1.Gene mutation type analysis of thalassemia in Taishan area

Objective To learn the gene mutation type and prevalence of thalassemia in the Taishan area and to provide basis for the prevention and treatment of local thalassemia .Methods 1 383 patients who visited the hospital for thalassemia genetic test from January to December 2014 were enrolled for this study .α‐andβ‐genes were determined by using PCR and membrane hybridization methods .Results Of the 1 383 patients ,595 were diagnosed with thalassemia (positive rate was 43 .02% ) ,including 386 cases (27 .91% ) of α‐thalassemia ,183 cases(13 .23% ) of β‐thalassemia and 26 cases(1 .88% ) of αβ‐thalassemia .- -SEA/αα(accounted for 64 .25% ) was the major genotype of α‐thalassemia ,the major genotype of β‐thalassemia wasβ41 -42/β(accounted for 28 .42% ) , and - -SEA/αα,β41 -42/βwas the major genotype of αβ‐thalassemia (accounted for 19 .23% ) .Conclusion Taishan is located in a high risk area of thalassemia ,it is of great significance to perform genetic diagnosis of thalassemia among reproductive population so as to reduce the birth rate of children with thalassemia in the area .
2. Protective effect of curcumin against reperfusion injury of mouse renal tubular epithelial cells in vitro

Objective To evaluate the protective effect of curcumin against reperfusion injury of mouse renaltubular epithelial cells in vitro and the related mechanisms. Methods The renal tubular epithelial cells(TECs)were isolated by an improved Percoll density gradient centrifugation method. The cultured TECs were divided into blank group, hypoxia reperfusion group, low-concentration curcumin (5μmol/L) and high? Concentration curcumin (25μmol/L) groups. Apoptoticrate of TECs was examined with FACS, and them RNA and protein levels of TLR4 were detected with RT-PCR and Westernblotting analysis, respectively. Meanwhile, expression of IL-6, IL-1ß, TNF-α and MCP-1 mRNA in TECs was examined by RT-PCR, and their contents in the supernatants of TECs were measured by ELISA. Results The purity of the isolated TECs washigher than 95%, and the cell viability was higher than 93%. Compared with hypoxia reperfusion group, the two curcumin groups exhibited significantly lower apoptotic rates (P<0.01) and lower levels of TLR 4m RNA and protein (P<0.01), inhibited P65 phosphorylation, and significantly decreased levels of IL-6, IL-1ß, TNF-α and MCP-1 in the TECs (at both mRNA and protein levels) and their supernatant (P<0.05orP<0.01). Conclusion Curcumin can protect TECs from reperfusion injury in vitro, which might be associated with the inhibition of TLR4 expression and the consequent attenuation of TLR4- triggered inflammatory damages.
3.Clinical result of phacoemulsification after implantation of an apodized diffractive multifocal IOL(Acrysof ReSTOR)

Objective To evaluate the visual outcomes,depth of focus and safety after implantation of the Acrysof ReSTOR and the Acrysof Natural.Design Nonrandomized clinical trial.Participant 53 patients with age-related cataract(61 eyes)were divided into two groups:Acrysof ReSTOR group(MIOL)included 28 patients(31 eyes),and reference group-Acrysof Natural group(SIOL)included 25 patients(30 eyes).Method All patients underwent phacoemulsification and IOL implantation.At 1 week,1 month and 3 months postoperatively,distant and near visual acuities were observed,and depth of focus was measured 3 months postoperatively.Main Outcome Measure Uncorrected distant visual acuity(UCDVA),uncorrected near visual acuity(UCNVA),best corrected distant visual acuity(BCDVA),best corrected near visual acuity(BCNVA),distant corrected near visual acuity(DCNVA),depth of focus.Result At 3 months postoperatively,in MIOL group:UCDVA was 0.90?0.15,UCNVA was 0.70?0.19,DCNVA was 0.73?0.21 and depth of focus was 5.5D(+1.5~-4.0D),while in SIOL group:UCDVA was 0.84?0.14,UCNVA was 0.36?0.10,DCNVA was 0.26?0.08 and depth of focus was 2.5D(+1.0~-1.5D).In MIOL group,UCDVA and UCNVA at 1 month were markedly better than those at 1 week. Conclusion Acrysof ReSTOR can provide excellent outcomes both in distants and near visions and it may reduce the dependence on spectacles in near vision.(Ophthalmol CHN,2006,15:344-347)

Objective To investigate the influence of Levetiracetam(LEV) and Topiramate(TPM) on P-glycoprotein(P-gp) expression at brain in the epileptic rats.Methods The epileptic rat models were made with 1.5 ?g(3 ?l) Kainic acid stereotaxic injected into male SD rats' hippocampi(epilepsy group) and the control group rats were injected with 3 ?l normal saline(NS).The rats in epilepsy group(18 rats) and control group(15 rats) were randomized into TPM,LEV and NS subgroups(6 or 5 rats for each subgroup) to receive once-daily orally feeding of TPM(50 mg/kg),LEV(40 mg/kg) and NS(same amount NS) for 30 d.Immunohistochemistry(EnVision) was used to measure the level of P-gp exprssion in the rats brain.The level of P-gp expression was semi-quantitatively analyzed by mean integrated blue(MIB) value in Leica Qwin imaging analysis & measuring system.Results The expression levels(MIB value) of P-gp in hippocampus and temporal lobe in epilepsy subgroups were significantly higher than those in the corresponding control subgroups(P
5.Non-control Study Systematic Review of Safety and Effectiveness of Three-Dimensional Printing Technology in Orthopedic

Objective To evaluate the safety and efficacy of clinical application of three-dimensional printing technology (3D printing) in Orthopedics.Methods The safety and effectiveness of clinical application of three-dimensional printing technology in Orthopedics were analyzed by systematic review of non-control studies.Results Non-control studies showed that cortical perforation rate of screws was 3.83% (calculated by the number of screw).Complication rate was 2.13%,surgical infection rate was 0.28%,prosthesis problematic rate was 0.79%,the average operation time was 139.23 minutes,and some results of the indicators for safety and effectiveness were similar between studies with and without control groups.Conclusion Although 3D printing application in Orthopedics has some clinical value,and its extension needs economic assessment.
6.Systematic Review of the Effect of Clinical Application of Three-dimensional Printing Technology in Orthopedics

Objective To assess the effect of clinical application of three-dimensional printing technology (3D printing) in Orthopedics.Methods Comparing the effect of clinical application of three-dimensional printing technology with traditional Standardized technology,through using a systematic review and Meta Analysis.Results Meta analysis of 25 studies included in the research showed that application of 3D printing reduced operation time by 26 minutes,decreased intraoperative blood loss by 77 ml,and increased accuracy rate or success rate of screw implanting(increased by 2.10 times compared with tradit onal standardized technology).Conclusion The application of 3D printing in Orthopedics has good short-term effect But its deVelopment in clinical application should be cautious.
7.Analysis of the Clinical Research of Three-Dimensional Printing Technology in Orthopedics

Objective To study onclinical researches of 3D printing technology in Orthopedics.Methods Through literature search,107 literatures were included and systematically reviewed,and the status of clinical researches of 3D printing technology in orthopedics was described.Results The clinical researches of 3D printing technology in Orthopedics were mainly focused on males and adults population conducted from 2007 to 2012.3D printing technology frequently used in Orthopedics were reduction and internal fixation,deformity correction,pedicle screw placement,tumor resection,revision and reconstruction,and knee replacement.Conclusion 3D printing technology in Orthopedics is still at an initial stage of clinical development.
8.Clinical Analysis of 87 Cases of Pregnant Women Complicated with Heart Failure

Objective To investigate the dangers of heart failure to the mother and fetal,and the diagnosis and treatment of pregnancy complicated with heart failure. Methods The clinical data of the causes,timing,management and outcomes of 87 cases of pregnant women complicated with heart failure from March 1993 to January 2006 were analyzed retrospectively. Results ①Thirty-one cases(35.6%) of heart failure in pregnancy were caused by rheumatic heart disease,26(29.9%) by pregnancy-induced hypertension(PIH),16(18.4%) by congenital heart disease,and 14(16.1%) by peripartum cardiomyopathy.②The heart failure appeared at the average of(32.69?5.57) weeks.③The average terminal time of pregnancy was(34.66?4.52) weeks.④There were 79 newborns with 6 cases of twins,and the perinatal mortality was 8.6%(8 cases).Hysterotomy was performed in 6 cases in mid-pregnancy.The average weight of babies was(2419.56?786.08) g.⑤The maternal mortality was 6.9%(6 cases).(Conclusion The main) causes of heart failure in pregnancy are rheumatic heart disease,PIH and congenital heart disease.Although the standard management of pregnancy complicated with heart failure is inotropic agents,diurectics and vasodilators,the treatment should be individualized according to the specific etiology and with the consideration of the safety of fetus as well.Pinpointing the causes of heart failure in pregnant patients and treating accordingly may be conducive to reduce the maternal and perinatal mortality.
9.Effect of cyclooxygenase-2 on bone loss in ovariectomized rats

Objective To investigate mechanism of cyclooxygenase-2 ( COX-2) in bone loss in a postmenopausal osteoporosis (PMOP) rat mode with ovarietomy (OVX).Methods Forty female Sprague Dawley adult rats at age of 3 months were randomly divided into 4 groups,10 in each group,including shamoperated (sham) group,OVX group,OVX treated with nilesteriol (OVX + E) group and OVX treated with aspirin ( OVX + P) group.All rats in OVX,OVX + E and OVX + P groups underwent ovarietomy under abdominal anesthesia with 10% chloral hydrate.Rats in sham group were only taken with fat tissue with same weight under bilateral ovary.After surgery,penicillin was administered to prevent infection.At day 7 after surgery,agents were given by intragastric administration for 12 weeks.Nilestriol at 1.0 mg/kg was used in OVX + E group once a week,aspirin at 45 mg · kg - 1 · d- 1 was used in OVX + P group once a day.Saline with same volume was used in rats in sham and OVX groups.All agents were administered one time per day.Dose of agents were adjusted by weight per week.At end of study,bone mineral density (BMD) of right femurs and lumbar vertebrae 3 -5 (L3-5) were measured.Morphology of bone was detected by hematoxylineosin,and expression of COX-2 was determined by immunohistochemistry staining.Results ( 1 ) BMD:BMD of right femur and L3-5 was (0.209 ±0.010) g/cm2 and (0.230 ±0.012) g/cm2 in sham group and (0.181 ±0.008) g/cm2 and (0.201 ± 0.016) g/cm2 in OVX group,which reached statistical difference (P<0.01).BMD of right femur and L3-5 was (0.203 ±0.009) g/cm2 and (0.224 ±0.028) g/cm2 in OVX + E group and (0.200 ± 0.011 ) g/cm2 and (0.204 ± 0.003 ) g/cm2 in OVX + P group,which were all higher than those in OVX group (P <0.01,P <0.05).However,there was no statistical difference in BMD between OVX + E and OVX + P group ( P > 0.05).(2) Morphology of bone:bone trabeculae became fewer and degenerated in OVX group.However,bone trabeculae were regular and dense in OVX + P group and OVX + E group,which were similar to those in sham group.(3) Expression of COX-2:cells with COX-2 positive and expression of COX-2 around bone trabeculae in OVX group were more than those in sham,OVX + E and OVX + P group.Conclusion COX-2 plays an important role in PMOP.Aspirin could prevent bone loss by decreasing COX-2 expression in OVX rats.
