Knowledge management, as a new managerial concept, is the specific embodiment of knowledge based economy in the field of management while technological innovation, constituting the core of technological progress, is the effective extension and application of scientific and technological knowledge. Hospitals, which are knowledge based institutions, possess good foundations and advantages of implementing knowledge management and technological innovation. There is a close link between knowledge management and technological innovation in hospitals, the former being the basis and primary guarantee of the latter and the latter being the result and manifestation of the former. As a classic fluorescent detect technique, fluorescence resonance energy transfer (FRET) has been widely used in biological researches. Researchers have developed a series of fluorescence detect probes which were based on FRET. Caspase family plays an important role in apoptosis pathway, especially Caspase-8 which located, at the initial of death receptor mediated apoptosis pathway, whose its activation can trigger subsequent precaspases' activation and lead to apoptosis. So it is of great significance to detect the activation of Caspase-8 in apoptosis assay. In this study, a fluorescent probe based on FRET has been designed which can detect the activity change of Caspase-8 in cells. To identify the effectiveness and specificity of the probe, we measure the Caspase-8 activity under the Caspase-8 specifically activated apoptosis inducer RGD-TRAIL with the flow cytometry FRET detection platform. The results show that the probe can respond to the activity change of Caspase-8 in apoptotic cells, and the change can be quantified rapidly by flow cytometry. The study provides a more efficient and convenient detection method of Caspase-8 activity in living cells.

ive] To isolate the active constituents with analgesic and antemetic actions from Rhizo-ma Alpiniae Officinarum. [Methods] Polyamide and silica gel column chromatography was used to isolate and extract the analgesic and antemetic constituents. The structure of compounds was identified by ultra violet spectroscopy, infrared spectroscopy, mass spectroscopy and nuclear magnetic resonance spectrosco-py. [ Results ] Galangin and kaempferide are identified as the analgesic and antemetic constituents. [Conclusion] Galangin can be used as the quality control for Rhizoma Alpiniae Officinarum.

Objective To investigate and evaluate the polymorphism distribution of arsenic(+3 oxidation state)methyhransferase(AS3MT)5'-UTR VNTR in Chinese populations.Methods Genomic DNA was extracted from peripheral blood anti-coagulated with ACD of 1440 individuals in a standard phenol-chloroform protocol.The phenotypes of AS3MT 5'-UTR VNTR were determined by polymerase chain reaction(PCR)associated with agarose gel electrophoresis.Results Of the 1440 individuals,771(53.5%),426(29.6%),211(14.7%),16(1.1%)and 16(1.1%)were carriers of the V2/V3(AB/A2B),V3/V3(A2B/A2B),V2/V2(AB/AB),V2/V4(AB/A3B)and V3/V4(A2B/A3B)genotype,respectively.The AB(V2),A2B(V3)and A3B(V4)allele frequency was 41.9%,57.0%,1.1%respectively.The differences of AB(V2)and A2B(V3)allele frequency were all significant between the northern and southern populations respectively(χ2=23.39,χ2=33.28,P＜0.007).Conclusions In different regions the AB(V2)and A2B(V3)allele frequency is different,the AS3MT 5'-UTR VNTR polymorphism can be used to evaluate the susceptivity of arsenieosis.

BACKGROUND: Various types of collagen affect differentiation and expression of chondrocytes in the process of tissue engineered culture for cartilage. The reliability of cartilage tissue engineering is determined by resource, structure and property of collagen.OBJECTIVE: To characterizing the structure of collagen type Ⅰ,Ⅱ and Ⅲ, to clarify the structural characteristics of collagen for cartilage tissue repairing and provide guidance for choosing cartilage tissue engineering scaffolds. METHODS: The type Ⅰ and Ⅲ collagen was obtained from neonatal calf skins, and type Ⅱ collagen was obtained from porcine articular cartilage. The differences of structures of collagen type Ⅰ,Ⅱ and Ⅲ were analyzed and compared by FTIR, DSC, SDS-PAGE and picrosirius-red staining.RESULTS AND CONCLUSION: The neonatal calf collagen type Ⅰ,Ⅱ and porcine collagen type Ⅱ had the similar functional group. The thermal denaturation temperature of the three type collagen was 104.2 ℃, 99.7 ℃ and 92.5 ℃. SDS-PAGE analysis showed the collagen type Ⅲ percentage in collagen type Ⅰ was (11.29±0.91)%. Picrosirius-red staining exhibited that the collagen type Ⅰ was comprised red and orange raw fibers, with strongly birefringent. Collagen type Ⅲ was lax and thin, greenish fibers, but collagen type Ⅱ was red, loosely cancellated fibers. The functional groups and important characteristics of the three of collagen are similar. However, there are some differences in advanced structure. To analyze the relationships of collagen type Ⅰ,Ⅱ and Ⅲ is important to choose and prepare scaffolds for cartilage tissue engineering.

[Objective] To analyze the constituents of volatile oil from Nervilla fordii (NF) by gas chromatography-mass spectrometry (GM-MS). [Methods] Volatile oil was extracted from NF by steam distillation and the constituents of volatile oil were isolated and identified by GM-MS. The relative contents of the volatile constituents were calculated by ionic flow chromatographic area normalization method. [Results] Eighty-nine components were separated. Among them, fifty-three components were identified, accounting 91.82% of the total volatile oil. [Conclusion] The main constituents identified from NF were: 2-pentadecanone, 6,10,14-trimethyl (13.55%), 4-ethyl-trans-3-thiobicyclo [4,4,0] decane (6.54%), benzenesulfonamide, 4-methyl-N- (2-oxo-2 phenylethyl) (6.33%), phytol (6.32%),?-carinol (4.54%), ?-ionone (4.43%), caryophyllene oxide (4.13%). The amount of the above constituents accounts 45.84% of the total volatile oil.
