Objective To investigate the possibility of anti-liver fibrosis of 13-estradiol nanoparticle prepared by interfacial polymeri-zation method with butylcyanoacylate as carrier material (E2-PBCA-NP) and its effect on the expression of transforming growth factor β1 and connective tissue growth factor in pig serum induced animal fibrotic model. Methods Male Sprague-Dawley (SD) rats were random divided into five groups. Except normal control group, other four groups were all given intraperitoneal injection with pig serum. Therapeutic drugs were administered to rats from the ninth week after injection of pig serum. All rats were killed at the end of the twelfth week. Several experi-ments were done as below, the tissues of liver were observed by Masson staining, and the mRNA of TGF-β1 and CTGF of liver samples were detected by RT-PCR. Meanwhile, the expression of TGF-β1 and CTGF protein were detected by immunohistochemistry. Results It showed that both E2 and E2-PBCA-NP treatment groups had lower stage of liver fibrosis, according to the observation of pathology by Masson staining (P < 0.05). The anti-liver fibrosis effect of E2-PBCA-NP treatment group was better than that of E2 treatment group (P < 0.05). The mRNA and protein level of TGF-β1 and CTGF were markedly reduced by E2 and E2-PBCA-NP treatment, compared with liver fibrotic model groups (P <0.01). There was no statistical difference between E2-PBCA-NP and E2 treatment (P >0. 05), while no significant change was observed in blank nano -particle group (P > 0.05). Conclusion Both E2-PBCA-NP and E2 had anti-liver fibrosis activity. E2-PB-CA-NP has stronger anti - liver fibrosis activity than E2, which could be resulted from the inhibition of TGF-β1 and CTGF expression.

Prion diseases are infectious and fatal neurodegenerative diseases. The pathogenic agent is an abnormal prion protein aggregate. Microglial activation in the centre nervous system is a characteristic feature of prion disease. In this study, we examined the effect of PrP 106-126 on PrP mRNA gene expression in Mouse microglia cells BV-2 by real-time quantitative PCR. PrP mRNA expression level was found to be significantly increased after 18 h exposure of BV-2 cells to PrP 106-126, with 3-fold increase after 18 h and 4.5-fold increase after 24 h and BV-2 cells proliferating occurred correspondingly. Our results provide the first in vitro evidence of the increase of PrP mRNA levels in microglial cells exposed to PrP 106-126, and indicate that microglial cells might play a critical role in prion pathogenesis.

OBJECTIVESr-HA, a new type of hydroxyapatite biomaterial, was implanted into animals to study the bioreaction and character, which would be helpful for the further clinical applications in the future.
METHODSTotally 24 rabbits were divided into 3 groups. The bone defect of 6 mm x 12 mm x 4 mm was made at both mandibular angles of rabbits and Sr-HA of different proportion (10%, 5%, 0) was applied to reform the defects. One group of animals were killed randomly at 1, 3 and 6 months after operation to evaluate the material biological compatibility using anatomic, X-ray examination, histological and ECT methods.
RESULTSThe histological photographs showed that Sr-HA caused little infection around implanted area and, almost was not repulsed by hosts. With the degradation of biomaterial, there was more apparent new bone growth in the area around Sr-HA than that around HA and some ossification can be found in soft tissue nearby. Also a tight osteointegrity was gradually got after the operation, according to the results of X-ray and, the border between Sr-HA and bone was hardly discovered at the 6th month after the operation. A more obvious nuclide assembling was observed at the side of Sr-HA by ECT images. With the biodegradation of Sr-HA, more new bone was intruded into the spare space of the biomaterial.
CONCLUSIONSr-HA has better biocompatibility and higher biodegradation than that of pure HA. It holds an excellent osteoinductivity and fair osteoconductivity to some degree too. So a more satisfying effect of bone defect rehabilitation was gained with the increasing new bone depositing in the free space of the material, when it degraded gradually.

Chinese Journal of Lung Cancer 2004;7(3):230-235
BACKGROUNDTo evaluate the quality of life (QoL) of lung cancer patients and to investigate the differences in QoL with respect to social and medical characteristics.
METHODSStructured questionnaires (EORTC QLQ-C30 and QLQ-LC13) were used in 106 consecutive lung cancer patients for data collection during January 1, 2002 to December 31, 2002. The t test and one-way analysis of variance (ANOVA) were used to compare the differences in QoL at a 5% level of significance.
RESULTSThe QoL of lung cancer patients were worse than reference value. The QoL of young, male and married patient groups was better than that in old, female and single groups. The QoL in patients with lower education or income was worse than that in patients with high education and income. Small cell lung cancer patients showed poorer QoL than non-small cell lung cancer patients did. The locally advanced or metastatic patients had worse QoL. All the kinds of treatments could worsen the QoL, however, the patients receiving surgical treatment had best QoL.
CONCLUSIONSThe results suggest that the establishment of QoL judgment standard in Chinese lung cancer patients might be useful for clinicians, researchers and policy-makers.

Objective To explore the diagnostic value of CT cerebral perfusion imaging on the brain injury of the high +Gx in Rhesus. Methods Seven healthy male adult Rhesus were randomly divided into control group and +15 Gx group. The +15 Gx group underwent parabolic G curve in animal centrifuge. The animals were all examined by CT cerebral perfusion before +Gx exposure, 2 h, 24 h, and 1 week after ~+Gx exposure. The results were compared with pathologic examination. Results 2 h and 24 h after +15 Gx exposure, brain ischemia was showed on CT cerebral perfusion imaging. After 1 week, the brain ischemia was almost recovered to normal. Mild ischemic atrophy was observed in pyramidal neurons in cerebral cortex by light microscopy. Electron microscopic observation showed chromatin marginating and mitochondria cristae blurring in pyramidal cells after +Gx overload. Conclusion High G from simulating spaceship emergency return can cause ischemic injuries of the brain in Rhesus, and CT brain perfusion imaging can provide valuable diagnostic information.

Objective:To observe the sensitivity of transcription mediated amplification (TMA),and to compare its performance with real-time reverse transcription polymerase chain reaction (real-time RT-PCR) in detecting human immunodeficiency virus RNA (HIV RNA).Methods:TMA system was established with TaqMan probes, specific primers, moloney murine leukemia virus (MMLV) reverse transcriptase,T7 RNA polymerase, and reaction substrates. The sensitivity of TMA was evaluated by amplifying a group of 10-fold diluted HIV RNA standards which were transcribed in vitro.A total of 60 plasma of HIV infected patients were measured by TMA and Cobas Amplicor HIV-1 Monitor test to observe the positive rate. The correlation and concordance of the above two technologies were investigated by linear regression and BlandAltman analysis. Results:TMA system was established successfully and HIV RNA transcribed standards at concentration of equal or more than 10 copies/mL could be detected by TMA technology.Among 60 samples of plasma from HIV infected patients,46 were positively detected and 12 were negatively amplified by both TMA and Cobas reagents;2 samples were positively tested by Cobas reagent but negatively tested by TMA system. The concordance rate of the two methods was 97.1% and the difference of positive detection rate between the two methods was not statistically significant (P>0.05).Linear regression was used for 46 samples which were positively detected by both TMA and Cobas reagents and showed an excellent correlation between the two reagents (r=0.997,P<0.001).Bland-Altma analysis revealed that the mean different value ofHIV RNA levels for denary logarithm was 0.02.Forty-four samples were included in 95% of credibility interval of concordance. Conclusion:TMA system has the potential of high sensitivity.TMA and real-time RT-PCR keep an excellent correlation and consistency in detecting HIV RNA.
