1.Clinical Study on the Post Cerebral Infarction Depression Patients Treated with Acupuncture and Acupoint Injection of Baihui with the Injection of Herba Erigerontis

[Objective]To observe the clinical effect of the treatment with Acupuncture Baihui and Acupoint Injection of Baihui with the injection of Herba Erigerontis on the post cerebral infarction depression(PCID).[Methods] 260 PCID patients were randomly divided into 3 groups.The first group was the amitriptyline group (80 cases),the treatment with the basis of service on the cerebral infarction and plus oral amitriptyline.The second group was acupuncture group (80 cases),treated with the basis of service on the cerebral infarction and plus acupuncture Baihui treatment.The third group was acupuncture and acupoint injection group (100 cases),the treatment with the basis of service on the cerebral infarction and plus acupuncture and acupoint injection of Baihui with the injection of Herba Erigerontis.Observe the changes of HAMD,SDS,MeSSS,MMSE,Barthel Index (BI) score of 3 groups before and after treatment for 4 w,8 w,and assess the efficacy of the treatment.[Results] After treatment,HAMD,SDS,MeSSS,MMSE,Barthel Index (BI) score were significantly improved in 3 groups,among them,the acupuncture group and amitriptyline group were improved similar on the observation score,acupuncture and acupoint injection group was significantly better than acupuncture group and amitriptyline.[Conclusion] Initial examinations show that it can significantly relieve the symptoms of PCID depression by Baihui,and worthy of clinical application.
2.Effect of acupuncture of Baihui on neurotransmitter of post cerebral infarction depression

Objective:To observe neurotransmitter changes of post cerebral infarction depression (PCID) before and after being treated with amitriptyline or acupuncture of Baihui,in order to explore a good way with a less toxicity and good effect of treatment on PCID.Methods:The 60 patients were divided into treatment group and control group.Besides the basic treatment,the treatment group was given acupuncture of Baihui (DV20) and point injection therapy,the control group was given amitriptyline. Both before and after treatment,compared the two groups with HAMD score,neurologic impairment score,plasma 5-HT and NE,and observed the adverse events and side effects.Results:The results of two groups in monoamine neurotransmitter 5-HT(5-HT),norepinephrine (NE) and the HAMD scale were significantly improved.Conclusion:Acupunture of Baihui and point injection of Herba Erigerontis had equal effect to the amitriptyline on PCID.Furthermore,it had fewer venenosus side effects.It provided a new way for the use of the traditional Chinese medicine in treating PICD.
3.Health management of risk factors of chronic diseases in Nanhong community

Objective Chronic diseases are recognized as a major health problem in the 21st century.This project was to assess the effects of self-awareness health management service on optimization of life-style of community residents so as to effectively control the occurrence and development of chronic diseases.Method A survey of health information,an evaluation of health status,health intervention were conducted among 231 people aged between 29-65 years in a community for a half year,and the health management was effectively assessed of them.38 subjects with chronic diseases were subjected to diet friends used and exercise quantitative management model.Results The way of life of the 231 people has been optimized,and in the 38 patients with chronic diseases who had adopted the management mode of friends six months later their average body weight dropped by 1.3 kg,body mass index dropped by 0.5,and waist circumference reduced by 1.8 cm(P<0.05).Conclusion It is feasible to conduct health management in residents of communities,especially in patients with chronic diseases,which COnforms to the basic principles and requirements of prevention and treatment of chronic diseases advocated by WHO.
4.Effects of total glucosides of peony on expression of inflammatory cytokines and phosphorylated MAPK signal molecules in hippocampus induced by fibrillar Aβ42.

OBJECTIVETo observe the effects of hippocampal Abeta42 deposition on the expression of inflammatory cytokines and phosphorylated MAPK signal molecules as well as the intervention of AD by total glucosides of paeony (TGP).
METHOD12 week-old female SD rats were stereotactic injected one-time with a fibrillar Abeta42 positioning hippocampus to replicate AD pathology model and interfered with TGP. The expression of inflammatory cytokines and phosphorylated MAPK pathway signaling molecules were observed by immunohistochemistry (SABC), and SABC images were analyzed by image analysis software.
RESULTCompared with the control group, the IL-1beta, IL-6 and p-p38, p-JNK, p-MEK3/6 positive stained areas of AD pathology model group increased and their staining intensity decreased (the protein expression quantity inversely proportional to the staining intensity), while the IL-1beta, IL-6 and p-p38, p-JNK, p-MEK3/6 positive stained areas of the treatment groups decreased and their staining intensity increased compared with AD pathology model group.
CONCLUSIONAbeta42 deposition in hippocampus can induce the brain inflammation and the over-expression of IL-1beta, IL-6 and p-p38, p-JNK, p-MEK3/6. Inhibiting the over-expression of inflammatory cytokines and phosphorylated MAPK signaling molecules may be a major antagonistic mechanism of TGP against AD.

5.Detection of ascarid ITS-2 sequence out of intrahepatic bile duct stones by PCR technique

Objective To search for the molecular figngerprint of ascaris lumbricoides in intrahepatic stones. Methods In 56 cases deoxyribonucleic acid (DNA) was exstracted from intrahepatic stones and surgically obtained bile duct tissues, stool and sercum examinations were performed. Based on the sequence and designed primers of ITS-2 of ascaris lumbricoides, polymerase chain reaction ( PCR) was performed. Results There were 12 positive results in 56 (21.4% ) cases of intrahepatic stones. Positive reaction appeared on the examinations of the stool on ascaris lumbricoides and the blood serum among 12 cases. Conclusions Ascaris lumbricoides DNA was found in human of intrahepatic stones, indicating that ascaris lumbricoides infection may play a role in the formation of intrahepatic stones.
6.Simulation and measurement of diagnostic X-ray radiation qualities

Objective To validate the feasibility of the simulationmethod and the reliability of thesimulationresult through comparison between simulation and measurement of the energy spectrum from medical diagnostic X-ray (RQR-Radiation qualities in radiation beans emerging from the X-ray source assembly).Methods A simplified model of the medical diagnostic X-ray RQR radiation quality was established using code of BEAMnrc.The energy spectrum of the same RQR radiation quality were measured through a plane high-purity germanium spectrometer,and compared with the simulationresult.Results The difference of spectral distribution between measurement and simulation was less than 3%,in spite of the convolution processing not happened to the pulse height distribution measured by the spectrometer.And the spectral distribution,fluence,energy fluence,means energy distribution of the radiation was obtained using the code of BEAMDP.Conclusions As indicated above,it is possible to use the simulation of the energy distribution as a foundation for the establishment of X-ray RQR radiation quality.
7.A method for radiation dose assessment of β-rays and γ-rays in mixed β-γ fields

Objective:To test a new method with thermoluminescent dosimeters (TLDs) to determine the β-ray and γ-ray doses of β-γ mixed radiation fields.Methods:TLDs for personal dose monitoring were irradiated in the reference radiation fields of β-rays ( 90Sr/ 90Y, 85Kr) and γ-rays ( 137Cs). Across the range of 2.0-15.0 mSv, the linearity of TLD response and normalized response with respect to 137Cs were determined at the depths of Hp(10) and Hp(0.07). Using TLD detector readings at the depths of Hp(10) and Hp(0.07), β- and γ-ray doses in the mixed radiation fields were determined, and the result were verified. Results:For Hp(10) and Hp(0.07) under γ-ray exposure and Hp(0.07) under β-ray exposure, the coefficient of determination ( R2) were all >0.998. For the 90Sr/ 90Y source and 85Kr source, the average values of response values normalized with respect to 137Cs at different doses were 1.14 and 0.18, respectively; and the normalized response values derived from the slope values of the dose-response curves for the two sources were 1.17 and 0.18, respectively. The ratios of measurements of Hp(10) to Hp(0.07), kR, for the 85Kr source were close to 0, while the kR values for the 137Cs source were close to 1. Using the average value of kR and the slope value of kR for calculation, the maximum relative deviations between the calculated values and conventional values for Hp(10) γ, Hp(0.07) γ, and Hp(0.07) β were 6.1% and 6.0%, respectively. Conclusions:This method can be applied for the assessment of β-ray and γ-ray doses in β-γ mixed radiation fields of a single β source and single γ source.
8.Generation and identification of pluripotent stem cells from NOD mouse

Three factors ( Oct4,Sox2,and Klf4) were introduced into tail tip-derived fibroblasts of NOD mouse by retrovirus infection.The induced pluripotent stem cells ( iPSCs ) generated from this method were analyzed in several aspects,including surface antigens,gene expression,alkaline phosphatase activity,and teratoma formation assays.The NOD mouse iPSCs generated from this study had ES-like characters,expressed ES-specific surface antigens,and possessed the ability to differentiate into 3-germ layers.Such NOD mouse-iPSCs should be useful in tyre 1 dialectic disease modeling,as well as for cell replacement therapy.
9.Optimization of Extraction Technology of Total Polyphenols and Total Flavonoids in Gardenia jasminoides by Response Surface Method

OBJECTIVE:To optimize the extraction technology of total polyphenol and total flavonoid in Gardenia jasminoi-des. METHODS:Plackett-burman(PB)design was used to select the ethanol volume fraction,liquid-solid ratio,particle size,ex-traction time and extraction temperature to determine the key factor affecting the extraction of total polyphenol and total flavonoids in G. jasminoides. Then central composite design (CCD) was combined with response surface method to optimize the extraction technology,and verification test was conducted. RESULTS:The optimal extraction conditions of total polyphenol were 40% etha-nol,particle size of 0.20 mm,extraction temperature of 60 ℃,liquid-solid ratio of 20,and extraction time of 20 min;the optimal extraction conditions of total flavonoids were 40% ethanol,particle size of 0.20 mm,extraction temperature of 30 ℃,liquid-solid ratio of 20,and extraction time of 20 min. In verification test,the contents of total polyphenol and total flavonoids in G. jasminoi-des were 1.70%(RSD=1.43%,n=3),3.23%(RSD=3.72%,n=3),with relative error of 1.80%,8.75% with predicted val-ues,respectively. CONCLUSIONS:The response surface method based on PB and CCD is simple,reasonable and feasible to opti-mize the extraction technology of total polyphenol and total flavonoids in G. jasminoides. The method can provide reference for its industrial extraction.
10.Effects of signaling-selective parathyroid hormone peptide analog on fracture healing in orchiectomized mouse models.

OBJECTIVETo assess the effect of intermittent subcutaneous injections of signal-selective parathyroid hormone (PTH) peptide analog on fracture healing in orchiectomized mouse models.
METHODSThirty-six 7-week-old C57/BL male mice were orchiectomized and injected with hPTH(1-34), the signal-selective PTH peptide analog [Gly(1), Arg(19)]hPTH (1-34), or an identical volume of vehicle 1 week after induction of femoral fracture. At 14 and 28 days after the operation, the mice were sacrificed for measurement of bone mineral density (BMD) and bone mineral content (BMC) of the callus using by dual energy X-ray absorptiometry. The bone healing was evaluated by radiography, biomechanical testing, micro-computed tomography (Micro-CT) and histological examination.
RESULTSAt 14 days after the operation, BMD in PTH peptide analog group was significantly increased (P<0.05). The mouse models treated with the PTH peptide analog showed significantly lower ultimate bending force and bending rigidity than those with hPTH(1-34) treatment. X-ray and Micro-CT scanning showed that callus transformation and remodeling was better in PTH peptide analog group than in the vehicle control group but poorer than in hPTH(1-34) group.
CONCLUSIONThe signaling-selective PTH peptide analog G1, R19 (1-28) can accelerate fracture healing in orchiectomized mouse models, in which process cAMP/PKA pathway plays an important role.
