1.Brushing abrasion of eroded enamel using bioactive glass toothpaste in different time after acid etching

BACKGROUND:Bioactive glass has good biocompatibility, which can promote the remineralization effects of demineralized enamel and enhance the resistance of enamel to erosion. OBJECTIVE:To evaluate the effect of bioactive glass toothpaste versus fluoride toothpaste on brushing abrasion of enamel after acid etching. METHODS:Fifty-six enamel specimens were prepared from bovine incisors, and were embedded in acrylic resin with exposed buccal surface. The exposed area was 6 mm×6 mm and two amalgam fil ings were placed into the enamel and polished. Al specimens were divided into seven groups with eight specimens each. In six groups, specimens were brushed with ordinary toothpaste, fluoride toothpaste, bioactive glass toothpaste immediately and 30 minutes after being eroded by the Sprite. The last one group was brushed with ordinary toothpaste without acid etching as control. The treatment course was cycled 60 times. The surface abrasion loss of the specimens between two amalgam fil ings compared with respective amalgam reference surfaces was final y observed and calculated under laser scanning confocal microscope. RESULTS AND CONCLUSION:The abrasion loss amount of teeth with no acid etching was significantly lower than that with acid etching (P<0.05). The abrasion loss amount of teeth brushed with bioactive glass toothpaste was less than that with fluoride toothpaste at the same brushing time (P<0.05). For the same toothpaste, the abrasion loss amount of teeth brushed 30 minutes after eroded was less than that bushed instantly after eroded (P<0.05). Use of bioactive glass toothpaste can effectively reduce enamel loss caused by acid etching and brushing. The loss amount can also be reduced by brushing teeth 30 minutes after acid etching.
2.Effects of antonymous mutation of 17th condon of salivary HRP5 on its expression in Pichia pastoris

Objective To induce the the 17th condon antonymous mutagenesis of salivary histatin 5(HRP5) cDNA,express the mutant and hrp5 in Pichia pastoris,and to study the effects of mutation on expression.Methods According to the Pichia pastoris' codon bias,two pairs of primers(H1 and H2,H3 and H4) were designed.H1 and H2,H3 and H4 have complementary 3' end,and the EcoR I site was added to the 5' end of H1 and H3,Sal I site to H2,H4.The cDNA of hrp5 and hrp5' was generated with PCR by H1 and H2,H3 and H4,respectively.The secrete vector pPICZ?-A,hrp5 and hrp5' were digested with EcoR I +Sal I,linkede by T4 DNA ligase and transformed to E.coli TOP10 comptetent cell,positive colonies were screened on LB plates with Zeocin.The recombinant plasmids pPICZ?-A-hrp5 and pPICZ?-A-hrp5' identified by digestion and DNA sequencing were amplified largely,linearized by Sac I and transformed to GS115 comptetent cell by electroporation,positive colonies were screened on YEPD plates with Zeocin,the recombinant GS115 were confirmed by PCR,cultured and induced expression by methanol.The amount and anticandidal activity of the expressed products was compared with synthetic HRP5.Results Both hrp5 and hrp5' were integrated into the genome of GS115 and expressed successfully,the anticandidal activity of the recombinant HRP5 and HRP5' was identical with synthetic HRP5,the amount of expressed HRP5 and HRP5' was 4?mol/L and 5?mol/L,respectively.Conclusion Both the recombinant HRP5 and HRP5' showed better anticandidal activity.The amount of expressed prodcts increased 25% by substituting Asn for Lys17 without changing anticandidal activity.
3.Role of miR-122 in bone marrow derived stem cell therapy for acute liver injury in rats

Objective To investigatet the effects of miR-122 in the therapy of bone marrow derived stem cells (BMSCs)for acute liver injury in the rats,and to clarify the mechanism.Methods The BMSCs were isolated from the bone marrow of male rats by density gradient centrifugation.The BMSCs were divided into transfection group and control group.The BMSCs in transfection group were transfected with miR-122 mimics by liposome,while the BMSCs in control group were not.60 SD rats with acute liver injury induced by 10%CCl4 were randomly divided into control group (the saline was injected through mainline),normal treatment group (the normal BMSCs were injected through mainline) and experimental therapy group (the BMSCs transfected with miR-122 mimics by liposome were injected through mainline)(n=20).The liver function and tissue pathology were examined at 1 d, 7 d and 14 d after transplantation.Results The expression level of ALB in BMSCs was up-regulated,while the AFP expression level was down-regulated after the transfection of miR-122 mimics.At 1 d after transfection of BMSCs,the serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities had no significant difference between normal treatment group and experimental therapy group.At 7 d and 14 d after transfection of BMSCs,the serum ALT and AST activities in experimental therapy group were obviously lower than those in normal treatment group (P <0.05).The liver congestion,cytoplasm degeneration and liver cell necrosis in experimental therapy group were improved compared with normal treatment group.Conclusion The up-regulation of miR-122 expression in BMSCs would promote its differentiation into hepatocyte like cells,which plays a role in promoting the recovery of liver injury.

5.The study of focal mild hypothermia on prolonging the therapeutic time window after cerebral infarct in rats

Objective To study whether focal mild hypothermia can prolong the therapeutic time window in ischemic stroke, and inquire into the protection mechanisms of focal mild hypothermia on cerebral ischemia. Methods The focal cerebral ischemia and reperfusion model of rats was achieved by intraluminal middle cerebral artery occlusion (MCAO). The adult rats were randomly divided into sham-operation group, normothermia ischemia and reperfusion group and hypothermia ischemia and reperfusion group. The two ischemic groups were respectively further divided into 5 subgroups (subjected ischemia for 2, 3, 6, 8 and 12 h respectively and then all reperfused for 4 h, n=14 per time point). Brain mild hypothermia was achieved 30 minutes after ischemia and maintained to reperfusion period. The neurological deficit scores were assessed. The infarct volumes were evaluated by TTC staining and measured with a computerized image analysis system. TUNEL method was used to observe the neuron apoptosis. The expressions of NF-?Bp65 and NF-?BP65mRNA were detected by immunohistochemistry and hybridation in situ. Results The infarct volumes of hypothermia group were respectively (57.39?37.62) mm~3, (74.09?61.00) mm~3, (81.26?25.46) mm~3, (87.43?54.81) mm~3 and (111.10?43.67) mm~3, which saw significant reductions of 63%, 55%, 56%, 70% and 63% as compared with the normothermia group. 6.Effect of Xiaoying Decoction on Changes of Regulatory T Cells and Th17 Cells in Experimental Autoimmune Thyroiditis Rats

Objective To explore the mechanism of Xiaoying decoction on experimental autoimmune thyroiditis (EAT) rats in view of regulatory T cells and Th17 cells.Methods SD rats were divided into five groups,normal control group,model control group,tripterygium glycoside group,Xiaoying decoction low and high dose groups.Except for normal control group,the other groups were established the model of EAT.Rats in the Xiaoying decoction low,and high dose groups were given Xiaoying decoction of 17.24 and 68.95 g·kg-1;rats in the tripterygium glycoside group were given tripterygium glycoside 6.25 mg·kg-1.The serum free triiodothyronine(FT3),free thyrocyte(FT4) and thyroglobulin antibody were detected by RIA method.Thyrocyte morphology was observed under optical microscope.The expression levels of Foxp3 mRNA and IL-17 mRNA were detected by real-time PCR.The changes of Treg cells and Th17 cells were analyzed by flow cytometry.Results Compared with the normal control group,FT3,FT4 and TgAb were increased in the model control group (P <0.01,P <0.01,P <0.05).Compared with the model control group,FT3,FT4 and TgAb were decreased in tripterygium glycoside group and Xiaoying decoction high dose group (P < 0.05).The infiltration score in the normal control group,model control group,tripterygium glycoside group,Xiaoying decoction low and high dose groups were 0,4,4,3.5,2.Compared with model control group,the infiltration was improved (P <0.01).Foxp3 mRNA expression was decreased while IL-17 mRNA increased in the model control group as compared with the normal control group(P < 0.01).In contrast,the expression of Foxp3 mRNA was increased and IL-17 mRNA expression was decreased in Xiaoying decoction low and high dose groups,tripterygium glycoside group as compared with the model control group (P <0.05).Compared with the normal control group,rats in the model control group had fewer Treg cells and more Th17 cells (P <0.01).Compared with the model control group,the percentage of Treg cells was elevated and Th17 cells was reduced in tripterygium glycoside group and Xiaoying decoction high dose groups (P < 0.01).Conclusion The therapeutic mechanism of Xiaoying decoction on EAT rats may be related to changing the percentage of regulatory T cells and Th17 cells with up-regulating the expression of Foxp3 mRNA and down-regulating the expression of IL17mRNA.

9.Treatment of cervical whiplash injuries: a report of 88 cases

Objective To compare the efficacy of operation and non-operatlon in treatment of cervical whiplash injuries. Methods A retrospective sturly was carried out in 88 cases of cervical whiplash injuries, of whom 24 cases were treated with non-operation and 64 with operation. There were 44 cases treated via anterior approach and 20 via posterior approach. NNeural function was evaluated by using Frankh classifications and ASIA (American Spinal Injury Association) grades. Results All cases were followed up for 6-24 months, which showed that neural function was improved significantly in both operation and non-operation groups. However, the neural function was better improved in the operation group compared with the non-operation, with statistical difference. The anterior approach group had significantly better outcome in neural function than the posterior approach group (P<0.05). Conclusion Operation is a better method and anterior approach should be the choice of treatment for treatment of cervical whiplash injuries.
10.Application of factor analysis to evaluate deformation behaviors of frequently-used tablet excipients.

The present work is to investigate the correlation between physical properties and deformation behaviors of tablet excipients, and rank them according to their plastic performances during compaction. The excipients selected were compacted using Korsch XP1 after measuring their physical properties where the compression parameters for evaluating deformation behaviors were Heckle equation, compression work and elastic stretch in die. The correlations between compaction descriptors and physical parameters were analyzed by canonical correlation analysis, and factor analysis was simultaneously employed to synthetically assess deformation behaviors for all our samples. The canonical variables show that true density (Pa) correlated negatively with plastic coefficient (PL) and positively with yield pressure (YP); compression degree (Cp) correlated negatively with fast elastic stretch (FES) as well as YP and positively with PL. When factor scores were used in combination with original data, the plasticity of our samples was sorted and ranked as high (-0.56 < F' < 0.21), intermediate (-0.16 < F' < 0.36), or low (0.38 < F' < 0.84), which are in accord with plasticity rankings previously reported in literature. This study indicates factor analysis can be an approach to evaluate deformation behaviors of pharmaceutical powders.
