Objective To study the effect of vitro thrombolytic test on the consumptions of urokinase(UK) in thrombolytic therapy of acute cerebral infarction.Methods The patients of acute cerebral infarction were treated with UK at the dose determined by vitro thrombolytic test,and then were given UK 40MU each day in the later 7 days.The degree of neurologic impairment,fibrinogen(Fbg) level,polyreaction speed(FMPV) and maximum luminosity(ODmax) were examined at different time point.Results Thrombolytic therapy could significantly reduce neurologic impairment as compared with control group((P
2.Impact of probe orientations on shear wave velocity of breast tumors

Objective To obtain the shear wave velocity(SWV) in breast tumors at different probe orientations.Methods SWV was measured on 92 breast tumors in 48 female patients with the probe placed on transverse,longitudinal and 45 degree planes.Pathological test results were followed up.Results Pathological tests showed benign tumors in 48 patients (92 tumors).Of the three planes,values of the biggest and the smallest SWV showed significant difference (P <0.001).Conclusions SWV differs on different planes of breast tumors.So the effect in different probe orientations should be considered using SWV for the differential diagnosis of breast tumors.
3.Effect of Early Intervention on Expression of NB-3 in Neonatal Rats with Cerebral Injury Induced with Intrauterine Infection

Objective To investigate the effect of early intervention on adhesion molecule NB-3 expressing in cerebral injured neonatal rat induced with intrauterine infection. Methods Pregnant rats were consecutively injected with lipopolysaccharide (LPS) 380 μg/kg or saline 17 d after gestation. The offspring from the LPS group accepted early intervention (I group, n=40) or not (NI group, n=40), and those for saline group were as control group (n=40). They were assessed with suspension test 14 d and 28 d after born, and the expression of were NB-3 were detected with immunohistochemistry 1 d, 7 d, 14 d, 21 d, 28 d after born. Results The expression of NB-3 was different among these 3 groups, as well as the scores of suspension test (P<0.001). Conclusion Intrauterine infection increases the expression of NB-3 of neonatal rats with brain damage. The early intervention may increase persistently the expression of NB-3, and improve the motor function.
4.Effects of acupuncture on NO and NOS in restoration environment of interstitial cells of Cajal after colonic anastomosis.

OBJECTIVETo explore the mechanism of acupuncture on promoting the restoration of interstitial cells of Cajal (ICCs).
METHODSThirty SD rats were randomly divided into a blank group, a model group and an acupuncture group, ten rats in each one. The rats in the model group and acupuncture group were treated with colonic anastomosis to establish the model. After successful establishment of the model, the rats in the acupuncture group were treated with acupuncture at bilateral "Zusanli" (ST 36), "Sanyinjiao" (SP 6) and "Taichong" (LR 3) for 15 min, once a day for 10 days. Rats in the model group and blank group were put into the fixator for 15 min at the same time daily. The propulsive rate of small intestine was measured in each group. Colonic tissues were collected to detect c-kit expression by using immunohistochemistry. The nitricoxide (NO) content was measured by nitrate reductase method and nitric oxide synthase (NOS) activity was measured by method of L-arginine.
RESULTSCompared with the blank group, the propulsive rate of small intestine in the model group was decreased; NO content was increased; iNOS activity was elevated; cNOS activity was declined; total NOS (tNOS) activity was increased and the counting of c-kit positive ICCs was decreased (all P < 0.05). Compared with the model group, the propulsive rate of small intestine in the acupuncture group was increased; NO content was decreased; iNOS activity was reduced; cNOS activity was elevated; NOS activity was decreased and the counting of c-kit positive ICCs was increased (all P < 0.05).
CONCLUSIONAcupuncture can regulate NO content and NOS activity in postoperative restoration environment of ICCs, which may participate in the process of acupuncture promoting the restoration of ICCs.

5.Expression of MIF in molecular subtypes of breast cancer and its clinical significance

Objective To determine the expression of macrophage migration inhibitory factor (MIF)in different molecular subtypes of breast cancer and its clinical significance so as to detect the biological markers of different molecular subtypes of breast cancer.Methods We divided 100 breast cancer patients into four molecular subtypes by immunostaining:luminal subtype,HER-2(+)subtype,basal-like (BLs)subtype and normal breast-like (NBLs)subtype,and then compared the expression of MIF in the groups.We analyzed the associations of MIF-positive expression rate with age,menstruation,tumor size,auxiliary lymph node metastasis,histological type and grade,and clinical stage of the breast cancer patients.We also compared MVD level and 5-year overall survival rate between MIF-positive patients and MIF-negative ones.Results The positive expression of MIF was correlated with HER2(+)subtype breast cancer and auxiliary lymph node metastasis (P < 0.05 ).The patients with MIF-positive expression had a significantly higher level of MVD than those with MIF-negative expression (P < 0.05 ). Kaplan-Meier method showed that MIF-positive patients had a poor prognosis than MIF-negative ones (Log-rank=1 9.5 1 6,P = 0.000).Conclusion Breast cancer patients with MIF-positive expression may be mostly of HER2 (+)subtype,and tend to develop auxiliary lymph node metastasis.These patients have a significantly higher level of MVD and poor prognosis than those with MIF-negative expression.
6.Mitochondria Injury due to Viral Myocarditis Improved by Different Doses of Captopril in Mice

Objective To further understand the cellular events of myocarditis,we have examined the myocardial mitochondria structure and activity of ATPase in mice with myocarditis,and observe the interventional effect of different doses of carptopril.Methods Sixty male Balb/c mice were randomly divided into 5 groups:infected coxsackie B3 virus(CVB3)group,infected CVB3 and treated with carptopril(in a dose of 10,30,or 100 mg/kg,twice a day)groups and control group.Captopril was administered after infection.The activity of Na+-K+-adenosine triphosphatase(Na+-K+-ATPase)and Ca2+-adenosine triphosphatase(Ca2+-ATPase)of myocardial mitochondrial as well as the morphological of myocardial mitochondrial were investigated on day 14.Results The activity of mitochondrial Na+-K+-ATPase and Ca2+-ATPase were significantly higher(Pa0.05).Conclusions The favorable effects of captopril exerts on myocyte mitochondria were shown in a dose-dependent manner.Thirty and 100 mg/kg,twice a day captopril protecs the membrane integrity and thus plays a role at the recovery of depressed mitochondrial Na+-K+-ATPase and Ca2+-ATPase activity and also in myocytes injury.
7.The inhibitory effects of rh-endostatin (YH-16) in combination with radiotherapy on lung adenocarcinoma A549 in mice and the underlying mechanisms.

In order to investigate the inhibitory effects of Endostar (rh-endostatin, YH-16) in combination with radiotherapy on lung adenocarcinoma A549 in mice and the interaction mechanisms of combined therapy, the transplantation tumor models of A549 lung adenocarcinoma were established. When the largest diameter of tumor reached 1.0 cm, all nude mice were randomly divided into 4 groups: Endostar group, radiotherapy group, radiotherapy plus Endostar (combined treatment) group, and control group (n=6 in each group). The largest diameter and the vertical diameter of tumor were measured at different time points. At the 16th day, mice were executed, and the tumors were applied to analysis of rate of tumor cell apoptosis, and the expression levels of basic fibroblast growth factor (bFGF) mRNA were detected by reverse transcription-polymerase chain reaction (RT-PCR) and those of vascular endothelial growth factor (VEGF) by immunohistochemistry. The results demonstrated that the rate of tumor inhibition in combined treatment group was higher than that in other groups. And the rate of tumor cell apoptosis in combined treatment group was also higher than that in other groups. Meanwhile, the levels of bFGF mRNA and VEGF expression in combined treatment group were lower than those in other groups. It was concluded that Endostar obviously enhanced the curative effectiveness of radiotherapy on lung adenocarcinoma A549 in mice. The underlying mechanisms may involve the down-regulation of bFGF mRNA and VEGF expression to inhibit angiogenesis by Endostar and the cooperative effect of Endostar and radiotherapy to synergistically promote tumor cell apoptosis. And Endostar inhibits angiogenesis by down-regulating the expression of bFGF mRNA and VEGF.
8.The effect and mechanism of GGNBP2 regulates the proliferation, invasion and migration of human U251 glioma cells

Objective To investigate the effects of GGNBP2 on proliferation,migration and invasion of human gli-oma U251 cells and the potential mechanism. Methods U251 glioma cells were transfected with lentiviral vector carrying GGNBP2 to establish a stable overexperssion of GGNBP2 in U251 cell line. After verification of the efficiency of transfec-tion by real-time PCR, Western Blot, PCR and CCK-8 were applied to detect the proliferation of U251 cells. The Tran-swell chamber assay was applied to measure the migration and invasion of human U251 glioma cells. Western blot was applied to measure protein levels of AKT, p-AKT, PCNA and MMP9. Result The stable U251 glioma cell line overex-pressing GGNBP2 was successfully established. Compared with the control group, overexpression of GGNBP2 significant-ly inhibited the proliferation(P<0.05),invasion(57±6 vs. 203±6,205±7,F=512.4,P < 0.05)and migration (74±7 vs. 254±14,248±13,F=242.5,P<0.05) of U251 cells. The protein expression levels of p-AKT (F=45.4, P<0.05), PCNA (F=348.5, P<0.05) and MMP-9 (F=88.7, P<0.05) in GGNBP2 group were markedly decreased compared with the control group. Conclusion GGNBP2 may suppress the proliferation, invasion and migration of glioma though down-regulation of p-AKT, which in turn inhibits PCNA and MMP-9 expression.
9.Study on Decreasing Effects of Polydatin on Blood Viscosity in the Rat Model of Acute Blood Stasis

OBJECTIVE:To observe the decreasing effects of Polydatin on blood viscosity and microcirculation in the rat model of acute blood stasis.METHODS:The rat model of acute blood stasis was established with high dose of adrenaline sub?cutaneous injection in combination with ice-water socking.The parameters such as whole blood viscosity?bat low and high shear rates,hemotocrit,platelet adhesiveness testPAdT,fibrinogenFIB,plasma viscosity?petc.were used.RESULTS:The results showed that Polydatin reduced FIB,PAdT in rat model remarkably.It obviously caused a reduction of?p,which induced decrease of?b.The effect was more significant than any other factors in reducing blood viscosity and improving mi?crocirculation.CONCLUSION:Polydatin is helpful to reduce blood viscosity and improve microcirculation.The data provided would be useful for rational use of drug.
10.The Use of Multiphasic Contrast-Enhanced CT Scan in Diagnosis of Focal Nodular Hyperplasia of Liver

Objective To study the value of multiphasic contrast-enhanced CT in diagnosing focal noudlar hyperplasia(FNH).Methods 12 patients with FNH underwent plain CT scan and multiphasic contrast-enhanced CT scan.Arterial phases was at 25~30 s,venous phases was at 55~60 s,and delayed scans were taken after 180 s.Results The lesions showed hypodensity and slight hypodensity on plain CT in 10 cases,among them,irregular center lower density was found in 5 cases and isodensity was found in 2 cases.In the arterial phases,the lesions appeared obvious in 9 cases,among which 4 cases were enhanced homogeneously and 5 were heterogeneously.No obvious enhancement in the center scar.The supply arteries were seen in 3 cases,speckled enhancement in peripheral area of lesions was seen in 2 cases,no obvious enhancement in 1 case.In the venous phases,the lesions showed slightly hyperdensity in 6 cases,isodensity in 4 cases,and mixture of hyperdensity and hypodensity in 2 cases.12 lesions were hypodense as compared with liver in the delayed phases.Conclusion Multiphasic contrast-enhanced CT scan is specific and accurate in diagnosing FNH and can be used to guide the clinical treatment.
