1.Clinical and immunological studies on transplantation of total lamellar cornea with circular lamellar sclera on treating serious chemical burn of eye

Objective To observe the clinical results of total lamellar cornea with circular lamellar sclera transplantation to treat serious chemical burn of eye and study its immunological mechanisms.Methods The patients(55 cases, 55 eyes) with corneal tansplantation were retrospectively analyzed. GroupⅠ(42 cases) was the patients with serious chemical burn of eye transplantation of total lamellar cornea with circular lamellar sclera, group Ⅱ(13 cases) was the patients transplantation of total cornea corresponding period of the groupⅠ. The time of tear-film break-up their rate of rejection and the time of occurrence of corneal neovascularization were compared.Results The time of occurrence of corneal neovascularization and rejection of group Ⅰ were later than groupⅡ.Conclusion Total lamellar cornea with circular lamellar sclera transplantation is an important method for treating serious chemical burn of eye, its immunological mechanisms is complex. It's important to increase the result of operation keeping high standard in quality of tear-film and using high effective immunosuppressive drug after operation.

Germplasm Resources Information System for Chinese Medicines (GRIS-CM) is designed and realized for improving the management efficiency of the germplasm resources for Chinese materia medica. The system integrates and analyzes the germplasm resources data, realizes information management of the germplasm resources for Chinese materia medica, and provides better services for scientific research institutions, governments, enterprises and medicinal herb growers. It contains 3 databases and 13 function modules, including the basic information base, literature library and gene bank. GRIS-CM can help manage the related data of germplasm resources for Chinese materia medica, and is better for data integration, analysis and statistics to find the rules and patterns. GRIS-CM provides flexible data comparison and visualization functions, and has significant practical value and research value.
3.Performance of Binomial Forced-Choice Digit Memory Test of the Elderly with Cognitive Impairment

Objective:To explore the effects of cognitive impairment on the performance of Binomial Forced-Choice Digit Memory Test (BFDMT). Methods: Subjects were screened by Mini-Mental State Examination (MMSE) and the brief form of WAIS-RC.BFDMT was administered to 38 dementias and 19 controls.Results: (1) There was no significant difference in the scores of two dimensions and total score of BFDMT between dementias and controls. (2) There was significant difference in the difficult item scores and the total score among three groups of subjects with different levels of IQ. The difficult item scores and the total score of the subjects with IQ 70~84 were significantly higher than those with IQ 50~69 and IQ 34~49. There were no significant differences among all groups in the easy item scores. (3) The false positive was 15.8% by the cutoff scores of total scores of BFDMT and increased with the decline of IQ. Conclusion: The performance of BFDMT is somewhat influenced by intellectual defect. A high false positive of BFDMT particularly happens in the patients with severe intellectual decline.
4.Surgical management of liver carcinoma accompanied by portal hypertension

ObjectiveTo evaluate the effects of various surgical procedures on liver carcinoma accompanied by portal hypertension. MethodsOne stage surgical procedure was performed in 28 cases of liver carcinoma accompanied by portal hypertension in our department from January 1992 to June 2000. ResultsThere was no operative mortality. The postoperative complications developed in 39%(11/28). The postoperative survival rates of 1?2 and 3 years were respectively 91%, 61%, and 48%. Postoperative upper digestive tract hemorrhage developed in 9 cases, 13 cases died during follow-up period, of whom 6 cases died of the recurrence of liver carcinoma, 2 cases died of liver failure, 5 cases died of upper digestive tract hemorrhage.Conclusions The survival time can be prolonged and the postoperative complications reduced through prudent selection of surgical procedures in patients suffering from concurrent liver cancer and portal hypertension. Postoperative recurrent bleeding can be prevented by pericardial devascularization. In poor risk cases with huge carcinoma, simple surgical procedure should be applied,and with a discreet application of chemoembolization.
5.Clinical value of 18F-FDG PET-CT to patients with follicular lymphoma staging Ⅲ-Ⅳ: monitoring response to therapy

Objective To evaluate the clinical value of 18F-FDG PET-CT in monitoring response to therapy in patients with follicular lymphoma (FL) staging Ⅲ-Ⅳ.Methods The clinical data of 20 patients with FL were analyzed retrospectively.All patients underwent PET-CT at three times:baseline,after 3-4 cycles of chemotherapy,and end of treatment.Kaplan-Meier method was used to analyze the survival of patients.Results 14 patients achieved CR at the interim PET-CT,all patients achieved CR at post therapy PET-CT:all patients were alive with no evidence of disease in the follow-up.2 patiens achieved PR at the interim PET-CT,and 1 patient achieved CR,and 1 patient was PD,at post therapy PET-CT,all patients were relapsed in the follow-up.4 patients achieved PD,and all patients were changed the chemotherapy plan.At post therapy PET-CT,2 patients achieved CR,1 patient achieved PR,and 1 patient was PD,1 patient was no evidence of disease,2 patients were relapsed,and another was died in the follow-up.The negative predictive values (NPV) of the interim and the post therapy PET-CT were 100 % (14/14),88.2 % (15/17) respectively.The interim PET-CT was no significantly correlated with PFS (P =0.135),while the post therapy PET-CT was significantly correlated with PFS (P =0.0006).Conclusion 18F-FDG PET-CT is useful to monitor response to chemotherapy in patients with FL staging Ⅲ-Ⅳ,and set personalized treatment plan.
6.Experimental study of the oriented differentiation of bone marrow derived mesenchymal stem cells into chondrogenic phenotype in a specific culture fluid

BACKGROUND:To construct tissue engineering cartilage would open up a novel way for the repair of cartilage damage in avoidance of the disadvantages of traditional therapeutic method.OBJECTIVE: To probe the techniques for the isolation of mesenchymal stem cells (MSCs) from bone marrow, as well as the in vitro differentiation into chondrocytic phenotype in a specific culture fluid.DESIGN:A complete randomized experimentSETTING:The Department of Traumatic Orthopedics and Hand Surgery of the First Affiliated Hospital of Guangxi Medical University, and Teaching and Research Faculty of Histology and Embryology of Guangxi Medical University.METHODS: The experiment was carried out at Guangxi Medical University between August 2002 and April 2003. Twenty SD neonatal weaning rats were selected. Bone marrow was aspirated from the bones of rat limbs and was isolated by gradient centrifugation in Percoll, and MSCs could be obtained in combination with adherent screening method, which were then cultured in DMEM-LG with 15% fatal bovine serum (FBS) in the incubator of 37℃ with 5% CO2 for 10-14 days. The passage cells were induced in DMEM-HG with 15% FBS (containing TGF-β1 10 μg/L, 10-7 mol/L dexamethasone, 50 mg/L VitC).MAIN OUTCOME MEASURES :The morphology, growth, as well as proliferation and specific expression of chondrogenic matrix of in vitro cultured MSCs due to specific induction.RESULTS: Totally 20 SD rats were observed and analyzed with no loss SCs grew in visible symmetric colonies, displaying a long-spindle shape,and the morphological characteristics of marrow-derived MSCs had no obvious changes during passage-culture, but its proliferation time was found from a shuttle fibroblastic appearance to polygonal shape, displaying posiHC staining of type Ⅱ collagen of cartilage specific matrix.bronectin adherent screening technique is a convenient, effective and practical method to separate and collect MSCs from rat bone marrows in chondrogenic phenotype when induced by a specific medium and can secrete cartilage specific matrix, and they can be the optimal seed cells for cartilage tissue engineering.
7.Role of NF-κB in the progression of aldosterone-induced renal injury and its associated mechanisms

Objective To investigate the role of NF-κB in aldosterone-1%NaCl-induced renal injury in uninephrectimized SD rats and the potential mechanisms.Methods Thirty-teo male SD rats were uninephrectomized and treated for 4 weeks.Rats were divided into four groups randomly:control group (n=8),1%NaCl group (1%NaCl in chow,n=8),aldosterone group (1%NaCl in chow,0.75 μg/h aldosterone delayed relase by osmotic mini-pump,SC,n=8),PDTC group (1%NaCl in chow,0.75 μg/h aldosterone,SC,100 mg/kg PDTC,IG,n=8).Systolic blood pressure (SBP),urinary protein,renal function and renal morphologic were observed.The expression of intercellular cell adhesion molecule 1 (ICAM-1) and connective tissue growth factor (CTGF) were measured respectively by Western blotting and real-time PCR.The activity and location of NF-κB in renal cortex were detected by electrophoretic mobility shift assay (EMSA) and immunohistochemisty.Results Rats of aldosterone group exhibited higher blood pressure and more serious renal injury characterized by proteinuria,glomerular sclerosis compared with rats of the 1% NaCl group.Protein and mRNA levels of ICAM-1 and CTGF were significantly increased inaldosterone group rats than those in 1%NaCl group (all P<0.05).Moreover,all these changes were associated with an increase in NF-κB activity.Treatment with PDTC which is a specific inhibitor of NF-κB notably alleviated SBP,proteinuria and renal injury in aldosterone-infused rats.Furthermore,PDTC markedly reduced the expression of ICAM-1 and CTGF (all P<0.05).Conclusion PDTC can alleviate aldosterone-1%NaCl-induced renal injury in uninephrectimized SD rats by preventing the expression of ICAM-1 and CTGF.
8.Effect of aldosterone on glomerular mesangial cells apoptosis bothin vivo and in vitro

Objective To evaluate the effect of aldosterone (Ald) on glomerular mesangial cells apoptosis and to explore the possible mechanisms.Methods Twenty-four Sprngue-Dawley rats were subcutaneously embedded with osmotic mini-pumps and randomly divided into 3 groups.Aldosterone (1.5 μg/h) was administrated subcutaneouly by osmotic mini-pumps in Ald group,eplerenone (Epl,100 mg·kg-1·d-1) and Ald (1.5 μg/h) was given to Epl group.And normal saline was used in control group (Con group).Systolic blood pressure and urinary albumin excretion rate (UAER) were detected on day 0,7,14,21,28.Blood and kidney samples were harvested on day 28.Plasma creatinine,potassium and aldosterone were measured.Renal paraffin sections were stained by PAS and the morphological changes were evaluated by light microscopy.Apoptosis index of mesangial cells were detected by TUNEL assay.The glomerular mesangial cells (MCs) were cultured in a DMEM-F12 media.MCs apoptosis was evaluated by staining cells with Annexin V and propidium iodide (PI) using flow cytometer.Expression of Bcl-2 and Bax mRNA was examined by RT-PCR.The protein level of Bad or phospho-Bad was measured by Western blotting.Results Ald-infused rats developed hyperaldosteronemia and hypokalemia.Rats in Ald group exhibited significant hypertension and marked albuminuria.Ald group rats showed increased number of TUNEL-positive mesangial cells when compared with control rats (P<0.05).Aldosterone induced mesangial cells apoptosis in a time-dependent manner.Expression of Bcl-2 mRNA was decreased but Bax mRNA was increased in aldosterone treated MCs compared to that in Con group (P<0.05).Aldosterone promoted dephosphorylation of cytosolic phospho-Bad compared with vehicle treated cells (P< 0.05).However,eplerenone attenuated these effects of aldosterone.Conclusion Aldosterone directly promotes mesangial cells apoptosis,and eplerenone can attenuate this effect of aldosterone.Dephosphorylation of cytosolic phospho-Bad may be the key role in the progression of mesangial cells apoptosis induced by aldosterone.
9.Electroluminescence Biosensor for Determination of Pb2+Based on G-Quadruplex of Aptamer probe

A turn on model of electrochemiluminescence ( ECL)-Pb2+ biosensor based on G-quadruplex of aptamer probe was developed by using quantum dots ( QDs) as labels. The ECL-Pb2+ biosensor was prepared by immobilizing a hairpin aptamer probe with a thiol group at 5 '-end and an amino group at 3 '-end on gold electrode via an Au-S bond. In the presence of Pb2+, the G-rich hairpin aptamer probe opened the stem-loop and formed G-quadruplex, which exposed the amino group at 3'-end and then covalently linked to the carboxyl group on the surface of CdS quantum dots. The fabrication and reaction process of the Pb2+ biosensor were characterized. The responsive ECL intensity was linearly related to the Pb2+ concentration in the range of 2. 0×10-10-5. 0×10-8 mol/L, with the detection limit of 3. 74×10-11 mol/L. In addition, the ECL biosensor exhibited high sensitivity, good selectivity, satisfying stability, and repeatability.
10.Changes of intrahepatic hemodynamics after liver ischemia reperfusion in rats

Objective To study the changes of intrahepatic shunt flow(IHSF) and functional hepatic blood flow(FHBF) after liver ischemia reperfusion in rats.Methods 12 healthy male SD rats were anaesthetized and heparinised.The right carotid artery,jugular vein and two distal ileocolic veins were cannulated for blood(transfusion),volume compensation,drug infusion,and blood sampling respectively.Rats were randomly divided into 2 groups.Group 1 was the sham operation control group.Group 2 rats was ischmic/reperfusion(I/R) group,subjecting to 45min liver lobar ischemia and 60min reperfusion.Both groups received infusion of(D-sorbitol)(10mmol/l,0.2mL/min) via portal venous,and blood samples(1mL) were drawn simultaneously from the carotid,portal venous,and hepatic venous catheters.Portal venous flow(PVF) and hepatic arterial flow(HAF) were measured via flow probes.Hepatic sorbitol uptake rate,FHBF and IHSF were calculated.Results There were no significant differences between the two groups in PVF,HAF and total hepatic blood flow((THBF)).Compared to control group,hepatic sorbitol uptake rate as well as FHBF were decreased and IHSF was increased in I/R group(P
