1.Analysis of laboratory external quality control of iodine deficiency disorders in Guizhou province in 1999 -2008

Objective To analyze the assessment results of iodine deficiency disorders(IDD) External Quality Control Laboratories at all levels in Guizhou province and the network operation to further standardize and improve the laboratory, improve network performance, provide reliable laboratory quality assurance for epidemiological surveillance and control of IDD and reliable decision-making. Methods The results of different level of IDD labs in Guizhou that took part in the entire country's IDD lab exo-network quality control examination of 1999 - 2008, which organized by the nation iodine deficiency disorder reference lab were analyzed. Results In the past 10 years, except the provincial laboratory examination results of urinary iodine in 2000 was failed, the other results were all qualified in the rest 9 years;iodized salt examination results were qualified. The urinary iodine laboratory response rate of Guizhou provinces and municipalities(state, district), from 1999 to 2002, were 88.9%(8/9), 66.7%(6/9), 77.8%(7/9), 66.7%(6/9), respectively, and the rate was stable at 100% from 2003 to 2008. Qualifying rate reached 100% in 2007, the remaining 9 years were 33.0%(3/9) - 88.9%(8/9). The iodized salt laboratory response rates were 100%(9/9) in 2000 - 2008. The pass rates were 77.8%(7/9), 88.9%(8/9),77.8%(7/9) from 2001 to 2003, 88.9%(8/9) in 2007, and the remaining 5 years 100%. Response rate of iodized salt laboratory at the county level that participated in the External Quality Control were 66.7% (20/30), 90.0%(27/30), 80.0% (24/30), 96.7% (29/30) from 2000 to 2003, respectively, and 2006 was 96.7% (29/30), and the remaining four years were all 100% (30/30). The pass rates in 2000 - 2008 were between 53.3% (16/30) -93.3%(28/30). Conclusions The accuracy of test results of external quality controls and the normal operation of the network at all levels of laboratories is closely related to the IDD laboratory conditions, detection techniques, and the degree of attention of relevant departments and professional bodies.
2.Relationship between combined multigene detection and response to chemotherapy and prognosis in epithelial ovarian carcinomas

Objective To evaluate the relationship between combined muhigene detection and response to chemotherapy and prognosis in epithelial ovarian carcinomas(EOCS).Methods A total of 80 ovarian tissue samples taken from the surgical specimens of patients with EOCS of our hospital in the last two decades who had received chemotherapy "after surgery were paraffin-embedded.The samples were divided into 2 groups,good prognosis group(patients who survived more than 2 years,n=46)and poor prognosis group(patients who survived less than 2 years,n=34).The expression levels of ToPo-Ⅱ,Ki-67,MGMT, PCNA,p27,p53,pl6,P-gp,LRP,GST-?,bcl-2,C-myc,Fas,bax,MSH2,MRP and BCRP were investigated by the combination of tissue arrays and immunohistoehemical streptavidin-biotin peroxidase(SP) method in all samples.Data were analysed with SPSS 12.0 for windows.Results There were statistically significant differences in the positive expression levels of P-gp,BCRP,MGMT,MSH2,p27 and p16(62%, 50% and 50% in poor prognosis group vs 33%,28% and 28% respectively,P

Objective To investigate the relationship between the imbalance of suppressors of cytokine signaling 1(SOCS1)/SOCS3 and abnormal activation of monocytes/ macrophages in children with acute-phase asthma, and explore the molecular mechanism of chronic inflammatory process on airway.Methods The present study enrolled 20 asthmatic children and 20 age-matched normal children. Dual-color flow cytometric analysis was performal to detect the percentage of CD_ 80 、CD_ 86 expressing on CD_ 14 ~+ cell. 4.Prevention and treatment of erectile dysfunction during peri-operation period of radical retropubic prostatectomy.

The incidence of prostatic cancer rises with increasing age and early diagnosis rate of the disease keeps increasing at the same time. Early diagnosis provides opportunities for radical prostatectomy to cure prostatic cancer. However, the problem of erectile dysfunction (ED) post operation puzzles urologists and exerts negative impacts on the patients life quality. Study on the prevention and treatment of peri-operational ED is of great importance to dwindle the occurrence rate of ED and is the key to improve the life quality of such patients.

5.Combination of OCT and FFA for the research of pathogenesis of central serous chorioretinopathy

Background The diagnosis of central serous chorioretinopathy (CSC) is mainly dependent onfluorescine fundus angiography (FFA). However, the combination of optical coherence topography (OCT) with FFA offers a new approach to the research of the pathogenesis of CSC. Objective This clinical study was designed to study the combined application of the FFA and OCT for the research of the pathogenesis of central serous chorioretinopathy (CSC). Methods Forty-four eyes of 44 patients with CSC were included in this study with 36 cases of males and 8 cases of female. The patients were aged 39.3 ± 5.3 years and the visual acuity was 0. 64 ±0. 27. FFA and OCT examinations were performed in all patients and the FFA images were imported into the Topcon 3D OCT 1000 device to locate the conformity of OCT lesions with the leakages of FFA. The neuroepithelial layer thickness at the fovea and the height of the neuroepithelial layer detachment were measured using 3-D OCT. Results OCT showed serous REP detachment in 34 eyes (77.3%) and rough surfaces of RPE in 10 eyes (22. 7% ). In thirtyfour eyes with RPE detachment, the OCT lesions and FFA leakage spots conformed to the same locations in 31 eyes, but the other three eyes did not. The mean foveal neuroepithelial thickness was (138.5±19.4) μm in CSC eyes and that of normal eyes was ( 131.35±5. 01 ) μm ,showing a significant difference between them( t=0. 39 ,P>0. 05 ). The mean height of neuroepithelial detachment was (263.3 ± 126.7 ) μm in CSC eyes. Conclusion RPE detachment occurs in CSC eyes and further induces macular neuroepithelial detachment. Leakage lesion of fluorescine corresponds to RPE detachment. CSC without RPE detachment may be related to the increase in RPE permeability. OCT can accurately measure the thickness of the macular neuroepithelial layer and the height of the neuroepithelial detachment.

7.Determination of plasma EBV DNA in 91 children with EBV-associated diseases.

OBJECTIVETo determine the plasma level of Epstein Barr virus (EBV) DNA in children with EBV associated diseases, and to investigate the dynamic changes of EBV DNA level after initial infection as well as the relationship between EBV DNA level and the diseases severity.
METHODSThe subjects consisted of 73 children with primary EBV infection (infectious mononucleosis, pneumonia,etc.) and 18 children with severe EBV-associated diseases (chronic active EBV infection, hemophagocytic lymphohistiocytosis, etc.). The plasma EBV DNA level was detected by a real-time PCR assay.
RESULTSThe plasma EBV DNA level decreased with the infection time in children with primary EBV infection. Two weeks after infection, plasma EBV DNA was almost undetectable. The positive rate of plasma EBV DNA in children with severe EBV associated diseases increased significantly when compared with that in children with primary EBV infection (89% vs 16%; p<0.05).
CONCLUSIONSThe level of EBV replication may be reduced with the infection time. Dynamic determination of blood EBV DNA is useful for the evaluation of disease severity in children with EBV infection.

8.Construction of pcDNA3.1AM and expression of adrenomedullin in mammalian cells.

The newly discovered endogenous vasodilating and diuretic peptide adrenomedullin (AM) was considered to be of attractive value in clinical treatment of hypertension and congestive heart failure. In order to explore the treatment of cardiovascular diseases by expressing AM in vivo, AM cDNA was inserted into mammalian expressing vector pcDNA3.1, and in vitro expression of AM was carried out in cultured K(562) cell line. AM mRNA was amplified by RT-PCR from the total RNA isolated from the adrenal glands of rats and was inserted into pcDNA3.1 vector to form pcDNA3.1AM, the recombinant pcDNA3.1AM was then transferred into cultured K(562) cell line by liposome. The expression of AM in pcDNA3.1AM transferred cell was identified by RT-PCR and dot immunoblot assay. The results demonstrated that there were AM mRNA in the pcDNA3.1AM-transferred K(562) cell line and AM peptides in the culturing medium, indicating that the recombinant pcDNA3.1AM vector can express AM in mammalian cell line.

9.Hysterectomy after cardiopulmonary resuscitation in patients with obstetric hemorrhagic shock.

OBJECTIVETo explore the indication of hysterectomy after successful resuscitation of cardiac arrest due to obstetric hemorrhagic shock.
METHODSA retrospective analysis was conducted in 13 patients with cardiac arrest due to obstetric hemorrhagic shock in 7 hospitals of Guangzhou, including 12 patients undergoing hysterectomy and 1 undergoing uterine artery embolization.
RESULTSs After successful cardiopulmonary resuscitation, only 4 of the 13 patients undergoing hysterectomy or uterine artery embolization for continuing uterus hemorrhage survived.
CONCLUSIONDetailed plans and emergency measures should be formulated in the management of high-risk pregnancies. Early diagnosis and active treatment of obstetric hemorrhagic shock with hysterectomy or uterine artery embolization are critical in preventing cardiac arrest and improving the survival of the patients.

10.Study of sexual hormone changes in prostate cancer patients before and after androgen deprivation therapy.

OBJECTIVESTo investigate and observe serum sexual hormone (testosterone, T; free testosterone, FT; dihydrotestosterone, DHT) changes in patients with prostate cancer (PCa) before and after androgen deprivation.
METHODSSerum T, FT, DHT were measured with radioimmunoassay in 16 cases with PCa respectively one week before and the fifth day after androgen deprivation. The changes of T, FT, DHT were analyzed.
RESULTSSerum T, FT, DHT declined 92.27%, 92.26%, 58.36% respectively. They were statistically significant before and after androgen deprivation (P < 0.001).
CONCLUSIONSAfter androgen deprivation, most T and FT were deprived while DHT declined 58.36% only. Androgen receptor competitor shall be applied continually to prohibit the action of remnant androgen.
