1.Investigation of Phthalates in Disposable Plastic Products

Objective To explore the kinds and levels of phthalates leaching from disposable plastic products. Methods Samples of peritoneal dialysis solution, blood preservative solution, infusion instruments, preservative film, disposable plastic bags and water in plastic bottles were analyzed for phthalates by RP-HPLC after liquid-liquid extraction and/ or solid phase extraction. Results Di(2-ethylhexyl) phthalate (DEHP) was leached from all medical instruments, the maximum level of which reached 77.51?g/ L. Di-n-butyl phthalate was leached from disposable plastic bags, the level of which reached 91.45?g/ kg. Phthalates were not found in samples of preservative film and water in plastic bottles. Conclusion As DEHP leaching from the medical instruments might directly enter the human body, attention should be paid to its health hazards.
2.Toxicokinetics of Phthalates in Male Rabbits

Objective To study the toxicokinetics of phthalates in male rabbits. Methods Concentrations of DEHP or DBP in plasma of 6 healthy male rabbits were determined by RP-HPLC after constant rate infusion of 0.5 g/kg DEHP or DBP(IV). The toxicokinetic parameters were computed by 3P87 program. Results The toxicokinetic model of DEHP and DBP were both first-order elimination and two-compartment model with constant rate infusion. The main toxicokinetic parameters of DEHP were as follows: distribution phose t1/2(?)=0.101 h; elimination phase t1/2(?)=12.701 h; CLs=0.013 g?kg-1?h-1. The main toxicokinetic parameters of DBP were as follows: t1/2(?)=0.441 h; t1/2(?)=31.311 h; CLs=0.021 g?kg-1?h-1. Conclusion DEHP and DBP were both first order elmination and two-compartment medol with constand rate intusion(IV). DEHP and DBP could be rapidly eliminated in male rabbits.
3.Radiofrequency ablation therapy for breast cancer:case report and reference review

Objective To evaluate the effect of ultrasound guided percutaneous radiofrequent ablation treatment on breast cancer .Methods Two patients,were pathologically confirmed as breast cancer ,underwent percutaneous ultrasound guided radiofrequency ablation of breast lesions ,followed with radical excision of breast within 3 months after the ablation .Postoperative pathological examination was conducted .Patients were followed up for over 12 months by now.Results The postoperative pathology of case No .1,in which the original breast le-sion was within 2 cm in diameter,found no tumor cells left but filled with inflammatory cells instead .Case No.2 with the longer diameter as 27 mm showed majority necrotic tissues but the tissue around left with some ductal carcinoma in-situ components .One-year′s follow up found no local recurrence or distant metastasis of tumor . Conclusion Radiofrequency ablation is a promising therapy with minimally invasion and better outlooking for early breast cancer within 2 cm in diameter .
4.Role of stroma cell-derived factor-1 receptor in utero transplantation with hematopoietic stem/progenitor cells

Objective To explore the effect of stroma cell-derived factor receptor CXCR4 on the homing of the hematopoietic stem/progenitor cells in utero transplantation. Methods CD34+ cells were collected by Ficoll density gradient centrifugation and MiniMACS and then stimulated for 48 h by SCF and IL-6 cytokines prior to transplantation. CD184(CXCR4) expressions and transmigrate rates of the CD34+ cells were analysed by flow cytometer. Cells pretreated with different treatment were transplanted into the abdominal cavity of the fetal BALB/c mouse in the pregnant days 13~14. Human CD45 cells as the marker of graft were detected by flow cytometry after 1 month the fetus born. Results Expression changes of CD184 on CD34+ cells were from 9.58%?1.56% to 19.32%?3.64% after SCF and IL-6 stimulation. The CD34+/CXCR4high cells exhibited significant increases in SDF-1 mediated chemotaxis compared with the CD34+/CXCR4low cells. Transmigration of CD34+/CXCR4high was inhibited by pretreatment with antiCXCR4mAb and PTX. The positive rates of human CD45 cells detected in the fetal mouse were significantly higher in the SCF and IL-6 pretreatment group. This effects were significantly abrogated after the addition of antiCXCR4mAbor PTX. Conclusion Up-regulation of CXCR4 expression may be useful for improving hematopoietic stem/progenitor cells homing in utero transplantation. This homing process is mediated and depends on the CXCR4 receptors. The signal transduction is mediated by PTX-sensitive Gi protein.
5.Apoptin Inducing Apoptosis in the Multidrug-Resistant Model of Human Osteosarcoma Cell Line

Objective: To investigate the curative effects of apoptin on the multidrug-resistant model, of human osteosarcoma cell line (R-OS-732). Methods: 363 bpVP 3 gene was cloned into the vector pIRES1 and the recombinant eukaryon expression vector pIRVP3 was transfected into R-OS-732 cells with liposome. The expression of apoptin gene at transcription level was proved by RT-PCR. The pathological changes of the cells was observed by light microscope and electronmicroscope. The transfected R-OS-732 cells were collected after 48 hours. The genomic DNA extracted from the cells was observed by agarose gel electrophoresis. The apoptosis rate of the cells was analysed by flow cytometry. Results: The expression of apoptin gene at transcription level had been proved by RT-PCR. The construction changes of the two groups were obviously different under light microscope: most of R-OS-732 cells in the transfected group existed in form of being away from the bottom of the culture dish after 24 hours, in form of apoptosis after 48 hours and in form of necrosis after 72 hours; but those cells in the controlled group grow luxuriantly. And under electronmicroscope there was much change of cell nucleus between the two groups. DNA ladder of the transfected cells was observed through agarose gel electrophoresis only one band was observed in the controlled group; but bands of fragmented DNA observed in ositive control group. 7.Establishment of cerebrocardiac syndrome model in rats

Aim To establish cerebrocardiac syndrome model in rats. Methods Right middle cerebral artery of rat was occluded (MCAO) with nylon thread and Ⅱ lead ECG was monitored continuously within 2 h to record the emerging of arrhythmias. 30 min, 2 h, 24 h after middle cerebral artery occlusion or 6 h reperfusion after 2 h occlusion, left ventricular myocardiums were observed with transmission electron microscope to find the morphological damage and brains were marked with TTC to locate infarction area. Results (14.9?11.4) minutes after MCAO, 71%(60/85)model rats had abnormal ECG changes including 38 %(32/85)premature ventricular contraction, 27 %(23/85)atrial premature beats, 5%(4/85)ventricular tachycardia, 1.2%(1/85)sinus tachycardia. Arrhythmias sustained (29.0?23.2 )minutes. Ventricular myocardial cell injury was obvious: chaotic and broken mitochondria ridges, aggregated chromatin under the nucleus membrane, deposited glycogen granules in the cytoplasma. 30 min, 2h, 24h after MCAO the infarction rates were 8.7%?1.1%,11.4%?2.3% and 13.7%?3.1% respectively. Conclusion A stable cerebrocardiac syndrome model in rat might be induced by means of occluding right middle cerebral artery and the morphological bases of cerebrogenic cardiac arrhythmias are myocardial cell injury caused by cerebral infarction.
8.Changes in Vasoreactivity of Rat Pulmonary Artery after 7 d Tail-suspension

Objective To clarify the effects of simulated microgravity on the local regulating function of the pulmonary circulation. Method Rat tail-suspension(TS) was used as the animal model to simulate the effects of microgravity.The changes in responses of pulmonary artery to several vasoactive agents were determined by using in vitro vessel rings perfusion technique. Result As for the endothelium-intact vessel rings from TS group compared with those from the control(C) group,the contractile responses of pulmonary artery to potassium chloride (KCl,10~70 mmol/L) did not change obviously after 7 d tail-suspension,which were markedly diminished as to phenylephrine (PE,10-9~10-5mol/L).The vasodilatory responses to acetylcholine (ACh,10-9~10-5mol/L) were markedly enhanced,which did not change obviously as to captopril (Cap,10-5~3×10-3 mol/L).The vasoreactivity of endothelium-removed pulmonary artery rings to all vasoactive agents did not change after 7 d tail-suspension as compared with those from control rats. Conclusion The release of nitric oxide (NO) by rat pulmonary arterial endothelium was enhanced after 7 d tail-suspension.The vasodilatory function of pulmonary artery was enhanced.

10.Combined supracondylar dome osteotomy of femur with arthroscopy in the treatment of the adult valgus deformity of knees

Objective To investigate the method and efficiency of supracondylar dome osteotomy of femur combined with arthroscopy for the treatment of the adult valgus deformity of knees. Methods From September 2008 to May 2012, 32 patients (35 knees) with valgus deformity of knees were treated with supracondylar dome osteotomy of femur combined with arthroscopy. The study group included 7 male and 25 female (average age: 43 years). The pre-operative HSS knee score was an average of 60.46. The weight-bearing knee joint AP & LAT and the full-length AP view of the lower extremity were examined before and after operation. At the same time , the femoral angle and femoral-tibial angle were measured and evaluated. The internal fixation was taken out in an average in 12 months. All the patients were followed-up and the change of the correction angles , the function of the knee and the condition of bone healing was recorded. Result The follow-up time was an average of 24 months. All the patients got a satisfied correction of the deformity, good bone union and obvious improvement of joint function. The postoperative HSS knee score was an average of 85.40. Conclusion The supracondylar dome osteotomy of femur combined with arthroscopy is a safe, reliable and effective strategy for the treatment of the adult valgus deformity of knees.
