1.Research advances in biomarkers for early prediction of preeclampsia

Preeclampsia is a leading cause of maternal and infant morbidity and mortality. It is very important to explore the biomarkers for the early prediction of preeclampsia. Some peptides released from placenta, such as soluble Flt-1 and placenta growth factor (PlGF), have been revealed for definite prospects of application. Meanwhile, the recent advances in proteomics, metabolomics and microRNA shed light on searching of new biomarkers for preeclampsia prediction.
2.Interaction between CYP450 enzymes and metabolism of traditional Chinese medicine as well as enzyme activity assay.

Drugs are exogenous compounds for human bodies, and will be metabolized by many enzymes after administration. CYP450 enzyme, as a major metabolic enzyme, is an important phase I drug metabolizing enzyme. In human bodies, about 75% of drug metabolism is conducted by CYP450 enzymes, and CYP450 enzymes is the key factor for drug interactions between traditional Chinese medicine( TCM) -TCM, TCM-medicine and other drug combination. In order to make clear the interaction between metabolic enzymes and TCM metabolism, we generally chose the enzymatic activity as an evaluation index. That is to say, the enhancement or reduction of CYP450 enzyme activity was used to infer the inducing or inhibitory effect of active ingredients and extracts of traditional Chinese medicine on enzymes. At present, the common method for measuring metabolic enzyme activity is Cocktail probe drugs, and it is the key to select the suitable probe substrates. This is of great significance for study drug's absorption, distribution, metabolism and excretion (ADME) process in organisms. The study focuses on the interaction between TCMs, active ingredients, herbal extracts, cocktail probe substrates as well as CYP450 enzymes, in order to guide future studies.

3.Cloning of M and NP Gene of H5N1 Avian Influenza Virus and Immune Efficacy of their DNA Vaccines

The M and NP genes of H5N1 avian influenza virus (A/chicken/Hubei/489/2004) were amplified by RT-PCR from viral RNA,and cloned into pMD 18-T vector respectively.The expression plasmid containing the M gene (pHM6-m) or the NP gene (pHM6-np) was then constructed by inserting the M or NP gene into the pHM6 eukaryote expression vector; the constructed plasmid was then sequenced.32 BALB/c mice (6-week-old) were divided into four groups at random.Three groups of BALB/c mice were inoculated one time the intramuscular route with either 30 μg of plasmid pHM6-m,30 μg of plasmid pHM6-np or the mixture of plasmid pHM6-m (15 μg ) and pHM6-np(15 μg) respectively.A additional group of mice were injected with 100 μ1 PBS as controls.Two weeks later,all mice were challenged with homologous H5N1 avian influenza virus,and observed in the following 12 days.The survival rates of mice in the pHM6-m group,the pHM6-np group and mixed plasmids group were 62.5% ,25.0% and 50.0%,respectively.Results showed that effective protection could be provided by either pHM6-m or pHM6-np,but pHM6-m provided a better protective effect than pHM6-np.
4.Analysis of monitoring results on plague's host animal and its medium insects in Quanzhou city of Fujian province between 2000 and 2009

Objective To examine the composition and distribution of animal reservoir of plague in Quanzhou city, Fujian province, and so as to accumulate basic data for making the plague prevention and control measures. Methods Rodents were captured by cages in six monitoring spots of Quanzhou city between 2000 and 2009. Then rodents and flea species were identified. Rats liver and spleen homogenates were detected of plague F1 antigen by reverse indirect hemagglutination test (RIHA); rats serum were detected of plague F1 antibody by indirect hemagglutination test (IHA). ResultsA total of 26 264 rodents were captured between 2000 and 2009.Rattus norvegicus and Rattus flavipectus were dominant species, which were accounted for 45.97% (12 074/26 264)and 32.01%(8407/26 264), respectively. The account of Rattus flavipectus captured between 2005 and 2009 was (26.99 ± 2.46)% and (37.03 ± 3.79)% between 2000 and 2004. The difference was statistically significant (t =4.97, P < 0.05). Total rodent densities was (6.86 ± 1.44)%, including(5.36 ± 1.83)% in mountains and (6.81 ±1.66)% in coastal areas, respectively, and the difference was not statistically significant(t =1.01, P > 0.05). Total flea index and flea infection rate of rodents were 1.39 ± 0.34 and (35.90 ± 5.34)%, respectively. Xenopsylla Cheopis index was 1.20 and free flea index was 0.009. Flea infection rate was (32.36 ± 0.96)% between 2005 and 2009, which was lower than (39.44 ± 0.39)% between 2000 and 2004(t =2.76, P < 0.05). ConclusionsThe major rodent species found in Quanzhou city is Rattus norvegicus. Xenopsylla Cheopis is the dominant flea species.Spreading of plague among Rattus is not found.
5.Effect of Medicine Combined with Electric Stimulation Therapy on Acontractile Detrusor

ObjectiveTo investigate the effective approach for the treatment of detrusor underactivity. Methods98 patients with detrusor underactivity were randomly divided into electrotherapy group (electroacupuncture, interference current stimulation, n=32), medicine group (n=32) and electrotherapy-medicine group (n=34). The patients were assessed with examination of urodynamics and clinical parameters before and after treatment. ResultsThe detrusor strength were stronger after treatment in all the 3 groups (P<0.001), but electrotherapy-medicine group was more stronger than the other 2 groups (P<0.01). The cure rate in the 3 groups were 68.75%, 59.38% and 88.23% respectively, the patients in electrotherapy-medicine group gained significantly better results than the other 2 groups (x2=9.976, P<0.01). The difference of cure rate between the electrotherapy group and the medicine group was not significant (P>0.05). ConclusionElectric stimulation therapy combined with medicine is an effective intervention for treatment of detrusor underactivity and is better than electric stimulation therapy or drugs only.

Objective To compare the efficacy of 18F-FDG PET/CT, 99Tcm-MDP bone scintigraphy (BS), and combination of the two techniques (PET/CT + BS) for detecting bone metastasis by ROC curve analysis. Methods All 296 patients with various cancers, who underwent both 99Tcm-MDP BS and 18F-FDG PET/CT within two months, were retrospectively analyzed. These images were interpreted according to 5-point scale (0: definitely negative, 1: probably negative, 2: equivocal, 3: probably positive, 4:definitely positive for bone metastasis), and the scale of PET/CT + BS was the sum of PET/CT and BS. In light of the confirmed diagnosis derived from pathology or follow-up, ROC curve analysis was performed.The area under the ROC curve (AUC) was compared by z-test. Results Of 296 cases, 61 (20.6%) were confirmed as bone metastases and 235 (79.4%) were negative. The AUC were 0. 919 (95% confidence interval (95% CI) :0. 867 - 0. 971) for BS, 0. 949 (95% CI: 0. 906 - 0. 991) for PET/CT, and 0. 994 (95% CI: 0.988-0.999) for PET/CT + BS, rctrospectively. The AUC of PET/CT + BS was statistically significantly larger than that of BS (z=2. 866, P=0.004) or PET/CT (z =2.027, P=0.043), while the AUC of PET/CT was larger than that of BS, but no statistically significance (z = 0. 881, P = 0. 378) was showed. The optimal sensitivity, specificity, accuracy, positive predictive value (PPV), negative predictive value(NPV) were 90. 2% (55/61), 85. 1% (200/235), 86. 1% (255/296), 61. 1% (55/90), 97. 1%(200/206) for BS, 88.5% (54/61), 97.0% (228/235), 95.3% (282/296), 88.5% (54/61), 97.0% for PET/CT, and 98.4% (60/61), 95.7% (225/235), 96.3% (285/296), 85.7% (60/70) for PET/CT + BS,respectively. The specificity (χ2 = 19.862, P<0. 001), accuracy (χ2 = 23. 361, P<0.001) and PPV (χ2 =11. 791, P =0.001) of PET/CT + BS were significantly higher than those of BS, the sensitivity of PET/CT +BS was significantly higher than that of PET/CT (χ2 =4.167, P=0.031). Compared with BS, PET/CT had a higher specificity (χ2 = 19.600, P<0. 001), accuracy (χ2 = 13. 755, P <0. 001), PPV (χ2 = 13. 608, P <0. 001), but their sensitivity showed no statistically significant difference (χ2 = 0, P = 1. 000). Conclusions The efficacy of 18F-FDG PET/CT for detecting malignant bone metastasis was superior to that of 99Tcm-MDP BS alone. The detection ability can be obviously improved by combination of the two techniques.
8.Relationship between the polymorphism of lipoprotein lipase gene and serum lipid levels in the Guangxi Hei Yi Zhuang population

9.Influence of Ginkgo ketone ester dropping pills combined atorvastatin on vascular endothelial function , serum omentin-1 and Hcy levels in EH patients with carotid arteriosclerosis

Objective :To observe influence of Ginkgo ketone ester dropping pills (GKE) combined atorvastatin on vascular endothelial function ,serum levels of omentin-1 and homocysteine (Hcy) in hypertensive patients with ca-rotid arteriosclerosis (CAS).Methods : A total of 120 hypertensive patients with CAS treated in our hospital were selected ,randomly and equally divided into atorvastatin group and combined treatment group (received GKE com-bined atorvastatin ) ,both groups received corresponding therapy based on health education ,diet and exercise etc for 12 weeks.Blood pressure ,blood lipids ,carotid intima-media thickness (IMT ) ,plaque size ,levels of endothelin (ET)-1 ,nitric oxide (NO) ,serum omentin-1 and Hcy were compared between two groups before and after treat- ment .Results : Compared with before treatment ,there were significant reductions in blood pressure ,blood lipids (except significant rise in HDL-C) ,serum levels of ET-1 and Hcy ,IMT and plaque size ,and significant rise in ser-um levels of NO and omentin-1 in two groups after treatment , P<0-01 all.Compared with atorvastatin group after treatment ,there were significant reductions in blood pressure ,blood lipids (except significant rise in HDL-C) ,ser-um levels of ET-1 [ (136-42 ± 15-57) μg/ml vs.(116-57 ± 13-28) μg/ml] and Hcy [ (15-46 ± 3-25 ) μmol/L vs. (10-58 ± 3-11) μmol/L] ,IMT [ (1-43 ± 0-21) mm vs.(1-26 ± 0-19) mm] and plaque size [ (22-53 ± 1-28) mm2 vs.(20-67 ± 1-14 ) mm2 ] ,and significant rise in serum levels of NO [ (102-43 ± 12-37) μmol/L vs.(129-84 ± 13-36) μmol/L] and omentin-1 [ (176-57 ± 45-20 ) μg/ml vs.(198-73 ± 45-38) μg/ml] in combined treatment group , P<0-05 or <0-01- Conclusion : Ginkgo ketone ester dropping pills combined atorvastatin can significantly improve vascular endothelial function and arteriosclerosis while reducing blood pressure and blood lipids in hyperten-sive patients with carotid arteriosclerosis .
