1.A new anti-cancer target: ADAM10
Journal of International Oncology 2011;38(7):492-495
Gene ADAM10 (a disintegrin and metalloproteinase 10) is a member of ADAMs gene family. The corresponding membrane protein contains a disintegrin domain and a metalloproteinase domain. It is involved in various biological events such as cell adhesion, cell fusion, cell migration, and membrane protein shedding and proteolysis. ADAM10 protein is overexpressed in many tumors, which interacts with adhesion molecules and critical signaling molecules associated with cancer development and tumor progression, and promotes tumor invasion and metastasis. Regulating the expression of ADAM 10 can inhibit tumor invasion and metastasis , which may become a new strategy for anticancer therapy.

Deflazacort (DFZ, a prodrug) is well absorbed and rapidly metabolized into the active metabolite 21-hydroxydeflazacort (21-OH DFZ) after oral administration. The aim of this study is to evaluate the pharmacokinetic properties of 21-OH DFZ in healthy Chinese volunteers after a single and multiple oral administration of DFZ tablets under fed condition. Twelve volunteers (six males and six females) were administered a single dose of 6 mg or 12 mg or 24 mg of DFZ in three different periods separately, according to the 3 x 3 Latin square design. Between each administration period there was a washout period of one week. The multiple-dose study of 12 mg dose DFZ per day for 7 consecutive days was started after a 1 w washout period when the single-dose study completed. The pharmacokinetic parameters of 21-OH DFZ after the single oral administration of 6 mg, 12 mg and 24 mg DFZ tablets were as follows: (37.7 +/- 11.6), (61.5 +/- 17.7) and (123 +/- 23) ng x mL(-1) for C(max); (1.90 +/- 0.32), (1.96 +/- 0.27) and (2.13 +/- 0.34) h for t1/2; (96.6 +/- 25.9), (190 +/- 44) and (422 +/- 107) ng x h x mL(-1) for AUC(0-14 h), respectively. After the multiple dose administration, the mean plasma concentration at steady-state C(av) was (7.00 +/- 1.66) ng x mL(-1) and the degree of plasma concentration fluctuation DF was 7.7 +/- 1.2. The results showed that the pharmacokinetic characteristics of 21-OH DFZ in healthy Chinese volunteers were linear over the dose range of 6 to 24 mg. No significant gender differences were found in the pharmacokinetics of 21-OH DFZ in healthy Chinese volunteers. After the multiple dose administration of 12 mg DFZ for 7 d, no accumulation of 21-OH DFZ in healthy Chinese volunteers was observed.

Objective To investigate the clinical features and analyze the molecular genetics of a pedigree of limb girdle muscular dystrophy (LGMD).Methods Pedigree analysis and clinical examination were performed in one four-generation family with LGMD.Electrophysiology and muscle biopsy were done in the affected members. With an informed consent, gene mutation, genome screening and linkage analysis were conducted in 26 members of this pedigree. Results Seven patients were identified.Pedigree analysis was consistent with autosomal dominant inheritance.Affected members had early presentation.Main features included proximal muscle weakness without dysarthria nor spasticity; electrophysiology and muscle biopsy revealed myopathic changes.LGMD1 A, 1B, 1C and facioscapulohumeral dystrophy genes were not detected by gene mutation analysis.Genome screening and linkage analysis did not reveal any linkage with the disease-causing gene and the reported loci of LGMD1D and LGMD1F genes. Conclusions The clinical manifestations of this LGMD family are highly heterogeneous, and the disease-causing gene of this family is not linked to any of the reported sites, suggesting this may be a new disease-causing locus, or a new genetic type of LGMD.

Hepatocellular carcinoma(HCC)is a common world wide disease.Transcatheter arterial chemoembolization(TACE)has been the first choice for the unresectable midadvanced ones. However, TACE has its own limitation such as high recurrence rate and many complications, what's more, recurrence induced by neovascularization which is stimulated by activation of vascular endothelial growth factors promoted by ischemia and hypoxia in local tissues after the embolization always reduce the effect of treatment. And combination of TACE and anti-angiogenesis therapy is capable to make up the shortage of each other and reduce the recurrence rate, which has been a hot issue for non-surgical treatment of HCC.

Objective To explore environmental hazards of childhood leukemia and to determine hazardous suspects on childhood leukemia. Methods Using 1∶2 matched case-control study design,50 patients of leukemia and 100 controls selected from the Affiliated Hospital of Ningxia Medical College were investigated during Feb.2005-May 2007. Information was obtained by questionnaires, which included the general social demographic characteristics, disease history, the history of exposure to the materials during the child's mother pregnancy, indoor and outdoor environmental situation and the history of the families. Results The data showed that hazards for childhood leukemia were significantly associated with the history of infection(OR=0.452),diagnostic X-ray exposure (OR=0.157), the history of exposure to the chemical fertilizer and the pesticide during the pregnancy of mother(OR=3.150), the house type(OR=5.162), the nearest distance to the high voltage electricity (OR=2.383) and the radioactive source to the dwelling environment (OR=2.793) were important hazardous factors of the childhood leukemia. Conclusion The history of infection, diagnostic X-ray exposure, the history of exposure to the chemical fertilizer and the pesticide during the pregnancy of mother, the house type, the nearest distance to the high voltage electricity and the radioactive source to the dwelling environment might be the hazardous factors of the childhood leukemia.

Objective To explore the effect of human umbilical cord blood cells (HUCBCs) transplantation to treat cerebral ischemia of rats and the status of transplanted HUCBCs in the ischemic brain tissue of these rats. Methods The mononuclearcells abstracted from 60~100 ml of cord blood of full-term babies were cultured in vitro and marked with 5-bromodeoxyuridine (Brdu)(5 ?mol/L) for 2 days. The middle cerebral artery occlusion rat models were made and the HUCBCs (3?106) were transplanted into the lateral ventricular 1 day later. Neurological severity scores (NSS) tests were undertaken at different time point after transplantation, and iimmunohistochemistry method was used to check the migration and differentiation of HUCBCs. Results The HUCBCs had the capacity of proliferation in vitro and were induced to differentiate into astrocytes, oligodendrocytes and neurons in vivo. 3 weeks later, the neurological function of rats that received transplantation recovered much better than the rats without transplantation, as evidenced by NSS (all P

Objective To investigate the proliferation of neural stem cells by subcutaneous injection of basic fibroblast growth factor(bFGF),epidermal growth factor (EGF)or combined bFGF and EGF respectively after transient focal cerebral ischemia in rats. Methods The right middle cerebral arteries(MCA) of rats were occluded for 2 h to establish transient focal cerebral ischemia model. bFGF, EGF,combined bFGF and EGF or vehicle were administered subcutaneously at 24, 48 and 72 h after MCA occlusion(MCAO).Thereafter, one administration every 3 d. Proliferating neural stem cells were labeled with bromodeoxyuridine (Brdu) immunohistochemically. Brdu-labeled cells were calculated to characterize neural stem cells in SVZ and the dentate gyrus of the hippocampus at 7 d,14 d,21 d after MCAO.Results Brdu-labeled cells were detected in the bilateral SVZ and the dentate gyrus of the hippocampus after MCAO in each group, the number of positive cells decreased progressively. Compared with the control group, the number of Brdu-labeled cells in treatment group increased significantly after treatment( P
