1.DNA methyltransferases in cancer development, treatment and prognosis

Overexpression of DNA methyltransferases(DNMTs)is observed in many tumors. Overexpressed DNMTs silence tumor suppressor genes by catalyzing the methylation of CpG islands within the promoter regions, and therefore promote the neoplastic transformation of normal cells. Highly elevated DNMT activities have been observed in cancers including gastrointestinal cancers, breast cancer, and lung cancer and have been associated with cancer development and prognosis. Drugs and targeted therapies that inhibit DNMT activities can reactivate methylated tumor suppressor genes, promote apoptosis, and thus inhibit tumor growth.

Objective To analyse the personality characteristics of medical undergraduate and graduate newcomers. Methods Cattell 16 Personality Factor Questionnaires (16PF) were employed to assess the personality of 675 undergraduate newcomers (306 males and 369 females) and 155 graduate newcomers (76 males and 79 females),and the results were compared with the established findings of the country. 3.Protective Effect of Tongsaimai Pellets and Butylphthalide to PC12 Cell in Hypoxia/Hypoglycemia Model

Objective To discuss the protective effect of Tongsaimai pellets(TSMP) and butylphthalide(NBP) on PC12 cell in the hypoxia/hypoglycemia model.Methods Two ischemia models including hypoxia,hypoglycemia models were used to assay the anti-ischemic roles of TSMP and NBP in cultured PC12.Results TSMP and NBP possessed obvious protective effects on PC12 cells from two injured models.Both of them could increase the number of living cells and decrease LDH activity significantly,particularly in hypoglycemia injured model.Conclusion TSM and NBP have protective effect on PC12 cells from two injured models effectively in vitro.
4.Expression of miRNA-1247-3P in acute respiratory distress syndrome cell model and its function

Objective To detect the expression pattern of microRNA in A549 cells treated by lipopolysaccharide, study the expression of miRNA-1247-3P in A549 cells under LPS treatment and explore the possible mechanism of miRNA-1247-3P in A549 cells under LPS treatment. Methods A549 cells were divided into experimental and control groups. Immunocytochemical method and RT-PCR were used to detect the changes of SP-A and SP-C. The expression of miRNAs were detected using miRNAs array in different groups. The key miR-1247-3P was collected to detect the changes of miR-1247-3P in all groups using quantitative real-time polymerase chain reaction. Results Compared with control group, the expressions of SP-A and SP-C were significantly decreased in the experimental groups (P < 0.05). MiRNA array showed that 31 miRNAs were up-regulated and 3 miRNAs were down-regulated. Compared with control group, the expression of miR-1247-3P was significantly increased in the experimental groups (P < 0.05). Conclusion The increased expression of miR-1247-3P may play an important role in the pathogenesis of ARDS.
5.Effect of Jiechangyan Qixiao Granules in the Treatment of Rats with Ulcerative Colitis Induced by Dextran Sulfate Sodium

Objective:To study the preventive and therapeutic effects of Jiechangyan Qixiao granule ( JQX) in the rats with ulcer-ative colitis ( UC) induced by dextran sulfate sodium .Methods:The UC model was induced by drinking dextran sulfate sodium ( DSS, 4%) freely in Wistar rats weighting 180-220g.Guben Yichang tablets and sulfasalazine was used as the standard drugs for the compari -son.After 7-day intragastric administration of Jiechangyan Qixiao granules at the dose of 2.70, 1.35 and 0.68 g· kg-1 · d-1 , the se-rum levels of malondialdehyde (MDA), superoxide dismutase (SOD), nitric oxide (NO), interleukin -6(IL-6)and tumor necrosis factor-α(TNF-α), and the protein expression of myeloperoxidase (MPO) and intercellular adhesion molecule (ICAM-1)in colon and nuclear factor(NF-kBp65) were detected.Results:Compared with the model group, the granule at high and medium dose could sig-nificantly increase the activity of SOD in blood and decrease the contents of MDA , TNF-αand IL-6 (P<0.05 or P<0.01).The granule could also notably decrease the MOP activity in colonic mucous of UC rats (P<0.05 or P<0.01), and the contents of NF-kBp65 and ICAM-1 in the inflammation reaction(P<0.01).Conclusion: JQX shows promising efficacy in the treatment of UC rats induced by dextran sulfate sodium .
6.Analysis of the function of the UBE gene based on bioinformatics

[Abstract ] Objective Bioinformatics provides a lot of valuable information for online prediction of new genes.In this study, we predicted the biological function of ubiquitin-conjugating enzyme 2S ( UBE2S) based on bioinformatics. Methods The UBE2S gene was screened and cloned from the cDNA library of human HepG2 cells.The relationship of the structure and function of UBE2S was explored based on the full-length cDNA library.MEGA5.05, CLUSTALW2 and SWISS-MODEL were used to study the phylogeny, conservation, and 3D structure of UBE2S. Results The UBE2S gene encoded a polypeptide of 241 residues with a predicted molec-ular weight of 23 770 and an isoelectric point of 8.81.The UBE2S protein contained no transmembrane locus and the probabilities of their functions of growth factors, cation channel and structural protein were 8.904, 0.313, and 0.291.The analysis of BLASTp showed that the isolated UBE2S had a 90-97%identity with the other species. Conclusion Analysis of the structure and function of the UBE2S protein can not only provide more information about its gene family but also pave the way for further experimental studies on the molecular mechanism of the consequent hepatocellular carcinoma.
7.Moxibustion at Front-Mu Point of Abdomen for Intestinal Dysbacteriosis in Rats

Objective:To study the moxibustion at Guanyuan(CV 4)and Tianshu(ST 25)on intestinal dvsbacteriosis in rats.Methods:Fifty Wistar rats,clearing grade,were randomly divided into 5 groups,a normal group,a model group,a medical group,and a Guanyuan(CV 4)group and a Tianshu(ST 25)group,with 10 rats in each group.The rats were treated with Lincomycin Hydrochloride via intragastric administration for developing models.The model rats were treated with medicine and moxibustion respectively.0.1 g fresh rat feces in each group were cultured on the selective culture medium of bifidobacterium species(BS),lactobacillus(LBS),enterobactefiaceae(EB)and enterococcus(EC).The growth and quantity of the bacterial colony were tested by biochemical identification tubes and turbidimetry.Results:Moxibustion at Guanyuan(CV 4)increased BS and LBS while moxibustion at Tianshu(ST 25)improved EB and EC.Conclusion:Moxibustion at Front-Mu points Of different body parts selectively regulated advantaged probiotics for treating intestinal dysbactenosis.
9.Endovascular Stents Placement in the Treatment of Atherosclerotic Stenosis of the Iliac Arteries

Objective To investigate the security and effect of endovascular stents placement in treating atherosclerotic stenosis of the iliac arteries.Methods 15 cases with iliac arterial stenosis caused by atherosclerosis were treated by a big amount of thrombolysis in combination with iliac arterial stent placement.The length of lesions was from 4.3 cm to 9.5 cm(mean of 6.7 cm),the claudicant distance was from 100 m to 300 m(mean of 185 m).Clinical follow-up included CT angiography,color Doppler sonography and clinical evaluation with the ankle-brachial index(ABI).Results Eighteen stents were placed in 18 limbs of 15 patients.The technical successful rate was 100%.The clinical symptoms(claudication or the rest pain)were improved,and ABI increased from 0.25?0.18 to 0.85?0.15 after stent implantation in all cases.All the patients were followed up for 3~47 months,one patient was death during the follow-up periods because of brain stem ischemic stroke.All patients maintained uninterrupted stent patency until the final follow-up.Conclusion Iliac arterial stent placement is a safe treatment with favorable long term patency for treating arterial stenosis caused by atherosclerosis.
10.Association of Glu298Asp polymorphism of the endothelial nitric oxide synthase gene with myocardial infarction in the elderly patients

Objective To explore the association of Glu298Asp polymorphism of the endothelial nitric oxide synthase gene (eNOS) with myocardial infarction in Chinese aged people based on a case-control study. Methods Thirty-seven patients with myocardial infarction and 172 sex- and age-matched healthy control subjects were enrolled. Their height, weight and blood pressure were measured and the fasting plasma lipid concentrations were examined. Glu298Asp polymorphisms of the eNOS gene were examined using PCR and RFLP methods. Results Genotype Glu/Asp in the patient group was higher than that in the control group (32.4% vs 18.0%). The genotype composition of Glu298Asp polymorphism showed significant difference between the patient group and the control group ( =3.87, P 0.05). Among the normotensive subgroup, the allelic frequencies of 298Asp in the patient subgroup was higher than that in the control subgroup (17.5% vs 5.4%).
