3. Preparation and characterization of sodium methacrylate modified photocrosslinked alginate hydrogel scaffold

BACKGROUND: Photocrosslinked alginate hydrogel has been a popular bone tissue engineering material because of its excellent biocompatibility and minimally invasive injection, but there are still problems such as insufficient strength and poor cell adhesion. OBJECTIVE: To construct the negatively charged hydrogels by introducing sodium methacrylate into photocrosslinked alginate hydrogels, and to explore the changes in its physical performance and cell adhesion. METHODS: After preparation of methacrylated alginate by reacting sodium alginate with 2-aminoethyl methacrylate, methacrylated alginate, photoinitiator and sodium methacrylate (0, 20, 40, 60 mmol/L) were homogeneously mixed. The negatively charged photocrosslinked alginate hydrogels were prepared under ultraviolet light. The functional groups of the hydrogels were analyzed by fourier transform infrared spectroscopy. The surface morphology of the hydrogels was observed by scanning electron microscopy and the swelling ratio was measured. MC3T3-E1 cells were cultured with each group of hydrogels for 48 hours, and the cytotoxicity of the hydrogels was investigated by cell counting kit-8 assay. MC3T3-E1 cells were seeded on the surface of each group of hydrogels. The early adhesion of the cells was observed by live/dead staining at the 4th hour, and cell spreading was observed on the 3rd day. RESULTS AND CONCLUSION: (1) Fourier transform infrared spectroscopy demonstrated that the introduction of sodium methacrylate could lead to a new peak at wavenumber of about 1 600 cm-1 in the hydrogel infrared wave, which was from the sodium methacrylate. (2) Scanning electron microscope observed that the density of the negatively charged photocrosslinked alginate hydrogels increased and the pore size of the gels decreased with augment of concentrations of sodium methacrylate. (3) The swelling ratio of the hydrogel decreased with the increase of the concentration of sodium methacrylate. (4) The live/dead staining revealed that the cells grew well on the surface of each hydrogel, and the cell viability reached above 95%. The cell counting kit-8 assay results showed that the negatively charged photocrosslinked alginate hydrogels had no cytotoxicity. (5) The early cell adhesion rate increased gradually and the cell extension became better with the increase of concentration of sodium methacrylate. (6) In summary, the introduction of sodium methacryl into photocrosslinked alginate hydrogels can adjust its physical properties and significantly improve its cell adhesion.

5.Effect of cold and cool herbs on liver mitochondria proteome of rats with heat symptom.

In the 1960s, modern science began involving the essence of heat syndrome, but there have still no in-depth systematic studies on pathological mechanisms of heat syndrome and action mechanisms of cold and cool herbs. In this study, the animal model with heat syndrome was set up by feeding herbs with hot property, and then cold and cool herbs was applied in the experimental therapy. The two-dimensional electrophoresis and mass spectrometry technologies were adopted to compare the liver mitochondria proteome of the rats of the heat syndrome model and the ones treated with cold and cool herbs, so as to discover specificity-related proteins after heat syndrome and treatment with cold and cool herbs.

6. Research progress of the relationship between endothelial progenitor cells and cardiovascular complications in diabetes

70% to 80% of people with diabetes died from cardiovascular complications, which had become the leading cause of death in diabetic patients. Cardiovascular complications in diabetic patients are mainly associated with vascular endothelial dysfunction and poor neovascularization. Its important influencing factors are changes in the number of endothelial progenitor cells and dysfunction. Hyperglycemia in diabetic patients can cause endothelial progenitor cell dysfunction, inhibit its proliferation ability, angiogenesis ability and paracrine effect. In order to provide research ideas for the prevention and treatment of cardiovascular complications in diabetic patients, this article aims to review the relevant mechanism of the effect of hyperglycemia on the number and function of endothelial progenitor cells in patients with diabetes, and the application of endothelial progenitor cells in the treatment of diabetic cardiovascular disease.
7.Research progress on musk secretion mechanism of forest musk deer.

Forest musk deer (Moschus berezovskii), a rare wild medicinal animal, is listed under the category of the state key protected wildlife list of China. Musk, secreted by the musk glands, is with high economic and medicinal value and used as precious traditional medicine in China. In order to meet the needs of musk in Chinese traditional medicine, forest musk deer farming was conducted in 1950s, but the research progress on musk secretion mechanism was slow. Therefore, by reviewing the histological and anatomical structure of forest musk deer musk gland, the relationship between sex hormones and the musk secretion process, and the molecular mechanism of the musk secretion, the existing problems in investigating the musk secretion mechanism were analyzed and the development trends in this field were also discussed, in order to provide a reference for further studies on the musk secretion mechanism and improve musk production of forest musk deer.

8.Effect of ligustrazine on reverse cholesterol transport in foam cells.

OBJECTIVETo discuss the intervention effect of ligustrazine on ox-LDL-induced foam cells from the perspective of reverse cholesterol transport.
METHODRAW264.7 cultured in vitro was induced with 20 mg x L(-1) ox-LDL to establish the foam cell model, and intervened with ligustrazine. The lipid accumulation in cells was observed by the oil red O dyeing. The changes in total cholesterol and cholesterol ester in the cells were detected with the colorimetric method. The fluorescent quantitative PCR and Western blot were used to detect the mRNA expressions of PPARgamma, LXRalpha and ABCA1.
RESULTLigustrazine could reduce total cholesterol and cholesterol ester in foam cells, inhibit the lipid accumulation, and increase the mRNA and protein expressions of PPARgamma, LXRalpha and ABCA1.
CONCLUSIONLigustrazine can promote the reverse cholesterol transport by increasing the gene expressions of PPARgamma, LXRalpha and ABCA1.

9.The study of correlation between anti-cyclic citrnllinated peptide antibody and rheumatoid arthritis

Objective To explore the correlation between anti-cyclic citrullinated peptide(A-CCP) antibody and tumor necrosis factor(TNF)-?, rheumatoid factor(RF), ESR, PLT count and clinical features in patients with rheumatoid arthritis(RA), and the outcome of unclassified arthritis(arthralgia)patients after six months follow up. The value of A-CCP antibdy in the diagnosis of early RA and its pathogenetic roles is in- vestigated. Methods A-CCP antibody and TNF-?were detected by ELISA and the RF was tested by the rate scatting immunity method in 91 RA patients, 46 unclassified arthritis(arthralgia)patients and 45 other rheumatic diseases patients. Results A-CCP antibody levels in serum correlated significantly with TNF-?levels, PLT count and the degree of joint swelling in RA and unclassified arthritis(arthralgia)patients(r= 0.854, P=0.O00; r=0.882, P=0.000; r=0.318, P=0.002; r=0.486, P=0.001; r=0.291, P=0.005; r=0.731, P= 0.000 respectively). A-CCP antibody levels in serum was weakly negatively correlated with the gripping power in RA patients(r=0.228, P=0.030). And it was weakly correlated with ESR in unclassified arthritis(arthrai- gia)patients(r=0.365, P=0.013). Compared with other rheumatic diseases patients, A-CCP antibody levlels in serum increased significantly in RA and unclassified arthritis(arthralgia)patients(P=0.000). Compared with normal controls, it increased in other rheumatic diseases patients(P=0.011). Twenty-four patients had positive A-CCP antibody in 46 unclassified arthritis(arthralgia)patients. Thirty-two out of 46 unclassified arthritis(arthralgia)patients were early RA after 6 monthes follow up. 95.8%(23/24)unclassified arthritis (arthralgia)patients with positive A-CCP antibody were early RA. Conclusion A-CCP antibody reflects disease activity in certain extent. It's benefit to the diagnosis of early RA. High A-CCPantibody levels com- bined with high levels of TNF-?, ESR, PLT count and joint swelling can help the diagnosis of early RA.
10.Correlation between the expression of cytokeratin-18-mRNA and invasion and metastasis of oral squamous cell carcinoma.

OBJECTIVETo investigate the relationship between the expression of cytokeratin (CK)-18 and biological behavior of oral squamous cell carcinoma (OSCC).
METHODSTwenty-three patients with OSCC were investigated for the expression of CK-18-mRNA by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). Correlations between clinical stages, pathological differentiation, lymphatic metastasis and the expression of CK-18-mRNA were evaluated. CK-18-mRNA expression of peripheral blood from the 23 patients and 23 healthy people were also examined. During follow-up after operation, the peripheral blood was collected again for the expression of CK-19-mRNA.
RESULTSExpression of CK-18-mRNA was found in 16 patients. The expression of CK-18-mRNA was significantly associated with clinical stages, tumor differentiation and lymphatic metastasis. CK-18-mRNA was positive in 4 of 23 blood specimens before operation, but during follow-up only 1 of 23 patients was still positive in peripheral blood.
CONCLUSIONSCK-18 may provide additional information in forecasting the metastasis of OSCC and serve as a reference in monitoring recurrence.
