Objective To obtain the shear wave velocity(SWV) in breast tumors at different probe orientations.Methods SWV was measured on 92 breast tumors in 48 female patients with the probe placed on transverse,longitudinal and 45 degree planes.Pathological test results were followed up.Results Pathological tests showed benign tumors in 48 patients (92 tumors).Of the three planes,values of the biggest and the smallest SWV showed significant difference (P ＜0.001).Conclusions SWV differs on different planes of breast tumors.So the effect in different probe orientations should be considered using SWV for the differential diagnosis of breast tumors.

Objective To investigate the effect of early intervention on adhesion molecule NB-3 expressing in cerebral injured neonatal rat induced with intrauterine infection. Methods Pregnant rats were consecutively injected with lipopolysaccharide (LPS) 380 μg/kg or saline 17 d after gestation. The offspring from the LPS group accepted early intervention (I group, n=40) or not (NI group, n=40), and those for saline group were as control group (n=40). They were assessed with suspension test 14 d and 28 d after born, and the expression of were NB-3 were detected with immunohistochemistry 1 d, 7 d, 14 d, 21 d, 28 d after born. Results The expression of NB-3 was different among these 3 groups, as well as the scores of suspension test (P<0.001). Conclusion Intrauterine infection increases the expression of NB-3 of neonatal rats with brain damage. The early intervention may increase persistently the expression of NB-3, and improve the motor function.

OBJECTIVETo explore the mechanism of acupuncture on promoting the restoration of interstitial cells of Cajal (ICCs).

METHODSThirty SD rats were randomly divided into a blank group, a model group and an acupuncture group, ten rats in each one. The rats in the model group and acupuncture group were treated with colonic anastomosis to establish the model. After successful establishment of the model, the rats in the acupuncture group were treated with acupuncture at bilateral "Zusanli" (ST 36), "Sanyinjiao" (SP 6) and "Taichong" (LR 3) for 15 min, once a day for 10 days. Rats in the model group and blank group were put into the fixator for 15 min at the same time daily. The propulsive rate of small intestine was measured in each group. Colonic tissues were collected to detect c-kit expression by using immunohistochemistry. The nitricoxide (NO) content was measured by nitrate reductase method and nitric oxide synthase (NOS) activity was measured by method of L-arginine.

RESULTSCompared with the blank group, the propulsive rate of small intestine in the model group was decreased; NO content was increased; iNOS activity was elevated; cNOS activity was declined; total NOS (tNOS) activity was increased and the counting of c-kit positive ICCs was decreased (all P < 0.05). Compared with the model group, the propulsive rate of small intestine in the acupuncture group was increased; NO content was decreased; iNOS activity was reduced; cNOS activity was elevated; NOS activity was decreased and the counting of c-kit positive ICCs was increased (all P < 0.05).

CONCLUSIONAcupuncture can regulate NO content and NOS activity in postoperative restoration environment of ICCs, which may participate in the process of acupuncture promoting the restoration of ICCs.

Objective To evaluate the feasibility of prospective electrocardiogram(ECG)-gating computed tomography coronary angiography(CTCA).Methods Sixty patients with suspected or known coronary artery disease underwent 64-slice CTCA using prospective ECG-gating.Multi-planar reconstruction (MPR),curved-planar reconstruction(CPR),maximum intensity projection(MIP)and volume rendering (VR)were used to demonstrate the coronary arteries.The image quality and radiation dose was evaluated.Results The mean effective radiation dose was(2.7±0.2)mSv.93.3%(720/772)segments of all coronary arteries were of diagnostic image quality,44.2%(341/772)Was classified as excellent and 49.1%(379/772)was good.Non-diagnostic coronary segments were found in 6.7%(52/772)of all coronary arteries.Therewere 5(8.3%)cases with severe coronary stenosis(＞75%)or occlusion,17(28.4%)cases with moderate stenosis(50%-75%),18(30.0%)cases with mild stenosis(＜50%)or irregular lumen,20(33.3%)cases with normal coronary artery.Conclusion With a low radiation dose,prospective electrocardiogram(ECG)-gated coronary 64-MSCT angiography has a good potential for the detection of coronary stenosis,especially for excluding coronary artery disease.

Objective To investigate the effects of GGNBP2 on proliferation,migration and invasion of human gli-oma U251 cells and the potential mechanism. Methods U251 glioma cells were transfected with lentiviral vector carrying GGNBP2 to establish a stable overexperssion of GGNBP2 in U251 cell line. After verification of the efficiency of transfec-tion by real-time PCR, Western Blot, PCR and CCK-8 were applied to detect the proliferation of U251 cells. The Tran-swell chamber assay was applied to measure the migration and invasion of human U251 glioma cells. Western blot was applied to measure protein levels of AKT, p-AKT, PCNA and MMP9. Result The stable U251 glioma cell line overex-pressing GGNBP2 was successfully established. Compared with the control group, overexpression of GGNBP2 significant-ly inhibited the proliferation(P<0.05),invasion(57±6 vs. 203±6,205±7,F=512.4,P < 0.05)and migration (74±7 vs. 254±14,248±13,F=242.5,P<0.05) of U251 cells. The protein expression levels of p-AKT (F=45.4, P<0.05), PCNA (F=348.5, P<0.05) and MMP-9 (F=88.7, P<0.05) in GGNBP2 group were markedly decreased compared with the control group. Conclusion GGNBP2 may suppress the proliferation, invasion and migration of glioma though down-regulation of p-AKT, which in turn inhibits PCNA and MMP-9 expression.

Objective To explore whether adiponectin activates AMP-activated protein kinase(AMPK) via LKB1 pathway or not in skeletal muscle and liver tissues.Methods Male Sprague-Dawley rats ( n =28 ) were divided into normal control diet( NC,n =15 ) and high-fat diet( HF,n =13 ) groups.After 16 weeks feeding,fasting blood free fatty acids( FFA ),triglyceride( TG ),total cholesterol( TC ),fasting plasma glucose( FPG ),fasting insulin(FINS),and adiponectin were determined.The protein levels of AMPKα,phosphorylated AMPKα ( p-AMPK ),and LKB1 in the skeletal muscle and liver tissues were analyzed with Western blot.Cultured primary skeletal muscle cells and hepatic cells were incubated with aditonectin and radicicol.The expression of AMPKα,p-AMPKα,and LKB1 inthese cells were analyzed with immunofluorescence method.Results Compared with NC group,body weight,FFA,TG,FPG,and FINS in rats of HF group were significantly higher( all P＜0.05 ) while serum adiponeetin level was lower( P＜0.05 ).The levels of AMPKα phosphorylation and LKB1 expression in the skeletal muscle and liver tissues of HF group were lower than those in NC group. In primary skeletal muscle cells and hepatic cells,adiponectin significantly increased the levels of AMPKα phosphorylation and LKB1 expression ( all P＜ 0.05 ),which were decreased by radicicol ( P＜0.05 ).Conclusion Adiponectin may activate AMPK via LKB1 pathway in skeletal muscle and liver tissues of rats.

Objective To observe the correlation between contrast-enhanced sonography and microveesel density (MVD) in peripheral lung tumor.Methods Contrast-enhanced sonography was performed in 55 patients with peripheral lung tumor.Among of them,malignant tumor was 40 cases,benign tumor was 15 cases.Analyzed the quantitative parameters (peak intensity,enhanced strength index) by using time-intensity curve (TIC) with computer.MVD was measured by immunohistochemistry after operation.Results The level of MVD in malignant tumor was significantly higher than that in benign tumor [(24.57 ± 5.93)/HP vs.(15.50 ± 2.82)/HP] (P ＜ 0.05).The level of MVD had significant correlation with peak intensity and enhanced strength index (P ＜ 0.01).Conclusions The peak intensity and enhanced strength index have significant correlation with MVD.They are valuable indexes in evaluating angiogenesis in peripheral lung tumor.

Objective To observe the effects of gypenosides (GP) on nuclear factor κB (NF-κB) and p38 mitogen activated protein kinase (p38MAPK) signaling pathways in photodamaged skin of mice,and to explore the mechanisms underlying the protective effects of GP against photodamage.Methods Eighty Balb/C female mice were randomly divided into 8 groups: blank control group receiving no treatment,ultraviolet B (UVB) model group receiving UVB irradiation for 60 seconds,GP group Ⅰ receiving topical GP treatment followed by UVB irradiation,GP group Ⅱ receiving UVB irradiation followed by topical GP treatment,VitE group Ⅰ receiving topical VitE treatment followed by UVB irradiation,VitE group Ⅱ receiving UVB irradiation followed by topical VitE treatment,matrix group Ⅰ receiving topical matrix treatment followed by UVB irradiation,matrix group Ⅱ receiving UVB irradiation followed by topical matrix treatment.UVB irradiation lasted 60 seconds at one time,and was given once every other day for 7 times to establish a skin model of photodamage.The interval between irradiation and topical treatment was 30 minutes in all the groups except the control group and UVB model group.After the last treatment,mice were sacrificed.Western blot was performed to measure the protein expressions of inhibitor of NF-κB(IκB),inhibitor of NF-κB kinase (IKK),p38MAPK as well as phosphorylated p38MAPK (pp38) in skin tissue from the mice.Results No expressions of IκB or IKK were observed in the blank control group.The expression level of IκB was 0.40 ± 0.07 in UVB model group,significantly lower than that in GP group Ⅰ (1.63 ± 0.85,P ＜ 0.05) and GP group Ⅱ (0.90 ± 0.40,P ＜ 0.05),whereas the level of IKK protein was higher in UVB model group than in the GP group Ⅰ and Ⅱ (2.01 ± 1.75vs.0.23 ± 0.12 and 0.45 ± 0.29,both P ＜ 0.05).No significant difference was observed in the expression of IκB or IKK proteins between the GP group Ⅰ,Ⅱ,VitE group Ⅰ and Ⅱ or in the expression of p38MAPK between the 8 groups.The phosphorylated p38MAPK expression in UVB model group was significantly higher than that in GP group Ⅰ and Ⅱ (0.835 ± 0.049 vs.0.425 ± 0.054 and 0.571 ± 0.090,both P＜ 0.05),but similar to that in VitE groups.Conclusions UVB can activate NF-κB and phosphorylated p38MAPK signaling pathways; GP 1.5％ cream can inhibit UVB-induced activation of NF-κB and p38MAPK pathways,which may be one of the mechanisms underlying its protective effects against inflammation and photodamage.

Objective To study the value of multiphasic contrast-enhanced CT in diagnosing focal noudlar hyperplasia(FNH).Methods 12 patients with FNH underwent plain CT scan and multiphasic contrast-enhanced CT scan.Arterial phases was at 25~30 s,venous phases was at 55~60 s,and delayed scans were taken after 180 s.Results The lesions showed hypodensity and slight hypodensity on plain CT in 10 cases,among them,irregular center lower density was found in 5 cases and isodensity was found in 2 cases.In the arterial phases,the lesions appeared obvious in 9 cases,among which 4 cases were enhanced homogeneously and 5 were heterogeneously.No obvious enhancement in the center scar.The supply arteries were seen in 3 cases,speckled enhancement in peripheral area of lesions was seen in 2 cases,no obvious enhancement in 1 case.In the venous phases,the lesions showed slightly hyperdensity in 6 cases,isodensity in 4 cases,and mixture of hyperdensity and hypodensity in 2 cases.12 lesions were hypodense as compared with liver in the delayed phases.Conclusion Multiphasic contrast-enhanced CT scan is specific and accurate in diagnosing FNH and can be used to guide the clinical treatment.
