1.Puerarin Injection- induced Hemolytic Reactions:Literature Analysis of 44 Cases

OBJECTIVE:To probe into the regular pattern,related factors and prognosis of hemolytic reactions induced by Puerarin injection.METHODS:A total of 44 hemolytic cases induced by Puerarin injection reported in 26 papers of domestic journals published since 1994 were analyzed.RESULTS:Of the 44 cases collected,31 were males and 13 females,with a mean age of(67.5?9.6) years.The hemolytic reactions were induced either by Puerarin injection or Puerarin for injection with a mean dose of(445.2?123.8)mg and a mean concentration of puerarin(in infusion) of(1.55?0.52) mg?mL~(-1).The hemolytic reactions occurred chiefly after administration of puerarin for 7 consecutive days(7.96?3.84 days on average) and appeared as acute hemolytic anemia.The puerarin specific antibody was noted in immunological test.10 patients including 9 males and 1 male died at a mean age of(72.1?5.7) years.CONCLUSION:The occurrence and turnover of Puerarin Injection - induced hemolytic reactions are related to many factors,to which great attention should be paid.
2.Expression of Id3,VEGF and C-erbB-2 and clinical significance in patients with breast cancer

Objective To investigate the correlation between clinicopathological parameters and the expressions of inhibitor of differentiation/DNA binding 3 (Id3), vascular endothelial growth factor (VEGF) and human epidermal growth factor receptor-2 (HER-2/C-erbB-2) in patients with breast cancer. Methods The protein expressions of Id3, VEGF and C-erbB-2 were detected by SABC immunohistochemistry in 70 spceimens of breast cancer, 40 specimens of tissue adjacent to breast cancer, 20 specimens of fibroadenoma and 20 specimens of normal breast tissue. Objective To observe the influence of the coumarins from Radix Angelicae pubescentis (TCA) on the hypnotic effects of pentobarbital sodium and barbital sodium. Methods Mice were randomly divided into solvent (5% Tween-80, 20 mL/kg), chloromycetin (25 mg/kg) and TCA (50, 100 mg/kg) groups, which were administered orally with equivalent volume. One hour later, all the animals were given pentobarbital sodium (40 mg/kg, ip) or barbital sodium (200 mg/kg, ip), and then the latent period and duration of hypnosis were observed. 4.Study on the Antioxidative Effect of the Extracts of Codonopsis Lanceolata from Mount Taishan in Mice

Objective To study the antioxidative effect of the ethanol extract (e-EXT) and aqueous extract (a-EXT) of Codonopsis lanceolata from Mount Taishan in mice. Method The mice were divided into six groups at random. After the mice were orally treated with 5% Tween-80 (solvent,20 mL/kg),Vitamin C (0.1 g/kg),e-EXT (10,20 g/kg) and a-EXT (10,20 g/kg) once daily for 14 days respectively,the blood,liver and brain were collected for determining SOD,GSH-Px,GSH and MDA levels. Results The SOD,GSH-Px and GSH levels were enhanced significantly,and the MDA levels were reduced markedly in serum,liver and brain homogenate of the mice treated with 20 g/kg e-EXT and a-EXT. In the serum and liver homogenate of 10 g/kg a-EXT group,the levels of SOD,GSH-Px and GSH were increased,the contents of MDA were decreased. Conclusion The ethanol extract and aqueous extract of Codonopsis lanceolata from Mount Taishan could enhance the antioxidative ability of the mice,especially the large-dose aqueous extract.
5.Content Determination of Total Flavonoids and Polysaccharides in Radix Codonopsis Lanceolatae from Mount Taishan.

Objective To establish the method for content determination of total flavonoids and polysaccharides in Radix Codonopsis Lanceolatae from Mount Taishan.MethodsThe content of the total flavonoids was determined by UV spectrophotometry and the polysaccharide content was measured with the phenol-sulphuric acid method.ResultsThe optimal detective wavelength was 510nm for the total flavonoids.Satisfactroy linear relationship with the Absorbability was achieved when the mass concentration of rutin was within a range of 8~48?g/ml(r=0.9986).The average recovery rate was 99.31%(RSD=1.82%,n=5).The optimal detective wavelength was 490nm for the polysaccharides.There was good linear relationship between the Absorbability and the content of glucose in a range of 5~50?g/ml(r=0.9964) with an average recovery of 99.12%(RSD=1.49,n=4).ConclusionThis method is sensitive,simple and accurate,and it can be used for the quality control of Radix Codonopsis Lanceolatae from Mount Taishan.

Objective To investigate the association between the difference of specific cytotoxic lymphocyte (CTL) and liver function of patients with hepatitis B virus (HBV) genotype B and C infections and interleukin (IL)‐7 induced follicular helper T lymphocytes (Tfh) .Methods Sixty‐seven patients with chronic hepatitis B (CHB) hospitalized at Wuxi No .5 People′s Hospital from August 2013 to January 2015 were collected and 30 healthy blood donors were set as healthy control group .The peripheral blood IL‐7 , Tfh ,IL‐21 ,HBV specific‐CTL ,nonspecific CTL ,levels of HBV DNA ,alanine aminotransferase (ALT) and total bilirubin (TBil) were compared between patients with genotype B and C infection ,hepatitis B e antigen (HBeAg)‐positive and HBeAg‐negative CHB ,high ALT level and low ALT level .Multivariate regression analysis was performed to determine the factors associated with IL‐7 .The t test was used for quantitative data and chi‐square test was used for categorical data .Results Of the 67 CHB patients with average age of (35 .1 ± 11 .4) ,48 were male and 19 were female;32 were infected with genotype C and 35 were infected with genotype B ;40 were HBeAg‐positive CHB and 27 were HBeAg‐negative CHB ;17 were with high ALT levels and 50 were with low ALT levels .IL‐7 ,Tfh ,IL‐21 and HBV specific‐CTL levels in the peripheral blood of genotype C‐infected patients were (20 .79 ± 4 .82 ) ng/L , (3 .93 ± 0 .82)% ,(24 .77 ± 7 .52) ng/L and (0 .20 ± 0 .04)% ,respectively ,while in genotype B‐infected patients , those were (29 .13 ± 8 .17) ng/L ,(5 .92 ± 1 .92)% ,(39 .94 ± 24 .00) ng/L and (0 .40 ± 0 .06)% , respectively .Levels of IL‐7 , Tfh ,IL‐21 and HBV specific‐CTL in genotype C‐infected patients were significantly lower than those in genotype B‐infected patients (t= 5 .027 ,5 .595 ,3 .553 and 15 .133 , respectively ;all P<0 .01) .Nonspecific CTL ,HBV DNA ,ALT and TBil levels in the peripheral blood of genotype C‐infected patients were all significantly higher than those in genotype B infected‐patients (t=4 .899 ,6 .815 ,2 .763 and 4 .899 ,respectively ;all P< 0 .01) .IL‐7 ,Tfh ,IL‐21 ,HBV specific‐CTL levels in the peripheral blood of HBeAg‐positive patients were significantly lower than those in HBeAg‐negative patients (all P<0 .01) .Nonspecific CTL ,HBV DNA ,ALT and TBil levels in the peripheral blood of HBeAg‐positive patients were all significantly higher than those in HBeAg‐negative patients (all P<0 .05) .IL‐7 ,Tfh ,IL‐21 ,HBV specific‐CTL levels in the peripheral blood of patients with high ALT levels were all significantly lower than those in patients with low ALT levels (all P<0 .01) .Nonspecific CTL and HBV DNA levels in the peripheral blood of patients with high ALT levels were both significantly higher than those in patients with low ALT levels (both P<0 .01) .HBV DNA ,IL‐21 and nonspecific CTL were all correlated with IL‐7 (all P<0 .01) .Conclusion The differences of HBV specific‐CTL and liver function in CHB patients infected with genotype B and C may be correlated with interleukin‐7 induced Tfhcells.
7. Effects of ICAT silencing in Wnt signaling pathway and NSC67657 induced cell differentiation of HL-60 cells

Objectives:
To investigate the effect of β-catenin interacting protein 1 (ICAT) silence in Wnt signaling pathway and sterol drug NSC67657 induced cell differentiation of HL60 cell.
Methods:
HL-60 cells were treated with NSC67657, the cell surface antigen CD14 expression was detected by flow cytometry. Lentivirus LV-ICAT-RNAi vector was constructed and infected HL-60 cells. Then the ICAT gene and protein expression were analyzed using real-time qPCR and Western blot technique. Furthermore, Co-immunoprecipitation assay was used to confirm the interaction of β-catenin/ICAT proteins, and Western blot was employed to compare the expressions of Wnt signaling pathway downstream targets Cyclin D1, TCF-1 and c-Jun between Lentivirus LV-ICAT-RNAi vector infected HL-60 (HL-60i) cells and un-infected HL-60 (HL-60v) cells. The cellular differentiation of HL-60i and HL-60v cells treated with NSC67657 for 24 h was evaluated by Wright’s staining, transmission electron microscopy and flow cytometry analysis.
Results:
HL-60 cells could be induced to differentiate into monocytes by 10 μmol/L NSC67657. The CD14 positive cells could reach to (92.30±5.14) % after NSC67657 treatment for 5d. The co- immunoprecipitation assay demonstrated that ICAT protein did interact with β-catenin protein, and the absorbance of protein electrophoresis bands increased in differentiated cells. The expressions of Wnt signaling pathway downstream target proteins in HL-60i cells were higher than that in HL-60v cells when they were treated by 10 μmol/L NSC67657, but lower than NSC67657 untreated cells. 8.Establishment of LC-MS/MS method for the determination of forsklin in rat plasma and its pharmacokinetics

Objective To develop a sensitive liquid chromatography-tandem mass spectrometric (LC-MS/MS) method for the determination of forsklin in rat plasma.Methods After extraction with methyl tert-butyl ether,chromatographic separation was performed on a C18 column with the mobile phase consisting of water ( 0.1% formic acid)-acetonitrile in a gradient elution mode.A tandem mass spectrometer equipped with electrospray ionization (ESI) source was used as detector in the positive ion mode.Quantification was performed using multiple reaction monitoring (MRM) with the precursor product combination ions of m/z 411→375.3 and 285→193 for forsklin and diazepam.Results Good linearity was obtained in the 0.5-1000 ng/ml range for the analyte and the analytical method was validated in terms of specificity,precision,accuracy,recovery,stability and matrix effect.These assays gave RSD values always lower than 14.4% and RE values between -3.5 % and 3.8%.In addition,the specificity,extraction recovery,stability and matrix effect were satisfactory.Conclusion Due to its high sensitivity,specificity and simplicity,the method could be used for pharmacokinetic studies of forsklin.
9.Atlanto-axial pedicle screw internal fixation to reconstruct upper cervical spine instability

Objective To explore the clinical effect of the atlanto-axial pedicle screw internal fixation to reconstruct upper cervical spine instability. Methods Using atlanto-axial pedicle screw and internal fixation system combined with autogenous iliac bone graft to treat 21 patients with upper cervical spine instability. Type Ⅱ odontoid process old fracture was 13 patients, odontoid process nonunion was 5 patients, injury of the transverse ligament was 3 patients. They were placed in 84 atlanto-axial pedicle screws, autogenous bones were placed in the posterior arch of atlas and axis to fusion, so that atlanto-axial complex got stabilized. Results The left lateral cortical bone of atlas vertebral pedicle was broken by screws in 2 patients, but the spinal cord and vertebral artery was intact. X-ray film showed the atlas and the fracture of dens of axis was completely replaced in all patients. The position of screw and vertebral artery or spinal cord was good in CT image. According to JOA score standard, 15 cases of all were excellent, 3 cases were good,2 cases were common,1 case was bad, and the rate of excellent and good was 85.71%(18/21).Seventeen cases were followed up for 12-26 (12.83 ± 4.23) months, all patients had acquired bone fusion and found no screw and plate fracture. Conclusions Atlanto-axial pedicle screw combined with screw-plate system fixation to treat upper cervical spine instability, can significantly enhance the biomechanical stability of the atlanto-axial interbody, bone grafting fusion rate is higher, the application value of the atlanto-axialfusion is higher.
