Objective To investigate the effect of IGF-1 on the TGF-β3 induced chondrogenesis of MSCs encapsulated in alginate beads and its application in cartilage tissue engineering.Methods MSC chondrogenesis in alginate beads was induced by TGF-β3 and/or IGF-1.Collagen type Ⅱ,aggrecan and Sox-9 expression was evaluated by immunostaining,RT-PCR and Western blot,respectively.Scanning electron microscope and laser confocal microscope were used to observe the differentiated chondrocytes when cultured on the chitosan-based scaffold.Results TGF-β3 with IGF-1 induced MSCs in alginate beads to express the higher level of collagen type Ⅱ,aggrecan and Sox-9 than any other growth factor alone (P＜0.05).The correlation coefficient between Sox9 and collagen type Ⅱ or aggrecan was 0.95 and 0.91,respectively.The chitosan-based scaffold supported the cell's adhesion,migration and proliferation.Conlusion IGF-1 enhances the TGF-β3-induced MSC chondrogenesis via upregulating Sox9 expression.The chitosan-based scaffold is biocompatible with the differentiated chondrocytes.

Objective To explore the expression of anaplastic lymphoma kinase(ALK) in human ovarian serous adenocarcinoma cell line SKOV3 and to further investigate the effect of crizotinib on SKOV3 cells.Methods The expression of ALK in SKOV3 cells were examined by Western blotting and immunocytochemical methods,and the effect of ALK inhibitor crizotinib on proliferation of SKOV3 cells were evaluated by cell proliferation assay.Results Both Western blotting and immunocytochemical methods showed the expression of ALK in SKOV3 cells.The results of cell proliferation assay suggested that the cell viability of SKOV3 cells was close to 1 when the drug concentration was less than 106 nmol·L-1,and close to 64% when the drug concentration was up to 107nmol·L-1.Conclusion ALK is overexpressed in human ovarian serous adenocarcinoma cell line SKOV3,but SKOV3 cells were insensitive to the therapy of ALK inhibitor crizotinib.

Objective To explore the role of Tbx3 gene in the occurrence and development of breast cancer. Methods 44 cases of the fresh breast carcinoma tissues and peritumor tissues were collected individually, the gene expression of Tbx3 was detected by RT PCR and the relationship between the changes of Tbx3 gene expression and the related clinicopathological factors of breast carcinoma was investigated. Results Obviously, the expression level of Tbx3 gene in breast carcinoma tissues was higher than that in the peritumor tissues. Furthermore, Tbx3 gene expression level in breast carcinoma tissues was not associated with the tumor size and ages of patients. However, the histological grading of breast carcinoma, tumor infiltrating degree, TNM stagings, and axillary lymph node metastasis were positively correlated with the expression level of Tbx3 gene. Conclusion Tbx3 gene with high expression may promote the occurrence and development of breast cancer and it is also likely a symbol of crucial changes in the development of breast cancer

OBJECTIVE To compare the clinical efficacy between adenoidectomy combined with auripuncture (AT+A) and adenoidectomy combined with tympanostomy tubes (AT+T) in children wiht otitis media with effusion. METHODS 92 cases of children with otitis media with effusion were selected from September 2010 to September 2014 in our hospital which was divided into AT+T group and AT+A group, depending on therapy methods.There were 48 in AT+T group and 44 in AT+A group. Retrospective analysis were made on a series of clinical data ,including the therapeutic effect, hearing threshold before and after treatment, infection rate and recurrence rate. RESULTS The hearing threshold has been significantly reduced within one year of follow-up after treatment,however,there was no significant difference (P>0.05). Total efficiency rate was 97.9% in AT+T group and 95.4% in AT+A group, no significant difference was found (P>0.05). The average middle ear fluid duration, infection and recurrence rate of children in AT+T group and AT+A group was (7.3±0.8) days, 6.3%, 4.2% and (11.7±0.4) days, 15.9%, 11.4% respectively, there was significant differences (P<0.05). CONCLUSION Both AT+T and AT +A have a good therapeutic effect for children with otitis media with effusion, which can significantly improve the hearing. Compared to AT+A, AT+T have a higher recovery rate,significantly reduce the recurrence and infection rate,and may be more suitable for clinical treatment of children with otitis media with effusion.

Objective To assess the aorta elastic properties in the procession of atherosclerosis by ultra‐high frequency ultrasound ,and to detect the relationship between the aorta elastic properties and the atherosclerotic plaque burden .Methods Mice deficient for the apolipoprotein E (ApoE‐/‐) with high‐cholestrol diet were studied as an age‐dependent model of atherosclerosis .At 8 ,16 ,24 and 32 weeks of age , the blood pressure in the ascending aorta was measured by catheter ,and the aorta mechanical properties were assessed by measuring aortic elastic modulus of the ascending aorta with ultra‐high frequency ultrasound .The plaque burden was assessed by high‐frequency ultrasound and Masson′s trichrome stain , separately .Results Vessel thickness at the lesion‐prone sites of the lesser curvature of the aorta and the proximal brachiocephalic artery increased with age ,consistent with the Masson′s trichrome staining which showed age‐dependent worsening of atherosclerosis in the mice model .Elastic modulus of the aorta significantly increased from 8 to 32 weeks of age in E‐/‐mice .There was a statistically difference between any two groups .Conclusions With the progression of atherosclerosis and the increased plaque burden ,aorta mechanical properties deteriorated in Apo E‐/‐mice .Ultra‐high frequency ultrasound was a potential tool for assessment of plaque burden and aorta mechanical properties in mouse model .

Purpose To explore the value of spectral CT imaging in differentiating pancreatic carcinoma and mass-forming pancreatitis, and to improve the preoperative diagnosis accuracy. Materials and Methods The dual-phase spectral CT imaging data in patients with pathology-proven pancreatic carcinoma (n=62) and mass-forming pancreatitis (n=30) were retrospectively analyzed. At 40-140 keV (with 10 keV increment), the mean CT values, energy spectrum curves and iodine concentrations were measured and compared between pancreatic carcinoma and mass-forming pancreatitis in arterial phase and portal venous phase. Results The mean CT values of mass-forming pancreatitis at 40-140 keV were higher than pancreatic carcinoma in both arterial phase and portal venous phase with significant difference (t= -7.349- -4.810, P<0.05; t= -6.760- -5.496, P<0.05). The energy spectrum curves were significantly different between pancreatic carcinoma and mass-forming focal pancreatitis, especially at 40-70 keV. The iodine concentrations of pancreatic carcinoma were lower than mass-forming pancreatitis in arterial phase and portal venous phase with significant difference (t=-28.577, P<0.05; t= -28.451, P<0.05). Conclusion Pancreatic carcinoma and mass-forming pancreatitis can be differentiated by measuring the CT value at different energies, energy spectrum curves and iodine concentrations.

Objective To observe effect of environmental enrichment on the learning and memory ability and the expressions of hypoxia inducible factor 1α (HIF-1a) and vascular endothelial growth factor (VEGF) in rats with chronic cerebral hypoperfusion.Methods A total of 40 rats were randomly divided into 3 groups: bilateral vascular occlusion (2VO) of the common carotid arteries group (n=14, 2VO group), 2VO + enriched environment (EE) group (n=14, 2VO+EE group) and sham group (n=12, SHAM group).Morris water maze, novel object recognition test, real-time reverse transcription-polymerase chain reaction (real-time RT-PCR), immunohistochemistry methods and Western blotting were used to detect changes in learning and memory ability of rats and HIF-1α and VEGF expression levels in hippocampus.Results Morris water maze showed that the escape latency was longer in the 2VO group than in the SHAM group at 3, 4 and 5 day during the training (all P＜0.05), while the 2VO+EE group spent significantly less time in finding the platform as compared with the 2VO group at 4 and 5 day (both P＜0.05).The time for space exploration in target quadrant was less in 2VO group than in SHAM group (P＜0.05), while it was longer in 2VO +EE group than in 2VO group (P＜0.05).Novel object recognition test showed that the 2VO operation impaired the priority index (PI) of time spending at exploring the novel object (P＜0.05), and environmental enrichment could improve the PI in 2VO group (P＜0.05).The real-time RT-PCR and Western blotting showed that the HIF-1α mRNA expression was higher in 2VO group than in SHAM group (P＜0.05).The VEGF mRNA and protein expressions were higher in 2VO+EE group than in 2VO group (both P＜0.05).The expression of HIF-1α in hippocampal CA1 area was higher in 2VO group than in SHAM group (P＜0.05).Conclusions Environmental enrichment can alleviate the damages of spatial and non-spatial learning and memory ability which are caused by chronic cerebral hypoperfusion.And HIF-1α and its downstream gene VEGF may be involved in the restoration of cognitive function by enriched environment.

OBJECTIVE:To observe the efficacy and safety of eprosartan in the treatment of hypertensive patients with coro-nary heart disease. METHODS:160 hypertensive patients with coronary heart disease randomly divided into observation group and control group. All patients were given aspirin,nitroglycerin,low molecular weight heparin,statins and other conventional treat-ment;control group was additioanlly given 50 mg Losartan potassium tablet,orally,once a day. Observation group was additional-ly given 600 mg Eprosartan tablet,orally,once a day. The treatment course for both groups was 6 months. Clinical efficacy,sit-ting systolic blood pressure and diastolic blood pressure,alanine aminotransferase(ALT),aspartate aminotransferase(AST),urea (UREA),creatinine(Cr),uric acid(UA),total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C), low-density lipoprotein cholesterol (LDL-C),the Mini-Mental status (MMSE) scale and activities of daily living (ADL) scale scores before and after treatment and incidence of adverse reactions in 2 groups were observed. RESULTS:There was no signifi-cant difference in the total effective rate between 2 groups(P>0.05). After treatment,the sitting systolic blood pressure and diastol-ic blood pressure,MMSE and ADL scale scores in 2 groups were significantly lower than before,and sitting systolic blood pres-sure in observation group was lower than control group,the differences were statistically significant(P<0.05),and there were no significant differences in sitting diastolic blood pressure,MMSE and ADL scale scores between 2 groups(P>0.05),and no signifi-cant differences in ALT,AST,UREA,Cr,UA,TC,TG,HDL-C and LDL-C between 2 groups before and after treatment(P>0.05). There were no obvious adverse reactions during treatment. CONCLUSIONS:Eprosartan can effectively reduce sitting systol-ic blood pressure in hypertensive patients with coronary heart disease,and improve cognitive function,with good safety.

OBJECTIVE:To study the effects and mechanism of propranolol on the myocardial abnormal electrophysiology sta-tion in diabetic model rats. METHODS:SD rats were randomly divided into normal control(normal saline)group,diabetic(nor-mal saline)group,PD98059(ERK inhibitor,10 mg/kg)group and propranolol low-dose,medium-dose and high-dose(1,20,50 mg/kg)groups,with 8 rats in each group. Except for normal control group,rats were given alloxan(20 mg/kg)intravenously via tail vein to induce diabetic model. They were given relevant medicine intragastrically,once a day,for consecutive 42 days. The car-diac index,electrocardiogram and action potential durations (APD) of rats were analyzed;the expression of TNF-α,IL-2,IL-6 and IL-10 protein in serum were detected,and the expression of Ras,Raf,ERK kinase(MEK)and ERK1/2 in myocardial tissue were detected. RESULTS:Compared with normal control group,cardiac index increased in diabetes group;heart rate decreased;QT interval and APD were prolonged;the relative expression of TNF-α,IL-2,IL-6,IL-10,Ras,Raf,MEK and ERK1/2 protein increased (P<0.01). Compared with diabetes group,cardiac index decreased in propranolol medium-dose and high-dose groups and PD98059 group,heart rate increased,QT interval and APD were shortened;the relative expression of TNF-α,IL-2,IL-6, IL-10,Ras,Raf,MEK and ERK1/2 protein decreased(P<0.05 or P<0.01). CONCLUSIONS:Propranolol can improve myocar-dial abnormal electrophysiology station of diabetic model rats by down-regulating inflammatory reactions in serum and inhibiting the activation of MEK/ERK signaling pathway.

Objective To evaluate the safety and efficacy of pancreatic duct precutting by arciform knife combined with needle knife in difficult cannulation of ERCP.Methods The clinical data of 38 patients who were treated with pancreatic duct precutting by arciform knife combined with needle knife in our hospital from June 2005 to May 2012 were retrospectively reviewed.Results The success rate of ERCP was 86.8％ (33/38) without any complications of bleeding or perforation,while 4 cases were presented with hyperamylasemia after ERCP,4 with pancreatitis and 1 with cholangitis who all got recovered after 5 days with conservative treatment.Conclusion The application of pancreatic duct precutting by arciform knife combined with needle knife can improve the success rate of ERCP and is a kind of effective operation technique.
