1.Effect of Dingzhixiaowan on Hippocampal Formation in Depression Model Rats

Objective To study the anti-depression mechanism of Dingzhixiaowan concerning on histology of hippocampal formation.Methods 40 female rats were divided into control,depression model group(model group),distilled water group(DW group) and Dingzhixiaowan group(Chinese herb group).Behavior points were measured by open field method.Serum concentrations of cortisol and estradiol(E2) were checked with ELISA.HE staining and electron microscopy were applied to observe the histology of neurons in hippocampal formation.Immunohistochemistry method was employed to detect Nestin expression.Results Compared with the control,behavior points,serum E2 level and Nestin expression decreased obviously and serum cortisol level increased markedly,CA3 neurons were seriously damaged in model and DW groups.While behavior points,serum E2 level did not change,Nestin expression increased obviously and serum cortisol level decreased markedly,CA3 neurons were basically normal in Chinese herb groups.Conclusion Dingzhixiaowan can protect hippocampal formation,which might be one of the anti-depression mechnisms.
2.Study on Expression of ENaC and rBSCl of Kidney Tissue in Lung-Qi Deficiency Model Rats

Objective To study the relationship of lung and kidney in TCM by detecting the protein expression related to Na+ transportation in the kidney tissue of Lung-Qi deficiency model rats.Methods Lung-Qi deficiency model of rats were copied.Immunohistochemistry was applied to measure the expressions of ENaC and rBSCl in renal tubular epithelium.RIA was used to detect ALD and ANP levels in plasma and lung tissue.Results Compared with the control group,expressions of ENaC and rBSCl of renal tubule were upregulated markedly,ALD level increased and ANP level decreased obviously in the model group.Conclusion Lung can regulate the expressions of ENaC and rBSCl in the renal tubule by secreting ALD and ANP,which can regulate Na+ and water reabsorption in the renal tubule and then influence the water metabolism of the body.
3.Characteristics of Traditional Chinese Medical Syndrome Types of Dengue Fever Patients Admitted in Guangzhou and Xishuangbanna in the Year of 2013:An Analysis of 117 Cases

Objective To investigate the characteristics of traditional Chinese medical syndrome types of fulminant and epidemic dengue fever patients admitted in Guangzhou and Xishuang banna in the year of 2013,and to ex plore the differences of etiology and pathogenesis, illness, and treatment for the patients in the two regions. Methods We collected the clinical data of 78 cases receiving integrative Chinese and western medicine from 255 patients admitted in Guangzhou Municipal Eighth People’s Hospital, and the clinical data of 39 cases receiving integrative Chinese and western medicine from 120 patients admitted in the People’s Hospital of Xishuangbanna Dai Nationality Autonomous Prefecture in the year of 2013. The traditional Chinese medical syndrome types and the syndrome scores of the total of 117 cases were investigated. The method of phenology was used for the analysis of epidemic time and epidemic region of dengue fever, and the theory of defense-qi-nutrient-blood syndrome differentiation of seasonal febrile diseases was used for the analysis of etiology and pathogenesis of dengue fever. Results ( 1) Dengue fever was epidemic in the first ten days of July and in the middle ten days of November of the year 2013 in Guangzhou region, and was epidemic in the middle ten days of August and the first ten days of October in Xishuangbanna region. The epidemicity of dengue fever in Guangzhou covered the end of summer and the whole autumn, and then disappeared before the coming of winter. In Xishuangbanna , the epidemicity of dengue fever was obvious in autumn, and disappeared in late autumn. ( 2) In the two hospitals, dengue fever patients were dominated by the syndromes of excessive heat in both Qifen and Xuefen, blood stasis blended with toxicity, excessive heat in Qifen, and lingering pathogens in order. (3) Before treatment, the scores of fever were higher in patients of Xishuangbanna hospital than those in patients of Guangzhou Eighth People’s Hospital ( P<0.01) . After treatment for 3 days, fever scores were improved in both hospitals (P<0.05 or P<0.01), but the differences between the two hospitals were insignificant (P>0.05) . After treatment for 6 days, fever disappeared in patients of both hospitals. (4) Before treatment, the scores of syndromes were higher in patients of Xishuangbanna hospital than those in patients of Guangzhou Eighth People’s Hospital ( P<0.05) . After treatment for 3 days, syndorme scores were improved in both hospitals ( P<0.01) , but the syndrome scores were higher in Xishuangbanna hospital than those in patients of Guangzhou Eighth People’s Hospital. After treatment for 6 days, syndrome scores were much improved in patients of both hospitals compared with those after treatment for 3 days (P<0.01) . Conclusion In dengue fever patients admitted in Guangzhou and Xishuangbanna region, the syndrome of excessive heat in both Qifen and Xuefen is the leading type, and then comes blood stasis blended with toxicity. The illness state of patients in Guangzhou region is milder than that of the patients in Xishuangbanna region, the time for symptom relief is about one week, and similar therapeutic effect can be achieved in the two regions .
4.Detection of ALK, ROS1 and RET fusion genes in non-small cell lung cancer patients and its clinicopathologic correlation.

OBJECTIVETo study the prevalence of ALK, ROS1 and RET fusion genes in non-small cell lung cancer (NSCLC), and its correlation with clinicopathologic features.
METHODSFormalin-fixed and paraffin-embedded tissue sections from samples of 302 patients with NSCLC were screened for ALK, ROS1, RET fusions by real-time polymerase chain reaction (PCR). All of the cases were validated by Sanger DNA sequencing. The relationship between ALK, ROS1, RET fusion genes and clinicopathologic features were analyzed.
RESULTSIn the cohort of 302 NSCLC samples, 3.97% (12/302) were found to contain ALK fusion genes, including 3 cases with E13; A20 gene fusion, 3 cases with E6; A20 gene fusion and 3 cases with E20; A20 gene fusion. There was no statistically significant difference in patient's gender, age, smoking history and histologic type. Moreover, in the 302 NSCLC samples studied, 3.97% (12/302) were found to contain ROS1 fusion genes, with CD74-ROS1 fusion identified in 9 cases. There was no statistically significant difference in patients' gender, age, smoking history and histologic type. One non-smoking elderly female patient with pulmonary adenocarcinoma had RET gene fusion. None of the cases studied had concurrent ALK, ROS1 and RET mutations.
CONCLUSIONSThe ALK, ROS1 and RET fusion gene mutation rates in NSCLC are low, they represent some specific molecular subtypes of NSCLC. Genetic testing has significant meaning to guide clinical targeted therapy.

5.Effect of Si Junziwan on Expression of Lon Protein in Hippocampal CA3 Region of SAMP8 Mice

ObjectiveTo study the expression changes of Lon protein and mitochondrial dynamics-related protein in the hippocampus of SAMP8 mice and provide a theoretical basis for the treatment of Alzheimer's disease by invigorating the spleen and supplementing Qi. MethodEight 3-month-old SAMR1 mice were used as the normal group, and 32 3-month-old SAPM8 mice were divided into model group, western medicine group (0.013 g·kg-1), low-dose Si Junziwan group (3.24 g·kg-1), and high-dose Si Junziwan group (12.56 g·kg-1), with 8 mice in each group. The western medicine group was gavaged with donepezil, and the Si Junziwan low- and high-dose groups were gavaged with Si Junziwan for 30 days. The positioning navigation experiment of the water maze was started on the 25th day, and the space exploration experiment of the water maze was started on the 30th day. On the 30th day, the protein expression of mitofusin 2 (MFN2) was detected by immunohistochemistry, the expression of adenosine 5'-monophosphate (AMP)-activated protein kinase (AMPK) was detected by enzyme-linked immunosorbent assay (ELISA), the content of ATP was detected by colorimetry, the microstructure of neuron mitochondria was detected by electron microscope, and the expressions of Aβ protein, Lon protein, dynamin-related protein 1 (DRP1) protein, and MFN1 protein were detected by Western blot. ResultAs compared with the normal group, the latency escape period increased, the number of crossings decreased, the expression of AMPK increased, and the content of ATP decreased in the model group. The expressions of Aβ protein and DRP1 protein increased (P<0.01), whereas the expressions of Lon protein, MFN1 protein decreased in the model group (P<0.05,P<0.01), and MFN2 protein decreased. The vacuolation of mitochondria increased and the cristae broke in the model group. As compared with model group, the time of the latent escape period decreased (P<0.01), and the number of crossings increased in the low-dose and high-dose Si Junziwan groups (P<0.05). The expression of AMPK (P<0.01) decreased, the content of ATP increased (P<0.01), the expression of Aβ and DRP1 protein decreased (P<0.05, P<0.01), and the expression of MFN1 protein was up-regulated (P<0.05) in high-dose Si Junziwan groups. The vacuolation was more obvious in the low-dose Si Junziwan group, whereas the vacuolation was restored and the ridge was clear in the high-dose Si Junziwan group. ConclusionSi Junziwan treats Alzheimer's disease by up-regulating the protein expression of Lon, correcting the disorder of mitochondrial division and fusion protein, and changing the memory function of SAMP8 mice.
6.The prevalence of HIV, hepatitis C virus and syphilis and related factors among cross-border couples in Mangshi county, Dehong Dai and Jingpo Autonomous Prefecture of Yunnan province from 2017 to 2019.

Objective: To understand the prevalence of HIV, hepatitis C virus (HCV) and syphilis and related factors among cross-border couples in Mangshi county, Dehong autonomous prefecture, Yunnan province. Methods: From May, 2017 to April, 2019, 2 500 couples with 5 000 cross-border marriages were selected by using cluster sampling method. The demographic characteristics, AIDS-related health services, HIV, HCV, syphilis infection and other information were collected through questionnaires and laboratory tests. The influencing factors of HIV, HCV and syphilis infection were analyzed by multivariate logistic regression model. Results: A total of 2 500 couples with cross-border marriage were investigated, among which 2 438 (97.5%) couples were Chinese men with Myanmar women. The average age of 5 000 participants was (34.16±9.00) years. Most of them were minority groups (59.9%), farmers (98.5%), education years ≤6 years (81.4%), marriage years>3 years (80.0%), and from mountainous areas (61.7%). The HIV prevalence of Chinese and Myanmar populations was 1.7% (43/2 500) and 2.0% (49/2 500), respectively. The HCV infection rates were 2.0% (49/2 500) and 1.3% (32/2 500), respectively and the infection rates of syphilis were 0.4% (10/2 500) and 0.2% (4/2 500), respectively. There were no statistically significant differences in the prevalence of three diseases among Chinese and Myanmar populations (P>0.05). The multivariate analysis showed that compared with those aged ≤ 30 years, having lower AIDS awareness, never receiving HIV testing, without HCV and syphilis infection, HIV prevalence was higher among those aged>30 years (OR=3.21, 95%CI: 1.80-5.73), having higher AIDS awareness (OR=17.41, 95%CI: 4.27-70.91), receiving HIV testing (OR=4.93, 95%CI: 2.72-8.92), with HCV infection (OR=5.64, 95%CI: 2.72-11.70) and syphilis infection (OR=8.37, 95%CI: 1.63-43.08). Compared with those aged ≤ 30 years, having marriage years ≤ 3 years, and with HIV negatives, HCV infection rate was higher among those age>30 years (OR=3.02, 95%CI: 1.69-5.38), having marriage years>3 years (OR=2.24, 95%CI: 1.34-3.74), and with HIV positives (OR=6.69, 95%CI: 3.29-13.59). Compared with those having HIV negatives, the syphilis infection rate was relatively higher among participants with HIV positives (OR=9.07, 95%CI: 2.00-41.10). Conclusion: The prevalence of HIV, HCV, and syphilis among cross-border couples in Mangshi county, Dehong autonomous prefecture of Yunnan province is relatively high. Age, AIDS awareness, HIV testing history, and the length of marriage are associated with the HIV, HCV, and syphilis infection.
