OBJECTIVE:To establish an HPLC method for determination of ganciclovir in serum of dogs and to study pharmacokinetics and relative bioavailability of Ganciclovir dispersible tablets.METHODS:The determination was performed on Dikma DiamonsilTM C18 column.The mobile phase consisted of 0.015 mol?mL-1 monobasic potassium phosphate buffer solution (pH 2.50,containing 0.25% acetonitrile) with flow rate of 1.75 mL?min-1 at detection wavelength of 254 nm.The column temperature was set at 40 ℃.In a randomized cross-over study,a single dose of dispersible tablets and the capsule were given to 6 dogs.An HPLC method was used to determine plasma concentration of ganciclovir in dogs and the pharmacokinetic data were treated by DASver1.0 software.RESULTS:The linear range of ganciclovir were 0.050 25~100.5 ?g?mL-1;the methodological recovery was larger than 90% (RSD

OBJECTIVE:To study the anti-inflammatory effect of ethanol extract of Polygonum multiflorum METHODS:To observe the oedema severity and vaso-permeability in inflamed animal and the response of animal to pain induced by acetate acid after the ethanol extract of Polygonum multiflorum had been intragastrically given for 3 days RESULTS:The oedema severity and vaso-permeability were obviously decreased by the ethanol extract of Polygonum multiflorum and this effect could last 4 hours At the large dose,analgesic effect was observed CONCLUSION:The ethanol extract of Polygonum multiflorum exhibits intensive anti-inflammation effect and the mechanism may be related to its immuno-depression effect
5.The protective effects of quercetin et al on the intestinal mucosal injury induced by burn in mice

AIM To investigate the protective effects of three flavonoids quercetin,rutoside and flavone on the intestinal mucosal injury induced by burn in mice. METHOD The change of injury index of small intestine was assayed and the content of protein and DNA were measured. DTNB method were adopted to measure the GSH content in the intestinal mucosa. The level of DNA damaged were determined by measuring the fluorescence. RESULTS The condition of injury of intestinal mucosa were improved in varying degrees after these favonoids were given introgastrically for 3 days. The injury index of intestinal mucosa decreased and the content of protein,DNA and GSH increased. The degrees of DNA damage were reduced also.Quercetin was the most effective one among these three flavonoids. CONCLUSION The flavonoids of quercetin, rutoside and flavone have obvious protective effects on burn induced intestinal mucosal injury, and the mechanism may be related to its antioxydation function.
6.Determination of the Plasma Concentration of Dexketoprofen with HPLC in Rabbits

OBJECTIVE:To establish a HPLC method for determining the concentration of dexketoprofen in rabbit’s pla_ sma.METHODS:Dexketoprofen was extracted from samples with ether.HPLC was performed on C 18 column with methyl al?cohol-50mmol/L sodium dihydrogen phosphate as mobile phase.Detection wavelength was260nm.RESULTS:The linear range was0.05~25?g/ml.Recovery was99.92%~100.54%.Within-day and between-day RSDs were0.56%~2.94%and0.6%~2.89%respectively.CONCLUSION:This method is simple,accurate and good in repetitiveness.Satisfactory results have been obtained in determining plasma concentrations of dexketoprofen in6rabbits after taking dexketoprofen-?-cy?clodextrin inclusion microspheres.
7.Effects of quercetin on metabolism of cyclooxygenase in IEC-6 cells stimulated by the serum of burned rats

Objective To observe the effects of quercetin on PGE 2 release and cyclooxygenase activity in IEC 6 cells stimulated by the serum of burned rats. Methods The PGE 2 levels were measured by radioimmunoassay and the changes of cyclooxygenase activity were determined by substrate fluorescence analysis. Results Stimulated by the serum of burned rats, the PGE 2 level increased first and then obviously decreased. Quercetin at 0.1 and 1.0 ?mol/L enhanced the PGE 2 level after the stimulation of IEC 6 cells for 24 h. There was no obvious change in total cyclooxygenase activity, but the activity ratio of COX 1 to COX 2 significantly decreased. Conclusion Quercetin might influence the release of cyclooxygenase metabolite PGE 2 through the regulation of COXs activity in IEC 6 cells.
8.Preparation of slowly released microspheres of dextro-ketoprofen-?-cyclodextrin inclusion

Objective To prepare microspheres of dextro ketoprofen ? cyclodextrin (S KP ? CD) for prolonging the drug releasing time in vitro . Methods Microspheres containing S KP ? CD were prepared by complex coacervation method with gelatin and acacia. Trap efficiency, drug loading, and drug content were determined. The dissolubility of S KP in the intestinal liquid was compared with that in the gastric liquid. Results In comparison with S KP, microspheres of S KP ? CD possessed slow releasing property. Conclusion Drug microspheres prepared by this simple, easy, and accurate method are of slow releasing property.
9.Determination of Residual Organic Solvents in Clofarabine by GC

OBJECTIVE:To establish a gas chromatography(GC)method for determination of seven residual solvents in clofarabine.METHODS:The capillary column was AT-1301;the carrier gas was nitrogen gas and the detector was FID.By temperature programming was applied for the column temperature with an initial temperature of 40 ℃,which rose to 80 ℃ with an increase rate of 7 ℃?min-1,then rose to 200 ℃(kept at this temperature for 2 min)with the increase rate of 20 ℃?min-1.The injector temperature was 250 ℃ and the detector temperature was 280 ℃.The sample was injected directly for determination of residual levels of methanol,acetonitrile,dichlormethane,t-butyl alcohol,ethyl acetate,n-heptane and acetic acid in 3 batches of clofarabine.RESULTS:All the organic solvents were effectively separated and they showed good linearity within a concentration range(r=0.999 41~0.999 93)with average recovery rate ranged from 96.5% to 102.4%(RSDs of all were less than 4.0%).7 kinds of organic solvents in 3 batches of samples were all up to the standard specified in Chinese Pharmacopeia.CONCLUSION:The method is sensitive,accurate and reliable,and it is applicable for the determination of the residual solvents in clofarabine.
