AIM To set up a method which can determining the blood concentration of omeprazole and its metabolite 5′ omeprazole, omerpazole sulphone in order to study its cinical pharmacokinetics. METHODS The blood concentration of omerpazole was determinated by HPLC. RESULTS Calibrated standard curve of omeprazole in blood is Y=-0 004 499+0 001 909X (r =0 9990), the recoveries of three concentrations 50, 500, 2 000 mg?L -1 are 90 36, 109 62, 108 91%, respectively; and the precisions are 9 86,7 86, 15 52% , respectively. Calibrated standard curve of its metabolite 5′ OH omeprazole in plasma is Y=-0 003 659+0 001 328X(r =0 9970), the recoveries of three concentrations 20, 200, 1 000 mg?L -1 are 79 42%, 96 49%, 95 04%, respectively; and the precisions are 8 95%, 4 52%, 9 73%, respectively. Calibrated standard curve of its another metabolite omeprazole sulphone in plasma is Y=0 009 248+0 001 765X (r =0 999 2), the recoveries of three concentrations 20, 200, 1 000 mg?L -1 are 94 44%, 105 59%, 104 26%, respectively; and the precisions are 8 72, 8 58, 9 60%, respectively. After 20 mg omeprazole were administered by a voluteer via oral, C max of 5′ OH omeprazole, omeprazole, and omerpazole sulphone were 14 622 7, 408 433 2, 454 363 7 mg?L -1 . CONCLUSION The method is good enough to study pharmacokinetics of omeprazole.

3.MRI of Acute Pancreatitis

Objective To assess the value of magnetic resonance imaging for acute pancreatitis.Methods 77 patients of acute pancreatitis were examined with holding breath MRI sequence.Transverse scan:2D Flash T 1WI/T 1W+FS,TSE T 2WI+FS or HASTE+FS,Trufisp.Coronals scan:2D Flash T 1W+FS.MRCP was used in 34 patients.2D Flash dynamic contrast enhanced were used in 77 patients.Results 59 cases of acute edematous pancreatitis showed swelling and low signal intensity on T 1WI and higher signal intensity on T 2WI.Excepted the above findings,18 cases of hemorrhagic necrosis pancreatitis alos showed the higher signal intensity on T 1WI and peripancreatic exudative hydrops.Conclusion MRI is a reliable method for demonstrating acute pancreatitis.
4.Comparison of genotype polymorphism of cytochrome CYP2C19 between Chinese Han subjects and Chinese Meng subjects

AIM: To compare genotype polymorphism of cytochrome CYP2C19 between Chinese Meng subjects and Chinese Han subjects. METHODS: The genotype polymorphism was analyzed by PCR RFLP (Restriction Fragment Length Polymorphism) in 74 subjects of healthy Chinese Han and 6 subjects of Chinese Meng. RESULTS: Of the 74 genotyped healthy subjects, 31 ( 41.9 %) were homozygous for wildtype (wt/wt), 9 ( 12.2 %) were homozygous for CYP2C19m1 and CYP2C19m2 (m1/m1 or m1/m2 or m2/m2), and 34 ( 45.9 %) were heterozygous for CYP2C19m1 or CYP2C19m2 (m1/wt or m2/wt). Among the 6 genotyped Chinese Meng, 1 ( 16.7 %) was homozygous for wildtype (wt/wt), 2 ( 33.3 %) were homozygous for mutant allele CYP2C19m1 and CYP2C19m2 (m1/m1 or m1/m2 or m2/m2), and 3( 50.0 %) were heterozygous for CYP2C19m1 or CYP2C19m2 (m1/wt or m2/wt); no homozygous genotype for CYP2C19m2(m2/m2)was found in this study. CONCLUSION: There is no statistical difference in occurrence of wt/wt and m1/m1 between in 74 subjects of healthy Chinese Han and 6 subjects of Chinese Meng.
5. Expression of hypoxia-inducible factor 1α, glucose transporter 1 and lactate dehydrogenase 5 in colorectal cancer and clinicopathologic significance

Objective:
To investigate the expression and clinicopathological significance of hypoxia-inducible factor 1 alpha (HIF-1α), glucose transporter 1(GLUT-1) and lactate dehydrogenase(LDH)-5 in colorectal cancer.
Methods:
The expression levels of HIF-1α, GLUT-1 and LDH-5 were detected by immunohistochemical staining in 142 specimens of human carcinoma in comparison with adjacent normal tissues.
Results:
The expression levels of HIF-1α(78.2%, 111/142), GLUT-1(75.4%, 107/142) and LDH-5(68.3%, 97/142) were higher in tumor tissues than in adjacent normal tissues(14.8%, 21/142; 11.3%, 16/142; 7.0%, 10/142;
6.The dynamic functional connectivity of brain network during motor imagination and execution of upper limbs based on EEG signals

Objective:The electroencephalogram(EEG)signals were collected for analysis to define the differences in dy-namic functional connectivity of the brain network of related nodes in the primary motor area(M1)and pre-motor area(PMA)during motor imagination and motor execution.The relationship between muscle synergy and isolated movement was also explored. Method:Ten stroke patients with right hemiplegia and nineteen healthy adults participated in this study.All participants performed motor imagination(MI)and motor execution(ME)tasks according to visual instruc-tions.We recorded and analyzed the EEG signals at 12 sites located in Ml and PMA areas.The chosen EEG signals were filtered and analyzed based on the modified S-transform(MST).All data were normalized to avoid individual differences.Then we analyzed the data with Pearson correlation to identify the dynamic func-tional connectivity(FC)and the differences with Fisher's exact test for node degrees. Result:All the distribution trend of correlation degree of chosen node about left or right MI and ME of stroke patients was similar to that of healthy participants.Compared with the motion execution,the function connection strength and density of each node were elevated at MI,which was also consistent with healthy par-ticipants.When healthy adults underwent left hand MI,the degree of the C4 node in the Ml area was signifi-cantly higher than that of C3 on the opposite side(P<0.05),while at right hand MI,the sum of the node de-grees of FC3 and FC1 in the left PMA area was significantly higher than that of the lateral symmetric chan-nel FC4 and FC2(P<0.05).When the right upper limb isolated movement was performed,the node degree of C3 decreased significantly(P<0.05). Conclusion:The major region of function connectivity of the right hand MI was in the left PMA area,and the node degree at MI was higher than ME.The functional connectivity of each node at the left hand MI was dispersed.The main channels activated by the muscle synergy are different from the isolated movement.
7.Application effects of structured intervention model on the protective restriction in psychiatric inpatients

Objective To explore the application effects of the structured intervention model on the protective restriction in psychiatric inpatients.Methods The study investigated 91 8 psychiatric inpatients hospitalized from January 201 3 to December 201 4.The participants were randomly divided into the intervention group (n =469 )and the control group (n =449 ).The patients of intervention group received protective restriction based on the structured intervention model.A standard risk assessment was applied before and after the restriction.The patients of control group received the routine protective restriction.The protection rate, protection duration,and day-night protection time were compared between the two groups.Results The protection rate was 1 5.4% (71 /469)for the intervention group,which was lower than the rate of 20.49%(92 /449)for the control group (P <0.05 ).The difference of day-night protection was also statistically significant between the two arms (P <0.05).Conclusions The application of structured intervention model on the clinical practice can effectively reduce the protection rate and protection duration in psychiatric inpatients.
