHODSExpression of CXCR4 in different colon cancer cell lines and SDF-1 in different tissues were detected by using Western-blot technique. Effect of SDF-1 and anti-CXCR4 monoclonal antibody (McAb) on proliferation and migration of HT-29 cells were measured using MTT methods. Model mimicking liver metastasis of human colon cancer was established by injecting HT-29 cells intrasplenically into BALB/C nude mice. Mice were randomly divided into AMD3100 treated group and control group. Liver metastatic rate and tumor foci were measured 7 weeks after.
RESULTSHT-29 cells expressed higher level of CXCR4 protein, and liver tissue expressed higher level of SDF-1 protein. Compared with the control, SDF-1 could significantly induced the proliferation and migration of the HT-29 cells, and anti-CXCR4 McAb could inhibited both functions of SDF-1. The liver metastasis rate in the control group was 100%, and it was 40% in the AMD3100 treating group (P < 0.05). The mean liver metastasis number also significantly decreased by AMD3100 (7.8 +/- 2.6 vs 22.4 +/- 8.6, P < 0.05).
CONCLUSIONSSDF-1/CXCR4 biological axis play an important role in liver metastasis of human colon cancer. Arrest of CXCR4 can inhibit liver metastasis of colon cancer through blocking cell proliferation and migration induced by SDF-1.

9.Diagnostic value of pleural effusion cell block immunohistochemical examination in patients with suspected lung cancer

Objective To investigate the diagnostic value of pleural effusion cell block (PECB) immunohistochemical examination in patients with suspected lung cancer.Methods Eighty-six patients with suspected lung cancer and pleural effusion were selected from December 2014 to December 2016 in the First People's Hospital of Zaoyang City.Liquid based cytology (LBC) smear was used for the diagnosis of cell morphology.Immunohistochemical detection of cell blocks was carried out,including CK7,CK5/6,p53,thyroid transcription factor-1 (TTF-1),calretinin,CD56,estrogen receptor(ER),progesterone receptor(PR),human epidermal growth factor receptor-2 (HER-2),carcinoembryonic antigen (CEA),cancer antigen 125 (CA 125)and calretinin.Immunologic marker staining was further carried out according to the cell morphology and clinical data.Results LBC smear test showed that there were 61 cases of metastatic adenocarcinoma,11 cases of metastatic poorly differentiated carcinoma,3 cases of metastatic small cell carcinoma,2 cases of metastatic large cell carcinoma,3 cases of metastatic squamous cell carcinoma,2 cases of malignant tumor and 4 cases of suspicious malignant tumor in the 86 cases.Immunohistochemical results showed that there were 69 cases of primary lung metastases (including 61 cases of pulmonary adenocarcinoma,2 cases of lung squamous cell carcinoma,one case of lung adenosquamous carcinoma and 5 cases of small cell lung cancer),5 cases of metastatic adenocarcinoma of extrapulmonary origin (including 2 cases of breast cancer,one case of prostatic cancer,one case of ovarian cancer and one case of pancreatic cancer),3 cases of metastatic squamous cell carcinoma(including 2 cases of esophageal carcinoma and one case of cervical cancer),2 cases of metastatic adenosquamous carcinoma(including one case of adenosquamous carcinoma of cervix and one case of adenosquamous carcinoma of rectum),2 cases of metastatic large cell carcinoma,3 cases of large cell neuroendocrine carcinoma,1 case of mesothelioma,and 1 case without definite diagnosis in the 86 cases.In the 69 cases of primary lung tumor metastasis,there were 63 cases with positive expression of TTF-1 and CK7 protein,2 cases with positive expression of CK20 protein and 2 cases with positive expression of p53 protein.The expression of CK5/6,CD56 and calretinin protein was negative in the 69 cases.In the 5 cases of metastatic adenocarcinoma of extrapulmonary origin,there was 1 case of breast carcinoma with positive expression of ER,PR and HER-2 protein,1 cases of breast carcinoma with positive expression of ER and PR protein,1 cases of prostatic cancer with positive expression of CK5/6 protein,1 cases of ovarian cancer with positive expression of ER protein and 1 cases of pancreatic cancer with positive expression of CEA and CA125 protein.In the 3 cases of metastatic squamous cell carcinoma,there were 2 cases of esophageal cancer with positive expression of CEA and p53 protein and 1 case of cervical cancer with positive expression of ER protein.In the 2 metastatic adenosquamous carcinoma,there was 1 case of adenosquamous carcinoma of cervix with positive expression of PR and TTF-1 protein and 1 case of adenosquamous carcinoma of rectum with positive expression of CK7 and CEA protein.Conclusion PECB immunohistochemical examination can accurately diagnose primary focus and subtype lung cancer,and further help to determine the location of the exogenous tumor.
10.Repetitive Transcranial Magnetic Stimulation for the Treatment of Restless Legs Syndrome.

BACKGROUNDRepetitive transcranial magnetic stimulation (rTMS) is a noninvasive technique used to alter cortex excitability that has been proposed as an efficient method for treating brain hyperexcitability or hypoexcitability disorders. The aim of this study was to investigate whether high-frequency rTMS could have any beneficial effects in restless legs syndrome (RLS).
METHODSFourteen patients with RLS were given high-frequency rTMS (15 Hz, 100% motor threshold) to the leg representation motor cortex area of the frontal lobe for 14 sessions over 18 days. Patients were diagnosed according to the international criteria proposed by the International Restless Legs Syndrome Study Group in 2003. The International RLS Rating Scale (IRLS-RS), Pittsburgh Sleep Quality Index (PSQI), Hamilton Anxiety Scale (HAMA) and Hamilton Depression Scale were used to evaluate the severity of RLS, sleep quality, anxiety and depression, respectively. The scale scores were evaluated at four-time points (baseline, end of the 14 th session, and at 1- and 2-month posttreatment). One-way analysis of variance was used to compare scale scores at different time points.
RESULTSThere was significant improvement in the IRLS-RS (from 23.86 ± 5.88 to 11.21 ± 7.23, P < 0.05), PSQI (from 15.00 ± 4.88 to 9.29 ± 3.91, P < 0.05), and HAMA (from 17.93 ± 7.11 to 10.36 ± 7.13, P < 0.05) scale scores at the end of 14 th session, with ongoing effects lasting for at least 2 months.
CONCLUSIONSHigh-frequency rTMS can markedly alleviate the motor system symptoms, sleep disturbances, and anxiety in RLS patients. These results suggest that rTMS might be an option for treating RLS.
