2.Flocculation of Fermented Broth with 1,3 -Propanediol

Flocculation of fermented broth with 1,3 -propanediol using component B of natural clarifier-Ⅱ was studied. Firstly, single factor tests and orthogonal tests were carried out. The results showed the main factors which influenced the flocculation and the optimal conditions were: pH6.0, flocculant B 0.01g/L , NaCl 0g/L, and the FR was 95.97% . The following results by filtration experiments, compared with the sample without pretreatment, indicated that the filtration speed of the flocculated sample was improved dramatically, and the weights of net and dry filter cakes were increased by 41.13% and 51.88% , respectively. Electrodialysis experiments showed that flocculation could accelerate desalting process, and could take the place of centrifugal pretreatment when it combined with the filtration.
3.Study on Cleavage in vitro of HPVll/E2 Gene by Specific Ribozyme

In order to study the pathogenesis of HPVl1 and seek for a therapeutic approach of the disease caused by HPV1l. The HPVl1/E2 644bp was amplified by PCR with HPVll plasmid DNA. pGEM-T Easy was used as vector and a clone pTV-644 was obtained.The inserted DNA sequencing was carried out after selecting and identification. According to the hammerhead structure described by Symon's, the possible secondery cleavage sites on HPVll/E2 mRNA were analyzed and the secondery structure of substrate was predicted by computer and ribozyme, excluding analogous sequence of substrate combined with ribozyme were found in the mRNA.The hammerhead ribozyme of RZ2777 against HPV1l/E2 mRNA was seleted to carry out cleavage reaction in vitro. Results of the experiment showed that 644bp substrate derived from HPVll/E2 can be cleavaed site-specifically by ribozyme in vitro. The cleavage activity showed over 85% by choosing the optimun reaction condition, which was not affected by two cis-ribozymes on both 3′-and 5′-ends released by self cleavage,but two flan-king sequences of target RNA influenced the cleavage activity. Results demonstrated that the ribozyme will become a highly effective and specific therapy against HPVll infection.
4.Relation between drug release and the drug status within curcumin-loaded microsphere.

To study the relation between drug release and the drug status within curcumin-loaded microsphere, SPG (shirasu porous glass) membrane emulsification was used to prepare the curcumin-PLGA (polylactic-co-glycolic acid) microspheres with three levels of drug loading respectively, and the in vitro release was studied with high-performance liquid chromatography (HPLC). The morphology of microspheres was observed with scanning electron microscopy (SEM), and the drug status was studied with X-ray diffraction (XRD), differential scanning calorimetry (DSC) and infrared analysis (IR). The drug loading of microspheres was (5.85 ± 0.21)%, (11.71 ± 0.39)%, (15.41 ± 0.40)%, respectively. No chemical connection was found between curcumin and PLGA. According to the results of XRD, curcumin dispersed in PLGA as amorphous form within the microspheres of the lowest drug loading, while (2.12 ± 0.64)% and (5.66 ± 0.07)% curcumin crystals was detected in the other two kinds of microspheres, respectively, indicating that the drug status was different within three kinds of microspheres. In the data analysis, we found that PLGA had a limited capacity of dissolving curcumin. When the drug loading exceeded the limit, the excess curcumin would exist in the form of crystals in microspheres independently. Meanwhile, this factor contributes to the difference in drug release behavior of the three groups of microspheres.

5.Meta - analysis of clinical randomized controlled trials comparing ReZOOM with ReSTOR multifocal intraocular lenses in cataract surgery

AIM: To systematic review the effectiveness of refractive multifocal intraocular lens ( MIOL ) ReZOOM vs diffractive MIOL ReSTOR in the treatment of cataract.
METHODS: Randomized controlled trials comparing refractive MIOL ReZOOM with diffractive MIOL ReSTOR were identified by searching CENTRAL, MEDLINE, EMbase, WANFANG MED ONLINE, CMJD, SinoMed, and CNKI. Related journals also were hand-searched. Methodological quality of randomized controlled trials ( RCTs ) was evaluated by simple evaluate method that recommended by the Cochrane Collaboration. Data extracted by two reviewers with designed extraction form. RevMan software ( release 5. 2 ) was used for data management and analysis.
RESULTS:A total of 7 trials (846 eyes) were included for systematic review. Subgroup analyses were used according to different model comparison of ReSTOR MIOL. The results showed a significant difference in the mean of the best distance corrected intermediate visual acuity ( BDCIVA) in the ReZOOM MIOL group with WMD=-0.11, 95% CI (-0.16, -0.06) (P<0.0001). It showed a significantly difference in the mean of the uncorrected near visual acuity ( UCNVA ) , complete spectacle independent rate, halo rate and glare rate in the ReSTOR MIOL group with WMD= 0. 09, 95% CI (0. 05, 0. 14) (P<0-00001), WMD= 2. 62, 95%CI (1. 76, 3. 91) (P<0. 00001), WMD=1. 35, 95% CI (1. 15, 1. 60) (P=0. 0004) and WMD=1-29, 95% CI (1. 09, 1. 53) (P= 0. 003). There was no significant difference between the two groups in the mean of the uncorrected distance visual acuity ( UCDVA ) , the uncorrected intermediate visual acuity ( UCIVA) , the best corrected distance visual acuity ( BCDVA ) and the best distance corrected near visual acuity ( BDCNVA ) with WMD-0.03, 95% CI (-0.06, 0.01) (P=0.15), WMD= -0.04, 95% CI (-0. 09, 0. 01) (P=0. 10), WMD= -0. 01, 95%CI (-0. 04, 0-02) (P=0. 55) and WMD= 0. 06, 95% CI (-0. 06, 0. 17) (P=0. 32).
CONCLUSION: Patients implanted with ReZOOM MIOL can provide better BDCIVA; patients implanted with ReSTOR MIOL show better UCNVA, are less likely to appear light halo, glare and other visual adverse reactions; correction in spectacles cases, patients implanted with ReZOOM or ReSTOR MIOL have considerable performances in the far and near visual acuity.
6. Research progress of the relationship between endothelial progenitor cells and cardiovascular complications in diabetes

70% to 80% of people with diabetes died from cardiovascular complications, which had become the leading cause of death in diabetic patients. Cardiovascular complications in diabetic patients are mainly associated with vascular endothelial dysfunction and poor neovascularization. Its important influencing factors are changes in the number of endothelial progenitor cells and dysfunction. Hyperglycemia in diabetic patients can cause endothelial progenitor cell dysfunction, inhibit its proliferation ability, angiogenesis ability and paracrine effect. In order to provide research ideas for the prevention and treatment of cardiovascular complications in diabetic patients, this article aims to review the relevant mechanism of the effect of hyperglycemia on the number and function of endothelial progenitor cells in patients with diabetes, and the application of endothelial progenitor cells in the treatment of diabetic cardiovascular disease.
7.Improvment and Analysis in Experimental Technology of Microoganic Anxanography

Inoculant the examine-needed germ on a flat panel of synthetic medium, and placed paper slices, which had being saturated by different kinds of sugar, on the medium to carry out the experiment of microorganic auxanography. Its result was compared with that of the traditional method, the sugar-grain method. The comparison showed that the former was better than the latter for its more definite result and more convenient operation.
8.Cytotoxicity of pentoxifylline and its effect on human hepatoma cell line Hep3b radiosensitivity.

OBJECTIVETo investigate the effects of pentoxifylline (PTX) on radiation induced-cell cycle redistribution and radiosensitivity of human hepatocellular carcinoma cell line Hep3b.
METHODSMTT assay was performed to evaluate the cytotoxicity of PTX on p53-defective human hepatocellular carcinoma cell line Hep3b and clonogenic assay employed to observe its effects on the radiosensitivity of the cells quantified by calculating the sensitive enhancement ratio (SER). Flow cytometry was performed to observe the cell cycle changes of Hep3b cells in response to X-ray irradiation and the interventional effect of PTX.
RESULTSThe cytotoxicity of PTX on the cells increased in a dose-dependent manner following a 48-hour treatment, with the optimal dose range of 1-5 mmol/L. A sub-toxic dose of PTX at 2 mmol/L was then used in subsequent experiments. Clonogenic survival assays up to 12 Gy demonstrated that p53-defective Hep3b cells (SER of 2.68+/-0.24) were sensitized by PTX (2 mmol/L). PTX (2 mmol/L) treatment following exposure to irradiation (6 Gy) resulted in abrogation of radiation-induced G(2)/M arrest of Hep3b cells, and the proportions of Hep3b cells in G(2)/M phase were 86.8% and 14.8% after exposure to 6 Gy alone and 6 Gy plus 2 mmol/L PTX, respectively.
CONCLUSIONRadiosensitization by PTX is possibly associated with the abrogation of G(2)/M arrest in Hep3b cells following radiation exposure, suggesting that potential clinical application of PTX may enhance the efficacy of radiotherapy against hepatocellular carcinoma.

9.Effects of yinian jiangya decoction contained serum on endothelial cell proliferation and PPAR gamma mRNA expression in spontaneously hypertensive rat.

OBJECTIVETo observe the effects of Yinian Jiangya (YNJY) Decoction contained serum on cell proliferation of primary cultured endothelial cells of spontaneously hypertensive rat (SHR) and the peroxisome proliferator-activated receptor-gamma (PPAR-gamma) mRNA expression in the cells.
METHODSSD rats were fed with high-lipid diet and different doses (high, medium and low, containing crude drug of 5.2 g/mL, 2.6 g/mL, 1.3 g/mL respectively) of YNJY respectively and the serum contained different doses of YNJY (S-YNJY) was prepared by collecting the rats' serum after 20 days of feeding. The serum obtained from SD rats fed with normal diet, i.e. drug free serum (S-free) was taken for control. Primary cultured endothelial cells of SHR were treated respectively with S-YNJY in different doses (treated groups) and S-free (control group), the cell activity and the mRNA expression of PPAR-gamma in cells of all groups were detected by MTT and RT-PCR respectively at different time points: 2, 4, 8, 16 and 24 h after treatment.
RESULTSMTT test showed that at time points of 4 and 8 h, the OD value raised in the treated groups with insignificant difference among them (P>0.05), but was higher than that in the control group (P<0.05); at 16 h, it increased but showed a smaller increment in the medium-dose treated group (P<0.05); at 24 h, it decreased in all groups, but the decrement in the high- and medium-dose treated groups was more remarkable (P<0.05); at 48 h, it decreased continuously, with the decrement more significant in the high-dose group than in the control group (P<0.05). RT-PCR detection showed that at 4 h, the expression of PPAR-gamma mRNA was not changed significantly in all groups (P>0.05); at 8 h, it was remarkably lower in high-dose group than in others (P<0.05); at 16 h, it was higher in the three treated groups than in the control group (P<0.05), with a highest level shown in the high-dose treated group (P<0.01); at 24 h, the expression decreased in all groups, but the level in the low-dose treated group was still higher than that in the control group (P<0.05).
CONCLUSIONSYNJY Decoction shows a two-way regulation on endothelial cells proliferation, and which is negatively dose-dependent in the late stage. The regulation is perhaps relevant to the regulating of the PPAR-gamma mRNA expressions. The PPAR-gamma mRNA expression up-regulating and maintaining effects of YNJY Decoction may be one of the mechanisms for its vascular endothelial cell protection and blood pressure suppressing.
