1.The effects of sodium phenylbutyrate(SPB)on the expression of p21 and survivine genes on human tongue squamous cancer cell line Tca8113

Aim To investigate the effects of sodium phenylbutyrate on cell proliferation,apoptosis and its expression of p21 and survivin genes in human tongue squamous cancer Tca8113 cell line.Methods The cellular proliferation inhibitory ratio was evaluated by MTT assay and the apoptosis and cell cycle of the Tca8113 cell line was detected by FCM.The expression of p21 and survivin genes was analyzed with Western blot and RT-PCR.Results Sodium phenylbutyrate could inhibit the Tca8113 cells proliferation,promote cell apoptosis and arrest the cells at G_1/G_0 phase.The expression of p21 gene in Tca8113 cell line treated by sodium phenylbutyrate was increased,and one of survivin gene was decreased.Conclusions Sodium phenylbutyrate induces up-regulation of p21 gene and down-regulation of survivin gene,which inhibits Tca8113 cell proliferation and induces its apoptosis and arrests the cells at G_1/G_0 phase.And the increase of p21 mRNA expression is negatively correlated with the decrease of survivin mRNA expression(r_s=-0.548,P<0.01),and so is its protein expression(r_s=-0.514,P<0.01).
2.Expression of interleukin-7 mRNA and identification of its splice variant in human hepatic cancer

AIM: To investigate the expression of interleukin-7(IL-7) in human liver cancer and identify the splice variant of IL-7. METHODS: Reverse transcription-polymerase chain reaction (RT-PCR) was used for investigating the expression of IL-7. According to IL-7 gene cDNA, the primers were designed and synthesized, then the splice variant of IL-7 in human liver cancer cells was identified, cloned into vector and sequenced. RESULTS: The expression of IL-7 mRNA was equal between normal liver tissue and hepatic cancer cells.A new band of IL-7 was obtained from hepatic cancer cells, which lacks exon 4. CONCLUSION: IL-7 gene is expressed highly in both normal liver tissue and hepatic cancer cells. In addition, hepatic cancer cells can produce splicing variant by alternative splicing.

4.Brain Micro-structural Alterations of Cough Syrup Abuse Addiction Patients Containing Codeine Under Resting State

[Objective]To investigate the micro-structural alterations within whole brain white matter(WM) in cough syrup abuse addiction patients containing codeine,and to explore the correlation between aberrant WH and abuse time of cough medicine abuse patients.[Methods]Thirty cough syrup abuse addiction patients containing codeine and 30 controls participated in the study. Structural changes in FA and(mean diffusivity)MD were examined in cough syrup abuse addiction patients containing codeine which derived from DTI tractography. Pearson correlation analysis was performed to compare the mean FA value and duration of cough syrup abuse addiction in patients.[Results]Cough syrup abuse addiction patients containing codeine had lower FA value in bilateral anterior limb of internal capsule(ALIC)and higher MD in the bilateral hippocampus and insula,right anterior cingulate cortex(ACC)and superior temporal gyrus,compared to the controls. Cough syrup abuse addiction group also had positive correlation between mean FAvalues and duration of cough syrup abuse addiction in patients.[Conclusion]Micro-structural alterations within whole brain white matter(WM)are found in cough syrup abuse addiction patients containing codeine. This disturbance progresses as duration increases of cough syrup abuse addiction in patients.
