1.Cardiac function change of healthy people after migration from plain to plateau by color Doppler echocardiography

Objective To investigate the relationship of cardiac function and ventricular structure with the training intensity and the periods lived in Tibet.Methods Sixty recruits to Tibet were selected and examined of their cardiac function,cardiac ventricular structure and the changes of elctrophysiology by color Doppler echocardiography and electro cardiogram.They were examined continuously in two years.After their training for three months together,they were divided into the different training intensity groups,Group A and Group B.The 30 recruits in Group A had high-intensity training,and the others in Group B had normal training.Results After three months,their cardiac function and ventricular structure had obvious difference in comparison with the level on plain( P 0.05 ).But there were obvious differences between in Tibet and on plain in the parameters of cardiac function and ventricular structure two years later( P 0.05 ).Conclusions Hypoxia can effect the cardiac function and ventricular structure directly.After three months in Tibet,cardiac function can fit the condition of hypoxia.The parameters of cardiac function are still less than those on plain.The right ventricules are bigger and the pulmonary arteries wider as the main character of the ventricular structure changes.The indexes of cardiac function showed no significant difference with the training intensity and the periods in Tibet.
4.Rehabilitation of diabetic peripheral neuropathy with integrated traditional Chinese medicine therapy

ObjectiveTo observe the effect of integrated traditional Chinese medicine therapy on diabetic peripheral neuropathy.Methods22 patients with diabetic peripheral neuropathy were as the group of integrated traditional Chinese medicine therapy ,who received Chinese herbal medicine treatment(orally),acupuncture,etc. Other 20 cases were as the control group,disease natural course. The effect of two groups were compared with the means.ResultsThe patients in group of integrated traditional Chinese medicine therapy got obviously better results than that of the control group(P<0.05).ConclusionThe integrated traditional Chinese medicine therapy is effective on diabetic peripheral neuropathy.
5.Mutation detection in the interaction domains of NRF2 and KEAP1 in lung cancer cell lines

Objective To detect the interaction domains' gene mutations of KEAP1 and NRF2 in non-small-cell lung cancer cell lines,and to analyze the significance of these mutations on the study of lung cancer cell lines.Methods Six non-small-cell lung cancer cell lines were used as the research materials.The 2nd to 6th exons of KEAP1 and the coding sequences of DLG and ETGE motif were amplified by PCR,and the products were used for gene sequencing analysis.Obtained gene sequences were analyzed using NCBI databases to get the base locations of gene mutations,as well as the subsequent amino acid sequence changes.Meanwhile,the relative intracellular reactive oxygen species (ROS) concentrations in the tested cell lines were detected and used in the analysis of abnormal NRF2 transcriptional activity in lung cancer cell.Results Four mutations were detected in the 4th exon of KEAP1 gene from all the 6 cancer cell lines,several other missense mutations were also investigated in the 3rd exon of KEAP1 from some cancer cell lines.Multiple genetic variations were found in all the NRF2-ETGE motif-encoding sequences of the 6 cancer cell lines.All 6 cancer cell lines were found to have higher ROS concentration than the lung germ cell line.Conclusion Lung cancer cells generally contain high levels of ROS as well as gene mutations in KEAP1 and NRF2 genes which lead to abnormal transcriptional activity of NRF2 in lung cancer cell lines.

8.Injury of fetal brain, lung and liver caused by maternal human cytomegalovirus infection before pregnancy in mice

Objective To investigate the injury of the brain, lung and liver of the pups after maternal infection of human eytomegalovirus (HCMV) 3 days before pregnancy. Methods Thirty kunming mice aged 10-12 weeks were mated and divided into infection group and control group. The female mice in the infection group were given intraperitonealty 1×106 50% tissue culture infective dose of HCMV suspension 0. 5 ml 3 days before pregnancy, while those in the control group were given intraperitoncally supernatant of human fibroblasts. At 19 days of gestation, cesarean sections were performed. Pathology and HCMV DNA of the brain, lung and liver tissue of the fetus were tested through microscope and PCR. Kai-square test was used to compared the positive rates of HCMV DNA and viral isolation between these two groups. Results The positive rates of HCMV viral isolation and DNA in the fetal mice of infection group were as follow: brain (73.3% and 93.3%), liver (53.3% and 93.3%)and lung (60. 0% and 86.7%), while HCMV DNA and viral isolation was not detected in the control group (P<0.05). Under microscope, viromicrosome and tissue injury including necrosis, degeneration and bleeding were found in the tissues of brain, lung and liver in the infection group. Conclusions Maternal mice HCMV infection before pregnancy might vertically transmitted to the fetus and cause multi-organ injury.
9.Assessment of Complications after Liver Surgery: Two Novel Grading Systems Applied to Patients Undergoing Hepatectomy

Although quality assessment is gaining increasing attention,there is still no consensus on how to define and grade postoperative complications.The absence of a defimition and a widely accepted ranking system to classify surgical complications has hampered proper interpretation of the surgical outcome.This study aimed to define and search the simple and reproducible classification of complications following hepatectomy based on two therapy-oriented severity grading system:Clavien-Dindo classification of surgical complications and Accordion severity grading of postoperative complications.Two classifications were tested in a cohort of 2008 patients who underwent elective liver surgery at our institution between January 1986 and December 2005.Univariate and multivariate analyses were performed to link respective complications with perioperative parameters,length of hospital stay and the quality of life.A total of 1716 (85.46%) patients did not develop any complication,while 292 (14.54%)patients had at least one complication.According to Clavien-Dindo classification of surgical complications system,grade Ⅰ complications occurred in 150 patients (7.47%),grade Ⅱ in 47 patients (2.34%),grade Ⅲa in 59 patients (2.94%),grade Ⅲb in 13 patients (0.65%),grade Ⅳa in 7 patients (0.35%),grade Ⅳb in 1 patient (0.05%),and grade Ⅴ in 15 patients (0.75%).According to Accordion severity grading of postoperative complications system,mild complications occurred in 160 patients (7.97%),moderate complications in 48 patients (2.39%),severe complications (invasive procedure/no general anesthesia) in 48 patients (2.39%),severe complications (invasive procedure under general anesthesia or single organ system failure) in 20 patients (1.00%),severe complications (organ system failure and invasive procedure under general anesthesia or multisystem organ failure) in 1 patient (0.05%),and mortality was 0.75% (n=15).Complication severity of Clavien-Dindo system and Accordion system were all correlated with the length of hospital stay,the number of hepatic segments resected,the blood transfusion and the Hospital Anxiety and Depression Scale-Anxiety (HADS-A).The Clavien-Dindo classification system and Accordion classification system are the simple ways of reporting all complications following the liver surgery.
10.Rapidly detect and distinguish between norovirus G I and G II type with a pair of primers.

OBJECTIVEThe purpose of this study was to develop RT- PCR assay for Rapidly detect and distinguish between Norovirus genogroup I and genogroup II with a pair of primers.
METHODSA pairs of primers specific to capsid prote in ORF2 gene of G I and G II Norovirus were dsigned according to the published complete genome sequence, with which the RNA of Norovirus was extracted and RT-PCR amplification. The sensitivity, specificity of the RT- PCR assay was estimated and apply it to the detection of Norovirus in clinical specimens.
RESULTSThe results showed that the assay possessed high specificity for Norovirus detection and without any evident cross-reaction with other viruses, including rotavirus, enteric adenovirus and hepatitis A virus. The detection limit of RT-PCR assay for Norovirus G I and G II were up to 100 pg/ml and 10 pg/ml respectively.
CONCLUSIONThe RT- PCR assay provide rapid and sensitive detection of Norovirus G I and G II and should prove to be useful for Norovirus diagnosis in the outbreaks of acute gastroenteritis.
