1.A new anti-cancer target: ADAM10

Gene ADAM10 (a disintegrin and metalloproteinase 10) is a member of ADAMs gene family. The corresponding membrane protein contains a disintegrin domain and a metalloproteinase domain. It is involved in various biological events such as cell adhesion, cell fusion, cell migration, and membrane protein shedding and proteolysis. ADAM10 protein is overexpressed in many tumors, which interacts with adhesion molecules and critical signaling molecules associated with cancer development and tumor progression, and promotes tumor invasion and metastasis. Regulating the expression of ADAM 10 can inhibit tumor invasion and metastasis , which may become a new strategy for anticancer therapy.
2.The expression of P16,Ki-67 in cervical squamous epithelial lesions and the significance of their detection combined with HPV

Objective To study the expression of P16 ,Ki‐67 in cervical squamous epithelial lesions and the significance of their detection combined with human papillomavirus(HPV) .Methods 188 cases of cervical tissues from surgical excision were collected , including 48 cases of cervical cancer ,100 cases of cervical intraepithelial neoplasia(CIN) ,and 40 cases of chronic cervicitis ,which were set as cervical cancer group ,CIN group and cervicitis group ,respectively .Expression of P16 ,Ki‐67 in cervical tissue were de‐tected by using immunohistochemistry ,and the HPV infection were detected by using PCR technology .Results The positive ex‐pression rates of P16 and Ki‐67 in cervical cancer ,CIN group was significantly higher than those in cervicitis group(P<0 .05) ,and the positive expression rates of P16 and Ki‐67 in cervical cancer group were higher than those in CINⅠ and CINⅡ group (P<0 . 05) ,but between the cervical cancer group and CIN Ⅲ group there were no statistically significant difference(P>0 .05) .With the increase of CIN level ,the positive expression rates of P16 and Ki‐67 increased ,among the three CIN groups there were significant difference(P<0 .05) .In cervical cancer group and CINⅢ group ,the expression rates of P16 and Ki‐67 were both significantly high‐er than the other 3 groups(P<0 .05) .In CINⅠ - Ⅲ groups ,the over expression rates of P16 and Ki‐67 were statistically different (P<0 .05) .The expression of P16 positively correlated with the positive rates of HPV infection (rs =0 .706 ,P=0 .011);the ex‐pression of Ki‐67 positively correlated with the positive rates of HPV infection(rs=0 .695 ,P=0 .021) .Conclusion CIN and cervi‐cal cancer of early stage could be diagnosed and the pathological progress of CIN could be predicted by using the combined detection of P16 ,Ki‐67 and HPV .
3.Study on human umbilical cord blood cells transplantation for treatment of experimental autoimmune encephalomyelitis

Objective To explore the effects of human umbilical cord blood cells(HUCBCs) transplantation to treat experimental autoimmune encephalomyelitis(EAE) rats and the status of transplanted HUCBCs in the brain and spinal cord tissue of EAE rats.Methods The mononuclear cells abstracted from cord blood of infants were cultured in vitro and marked with 5-bromodeoxyuridine(Brdu) for 48 h.EAE rat models were made and the HUCBCs(3?106) were transplanted into the tail vein(transplanted group) 14 d later.The score of neurological function dificit and the number of the demyelinated foci in brain and spinal cord were undertaken at different time point after transplantation.The statue of survival,differentiation and migration of HUCBCs in vivo were determined by immunohistochemical technique,and compared with control group.Results The scores of neurological function dificit at 21 d,28 d post transplantation in transplanted group were much lower than those in the control group(all P
4.Clinical features and molecular genetic analysis of a pedigree of limb girdle muscular dystrophy

Objective To investigate the clinical features and analyze the molecular genetics of a pedigree of limb girdle muscular dystrophy (LGMD).Methods Pedigree analysis and clinical examination were performed in one four-generation family with LGMD.Electrophysiology and muscle biopsy were done in the affected members.With an informed consent, gene mutation, genome screening and linkage analysis were conducted in 26 members of this pedigree.Results Seven patients were identified.Pedigree analysis was consistent with autosomal dominant inheritance.Affected members had early presentation.Main features included proximal muscle weakness without dysarthria nor spasticity; electrophysiology and muscle biopsy revealed myopathic changes.LGMD1 A, 1B, 1C and facioscapulohumeral dystrophy genes were not detected by gene mutation analysis.Genome screening and linkage analysis did not reveal any linkage with the disease-causing gene and the reported loci of LGMD1D and LGMD1F genes.Conclusions The clinical manifestations of this LGMD family are highly heterogeneous, and the disease-causing gene of this family is not linked to any of the reported sites, suggesting this may be a new disease-causing locus, or a new genetic type of LGMD.
5.Pharmacokinetics of deflazacort tablets in healthy Chinese volunteers.

Deflazacort (DFZ, a prodrug) is well absorbed and rapidly metabolized into the active metabolite 21-hydroxydeflazacort (21-OH DFZ) after oral administration. The aim of this study is to evaluate the pharmacokinetic properties of 21-OH DFZ in healthy Chinese volunteers after a single and multiple oral administration of DFZ tablets under fed condition. Twelve volunteers (six males and six females) were administered a single dose of 6 mg or 12 mg or 24 mg of DFZ in three different periods separately, according to the 3 x 3 Latin square design. Between each administration period there was a washout period of one week. The multiple-dose study of 12 mg dose DFZ per day for 7 consecutive days was started after a 1 w washout period when the single-dose study completed. The pharmacokinetic parameters of 21-OH DFZ after the single oral administration of 6 mg, 12 mg and 24 mg DFZ tablets were as follows: (37.7 +/- 11.6), (61.5 +/- 17.7) and (123 +/- 23) ng x mL(-1) for C(max); (1.90 +/- 0.32), (1.96 +/- 0.27) and (2.13 +/- 0.34) h for t1/2; (96.6 +/- 25.9), (190 +/- 44) and (422 +/- 107) ng x h x mL(-1) for AUC(0-14 h), respectively. After the multiple dose administration, the mean plasma concentration at steady-state C(av) was (7.00 +/- 1.66) ng x mL(-1) and the degree of plasma concentration fluctuation DF was 7.7 +/- 1.2. The results showed that the pharmacokinetic characteristics of 21-OH DFZ in healthy Chinese volunteers were linear over the dose range of 6 to 24 mg. No significant gender differences were found in the pharmacokinetics of 21-OH DFZ in healthy Chinese volunteers. After the multiple dose administration of 12 mg DFZ for 7 d, no accumulation of 21-OH DFZ in healthy Chinese volunteers was observed.
6.Application of sequential method in training of primary anesthesia nurse

Objective To investigate the effect of sequential method in the training of primary anesthesia nurses. Methods A phased, gradual sequential method was used to train 11 primary anesthesia nurses. One-year full-course training was divided in 4 stages, each lasting 3 months. The theory and operational examination were held at the end of each stage and the tutoring teachers reported their comments. Result The mean scores of 11 primary anesthesia nurses were more than 90 points in both theory and operations examination, and the mean scores of tutor′s evaluation more than 85 points in each stage. Conclusion The sequential method can help primary anesthesia nurses to get better anesthesia-related knowledge, skill and clinical comprehensive ability, which set a solid foundation for their work of anesthesia care.
7.Nursing about using the Worst lacrimal probe to repair the lacerateon of lacrimal canaliculus

Objective To explore the ideal postoperative nursing methods of repair the lacerateon of lacrimal canaliculus. Methods Using pigtail probe to repair the lacerateon of lacrimal canaliculus in 49 patients, observed the postoperative nursing points in different clinical phases. Results There were 2 patients had the anstomotic stoma split, 1 patient had the canaliculitis. There were 46 patients had successful extubation. Conclusion The Worst lacrimal probe is an satisfactory and reliable method to repair the lacerateon of lacrimal canaliculus. The detailed interpretation and the skillful nursing care are the guarantee for successful operation.
8.Combination of transcatheter arterial chemoembiolization and antitumor drugs in the treatment of Hepatocellular carcinoma

Hepatocellular carcinoma(HCC)is a common world wide disease.Transcatheter arterial chemoembolization(TACE)has been the first choice for the unresectable midadvanced ones.However, TACE has its own limitation such as high recurrence rate and many complications, what's more, recurrence induced by neovascularization which is stimulated by activation of vascular endothelial growth factors promoted by ischemia and hypoxia in local tissues after the embolization always reduce the effect of treatment.And combination of TACE and anti-angiogenesis therapy is capable to make up the shortage of each other and reduce the recurrence rate, which has been a hot issue for non-surgical treatment of HCC.

Objective To explore the effect of human neural stem cells(hNSCs) transplantation to treat cerebral ischemic rats and the status of transplanted hNSCs in the ischemic brain tissue of these rats.Methods Human neural stem cells were separated from 10~13 weeks brains of human embryo and were cultured and induced to differentiate. The middle cerebral artery occlusion rat models were made and the human neural stem cells were transplanted through tail vein 1 day later.
