1.Progress of tissue-engineered blood vessels

Vascular bypass grafting is the most common procedure for ischemic heart disease and peripheral vascular disease. Although synthetic grafts have been developed, replacement of vessels with purely synthetic polymeric conduits often leads to the failure of such graft, especially in the grafts less than 6 mm in diameter, mainly due to the early formation of thrombosis, infection and so on. Tissue engineering is a relatively new discipline that offers the potential to create vascular grafts from autologous cells and biodegradable polymer scaffolds. It has become a promising approach for generating a biocompatible vessel graft and contributing to the long-term patency rate of small-caliber grafts. This review describes the major progress in the tissue engineering vascular grafts, including the seeding cell sources, the biodegradable scaffolds, the construction technologies of tissue-engineered vessels, as well as nanotechnology.
3.Angiotensin Ⅱ and receptors in rheumatoid arthritis

Angiotensin Ⅱ and type 1 receptor ( AT1R) are ab-solutely therapeutic targets in the hypertension and cardiovascular disease, and angiotensin Ⅱ gets involved in monocytes chemo-taxis and migration, T lymphocytes activation and proliferation, Th1 and Th17 immune function, and synoviocytes apoptosis inhi-bition in patients with rheumatoid arthritis ( RA) through binding to AT1R in the autocrine and/or paracarine manner. Drugs that limit angiotensin Ⅱ production by angiotensin converting enzyme inhibitors ( ACEIs) or blockade of angiotensin Ⅱ-AT1R action by AT1R blocker ameliorate immune and inflammation responses in RA patients. Additionally, angiotensin Ⅱ type 2 receptor (AT2R) expression increases in RA and experimental model, and activation of AT2R exerts a therapeutic effect on adjuvant-in-duced arthritis. AngiotensinⅡand its receptors in RA and auto-immune diseases are retrospectively reviewed in this paper.
4.Effect of iodine supplement on thyroid of iodine-deficient rats

Objective To study the effect of chronic iodine supplement on thyroid of iodine-deficient rats.Methods Iodine-deficient rat models were made by giving 4- week-old rats iodine-deficient fodder for 3 months.These iodine-deficient rats were then supplemented for 8 months 1-,3- and 6- fold iodine which corresponded to the 3 levels of iodine intake found in our epidemiological study in 3 rural communities.Then changes of thyroid after iodine supplement were observed.Results After supplement of 1-,3- and 6- fold iodine to iodine-deficient rats for 8 months,thyroid relative weight did not recover,and iodine accumulated in thyroid.The ultrastructure injuries of swelling mitochondria and dilating endoplasmic reticulum in epithelial cells of iodine-deficient thyroid did not recover,but became worse with iodine supplement increase.Conclusion Iodine supplement alone can not correct the injuries resulting from iodine deficiency;moreover,a sub-pathological state might occur.
5.Preparation and Clinical Application of Sophora Alopecurodies Suppository

OBJECTIVE:To prepare Sophora alopecurodies suppository for treating prostatitis chronica and observe its clinical therapeutic effect METHODS:The content of the total alkaloid in the suppository was determined with acid dye spectrophotometry and the therapeutic effect was observed in comparison with qianliean suppository RESULTS:The linear range of TASa was 0 14~15 4?g/ml,standard oxymatriner's recovery was 101 2%,RSD=0 55% CONCLUSION:This suppository is simple in preparation,reliable in quality-control It is slight in irritation to rectum and effective in clinical use The preparation is worth popularizing in clinical practice
6.Culture of Donor Cells in Extracranial Pituitary Implantation

Objective To culture human pituitary adenoma cells to provide some useful experimental foundations to practice pituitary adenoma cells as donor cells in the extracranial implantation. Methods Human pituitary adenoma cells obtained by operation were cultured in vitro, and their growth state was dynamically observed. Immunohistochemical staining and RIA were used to evaluate their functional status. Results The cultured pituitary adenoma cells still had secretory function in vitro. 6 cases of GH adenoma cells secreted a large amount of GH, 5 cases of PRL adenoma cells secreted a large amount of PRL, and 3 cases of pituitary adenoma cells without function could also secrete a small quantity of FSH, LH and PRL. Conclusion 14 cases of cultured human pituitary adenoma cells had secretory function in vitro, which provides the useful experimental data to perform extracranial implantation with pituitary adenoma cells as donor cells.
7.A discussion of the combined application value of FMEA and RCA in the IV infusion risk management

Objective:To explore the combined application effect of failure mode and effect analysis (FMEA) and root cause analysis (RCA) in IV infusion risk management of pediatric outpatient so as to enhance the quality of care and ensure the safety of outpatients.Methods: For this exploration, a special analysis team was established and the flowchart was drawn up, and the potential failure modes were listed by using FMEA. And the RCA was used to analyze and evaluate the key mode in these potential failure modes so as to find the root cause might affect the safety of IV infusion, and then the improved measures were implemented and the effects of new measurement were evaluated. Results:After implemented the improved measures, the risk of top 4 risk priority number (RPN) in intravenous infusion flow decreased obviously. For the transfusion management, patients' satisfaction increased from 78.09% to 95.03% (P<0.05), and nurses' satisfaction increased from 83.79% to 97.80%(P<0.05).Conclusion: The combined application of FMEA and RCA in the IV infusion risk management of pediatric outpatient can effectively reduce the occurrence of risk events, reduce nursing complaints and disputes, and enhance the quality of nursing care.
8.Suppression effect of RNA interference on expression of EGFR of SKOV3 cell line

Objective To investigate the suppression effect of RNA interference(RNAi) on the expression of epidermal growth factor receptor(EGFR) of ovarian cancer SKOV3 cell line. Methods SKOV3 cells were transfected with synthetic EGFR sequence-specific dsRNA by lipofectamin 2000.The expression level of EGFR mRNA and protein were detected by RT-PCR and Western blotting,respectively.The proliferation of transfected SKOV3 cells was detected by WST-1 cell proliferation assay.The abilities of adhesion,migration and invasion were detected by cell adhesion assay,cell migration assay and Matrigel invasion assay,respectively. Results The suppression rates on EGFR mRNA and protein by sequence-specific dsRNA-EGFR were 73.65% and 58.94%,respectively.The cell proliferation was inhibited by 22.54%,35.76% and 31.07%,respectively at 24,48 and 72 h after transfection.The adhesion of sequence-specific dsRNA transfected SKOV3 cells for 30 min and 90 min were reduced by 12.11% and 18.66%,respectively.The migration and invasion of SKOV3 cells were decreased by 25.47% and 22.08%,respectively. Conclusion The down-regulation of EGFR expression of ovarian cancer by sequence-specific dsRNA can lead to the suppression of proliferation,adhesion,migration and invasion of ovarian cancer cells.
