1.Feasibility of TCI etomidate for total intravenous anesthesia

Objective To study the feasibility of TCI etomidate for total intravenous anesthesia (TIVA). Methods Forty patients scheduled for abdominal operation were divided into two groups with 20 cases each. Anesthesia in group E was induced and maintained with TCI etomidate 1-2 μg/ml and remifentanil 6 ng/ml,which in group P with TCI propofol 2-4 μg/ml and remifentanil 6 ng/ml to keep BIS 40-60. The perioperative changes of plasma glucose, cortisol, aldosterone and ACTH were Observed. Recovery from anethesia was recorded as well Results Both groups had the stabilized hemodynamics and glucose concentration. The plasma cortisol and ACTH concentrations in group E were decreased at the end of operation(P<0.05), but returned to preoperative level on the next day, which were not significantly changed perioperatively in group P. The time of eye opening was longer in group E than that in group P. Conclusion Etomidate has an inhibition on the function of the adrenal cortex temporally and can be used safely togather with remifentanil for TIVA in patients without hypoadrenocorticism.
2.Effects of Crizotinib on Proliferation of Human Ovarian Cancer Cell Line SKOV3

Objective To explore the expression of anaplastic lymphoma kinase(ALK) in human ovarian serous adenocarcinoma cell line SKOV3 and to further investigate the effect of crizotinib on SKOV3 cells.Methods The expression of ALK in SKOV3 cells were examined by Western blotting and immunocytochemical methods,and the effect of ALK inhibitor crizotinib on proliferation of SKOV3 cells were evaluated by cell proliferation assay.Results Both Western blotting and immunocytochemical methods showed the expression of ALK in SKOV3 cells.The results of cell proliferation assay suggested that the cell viability of SKOV3 cells was close to 1 when the drug concentration was less than 106 nmol·L-1,and close to 64% when the drug concentration was up to 107nmol·L-1.Conclusion ALK is overexpressed in human ovarian serous adenocarcinoma cell line SKOV3,but SKOV3 cells were insensitive to the therapy of ALK inhibitor crizotinib.

Objective To investigate the effect of chronic electrical stimulus (CES) of low frequency and physiological frequency on diaphragmatic subtype of myosin heavy chain (MHC) and mechanical characteristic in rabbits with emphysema. Methods The rabbit model of emphysema was made by inhaling papain of ultrasonic atomization once a week for 3 weeks in 30 adult Japanese rabbits, 6 receiving 10 Hz chronic low-frequency electrical stimulation, 6 receiving 40 Hz, 6 receiving 10 Hz plus 40 Hz. Another 6 emphysema rabbits and 6 normal rabbits served as control. The twitch tension (Pt), titanic tension (Po), time to peak tension (TPT), half-relaxation time (1/2Rt), fatigue index (FI) and fatigue recovery index (FRI) were measured and SDS-polyacrylamide gel electrophoresis was used to observe the changes in subtype of MHC in control group, emphysema group, CES groups respectively. Results ①Following chronic electrical stimulus of 40 Hz, 10 Hz, and (10+40) Hz, the relative contents of MHC were increased in diaphragmatic muscle of rabbits with emphysema. The percentage of MHC-I was significantly increased in the 10 Hz group, while that of MHC-I and MHC-IIa were significantly increased in the (10+40) Hz group. ②Following chronic electrical stimulation using 40 Hz and (10+40)Hz, diaphragmatic muscle Pt and Po were significantly increased in rabbits with emphysema, TPT and 1/2Rt were significantly reduced, and FI and FRI were significantly decreased (P
6.Induction of L1 and L2 ?-lactamase by common antimicrobial agents and their modulation

Objective To compare the induction of L1 and L2 ?-lactamase stenotraphomonas maltoptilia by common antimicrobial agents, including imipenem, meropenem, cefotaxime and ceftazidime, and to survey the modulation of L1 and L2 ?-lactamase expression. Methods One clinical strain of S. maltophilia was isolated and identified with VITEK automatic microbic system. L1 and L2 ?-lactamase genes were amplified, cloned and sequenced by PCR method. Minimal inhibitory concentrations (MICs) of four antimicrobial agents against the clinical isolates were determined by agar dilution method. Two hours after being induced by different concentrations of four antimicrobial agents, total RNA was extracted, and RT-PCR method was used to determine the induction of L1 and L2 ?-lactamase by different concentrations (0.25, 1 or 4?MIC) of common antimicrobial agents. Electrophoresis strips of L1 and L2 ?-lactamase were quantified by Image J software. Results The clinical isolates of S. maltophilia with simultaneous production of L1 and L2 ?-lactamse were identified. When different concentrations of four antimicrobial agents were used as inductors, electrophoresis strips of L1 and L2 amplicons were not found in strains of blank control and those in which imipenem, meropenem or cefotaxime (4?MIC) was added to the culture mediam, while light electrophoresis strips were exhibited by the isolates with ceftazidime (0.25, 1 or 4?MIC) or cefotaxime (0.25?MIC) added to the medium. The strongest electrophoresis strips and the strongest expression were found in the isolates with cefotaxime (1?MIC) added to the medium. Conclusions Clinical common antimicrobial agents, e.g. ceftazidime and cefotaxime, are able to induce production of L1 and L2 ?-lactamase, and cefotaxime (1?MIC) is the strongest inductor. Cefotaxime can exert an effect on transcription of L1, L2 genes simultaneously, implying that a significant overlap might exist between the mechanism of modulation of two ?-lactamases.

8.Evaluation of left ventricular rotation and twist in patients with constrictive pericarditis after pericardiectomy with speckle tracking echocardiography

Objective:To evaluate the left ventricular rotation and twist in patients with constrictive pericarditis (CP) after pericardiectomy by using speckle tracking echocardiography (STE), and observe its trend over time.Methods:A total of 29 patients with CP from Department of Medical Ultrasound, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology from September 2014 to December 2017 underwent echocardiography 1 week before and 1 month, 6 months, 12 months after pericardiectomy. STE was performed to obtain peak values of basal and apical rotation and left ventricular twist. Twenty-nine healthy subjects were recruited as controls in the same period.Results:The left ventricu1ar apical rotation[before surgery(6.62±3.19)°, 1 month after surgery(7.07±4.02)°, 6 months after surgery(7.88±4.46)°, 12 months after surgery(7.85±4.51)°], the left ventricu1ar twist [before surgery(10.50±4.94)°, 1 month after surgery(9.42±5.40)°, 6 months after surgery(9.59±4.62)°, 12 months after surgery(9.70±4.45)°] were significantly lower in patients with CP than those in controls [(11.22±5.17)°, (16.35±5.21)°](all P<0.05); while basal rotation in patients with CP after surgery among different time points were all significantly lower than those in controls[1 month after surgery(-3.85±3.20)°, 6 months after surgery(-3.49±2.09)°, 12 months after surgery(-3.53±2.01)°; controls(-5.57±2.78)°] (all P<0.05), with no significant difference between patients with CP before surgery (-5.22±3.14)° and controls (-5.57±2.78)°( P>0.05). There were no significant differences in left ventricular twist, basal and apical rotation in CP groups before and after surgery among different time points (all P>0.05). Conclusions:Although the left ventricular global function of patients with CP in the long postoperative period seems "normal" , the left ventricular twist, basal and apical rotation are still significantly lower than those in controls. STE can be used to assess the changes of left ventricular twist and rotation in patients with CP long-term after pericardiectomy.

Objective Our purpose is to investigate the expression of natriuretic peptide receptor A (NPR-A) in the retina and to understand the NPR-A’ s functions during the mouse development .Methods Mice eyes were harvested from E16 ( embryonic day 16 ) to P90 ( postnatal day 90 ) . Total of 127 eyes were used in the study . Immunohistochemistries of NPR-A were carried out .Results During development , NPR-A was widely expressed in the retinal neurons .In the outer nuclear layer , NPR-A began to appear in the inner and outer projections of cone and rod cells at P7, but decreased at P14.From P30 afterward, it continued to express weakly .In the inner nuclear layer , NPR-A expressed in the dendrites of bipolar cells weakly from P 7 to adulthood , whereas no expression in horizontal cells .In the ganglion cell layer, NPR-A started highly to express in the ganglion cell bodies at E 16, and in the meantime, in the nerve fiber layer , ganglion cell axons , NPR-A was expressed highly from embryonic to adult .In the inner and outer plexiform layers, NPR-A was highly expressed at P14, but decreased gradually after P30.In addition, NPR-A also widely expressed in the inner protrusions of Müller cells.Conclusion NPR-A participates in the development of the retina , and may be the key molecule in the developing retinal neurons .Moreover, it plays an important regulatory role in the functional activity of Müller cells .
10.Effects of gypenosides on the expressions of p53 and p21 protein in photodamaged BALB/c mouse skin

Objective To explore the mechanism underlying the effects of gypenosides (GP) against photodamage. Methods Eighty BALB/c mice were equally divided into 8 groups, i.e., blank control group (untreated), UVB model group (irradiated with UVB), GP I group (irradiated with UVB before topical application of GP), GPⅡ group (irradiated with UVB followed by topical application of GP), VitE I group (irradiated with UVB after topical application of Vitamine E cream), VitE Ⅱ group (irradiated with UVB followed by topical application of Vitamine E cream), Vehicle group Ⅰ (irradiated with UVB after application of the drug vehicle),and Vehicle group Ⅱ (irradiated with UVB before application of the drug vehicle). UVB irradiation was performed once every other day for 14 days. Mice were sacrificed after the last irradiation and skin specimens were obtained from the irradiated sites, and the levels of p53 and p21 protein were measured by Western blot in the specimens. Results The expression level of p53 protein was significantly lower in the blank control group than in the UVB model group (0.11 ± 0.08 vs. 0.22 ± 0.12) and GP Ⅰ group (0.44 ± 0.23, P < 0.01),in the blank control group and UVB model group than in the GP Ⅱ group (0.48 ± 0.24, P < 0.01, 0.05). VitE Ⅰ group (0.49 ± 0.29) and VitE II group (0.50 ± 0.27) were similar to the GP groups in the expression of p53 protein. No statistical difference was observed in the expression of p21 protein between the eight groups. Conclusion The upregulation of p53 protein expression in epidermal cells may be related to the mechanisms underlying the protective effect of 1.5% GP cream against photodamage.
