Objective To investigate mRNA and protein expression of aproliferation-inducing ligand (APRIL) in peripheral blood mononuclear cell and plasma of patients with B cells non-Hodgkin lymphoma (B-NHL) pre- or post- chemotherapy and to explore the role of APRIL in the B-NHL. Methods The mRNA and protein expression of APRIL were detected by real-time fluorescence quantitative polymerase chain reaction (RFQ-PCR) and enzyme-linked immunosorbent assay(ELISA), respectively. According to the standard curves, the quantitative levels of target mRNA and protein were determined. Results The detection linear range of targeted mRNA by RFQ-PCR was 101-109 pg/ml, and the coefficient of variation values for both intra- and inter-experimental reproducibility ranged 1.69 %-5.99 % and 6.35 %-10.12 %, respectively. To detect expression of APRIL protein by ELISA, the correlation coefficient of standard curves reached 0.9922. Before or after chemical treatment, the expression levels of APRIL mRNA and protein in patients with B-NHL were significantly higher than those in normal control (P <0.01), while the expression levels in post-treatment patients with Ⅲ and Ⅳ stage were lower than those of pre-treatment patients with corresponding stage (P <0.05, respectively), but those of pre- and post-treatment patients with Ⅰ / Ⅱ stage were not different (P >0.05).Conclusion The expression level of APRIL mRNA is similar to that of APRIL protein. APRIL may be involved in pathogenesis and development of B-NHL. Moreover, APRIL may be related to the burden of B-NHL and it may be considered as a targeted molecule for B-NHL.

Journal of Clinical Medicine in Practice 2014;(16):116-119
Objective To discuss curative effect of Ganglioside on acute intracerebral hem-orrhage (AICH)and its influence on serum interleukin-1β,IL-6 and IL-8 levels. Methods 76 pa-tients with AICH were divided into observation group and control group by random digital table method with 38 patients in each group. The patients in two groups were given routine medical treatment, such as reducing intracranial pressure and blood pressure,hemostasis,anti-infection, brain cells nutrition and so on.The patients in observation group were additionally given 60 mg Ganglioside by intravenous infusion for once a day, while the patients in control group were addi-tionally given 0.5 g Citicoline Sodium Injection by intravenous infusion, once a day for 10 days. The changes of serum IL-1β,IL-6 and IL-8 levels of patients in two groups before and after medi-cal treatment were observed, and the clinical curative effect and drug adverse reaction (DAR)were compared as well. Results After 10 days′medical treatment, the serum IL-1β,IL-6 and IL-8 lev-els of patients in two groups obviously declined than before (P <0.05 and P <0.01),and the de-clining rate in observation group was much higher than that in control group (P <0.05).The total clinical efficiency of patients in observation group was much higher than that in control group (94.74% vs.78.95%)(χ2=4.15,P <0.05).Adverse effect appeared in 2 and 4 patients in control group and observation group respectively during the medical treatment with light symptom and no serious DAR.After comparing the occurrence rates of adverse effect of patients in two groups, there was no statistical differences (χ2=0.18,P >0.05).Conclusion Ganglioside has favorable curative effect on AICH with high security, whose mechanism of action may inhibit lo-cal inflammatory response to protect the secondary brain injury after intracerebral hemorrhage by reducing the serum serum IL-1β,IL-6 and IL-8 levels.

According to the framework of role transition theory, the factors could be classified as individual factors (37 items, 23.72%), organizational factors (105 items, 67.31%), effectiveness of role definition (11 items, 7.05%), and problems of role definition (3 items, 1.92%).
Conclusions
Role definition in nursing handoff helping the coming nurse check the performance of the last shift and transfer the nursing responsibility, contributes to the improvement of nursing quality, but there are still many factors affecting the role definition in nursing handoff.

Objective:
To test the reliability and validity of the Chinese version of the Handoff Clinical Examination Exercise (Handoff CEX).
Methods:
The Chinese version of the Handoff CEX was composed of two parts, one to assess the nurse providing the handoff and another to assess the nurse receiving the handoff. A total of 80 handoffs was evaluated to test the reliability and validity of the Chinese version of the Handoff CEX.
Results:
A total of 320 evaluations of handoff was obtained. The Cronbach α coefficient was 0.85 for the handoff provider, and that was 0.80 for the handoff recipient. Weighted kappa scores for provider evaluations ranged from 0.31-0.52, and that for recipient evaluations ranged from 0.35-0.55. The correlation coefficients between each factor ranged from 0.23 to 0.62, and that between each factor and the total scale ranged from 0.33 to 0.64 (

Scaffold hopping refers to computer-aided screening for active compounds with different structures against the same receptor to enrich privileged scaffolds, which is a topic of high interest in organic and medicinal chemistry. However, most approaches cannot efficiently predict the potency level of candidates after scaffold hopping. Herein, we identified potent PDE5 inhibitors with a novel scaffold via a free energy perturbation (FEP)-guided scaffold-hopping strategy, and FEP shows great advantages to precisely predict the theoretical binding potencies ΔG FEP between ligands and their target, which were more consistent with the experimental binding potencies ΔG EXP (the mean absolute deviations
