2.Artery stiffness is an independent factor for left ventricular remodeling and cardiac function in coronary heart disease patients

Objective To study the correlation of artery stiffness with left ventricular remodeling and cardiac function in coronary heart disease (CHD) patients .Methods Four hundred and eighty patients were divided into CHD group (n=373) and non-CHD group(n=107)according to their coronary angiography .Patients in CHD group were further divided into normal left ventricular re-modeling group (n=139) and abnormal left ventricular remodeling group (n=243) .Their brachi-al-ankle pulse wave velocity (BaPWV ) was measured .The patients underwent echocardiography and 42 patients in CHD group received Doppler tissue imaging .Results The BaPWV was signifi-cantly higher in CHD group than in non-CHD group (P<0 .01) and in abnormal left ventricular remodeling group than in normal left ventricular remodeling group (1689 ± 305 cm/s vs 1508 ± 280 cm/s ,P< 0 .05) .The BaPWV was positively related with the IVST ,PWT ,RWT ,LVMI ,E/Ea and negatively related with the LVEF ,Ea ,Ea/Aa in CHD group (P<0 .05 ,P<0 .01) .Partial cor-relation analysis showed that the BaPWV was significantly related with the IVST ,PWT ,RWT , LVMI ,LVEF ,Ea ,Ea/Aa and E/Ea when the effect of hypertension ,etc ,was excluded (P<0.01) .Multivariate regression analysis demonstrated that the BaPWV was an independent factor for LVMI and LVEF .Conclusion BaPWV is an independent factor for left ventricular remodeling and cardiac function in CHD patients .
3.miR-193 regulates proliferation of rat marrow mesenchymal stem cells by cell cycle-related proteins

AIM:To investigate the effects of microRNA-193 (miR-193) on the proliferation and apoptosis of rat bone marrow mesenchymal stem cells (MSCs).METHODS:Cultured rat MSCs were transfected with pre-miR-193 or anti-miR-193 to regulate the expression of miR-193.The proliferation of the MSCs after transfection was evaluated by MTS assay, colorimetric BrdU cell proliferation assay and Ki-67 immunostaining.Cell apoptosis was analyzed by flow cytometry with Annexin V/PI staining.The effect of miR-193 on the expression of cell cycle-related proteins was evaluated by qRT-PCR.RESULTS:Transfection of pre-miR-193 or anti-miR-193 regulated the expression of miR-193 in MSCs effectively . Over-expression of miR-193 significantly promoted the proliferation of MSCs ( P<0.05 ) , and inhibition of miR-193 re-duced the proliferation of MSCs (P<0.05).miR-193 had no significant effect on the apoptosis of MSCs (P>0.05).The result of qRT-PCR indicated miR-193 promoted the expression of cyclin-dependent kinase 2 ( CDK2 ) significantly ( P<0.01).CONCLUSION:miR-193 promotes the proliferation of MSCs possibly through the CDK 2 pathway.
4.Study on quality standard of Huganbadu Ointment

Objective: To develop the quality standard of Huganbadu Ointment(Radix Sophorae Flavescentis, Herba Artemisiae Annuae, Radix Astragali, Radix Paeoniae Rubra, etc.) Methods: Radix Sophorae Flavescentis, Herba Artemisiae Annuae, Radix Astragali and Radix Paeoniae Rubra were identified by TLC and the content of matrine was determined by TLC Scanning. Results: The identification study showed strong specificity. The average recovery of matrine was 97.11%, and RSD was 2.50%. Conclusion: The methods established can be used for the quality control of the preparation.
5.Study on preparation of ?-cyclodextrin inclusion compound of volatile oil from Curcuma wenyujin

Objective:To study optimum inclusion process conditions for volatile oil from Curcuma wenyujin. Methods: The study was carried out with orthogonal design. The process conditions were studied by determining the ultizision ratio of volatile oil from Curcuma wenyujin , the oil bearing rate and extract ratio of inclusion compound. Results: The optimum preparation conditions for inclusion were established as∶oil∶? CD was 1∶9, The inclusion temperature and time were at 40 ?C and for 1.5h , The ultizision ratio of oil was 86.5% . Conclusions: The rate is high and the method is applicable.
6.The effects of inhaled budesonide on interleukin-8, C-reactive protein and procalcitonin in bronchoalveolar lavage fluid

ObjectiveTo investigate the effects of budesonide inhaled on the levels of interleukin-8 (IL-8), C-reactive protein (CRP) and procalcitonin (PCT) in bronchoalveolar lavage fluid (BALF) of patients with AECOPD treated by using inhalation of budesonide in different doses or by using injection of dexamethasone. MethodsNinety AECOPD patients with mechanical ventilation in ICU ward were enrolled from Jan. 2008 through Sep. 2010. All patients were selected in this study as per the criteria of AECOPD set by the Chinese Medical Association. Ninety AECOPD patients treated with routine therapy were randomly (random number) divided into 3 groups. The patients of group A were given budesonide 2 mg inhaled 1 time/d. The patients of group B was given budesonide 4 mg inhaled 1 time/d. The patients of group C had dexamethasone 2. 5 mg injected 1 time/12h. The changes of IL-8, PCT and CRP in both BALF and serum were respectively detected in three groups at the beginning of treatment, and 3 days and 7days after treatment. The detected data of variables were analyzed by SPSS 13.0 package. ResultsThe level of IL-8 in BALF declined in 3days and 7days after treatment, and the magnitude of decrease in the following order:Group B ( budesonide 4mg/d) > Group A ( budesonide 2 mg/d) > Group C ( intravenous dexamethasone)with significant differences among them ( P < 0. 05 ). The decrease in IL-8 in serum showed the similar trend, but there were no statistical differences among them. The CRP and PCT in both BALF and serum had no significant changes. There was a correlation between the concentration of IL-8 in BALF and the duration of mechanical ventilation.Conclusions The treatment with the inhalation of budesonide could more significantly reduced the IL-8 level in BALF compared with intravenous dexamethasone, whereas the CRP and PCT were not changed. The dynamic changes of IL-8 in BALF might be used as an indicator of prognosis in AECOPD patients with mechanical ventilation.

Objective To study the effect on Henoch-Schnlein purpura nephritis with cyclophosphamide (CTX) intravenous pulse therapy.Methods Fourty cases of Henoch-Schonlein purpura nephritis were divided into observation group and control group.The observation group:8~10 mg/(kg?time) of CTX was added into 100 mL of saline which was dropped intravenously,2 days was a period of treatment, intermission of 30 d, and the total duration was 4-6 months. 9.Preparation of internal quality control substance of real-time PCR to analyze UU-DNA and the design quality control programs

Objective Developing an internal quality control substance of Ureaplasma urealyticum(UU)‐DNA for real‐time PCR to establish an internal quality control system and preliminary evaluation its clinical value .Methods Internal quality control sub‐stance was prepared by mixing samples which Ct value were 24-25(positive sample) and 32 -33(weak positive sample) ,respec‐tively .At the same time ,selecting samples that test results were negative as negative control .The target value ,standard deviation (s) and coefficient of variation(CV) of internal quality control substance were defined by“instant method”for the first 20 runs and Levey‐Jennings quality control(QC) chart after the first 20 runs .Using the“Westdard” multi‐rule quality control methods to ana‐lyze the detection results .Exporting OPSPecs chart by quality control rules in Unity Real Time (URT ) system and setting up new quality control rules according with OPSPecs chart .Results 131 times of the detection of quality control substance were performed totally .The first 20 runs were defined by“instant method”and later 111 runs were defined by Levey‐Jennings QC chart ,the results were stable of quality control substance and reasonable quality control rules .Conclusion Preparing of internal quality control sub‐stance of UU‐DNA used in real‐time PCR might be easy and stable .So ,the internal quality control substance of UU‐DNA could be worthy for practical application in this PCR laboratory .Design internal quality control rules based OPSPecs chart in molecular de‐tection is very simple and practical .
10.Zebrafish embryonic brain cell apoptosis and c-fos gene expression after hypoxia reperfusion

BACKGROUND:Foreign scholars have researched hypoxia reperfusion in zebrafish embryos, but there is no research on c-fos gene expression and the mechanism during zebrafish cerebral hypoxia reperfusion.
OBJECTIVE:To observe the zebrafish embryonic brain cel apoptosis and expression of c-fos gene in brain tissues after hypoxia reperfusion.
METHODS:Zebrafish embryos were selected at 48 hours post fertilization. Neonatal hypoxia reperfusion injury was simulated by gradual y leading nitrogen (99.999%) into the device. After hypoxia treatment for 6, 12 and 24 hours, the embryos received reperfusion for 6 hours under normal oxygen concentration. The embryos in the control group received normoventilation (the dissolved oxygen concentration was about 7.0 mg/L). Acridine orange staining was performed to observe the effect of different hypoxia durations on the apoptosis of neurons in zebrafish, and then the c-fos gene expression was quantitative analyzed with real-time quantitative nucleic acid amplification detection system. And the expression level of c-fos gene was compared before and after hypoxia reperfusion.
RESULTS AND CONCLUSION:A smal amount of apoptotic brain cel s could be detected in the control group, and the c-fos gene expression level was decreased;in the experimental group, the number of apoptotic cel s was increased after hypoxia for 6, 12 and 24 hours, and the gene expression after hypoxia for 6 hours was increased distinctly. The results indicate that hypoxia can increase the c-fos gene expression in brain cel s of zebrafish embryos, which may be one of the mechanisms of brain cel apoptosis increasing after hypoxia.
