1.Low expression of cystic fibrosis transmembrane conductance regulator gene allele IVS8-5T may be associated with decreased prostate cancer risk

Objective To investigate the association between functional polymorphism IVS8-poly (T) in cystic fibrosis transmembrane conductance regulator (CFTR) gene and prostate cancer risk.Methods A population-based case-control study was conducted including 230 prostate cancer patients and 230 agematched controls.PCR-STRP was used to analysis the IVS8-poly (T) tract in CFTR gene.Results The prevalences of the low expression allele,the IVS8-5T,in prostate cancer patients and controls were 1.52 % (7/460) and 5.22 % (24/460),respectively (x2 =12.489,P =0.002).Logistic regression analysis confirmed the 5T/7T genotype was in protective association with prostate cancer risk (OR =0.232,P =0.003,95 % CI0.090-0.599).Conclusion The low expression CFTR IVS8-5T allele contributes to a reduced risk of prostate cancer in Chinese Han population,and may be a protective factor against prostate cancer.
2.A correlative study of ultrashortwave therapy affecting survival of rat tail replantation

Objective To research the effect mechanism of ultrashortwave in the correlation of ultrashortwave and the tail replantation, provide the experiment basis of clinical practice of prevention and cure for the vascular crisis after micromodule anastomosis. Methods Eighty Sprague-Dawley(SD) rats of clean grade were 3-month-old,female,and were divided into four groups:control group (group 0),model group (group 1 ),contrast group (group 2),ultrashortwave (USW) group (group 3).The preparation of tail replantation model was cut off soft tissue except for caudal veins on both sides of the tail. The coccyx was not broken away from tail.At last,the audal artery under abdominal main centre ditch was anastomosed.In experiment process, the USW group was divided into high dosage group (group 3A) and low dosage group (group 3B). The caudal arterys were ligated and not anastomosed in the group 0. Caudal arterys in other groups were anastomosed.Rats in the group 0 and group 1 received no treatment,normal management after the operation. Rats in the group 2 were given abdominal cavity injection of papaverin liquid immediately,then once a day to 5 days after the tail replantation.Rats in the group 3 were immediately given USW therapy of twenty minutes on the anastomosis section,and then once a day for 5 days after the tail replantation.The USW dosage of group 3A was 3th grade and 50mA. The USW dosage of group 3B was 2th grade and 28mA.The survival rate of the rat tails was observed after the tail replantation for 10 day.Before being grouped,it was measured that the tail skin temperature diference between near and far side of anastomosis section.After the tail relpantation, the temperature diference was inspected daily for 10 postoperative days hence. Before rats were grouped and the eighth hour after the tail replantation, rats were collected blood plasma specimens and measured contents of nitric oxide with destination colorimetric mathods of nitric oxide.Results Carrying out comparison of survival rate of every group,the output weve:between tail cutting off group (group 0,0) and tail replantation group (group 1,2,3,43.94%) to compare P > 0.05; between each group of the tail replantation groups (1,2,3A,3B group) to compare P> 0.05,group 3B < 2 < 1 < 3A; between group 3B and group 1 to compare P > 0.05; between group 2 and group 1 to compare P > 0.05. Each group were compared with the change daily between postoperative and preceding operative the skin temperature diference,single-factor analysis of variance (One-Way ANOVA) analysis:Postoperative 1 day,group 3A < 1,P < 0.05.Postoperative 6 day:3A < 3B < 1 < 2,P > 0.05.Postoperative 7 day:group 3B < 1 < 3A < 2,P < 0.05.Each group were compared with the change of the content of nitric oxide between postoperative 8 hour and preceding operative,with rank-sum test:group 3B > 3A > 2 > 1 > 0,H =33.760,P < 0.05,shows statistically significant.Conclusions USW therapy,especially USW low-dose therapy,can reduce vascular crisis and improve the survival rate of replanted rat tails,after the postoperative 1,6,7 days,reduce skin temperature,improve blood supply,improve nitric oxide at postoperative eighth hour,prevent vascular crisis.Rat tail replantation model in this experiment is feasible.
3.Clinical study on effect of Shenmai Injection in treating congestive heart failure.

OBJECTIVETo observe the therapeutic effect of Shenmai Injection (SI) in treating congestive heart failure (CHF).
METHODSThe changes in cAMP, cGMP, serum cardiac troponin T (cTnT, a specific marker reflecting myocardial injury), creatine kinase (CK) and creatine kinase isoenzyme (CK-MB) were simultaneously monitored in 62 chronic CHF patients, distributed in the two groups, the routine treatment group and the routine treatment + SI group, by randomized grouping method, and the therapeutic effect of the two groups was compared.
RESULTSThe plasma cAMP/cGMP ratio increased in early stage and decreased in late stage of the course of CHF. The serum cTnT level was progressively increased along with heart function deterioration. After treated with SI for 2 weeks, the CHF patients' hemodynamics got stable and heart function obviously improved. No serious adverse reaction was found in the therapeutic course.
CONCLUSIONThe level of serum cTnT might be taken as a reliable biochemical parameter to predict the prognosis of CHF patients. SI is an effective and safe agent in treating CHF.

4.Degradation of acenaphthene by ozone.

OBJECTIVETo investigate the oxidation of acenaphthene (Ace), a polycyclic aromatic hydrocarbon (PAH) with a saturated C-C bond by ozone and to characterize the intermediate products of ozonation.
METHODSOzone was generated from filtered dry oxygen by an ozone generator and continually bubbled into a reactor containing 1g/L Ace dissolved in an acetonitrile/water solvent mixture (90/10, v/v) at a rate of 0.5 mg/s. HPLC was used to analyze the Ace concentration. Total organic carbon (TOC) was used to measure the amount of water soluble organic compounds. GC-MS was used to identify the ozonized products. Oxygen uptake rate (OUR) of activated sludge was used to characterize the biodegradability of ozonized products.
RESULTSDuring the ozonation process, Ace was degraded, new organic compounds were produced and these intermediate products were difficult mineralize by ozone, with increasing TOC of soluble organics. The ozonized products were degraded by activated sludge more easily than Ace.
CONCLUSIONOzonation decomposes the Ace and improves its biodegradability. The ozonation combined with biological treatment is probably an efficient and economical way to mineralize acenaphthene in wastewater.

5.Anaerobic degradation of tetrachloroethylene using different co-substrates as electron donors.

OBJECTIVETo investigate the biodegradation of tetrachloroethylene (PCE) by acclimated anaerobic sludge using different co-substrates, i.e., glucose, acetate, and lactate as electron donors.
METHODSHP-6890 gas chromatograph (GC) in combination with auto-sampler was used to analyze the concentration of PCE and its intermediates. Results PCE could be degraded by reductive dechlorination and the degradation reaction conformed to the first-order kinetic equation. The rate constants are k(lactate) > k(glucose) > k(acetate). The PCE degradation rate was the highest in the presence of lactate as an electron donor.
CONCLUSIONLactate is the most suitable electron donor for PCE degradation and the electron donors supplied by co-metabolic substrates are not the limiting factors for PCE degradation.

6. Anatomic study and clinical practice of mesopancreas and total mesopancreatic excision

Objective:
To explore the anatomical characteristics of the mesopancreas, to define the range of the total mesopancreas excision and to evaluate the feasibility, safety and effectiveness in the treatment of pancreatic cancer.
Methods:
A regional anatomical and pathological study was performed on 14 cadavers with large slices and paraffin sections. The clinical and pathological data of 58 consecutive patients underwent total mesopancreas excision for pancreatic head carcinoma from January 2013 to December 2015 were prospectively collected and analysed. The perioperative morbidity, mortality and clinical outcomes of patients underwent total mesopancreas excision were compared with the patients underwent conventional pancreaticoduodenectomy from January 2010 to December 2012.
Results:
The mesopancreas located in the retropancreatic area, extending from the head, neck, and uncinated process of pancreas to the aorto-caval groove, in which there were loose areolar tissue, adipose tissue, nerve plexus, lymphatic and capillaries. Although no fibrous sheath or fascia like mesocolorectum was found around the structures, a relatively fixed extent could be defined according to its embryologic and anatomic characters. In clinical practice, total mesopancreas excision was classified into two levels according to the extent of resection in this series: level Ⅰ was a"standard total mesopancreas excision" or"total mesopancreas excision in a narrow sense" , which was similar to the extent of standard resection from consensus statement of ISGPS. Level Ⅱ was defined as any procedure extending the range of level Ⅰ, called the"extended total mesopancreas excision" or"total mesopancreas excision in a broad sense". 7.Selective inhibition of endothelin receptor A improves white matter le-sions in hypertensive rats

AIM:To observe whether selective inhibition of endothelin receptor A(ETRA)improves white matter lesions(WMLs),and explore the mechanism.METHODS:Sprague-Dawley rats(n=33)were randomly divided into sham operation group(n=9),treatment group[stroke-prone renovascular hypertensive rats-modified 2 vessel occlu-sion(RHRSP-modified 2VO)+ambrisentan(n=12)]and placebo group[RHRSP-modified 2VO +vehicle(n =12)].Drug and vehicle administration was performed from 17th to 20th week and monitoring of systolic arterial pressure was performed weekly.Morris water maze test was conducted to evaluate the function of cognition.The protein levels of en-dothelin-1(ET-1)in the cortex,corpus callosum and caudate putamen were quantitatively analyzed respectively.The se-verity of WMLs and the relationship between ET-1 and vessels were observed by the method of histopathology.RESULTS:The difference of systolic arterial pressure between treatment group and placebo group was not significant.The animals in treatment group exhibited shorter escape latency(P<0.05),more times of crossing platform(P<0.05), lower level of ET-1 in corpus callosum and caudate putamen(P<0.05),respectively,improved WMLs severity(P<0.05)and lower binding level of ET-1 to vessels compared with the placebo group.CONCLUSION: Selective inhibition of endothelin receptor A improves the severity of WMLs and ameliorates the cognitive function.
8.Study on the extraction recovery of ginsenoside Re in plasma by different solid-phase cartridges.

OBJECTIVETo optimize the solid-phase extraction method by comparison of the extraction recovery of ginsenoside Re plasma samples.
METHODAfter extracted by different solid-phase cartridges with water, acetonitrile, and different content methanol elution, the plasma samples were analyzed on an Zorbax SB-C18 column with acetonitrile-water gradient elution. From the recovery achieved, the best solid phase cartridge was found.
RESULTThis method consists of using 40% methanol as the wash solvent, and 80% methanol for the elution. Among the three kinds of solid-phase being tested, Waters Oasis HLB cartridge was found to be the best one.
CONCLUSIONThe average extraction recovery of the Waters Oasis HLB cartridges was between 103%-113%, it can be used in the analysis of ginsenoside Re in plasma samples.

9.Biodegradation of tetrachlorothylene using methanol as co-metabolic substrate.

OBJECTIVETo investigate the biodegradation of tetrachloroethylene (PCE) using methanol as electron donor by acclimated anaerobic sludge.
METHODSHP-6890 gas chromatograph (GC), together with HP-7694 autosampler, was used to analyze the concentration of PCE and intermediates.
RESULTSPCE could be decholrinated reductively to DCE via TCE, and probably further to VC and ethylene. The degradation of PCE and TCE conformed to first-order reaction kinetics. The reaction rate constants were 0.8991 d(-1) and 0.068 d(-1), respectively, and the corresponding half-life were 0.77 d and 10.19 d, respectively. TCE production rate constant was 0.1333 d(-1), showing that PCE was degraded more rapidly than TCE.
CONCLUSIONMethanol is an electron donor suitable for PCE degradation and the cometabolic electron donors are not limiting factors for PCE degradation.

10.Preparation and property evaluation of a self-setting calcium phosphate cement

BACKGROUND: Preliminary studies have found that poly(lactic-co-glycolic acid) (PLGA) can improve the compressive strength and degradability of calcium phosphate cement. OBJECTIVE: To prepare a self-setting calcium phosphate cement which has better mechanical properties, biocompatibility and degradability on the basis of the previous findings. METHODS: Beta-tricalcium phosphate (β-TCP), pure calcium phosphate cement (CPC) and PLGA powder were mixed at different mixing ratios for preparation of PLGA/β-TCP/CPC. Setting time, compressive strength, elastic modulus and degradation properties of the composite bone cement were evaluated to screen the optimal level of β-TCP. MC3T3-E1 cells were cultured in CPC extract (control), PLGA/β-TCP/CPC extract (experimental), α-MEM containing 10% fetal bovine serum and 1% penicillin-streptomycin double antibody (negative control), and 6.4% phenol liquid (positive control). MTT method was used to detect cell proliferation at 1, 3, 5 days after culture, and lactate dehydrogenase activity in culture media was detected at 1 and 3 days after culture. MC3T3-E1 cells were seeded on the surface of PLGA/β-TCP/CPC and pure CPC respectively, and were then observed by scanning electron microscopy after 3 days. RESULTS AND CONCLUSION: Initial setting time and final setting time among of the composite bone cement were increased with the increasing of β-TCP content, but had no significant difference from those of the CPC (P > 0.05). The compressive strength and elastic modulus of the composite bone cement were higher than those of the CPC, and moreover, the composite bone cement with 20% β-TCP exhibited the highest compressive strength and higher elastic modulus as compared with the other groups. Therefore, PLGA/20% β-TCP/CPC was selected in the cell test. Moreover, the degradation properties of the composite bone cement were also better than those of the CPC. (3) With the growth of culture time, cell absorbance value and lactate dehydrogenase activity were gradually increased in the experimental group, and no difference existed between the experimental group and the negative control group. The cells in the experimental group also grew well. (4) MC3T3-E1 cells grew well and fully extended on the surface of PLGA/β-TCP/CPC, and cell pseudopodia on the material surface were tightly adhered to the material. To conclude, PLGA/20% β-TCP/CPC has better compressive strength, elastic modulus, degradation properties and cytocompatibility relative to the CPC, and moreover, the composite bone cement has no obvious cytotoxicity.
