Objective:To determine the role of lncRNA TUG1 in pancreatic β cells functioning both in vitro and in vivo. Methods:The lncRNA TUG1 expression in mice pancreas, brain, muscle and other different tissues was examined through qRT-PCR.MTT,flow cytometry,GSIS,ELISA and immunochemistry analyses were performed to detect the effect of lncRNA TUG1 on insulin secretion in vitro and in vivo. Results:lncRNA TUG1 was highly expressed in pancreatic tissue compared with other organ tissues.Knockdown of lncRNA TUG1 expression resulted in decreased insulin secretion in β cells both in vitro and in vivo.Immunochemistry analyses showed decreased relative islet area after treatment with lncRNA TUG1 siRNA.Conclusions:Downregulation of lncRNA TUG1 expression can affect insulin secretion in pancreatic β cells in vitro and in vivo, and lncRNA TUG1 may represent a factor that regulates the function of pancreatic β cells.

Objective To evaluate the effect of medicial and surgical treatment for juvenile hyperthyroidism. Methods Ortapazole was administ rated separately in drug therapy group for 1.5-2 years.Bilateral subtotal thyroidectomy was done in surgical therapy group. Results In drug therapy group, effective rate was 60 percent in 6 months and 70 percent in one year.Recurrence rate was 40 percent after drug withdrawal in 2 years curative rate was 60 percent. In surgical therapy group, the average stay in hospital was 16 days. There was no nerve injury, parathyroidal hypofunction, thyroid crisis or hypothyroidism complications, with 100 percent curative rate after 2 years′ followup.Postoperative growth and development were normal. Conclusions Surgical treatment may be suitable for those who have no response to drug therapy, with recurrence after drug withdrawal, whose compression symptom was obvious, with moderate and severe hyperthyroidism or those who could not take medicine persistently.Bilateral subtotal thyroidectomy applied in juvenile hyperthyroidism could achieve quick and better recovery, and has no influence on the juvenile growth and development.

Objective To study the life quality of 2 - 3 years old survivors in Neonatal Intensive Care Unit (NICU). Methods Severe neonates were randomly assigned to intervention group (group 1,30 cases) and non- intervention group (group 2,30 cases) depending on the early intervention applied or not,as well as 30 healthy newborns as normal controls. The physical, neurological conditions and intelligence test were taken regularly. To investigate the psychological state, actions, temperament and family conditions when they were2-3 years old. Results Mental development index(MDI) and physical development index(PDI) in early interventional group were significant higher than those in group 2(P

Objective To investigate plasma levels of total carnitine (TC) and free camitine (FC) in patients undergoing hemodialysis and peritoneal dialysis. Methods 200 cases of normal group came from physical examination in this hospital, all testing cases were the in-hospital patients in the department of nephropathy.TC and FC were determined by use of an enzymatic cycle assay on Hitachi 7170 automatic biochemical analyzer. Results In 200 cases of normal group,TC level was (56.52?9.61) ?mol/L,and FC was (46.60?8.23) ?mol/L.In 37 hemodialysis patients,TC and FC levels were (41.47?13.22) ?mol/L and (24.58?8.91)?mol/L before dialysis,a statistic difference was observed against the control group (P0.05).Conclusions Carnitine deficiency was seen in most patients undergoing hemodialysis and peritoneal dialysis.Furthermore, the deficiency status got worse along with the dialysis course in hemodialysis patients.Carnitine infusion can effectively improve the status of these patients.

AIMTo establish the method of HPLC-fingerprint analysis for the quality control of Dalbergia odorifera and identify its main constituents by HPLC-MS.
METHODSThe 37 hatches of samples were analyzed on a Phenomenex Luna C18 column with a gradient of acetonitrile and 0.3% aqueous acetic acid at a flow rate of 1.0 mL x min(-1) and detected at 275 nm. Furthermore, the typical samples were detected by HPLC-DAD-MS under negative ion mode.
RESULTS37 batches of D. odorifera samples were classified into three types based on the results of similarity analysis. According to the comparison of the tR, MS data and UV maximum absorbance (gamma(max)) values with the standards, 10, 7 and 2 phenolic components were identified in three types of D. odorifera extracts, separately.
CONCLUSIONThe method is repeatable and reliable, and it is capable of effectively controlling the quality of D. odorifera.

Objective To evaluate the application of non-bioartificial liver support system (ALSS) combined with liver transplantation(LT) in treating mid-or end-stage chronic severe hepati- tis. Methods ALSS plus liver transplantation were employed in treating 28 patients with mid-or end- stage chronic severe hepatitis.Clinical data from the patients before and after treatment were collect- ed.The survive rate of ALSS plus LT group were compared with that of medication group 99 cases and medication plus ALSS group 30 cases. The data were analyzed with t test and X~2 test. Results After 57 times ALSS treatment, the serum total bilirubin(TBil),prothromin time(PT),bile acid, blood urea nitrogen(BUN),creatnine(Cr) and ammonia of all the 28 patients got improved(P 0.05).The clinical symptoms and signs of the patients were ameliorated at median 3 d(1~153 d). All patients were bridged to liver transplantation successfully after median 20 d(1~153 d).The 3 and 6 months post-operation survival rate of ALSS plus LT group(71.4%,71.4%) were significantly higher than those in medication group(18.2%,11.1%) and medication plus ALSS group(36.7%, 26.6%)(P

OBJECTIVETo improve the recognition of the clinical features and results of laboratory examination for isolated pulmonary Langerhans cell histiocytosis (PLCH) in children.
METHODThe information of one case with isolated PLCH was analyzed and reports of 11 cases with isolated PLCH were reviewed.
RESULTThe patient we report is only 2 years old with 1 month of course of disease, manifesting with prominent pulmonary involvement: cough and short of breath; CT scan of the chest showed punctiform, nodular and reticular high density opacities involving all lobes of both lungs. Biopsy of the lung tissue showed expression of CD1a, CD68, S-100, consistent with the diagnosis of LCH. He received prednisolone, VP16 and Vindesine with good response. Ten of 11 cases of isolated PLCH reported before manifesting with cough and dyspnea, CT scan of the chest showed interstitial lung changes (5/8), cystic changes (5/8), small nodules (2/8) and pneumothorax (2/8). Langerhans cells were found in 9 cases on lung biopsy, part of biopsy lung tissues were stained with anti-CD1a, the alveolar lavage fluid of the other 2 cases were stained with S-100 and anti-CD1a.
CONCLUSIONIsolated PLCH is rarely reported in children. It manifested with prominent pulmonary involvement: cough and short of breath, and CT scan of the chest showed interstitial lung changes, small nodules or cysts involving the lung, Langerhans cell could be found in lung biopsy, and the immunohistochemical staining in lung biopsy lung and alveolar lavage fluid stained with S-100 and anti-CD1a antibodies.
