1.Purification of epidermal melanocytesin culture
Tianjin Medical Journal 2017;45(1):5-8
Objective To compare the influence of different coating materials and cultural conditions on the purification and growth of human epidermal melanocytes. Methods The full-thick foreskin, epidermis and cell suspension obtained from human foreskin were cultured in the plates, which were precoated with matrigel or laminin respectively. When having reached 80%-90%confluence, the cells were treated with 0.05%trypsin-EDTA for 4 minutes and resuspended in M254 medium, which were supplemented with G418 and 5-BrdU, respectively. The purity of melanocytes was observed by an immunofluorescence staining with melanocyte markers. Results During the primary culture, the cell suspension generated more cells at faster speed compared with those of skin explants and epidermal specimen. Moreover, the epidermis released cells earlier and proliferated quickly over skin explants. The melanocytes in the plates coated with laminin other than with matrigel displayed faster and better growth. The unwanted keratinocytes and fibroblasts were removed by using differentiation trypsinition combined with supplement of G418 or 5-BrdU. Conclusion Using a plate coated with laminin to culture cell suspension from human foreskin, and via a differentiation trypsinization combined with supplement of small doses of G418 to subculture the cells, is advantageous to the melanocyte purification, without affecting their growth.
2.Advances in chemotherapy of small cell lung cancer
Chinese Journal of Clinical Oncology 2016;43(10):418-423
Small cell lung cancer (SCLC) accounts for about 13%of lung cancer. SCLC exhibits an early metastatic potential and sensitivi-ty to first-line cytotoxic chemotherapy compared with non-small cell lung cancer. Current treatments include surgery, radiotherapy, and chemotherapy. Platinum plus etoposide is viewed as the standard first-line chemotherapy. However, recurrence rate of cancer af-ter platinum plus etoposide chemotherapy is high. Topotecan monotherapy is the standard second-line chemotherapy. Meanwhile, novel targeted immunotherapy showed poor effects. Hence, new breakthroughs in the treatment of SCLC are urgently needed.
3.Diagnosis and treatment of spontaneous colorectal rupture
Clinical Medicine of China 2010;26(9):969-971
Objective To retrospectively investigate the cause of spontaneous rupture of the large intestine, pathology, diagnosis, treatment and prognosis of the relevant factors. Methods The collection of domestic medical records of 155 cases of spontaneous rupture of the large intestine in last decades, combined with 5 cases collected in our hospital since 2002. All the cases underwent emergency surgery confirmed the preoperative diagnosis of straight, colon rupture in 25 cases. Surgical repair for rupture in 5 cases, rupture of an external repair in 5 cases, rupture of the proximal colostomy repair combined with single or dual-chamber in 43 cases of ruptured bowel resection and anastomosis of the proximal intestine fistula combined with single or dual-chamber in 61 cases ruptured bowel resection stump of dual-chamber fistula in 6 cases, rupture of bowel resection and anastomosis in 5 cases, rupture of intestine directly fistula 9 cases. Surgical presentation in the remained cases were unknown. Results One hundred and seventeen patients cured, 43 cases died. Conclusions Most spontaneous colon rupture occurred in elderly patients, and is rare disease. The diagnosis rate is low and easy to delay in the treatment. Moreover, the intraabdominal infections caused by pollution would leed to a direct life-threatening toxic shock. Therefore, prompt diagnosis and timely surgery is important to improve the cure rate.
4.Multi-mode administration of dexmedetomidine for thoracic surgery anesthesia
Chinese Journal of Postgraduates of Medicine 2013;36(33):39-42
Objective To investigate the reasonable and effective administration ofdexmedetomidine for thoracic surgery anesthesia. Methods Eighty ASA Ⅰ-Ⅱ patients, aged 18-60 years old scheduled for elective thoracotomy were randomly assigned to 4 groups(each 20 patients).Group A:dexmedetomidine before anesthesia induction + bupivacaine before closed thorax cavity.Group B:dexmedetomidine before anesthesia induction + dexmedetomidine and bupivacaine before closed thorax cavity.Group C:0.9% sodium chloride before anesthesia induction + bupivacaine before closed thorax cavity.Group D:0.9% sodium chloride before anesthesia induction + dexmedetomidine and bupivacaine before closed thorax cavity.Mean arterial pressure (MAP) and heart rate (HR) were measured before infusing dexmedetomidine or 0.9% sodium chloride (T0),after infusing dexmedetomidine or 0.9% sodium chloride (T1),instant time after intubation (T2) and 3 min after intubation (T3),5 min after intubation (T4).The scores of visual analogue scale(VAS) and the consumption of analgesics were compared. Results There was no significant difference including gender, age, weight and operation time among four groups (P > 0.05).Compared with T0,MAP and HR were significantly decreased at T1 in group A and group B (P < 0.05),and were significantly increased at T2 in group C and group D (P< 0.05).Respectively compared with group A and group B,MAP and HR were significantly increased at T2,T3 in group C and group D (P < 0.05).The scores of VAS in group B [(2.47 ± 1.43) scores] and group D [(2.00 ± 1.68) scores] were lower than those in group A [(4.78 ± 1.26) scores] and group C [(4.88 ± 1.62) scores] after operation 12 h.The times of using postoperative analgesics in group B [(0.6 ± 0.4) times] and group D [(0.8 ± 0.1) times] were significantly less than those in group A [(1.3 ± 0.5) times]and group C [(1.5 ± 0.4) times] (P < 0.05).Conclusions Intravenous dexmedetomidine before anesthesia induction can control the effect of double-lumen endobronchial tube responses and make hemodynamics stable.Intercostal nerve block with 0.5 μ g/kg bupivacaine and 0.375% dexmedetomidine can enhance the analgesia effect and prolong the analgesia time.
5.THE ACUTE EFFECT OF PHYSICAL EXERCISE ON PLASMA ?-THROMBOGLOBULIN LEVELS IN DIABETIC PATIENTS
Chinese Journal of Endocrinology and Metabolism 1985;0(02):-
The dynamic change of plasma B-TG as well as it's relations with blood glucose, serum insulin and growth hormone levels before and after a moderate physical-exercise of 30 minutes in 16 cases of NIDDM without demonstrable vascular complications were investigated. Results indicated that the average basal level of B-TG in NIDDM was significantly higher than that of control group (P
6.Effect of azithromycin and terbutaline on the improvement of clinical symptoms in children with pneumonia
Chinese Journal of Biochemical Pharmaceutics 2017;37(6):312-314
Objective To investigate the effect of Azithromycin combined with terbutaline on the treatment of pneumonia in children and its effect on symptom improvement.MethodsThe clinical data of children with pneumonia treated in Taizhou Hospital of traditional Chinese and Western medicine from January 2014 to December 2016 were retrospectively analyzed, according to the treatment methods, the patients were divided into control group and observation group, the control group was given azithromycin treatment, the observation group on this basis to give terbutaline atomization treatment. The therapeutic effects of the two groups were observed, and the differences of clinical symptom score, inflammatory factor level were compared between the two groups before and after treatment.ResultsThe effective rate of the observation group was 98%, which was significantly higher than that of the control group (84%);The clinical symptom score had no difference between the two groups before treatment, after treatment, the observation group of cough, expectoration, fever and rales were lower than those in the control group; The two groups had no difference between the levels of proinflammatory cytokines, after treatment, the observation group IL-6, IL-8, TNF-and CRP levels were lower than the control group.ConclusionAzithromycin combined with terbutaline in the treatment of children with pneumonia has a better therapeutic effect, can significantly improve the clinical symptoms, reduce the level of inflammatory factors, has a good clinical value.
