3.Research Development on Tibetan Medicine Prevention and Treatment of High Altitude Polycythemia

High altitude polycythemia (HAPC) had become one of the main common chronic diseases, which had seriously threatened the health of Plateau people. In the Tibetan medicine classic bookSi-Bu Yi-Dian, there were recordings on HAPC treatment methods and medications, which had the unique advantages of identified therapeutic effect with little side effect. This article analyzed Tibetan medicine in the prevention and treatment of HAPC from aspects such as etiology and pathogenesis, clinical treatment advantages and modern innovation study. Questions were also raised on lacking of standardization on HAPC clinical effectiveness, as well as Tibetan medicine compound material basis and action mechanisms were unclear. It was proposed that based on the inheritance of Tibetan medicine theoretical basis and clinical therapeutic effect, the Tibetan medicine original thinking should be combined with modern science and technology, in order to strengthen the analysis of ancient literature collection in HAPC treatment and data mining in medication experiences. The clinical treatment standards and medication plan should be standardized. Methods of systems biology, such as metabolomics, can be used in the further study of the scientific connotation of HAPC treatment by Tibetan medicine.
4.Optimization of genomic DNA extraction with magnetic bead- based semi-automatic system.

OBJECTIVETo develop a rapid and effective method for genomic DNA extraction with magnetic bead-based semi-automatic system.
METHODSDNA was extracted from whole blood samples semi-automatically with nucleic acid automatic extraction system.The concentration and purity of samples was determined by UV-spectrophotometer. Orthogonal design was used to analyze the main effect of lysis time, blood volume, magnetic bead quantity and ethanol concentration on the DNA yield; also the 2-way interaction of these factors.
RESULTSLysis time, blood volume, magnetic bead quantity and ethanol concentration were associated with DNA yield (P<0.05), but no interaction existed. DNA yield was higher under the condition with 15 min of lysis time, 100 μl of blood volume, 80 μl of magnetic beads and 80 % of ethanol. A significant association was found between the magnetic bead quantity and DNA purity OD260/OD280 (P=0.008). Interaction of blood volume and lysis time also existed (P=0.013). DNA purity was better when the extracting condition was 40 μl of magnetic beads, 15 min of lysis time and 100 μl of blood volume. Magnetic beads and ethanol concentration were associated with DNA purity OD260/OD230 (P=0.017 and P<0.05), the result was better when magnetic beads was 40 μl and ethanol concentration was 80 %.
CONCLUSIONThe results indicate that the optimized conditions with 40 μl magnetic beads will generate higher quality of genomic DNA from the whole blood samples.

5.Treatment of extremity infections with soft tissue defects with the ramified musculocutaneous flap pedicled with the descending branch of lateral circumflex femoral artery

Objective To discuss the clinical effect of the ramified musculocutaneous flap pedicled with the descending branch of lateral circumflex femoral artery treatment of bones and arthrsis of extremity in- fections with soft tissue defects.Methods The muscle flap blooded with the muscular branch of lateral fem- oral muscle and musculocutaneous flap blooded with the musculocutaneous branch were designed,all of which were pedicled with the descending branch of lateral circumflex femoral arteryIn clinic24 cases of bones and arthrosis of extremity infections with soft tissue defects were treated with this kind of ramified musculocutaneous flap.Results Of the 24 cases23 cases were survived while 1 case was lost16 cases were healed at stageⅠ8 cases were healed at stageⅡSinus has formated in 3 casesone of which twicebut they were healed in one year with the treatment of debridmentsFour cases with osteomvelitis and bone defect were treated with bone grafting in the later 6~8 months after the wound has healedTwenty-two cases were followed-up for 6~20 monthsinfcetiou didn't recur.Conclusion This kind of ramified musculocutaneous flap has such ad- vantages as longer blood vessel pediclefilling the defects completely flexible application and stronger anti-in- fectionthat it may be an effective way in treating bones and arthrosises of extremity infections with soft tissue defects.
6.Chin augmentation with bone transplantation from the mandible.

OBJECTIVETo explore a new method for mentoplasty.
METHODSThe bilateral prominent mandibular angle or outer lamina was resected through the intraoral approach. The resected bone fragments were shaped and rigid fixed to the chin with miniplates and screws.
RESULTSA total of 30 patients (28 females, 2 males) accepted chin augmentation with this method. The mandibular angle bone was used in 20 cases and the mandibular outer lamina was used in 10 cases. The operative results were satisfactory, and the patient's facial contour was improved substantially.
CONCLUSIONNo rejection reaction was found after this procedure. Chin augmentation with autogenous mandibular bone is an ideal method for genioplasty.

7.Determination of loganin in Pterocephalus hookeri by HPLC.

OBJECTIVETo develop an HPLC method for determination of loganin in Pterocephalus hookeri.
METHODThe analysis was performed on a Welchrom C18 column (4.6 mm x 250 mm, 5 microm) with acetonitrile-water (12:88) as the the mobile phase at a flow rate of 1 mL x min(-1) and the detection wavelength was 240 nm.
RESULTThe peak areas and injection amounts of loganin had a good linear relationship at the range of 0.06-1.2 microg. The average recovery was 97.0%, and the RSD was 1.4%.
CONCLUSIONThe method is simple, accurate, and can be used for its quality control.

9.Bioinformatics analysis to the differentially expressed genes of normal mucosa and carcinoma of colon.

OBJECTIVETo systemically analyze the differentially expressed genes from normal mucosa and carcinoma of colon obtained by SSH method.
METHODSAn automatic platform for the analysis of nucleotides based on Linux was constructed, and one of the three subtracted libraries from SSH (T -N) was systematically analyzed by this platform. Part of the results was verified by semi- quantity RT-PCR.
RESULTThe automatic platform for the analysis of nucleotides based on Linux was successfully constructed. There were 15 contigs from the subtracted T-N libraries, among which 2 had no match with known genes in the GenBank. The expressions of genes Homo Sapiens thymosin beta 4 (THY) and Homo Sapiens HbxAg transactivated protein 2 (XTP) had trends of increase with the progress of normal tissue-adenoma-adenocarcinoma when verified by semi- quantity RT-PCR.
CONCLUSIONLinux-based automatic platform provides an efficient way to systematically analyze the nucleotide sequences, which may be used in study on the mechanism of colorectal carcinogenesis.
