1.Activity of Ca~(2+).Mg~(2+)-ATPase and SDH in myocardium of rat induced by macleaya cordata and ultrastructures

Objective To study the mechanism of the rat’s myocardium induced by macleaya cordata. Methods36 SD rats were randomly assigned to 2 experimental groups treated by macleaya cordata decoction with a dose of 1/6 LD50, 1/3 LD50 respectively and a control group treated with distilled water by oral administration. The rats were killed 6 hours after the administration for taking the cardiac muscle to examine the Ca2+.Mg2+-ATPase, SDH while observing the structures. ResoultsIn the group treated by macleaya cordata with dose of 1/6 LD50, the activity of Ca2+.Mg2+-ATPase and SDH were increased significantly.However, the groups treated by macleaya cordata with dose of 1/3 LD50 and distilled water, the activity of Ca2+.Mg2+-ATPase,SDH showed no significant change. The apoptosise in myocardium cells of rat’s can be observed in the experimental groups,while the changes were not seen in control group. ConclusionThe results indicate that the macleaye cordata can induce and improve the apoptosis of myocardium cells in low dose, and these effects may be induced by stimulating the activity of Ca2+.Mg2+-ATPase,SDH in myocardium cells.
2.Giant benign symmetric lipomatosis of the neck: four case reports.

Benign symmetric lipomatosis is a rare disease and may appear as a huge tumor in the neck. Four benign symmetric lipomatosis associated with gigantic painless mass or neck motion limitation were reported. Operative technique of one-time radical resection or stage resection was used to remove these tumors. One patient had a postoperative complication of incision effusion and infection, and three patients had no significant complication. At more than one-year follow-up, the motion and appearance of a patient's neck returned to normal, and no recurrence was observed. The etiology, clinical manifestations, diagnosis, and treatment of the disease were discussed.

3.Lipocalin-type prostaglandin D synthase:preparation of monoclonal antibodies and localization on human sperm

Objective:To prepare monoclonal antibodies against lipocalin type prostaglandin D synthase and to explore the distribution of lipocalin type prostaglandin D synthase on human sperm.Methods:BALB/c mice were immunized with lipocalin type prostaglandin D synthase,monoclonal antibodies were prepared by hybridoma technique,the biological characteristics of antibodies were analyzed by ELISA and Western blot,the distribution of lipocalin type prostaglandin D synthase on sperm were performed by immunohistochemistry.Results:A strain of hybridoma cell was obtain,and the IgG isotype of the antibody was identified as IgG1,with affinity constat(K) of 2?108 and titer over 103.Western blot demonstrated specific recognition of lipocalin type prostaglandin D synthase by the obtained monoclonal antibody.The result of immunohistochemistry displayed that lipocalin type prostaglandin D synthase was mainly distributed on the surface of human sperm.Conclusion:A hybridoma cell secreting anti-lipocalin type prostaglandin D synthase monoclonal antibodies stably was established.On the surface of human sperm,there is lipocalin type prostaglandin D synthase's distribution. Objective To study the expression and clinical significance of miR-183-5p, TβRⅠ and TβRⅡ in esophageal squamous cell carcinoma (ESCC). Methods The mRNA and protein expression of miR-183-5p, TβRⅠ and TβRⅡ were examined in ESCC cell lines ECA-109, TE-1, normal esophageal epithelial cells, tumor tissues and tumor-free tissues from 72 ESCC patients. Their clinical significance and the relationship between miR-183-5p and the latter two were analyzed. The effects of miR-183-5p on the expression of TβRⅠand TβRⅡ in ECA-109 cells and the cell functions of ECA-109 were also investigated. Results Compared with the normal esophageal epithelia cells, ESCC cell lines TE-1 and ECA-109 were statistically characterized by a high expression of miR-183-5p (all P<0.05) and low expression of TβRⅠand TβRⅡ(all P<0.05). The expression of miR-183-5p in ESCC tissues was higher than that in adjacent normal tissues, while the expressions of TβRⅠ and TβRⅡ were lower (all P< 0.05). The expression of miR-183-5p was closely related to sex, tumor differentiation, tumor staging, distant metastasis, lymphatic metastasis, and tumor location (all P<0.05). TβRⅠlevel was associated with sex, lymph node metastasis and tumor size (all P<0.05). Experimental data showed the negative correlation between the expression of miR-183-5p and TβRⅠin ESCC tissues (r= -0.521, P< 0.05). Over expression of miR-183-5p significantly inhibited the expression of TβRⅠ in ECA-109 cells (P< 0.05) and promoted the growth, invasion and metastasis of ECA-109 cells (P< 0.05). Low expression of miR-183-5p significantly promoted the expression of TβRⅠ in ECA-109 cells (P< 0.05), and suppressed the growth, invasion and metastasis of ECA-109 cells (P< 0.05). There was no significant change in the expression of TβRⅡ in the transfection experiments. Conclusion MiR-183-5p is closely related to the abnormal expression of TβRⅠ, which may exert an important role in the progression of lymphatic metastasis.
5.Clinical efficacy of different operative techniques for reconstruction of vermilion tubercle after primary repair of cleft lip

Objective To evaluate the clinical repair effects of the reconstruction of different vermilion tubercle after primary cleft lip repair.Methods According to the tight lip deformity and dif ferent degrees of defect of the vermilion tubercle and exposing the gums and the crown of the anterior incisors,178 patients after primary repair of the cleft lip from Department of Oral and Maxillofacial Surgery of Xiangya Hospital,Central South University were classified into the tight lip deformity and four grades of whistling deformities.According to different deformities and defects,the different operative techniques were used as follows:classic Abbé flap and modified Abbé flap were used for tight lip deformity;the bilateral advanced myomucosal flaps of lip were used for the grade Ⅰ;modified Abbé flap was used for repairing the grade Ⅱ and the grade Ⅲ whistling deformities;for grade Ⅳ whistling deformity,we mainly used classic Abbé flap.The follow-up after operation was performed,all patient's Cupid's bow,Cupid's bow peak,the vermilion tubercle and the height and width of the upper lip were observed.The scarring of the upper lip was also observed.Results The follow-up after operation was performed from six months to ninteen years,all the 187 patient's anatomical structures of the upper lip were nearly normal after recovery.The all patients showed an obvious Cupid's bow,Cupid's bow peak,the vermilion tubercle and the height and width of the upper lip.The relation of the upper lip and lower lip was harmonious.The scarring of the upper lip was not obvious 2 years after operation.Conclusions The different repair methods are chosen to repair different deformity and defect that has an important clinical value.

7.Clinical Trials on Zhenjujiangya Tablets Combined with Long-acting Antihypertensive Drugs in the Treat-ment of Hypertension

To determine the efficacy and safety of Zhenjujiangya tablets combined with long-acting antihypertensive drugs in the patients with hypertension. Methods:A controlled study was carried out among 220 patients meeting the inclusion / ex-clusion criteria. All the patients were randomly divided into the experimental group and the control group. Both groups were treated with long-acting antihypertensive drugs as usual. The 110 patients in the experimental group were orally treated with Zhenjujiangya tab-lets ( bid) , while the other 110 patients in the control group were orally treated with Zhenjujiangya simulated tablets ( bid) , and the treatment course was 4 weeks. The efficacy was assessed using the reduction range of blood pressure, the clinical symptom score of hy-pertension and the indicators of ambulatory blood pressure monitoring as the indices, and the safety was assessed by the symptoms, signs and laboratory tests. Results:After the four-week treatment, the systolic blood pressure, diastolic blood pressure and hyperten-sion clinical symptom score were significantly decreased in the two groups, and the changes in the experimental group were more signifi-cant than those in the control group (P<0. 01). The effect on the clinical symptom of the experimental group was better than that of the control group (P<0. 01). There was no significant difference in systolic blood pressure, diastolic blood pressure and the number of abnormal blood pressure in 24h, daytime and nighttime (P>0. 05). After the treatment, the changes in average systolic blood pres-sure in 24h and daytime in the experimental group showed statistically significant difference (P<0.05),while those in the control group were without statistically significant difference (P>0. 05), and the number of abnormal blood pressure in 24h and daytime in the experimental group exhibited reduction trend. There were no significant difference in the incidence of adverse events between the two groups (P>0. 05). Conclusion:Long-acting antihypertensive drugs combined with Zhenjujiangya tablets can further improve the antihypertensive efficacy without significant adverse reactions, which is suitable for clinical application.
8.Fast edge extraction for ultrasound image of breast tumor based on fuzzy number.

An accurate edge extraction method for the ultrasound breast tumor image is useful for classifying tumors as benign or malignant. This paper refers to a fast technique to extract edge of breast tumor from ultrasound image. This method uses the triangular fuzzy number to build up a fuzzy number plane whose basic unit is the marching square. It is possible to visualize at once the results obtained using different presumption levels. Experiments of benign and malignant breast tumor in ultrasound images have shown that our method can extract the breast tumor edge faster than many conventional methods can do separately, and the results are reliable and credible. Our experiments demonstrate that it can be efficiently used to extract the edge of breast tumor from the ultrasound image.

9.Effects of Morroniside on Expression of Vascular Endothelial Growth Factor and Fibroblast Growth Factor-2 in Rats after Focal Cerebral Ischemia-reperfusion

Objective To investigate the effects of morroniside on the expression of vascular endothelial growth factor (VEGF) and fi-broblast growth factor-2 (FGF-2) in rat cortex after focal cerebral ischemia-reperfusion. Methods 30 male Sprague-Dawley rats were ran-domly divided into sham group, model group, morroniside-low group (30 mg/kg), morroniside-middle group (90 mg/kg) and morroni-side-high group (270 mg/kg). Middle cerebral arteries of rats were occluded for 30 minutes with Longa's method and re-perfused. The ex-pression of VEGF and FGF-2 in the ischemic ipsilateral cortex was detected with Western blotting 7 days after reperfusion. Results The ex-pression of both VEGF and FGF-2 increased in the ischemic ipsilateral cortexin in all the ischemic groups compared with the sham group (P<0.05). The expression of VEGF further increased in a dose-dependent manner in all the morroniside groups compared with that of model group (P<0.05), and the expression of FGF-2 increased in the morroniside-high group (P<0.001). Conclusion Morroniside could increase the expression of VEGF and FGF-2 after ischemia-reperfusion, which might promote angiogenesis.
10.Effects of Morroniside on Expression of CD34 in Ipsilateral Cortex of Focal Cerebral Ischemia-reperfusion Rats

Objective To explore the effects of morroniside on the expression of CD34 in ipsilateral cortex of rats after focal cerebral isch-emia-reperfusion. Methods 45 male Sprague-Dawley rats were divided into sham group (n=9), ischemia group (n=9), and morroniside groups (low, medium and high dosage groups, n=9). The middle cerebral artery were occluded for 30 minutes, and reperfused. Morroniside was administered intragastrically once a day at dose of 30 mg/kg, 90 mg/kg, 270 mg/kg after operation. The expression of CD34 in the isch-emic ipsilateral cortex were detected with immunohistochemistry (n=6) and Western blotting (n=3) 7 days after operation. Results The ex-pression of CD34 increased in the ischemia group compared with the sham group, and further increased in the morroniside groups of high dos-age compared with the ischemia group (F>14.865, P<0.001). Conclusion Morroniside could increase the expression of CD34 in the ischemic ipsilateral cortex after ischemia-reperfusion in rats, which may promote the angiogenesis and neurogenesis after ischemia.
