ATM: To prepare Shenfuqing Lyophilized Powder (Radix Ginsenp Rubra, Pericarpium Citri Reticulatae Viride, ect.) METHODS:Shenfuqing Lyophilized Powder was made by freeze drying. The concentration of Shenfuqing solution and the varieties and quantities of filling material were selected as well as the stability of Shenfuqing solution and powder for injection were examined primarily. RESULTS: The better prescription is 10% mannitol and 6% drug components. CONCLUSION: The quality of Shenfuqing Lyophilized Powder is more stable than that of solution for injection.

AIM: To study extraction process with the aid of cyclodextrin (EPAC) of Radix Salvias Miltiorrhizae. METHODS: Taking tanshinone Ⅱ_A, Danshensu, dry extract as the indexes, EPAC and water extraction process(WEP) were compared. RESULTS:The index of tanshinone Ⅱ_A of EPAC was better than WEP in extract, but there was not obvious difference in the indexes of Danshensu and dry extract. CONCLUSION:EPAC is worth researching further.

AIM: To use the increasing rate of hygroscopic weight added to represent the mechanical properties of microcapsules, and to study the influence of excipients on the mechanical properties of microcapsules. METHODS: To develop a method of processing the microcapsules, which simulates the rubbing, shearing effect on the microcapsules during manufacturing, and calculate the increasing rate of hygroscopic weight added to study the influence of excipients on the mechanical properties. RESULTS: The excipients could affect the mechanical properties of microcapsules. CONCLUSION: The mechanical properties of microcapsules is the important quality standard, We can use it for selecting excipients used for microcapsulation.

Objective To optimize the technique of isolating and extracting oligosaccharide from Radix Rehmanniae.Methods The optimum conditions for the water extracting technique of Radix Rehmanniae were studied by orthogonal test design.Then the water extract was isolated by macroporous resins and active charcoal successively,meanwhile the pattern of macroporous resins,the volume of the eluents,and the decolorizing methods of active charcoal were studied.And then the oligosaccharide was obtained after being purified by Sephadex gel column chromatography.The content of stachyose was determined by HPLC.Results The optimal preparation process was as follows: 12 fold water,three times,0.5 h for each time.Then the water extract was isolated and purified by D-101 macroporous resins,0.1% active charcoal and Sephadex G15 column chromatography were successively used to obtain the oligosaccharide.The analysis of oligosaccharide indicated its main constituents are stachyose,rafinose,and manninotriose,and the content of stachyose is more than 60%.Conclusion This preparation technique of extracting oligosaccharide from Radix Rehmanniae is reasonable and feasible.And the yield of the oligosaccharide from Radix Rehmanniae achieves to 26%.

Objective To induce the site-directed mutation of human cardiac troponin I (cTnI) gene, express the mutant in E. coli, and to study the effects of the mutation on the prokaryotic expression of cTnI. Methods The cDNA encoding cTnI was cloned with RT-PCR from the total RNA extracted from human myocardium tissues. A pair of primers was designed and, after the mutations were induced at the second and the fourth codons, inserted into prokaryotic vector pET-28c (+) and transform the recombinant to BL21 (DE3) bacteria. After purified with Ni-NTA resin, the histidine-tagged fusion protein expressed by IPTG-induced was identified by Western blotting and the expression yield of cTnI protein was investigated. Results The expression of the recombinant carrying processed cTnI cDNA was stronger than that in control group. Conclusion cDNA encoding cTnI was successfully cloned. The recombinant with mutations can be more efficient expressed in E. coli. The cTnI protein can be purified to near homogeneity.

Objective To establish HPLC method to determine the content and the recovery of rosmarinic acid in common selfheal fruit-spike effective component of colon target tablets.Methods HPLC column was Diamonsil C18,200 mm×4.6 mm,5μm, wavelength was 330 nm, mobile phase methanol was 0.1% formic acid aqueous solution (45:55), flow rate was 1.0 ml/min, and the sample size was 20μl.Results The fine linear relation was shown at the rosmarinic acid methanol concentration of 2.99-119.6μg/ml, with 72 h in stable state. The method has fine precision, the spotting recovery was 95%-105%, andRSD was less than 2%. Conclusion The method has high specificity, meeting the demands for content determination.

It is nonlinear relationship between breath sounds and respiratory flow rate, thus breath sounds might be a new solution for respiratory monitoring. The envelope of the breath sounds was created firstly. And the linear relationship between the logarithm of envelope and the respiratory flow rate was proved. Then model parameters were derived from every subject using regression analysis. Finally, using these parameters estimated respiratory flow rate was achieved by the logarithm of envelope with 14.9% error. Therefore, respiratory flow rate estimation and respiration monitoring based on breath sounds are feasible.

Objective To evaluate the effect of ultrasound guided percutaneous radiofrequent ablation treatment on breast cancer .Methods Two patients,were pathologically confirmed as breast cancer ,underwent percutaneous ultrasound guided radiofrequency ablation of breast lesions ,followed with radical excision of breast within 3 months after the ablation .Postoperative pathological examination was conducted .Patients were followed up for over 12 months by now.Results The postoperative pathology of case No .1,in which the original breast le-sion was within 2 cm in diameter,found no tumor cells left but filled with inflammatory cells instead .Case No.2 with the longer diameter as 27 mm showed majority necrotic tissues but the tissue around left with some ductal carcinoma in-situ components .One-year′s follow up found no local recurrence or distant metastasis of tumor . Conclusion Radiofrequency ablation is a promising therapy with minimally invasion and better outlooking for early breast cancer within 2 cm in diameter .

Objective To observe the expression changes of heme oxygenase-1(HO-1) and its mRNA after spinal cord injury (SCI). Methods SD rats were randomly divided into normal (n=5), control (n=5) and experimental group (n=10). The rats were given no operation in the normal group, laminectomy only in the control and both laminectomy and traumatic injury in the experimental group. The SCI animal models were made by using a modified Allen's weight-drop device (50 g.cm) on T 12 . Rats were sacrificed at 1 and 3 days after injury. The distribution and content changes of HO-1 and its mRNA in spinal cord segments were observed by means of immunohistochemistry and in situ hybridization. Results Expression of HO-1 was mainly in neurons in the normal spinal cord. After injury, HO-1 expressed primarily in astrocytes and microglia and the distribution of positive cells closely correlated with intraparenchymal hemorrhage. Protein began to increase at the first day after injury. The expression intensity and the area of positive cells were (148.26?11.39) and (90.50?8.70)?10 3 ?m 2 respectively. At the third day, protein expressed more significantly with expression intensity of (128.03? 12.59) and area of (112.99?10.01)?10 3 ?m 2. The expression intensity and area of positive cells expressed by mRNA were (106.02?9.10) and (70.05?9.26)?10 3 ?m 2 respectively at the first day after injury, (85.82?9.07) and (87.37?10.95)?10 3 ?m 2 respectively at the third day after injury. Objective To investigate the effects of p53 protein and mRNA expression in rat liver tumor promotion by phenobarbital(PB). Methods Male Wistar rats were randomly divided into 6 groups,i. e. higher dose group,middle dose group,lower dose group,tumor-initiating control group,tumor-promoting group and normal control group. The rat liver tumor DEN-initiating-PB-promoting model was established among higher dose group,middle dose group,lower dose group and tumor-initiating control group. The rats in higher dose group,middle dose group and lower dose group were fed with feed containing 500,100,50 mg/kg PB respectively. The rats in liver-tumor promoting control group were only fed with normal feed. The rats in tumor-promoting group weren't initiated by DEN,were only fed with feed containing 500 mg/kg PB. The rats in normal control groups weren't treated by any factors. At the 1st,5th,10th,15th,20th,30th week of the exposure to PB,the expression of p53 protein and p53 mRNA of the rats in every group were determined by immunohistochemistry and in situ hybridization respectively. Results The expression of the mutant type p53(mtp53) protein was found in liver tumor-initiated rats which revealed precancerous lesion. The expression of p53 protein of rats increased in higher and middle dose groups,and showed higher levels in lower dose group and liver tumor-initiating control group compared with those of normal control group which didn't variate significantly with the prolongation of period of exposure to PB. The expression of wild type p53 (wtp53) mRNA showed lower levels in rats of normal control group and liver tumor-promoting group,showed higher levels in higher dose group,middle dose group,lower dose group and liver tumor-initiating control group compared with those in normal control group. The expression of wtp53 mRNA decreased in higher dose group and middle dose group,increased a little in lower dose group and liver-tumor-initiating with the prolongation of period of exposure to PB. Conclusion During the promoting stage of rat liver tumorigenesis tumor promotor PB might reduce the expression of wtp53 and induce mtp53 expression,which affected the cell cycle arrest and apoptosis,and might favor clonal expansion of preneoplastic hepatocytes,which promoted the formation of liver tumor.
