1.The effects of rehabilitation training on cardiac function in cerebral infarct patients

Objective To study the effects of rehabilitation(RT)on cardiac function in cerebral infarct (CIF)patients with cardiac insufficiency(CIS).Methods Fifty-nine CIF patients with CIS were randomly divid- ed into a treatment group(T group,n=29)and a control group(n=30),and all patients were treated with routine pharmacotherapy for 2 months.In addition,RT was administrated in the T group at the same time.The grading of the New York Heart Association(NYHA)and the changes in cardiac function associated index were observed in both groups before and after treatment.Results Compared with the control group,NYHA grades,left ventricle ejection fraction(LVEF),the levels of brain natriuretic peptide(BNP)in the blood plasma,and the 6min walking range of the T group patients were all significantly improved after treatment(P<0.05).Conclusion RT can improve car- diac function in CIF patients with CIS.
2.Effect of prepared radix Polygoni multiflori on the elevation of extracellular hydroxyl radical in striatum of rats induced by intracerebral perfusion of 6-hydroxy dopamine.

OBJECTIVETo investigate the effect and mechanism of prepared Radix Polygoni Multiflori (RPM) on the elevation of extracellular hydroxyl radical in striatum of rats induced by intracerebral perfusion of 6-hydroxy dopamine (6-OHDA).
METHODSCerebral microdialysis was used to establish the model. Hydroxyl radical was captured by salicylic acid, and 2,3-dihydroxy benzyl acid (2,3-DHBA) and 2,5-dihydroxy benzyl acid (2,5-DHBA) formed by hydroxyl radical in vital brain were measured by high performance liquid chromatography-electrochemical detector (HPLC-ED).
RESULTSAfter perfusion of 6-OHDA in brain of rats, the levels of 2,3-DHBA and 2,5-DHBA in the model group increased rapidly. The former was higher during the whole course of observation (P<0.01), while the latter was higher at most time points than that in the control group (P<0.05 or P<0.01). The level of 2,3-DHBA in the RPM group was lower than that in the model group at 5 time points (P < 0.05, P < 0. 01).
CONCLUSIONRPM could inhibit the elevating of extracellular hydroxyl radical in striatum of rats induced by intracerebral perfusion of 6-OHDA, indicating one of the brain protective mechanisms of RPM may be related to its anti-oxidation effect.

4.The effect of shourong compound formula on levels of dopamine and its metabolites in brain of Parkinson's disease mice induced by reserpine.

OBJECTIVETo investigate the effect of Shourong compound formula on treating Parkinson's disease.
METHODParkinson's disease model mice induced by reserpine was used and by HPLC-ED the levels of Dopamine (DA) and its metabolites were determined.
RESULTMadopar could increase the levels of DA in brain of PD mice. The effect of madopar together with Sourong compound formula was better than that of madopar(P < 0.001).
CONCLUSIONShourong compound formula together with madopar has synergic effect on increase of DA level in brain and can reduce clinic dose of madopar so that side effect of madopar can be decreased.

6.Effect of transient forebrain ischemia-reperfusion on the binding of brain-derived neurotrophic factor promoters to histone deacetylase 3 in hippocampus of rat and its mechanism

Objective To evaluate the effect of transient forebrain ischemia-reperfusion(I/R)on the binding of brainderived neurotrophic factor(BDNF)promoters to histone deacetylase 3(HDAC3)in the hippocampus of rat and investigate its mechanism.Methods The I/R model of SD rats(I/R group)was established by Pulsinelli four-vessel clamping method,and sham operation group(Sham group)was set at the same time,which were observed for the survival of neurons in the hippocampus of rats by Nissl staining,detected for the binding of BDNF promoters(Bdnf-p1,Bdnf-p2,Bdnf-p4 and Bdnf-p6)to HDAC3 by chromatin immunoprecipitation(ChIP)and determined for the expression of brain derived neurotrophic factor antisense(BDNF-AS)by qPCR.Results Compared with Sham group,the quantity of neurons in hippocampal CA1 region of rats decreased significantly in I/R group,while those in CA3 region and DG region showed no significant changes.The binding levels of Bdnf-p1 and Bdnf-p2 to HDAC3 in hippocampal CA1 region decreased significantly in I/R Group(t = 2.575 and 2.241 respectively,each P<0.05),while there was no significant difference in the binding levels of Bdnf-p4 and Bdnf-p6 to HDAC3(t = 1.033 and 0.348 respectively,each P>0.05);The binding levels of Bdnf-p1 and Bdnf-p2 to HDAC3 in CA3 region increased significantly(t = 12.600 and 3.191,P<0.001 and<0.05,respectively),while the binding level of Bdnf-p6 to HDAC3 decreased significantly(t = 4.029,P<0.05)and no significant difference was observed in the binding level of Bdnf-p4 to HDAC3(t = 0.175,P>0.05);In DG region,the binding level of each BDNF promoter to HDAC3 showed no significantly difference(t = 0.630 ~ 1.687,each P>0.05).Meanwhile,the expression level of BDNF-AS in hippocampal CA1 region of rats decreased significantly(t = 2.560,P<0.05),but increased significantly in hippocampal CA3 and DG regions(t = 3.543 and 3.637 respectively,each P<0.01)in I/R group.Conclusion I/R showed a significant effect on the binding level of BDNF promoter to HDAC3 in rat hippocampus,which may play a role by changing the expression level of BDNF-AS.
7.Basic fibroblast growth factor in human detrusor muscle in bladder outlet obstruction due to benign prostatic hyperplasia

Objective To investigate the change of the basic fibroblast growth factor(bFGF) leve in human detrusor muscle(DM)in bladder outlet obstruction(BOO)due to benign prostatic hyperplasia(BPH)and its implication.Methods Fifty-four patients with BPH were divided into two groups:the obstructive DM stability and instability groups;and 15 men with bladder tumor who underwent operation in the same period were enrolled in the control group.The bFGF mRNA level in DM was measured by reverse transcription and polymerase chain reaction(RT-PCR)and the bFGF protein level was measured by immunohistochemical staining method.Results The bFGF-mRNA expression level of bladder smooth muscle cells was significantly lower in the control group than that in the obstructive DM stability and instability groups(all P<0.05),but there was no significant difference between the obstructive DM stability and instability groups(P>0.05). Conclusions The expression level of bFGF mRNA in bladder DM is elevated in BOO due to BPH,but there is little or no correlation between the increased expression of bFGF mRNA and detrusor muscle instability.
8.Analysis of salt resistance on the poplar transferred with salt tolerance gene.

Poplar(Pupulus x Xiao zhannica, cv. "balizhuang-yang") transferred with mtl-D gene was used as an experimental material. Through tissue culture, aquaculture and pot culture transgenic poplars are tested with variant Nacl salt gradient. In it transgenic poplar raises its initial days for differentiation, differentiation rate, bud intensity, bud height and growth potential than control plant. At the same time, the transgenic poplar has higher rooting rate and more top roots, side roots and higher root length than control under the same salt stress. The result shows that the transgenic poplar has better tolerance ability than control plant in the salt 4@1000 contensity mediums.

9.Prevalence and clinical characteristics of Coronavirus NL63 infection in children hospitalized for acute lower respiratory tract infections in Changsha

Objective The main objective of this study was to explore the prevalence and clinical characteristics of human coronavirus NL63 infection in hospitalized children with acute lower respiratory tract infection (ALRTI) in Changsha.Methods Nasopharyngeal aspirates (NPA) samples were collected from 1185 hospitalized children with ALRTI at the People's Hospital of Hunan province,between September 2008 and October 2010.Reverse transcriptase polymerase chain reaction (RT-PCR) was employed to screen for coronavirus NL63,which is a 255 bp fragment of a part of N gene.All positive amplification products were confirmed by sequencing and compared with those in GenBank.Results The overall frequency of coronavirus NL63 infection was 0.8%,6 (60%) out of the coronavirus NL63 positive patients were detected in summer,2 in autumn,1 in spring and winter,respectively.The patients were from 2 months to two and a half years old.The clinical diagnosis was bronchopneumonia ( 60% ),bronchiolitis ( 30% ),and acute laryngotracheal bronchitis( 10% ).Four of the 10 cases had critical illness,4 cases had underlying diseases,and 7 cases had mixed infection with other viruses. The homogeneity of coronavirus NL63 with those published in the GenBank at nucleotide levels was 97%-100%.Conclusion Coronavirus NL63 infection exists in hospitalized children with acute lower respiratory tract infection in Changsha.Coronavirus NL63 infections are common in children under 3 years of age.There is significant difference in the infection rate between the boys and the girls:the boys had higher rate than the girls.The peak of prevalence of the coronavirus NL63 was in summer.A single genetic lineage of coronavirus NL63 was revealed in human subjects in Changsha.Coronavirus NL63 may also be one of the lower respiratory pathogen in China.
10.Effect of prepared Polygonum multiflorum on striatum extracellular acetylcholine and choline in rats of intracerebral perfusion with sodium azide.

OBJECTIVETo investigate the protection mechanism of prepared Polygonum multiflorum (PPMT) in rat brain with sodium azide (NaN3) perfusion.
METHODRats were divided into six groups: control, model, PPMT, Duxil and PPMT + Duxil groups. The intracerebral microdialysis and high performance liquid chromatography-post column Immobilized enzyme reactor-electrochemical detection were used to continuously measure extracellular acetylcholine (ACh) and choline (Ch) levels in striatum of freely moving awake rats.
RESULTThe extracellular Ach, Ch levels of striatum stayed stable in the control group during the whole observing period, but the ACh levels in the model group were lower significant than that in the control group. The Ach levels of three drug groups were respectively higher significant than that of model group at some time points. While the extracellular Ch level in striatum of the model group increased singnificantly compared with the control group. The Ch levels of the three drug groups were lower significant than that of the model group respectively at certain time points. The effects of PPMT were similar with that of Duxil.
CONCLUSIONThe prepared P. multiflorum can improve the impaired cholinergic nerve function to exert the effects of brain protection by elevating extracellular Ach level and improving uptake of extracellular Ch. It may provide the experimental evidence to support the idea that P. multiflorum could be brain protective drug to treat retrogressive disease such as dementia.
