2.Research on ursolic acid production of Eriobotrya japonica cell suspension culture in WAVE bioreactor.

Through scale-up cultivation of Eriobotrya japonica suspension cells using WAVE bioreactor, the cell growth and ursolic acid (UA) accumulation were studied. The comparison test was carried out in the flask and the reactor with cell dry weight (DW) and UA content as evaluation indexes. The culture medium, DW and UA content were compared in 1 L and 5 L working volumes of bioreactor. The orthogonal test with main actors of inoculation amount, speed and angle of rotation was developed to find the optimal combination, in 1 L working volume of bioreactor. DW of the cell growth and the UA content in bioreactor were higher than those of the shaker by 105.5% and 27.65% respectively. In bioreactor, the dynamic changes of elements in the fluid culture, the dry weight of the cell growth and the UA content in 1 L and 5 L working volumes were similar. Inoculation of 80 g, rotational speed of 26 r · min(-1), and angle of 6 ° was the optimal combination, and the cell biomass of 19.01 g · L(-1) and the UA content of 27.750 mg · g(-1) were achieved after 100 h cultivation in 1 L working volume of bioreactor. WAVE Bioreactor is more suitable than flasks for the E. japonica cell suspension culture, and culture parameters can be achieved from 1 L to 5 L amplification.

4.Effects of maternal cypermethrin exposure during lactation on learning and memory ability in adult female offspring.

OBJECTIVETo explore the effect of maternal cypermethrin exposure during lactation on learning and memory ability in adult female offspring, as well as the possible mechanism.
METHODSTwelve maternal mice were randomly divided into 6.25 mg/kg cypermethrin, 25.0 mg/kg cypermethrin and control groups (four dams each group, ten pups each dam, half male half female, twenty female pups each group). Maternal mice were orally administered with different doses of cypermethrin (6.25 and 25 mg/kg/d) once daily from postnatal day1 (PND1) to PND21. Maternal mice in control group were treated with corn oil. The learning and memory ability of female offspring were observed by using water labyrinth task for continuously seven days on PND60. All the female offspring were killed and the brain and hippocampus were detached after the test. The expression level of NMDA receptor NR1 protein in hippocampus was detected by Western-blotting.
RESULTSThere were no statistically significant in the difference in weight of body and brain among three groups (P > 0.05). Through the Repeated one way ANOVA, the learning time of latency in the 25.0 mg/kg cypermethrin group [(31.3 ± 17.0) s] were significantly longer than that in the control group [(21.0 ± 14.0) s] (P < 0.05). The memory time of latency in the 25.0 mg/kg cypermethrin group [(24.6 ± 21.1) s] were significantly longer than that in the control group [(14.1 ± 16.3) s] (P < 0.05). However, the difference of the wrong number among groups was not statistically significant in the test (P > 0.05).
CONCLUSIONMaternal cypermethrin exposure during lactation disturbs learning and memory ability in adult female offspring in a degree, which maybe caused by the reduction of protein level of hippocampus NR1.

6.Genotyping of HBV DNA in Shenzhen and clinical manifestations.

OBJECTIVETo detect HBV DNA and its genotypes.
METHODSThe 6 isoforms of HBV DNA was detected out using of different probes by Polymerase Chain Reaction and Nucleic Acid hybridization.
RESULTSOf 150 HBV DNA positive patients who lived in Shenzhen, 50 samples (33%) are type B, 36 samples (24%) are type C, 13 samples (9%) are type D, 3 samples is type F, 1 sample is type A, 48 samples (31%) are mixed type. The ALT value was significantly higher in genotype B than in genotype C. HBe positivity were higher in genotype B than genotype C. HBeAg positivity were higher in genotype C than in genotype B. There are not obvious relations between genotype and age or sex.
CONCLUSIONIn the detected samples, the major genotype of HBV DNA is type B, several are type C, D. The type E haven't been found. There are some relations between all kinds of genotypes and the severity of hepatitis B.

7.Quantitative monomolecular coverage of barium glass by the silane coupling agent gamma-methacryloxypropyltrimethoxysilane.

OBJECTIVETo study the monomolecular coverage of the silane coupling agent gamma-methacryloxypropyltrimethoxysilane (gamma-MPS) on the barium glass filler surface.
METHODSKubelka-Munk (K-M) function values of the isolated OH-groups on the barium glass filler surface, which were based on the change of gamma-MPS concentrations, were measured using the quantitative analysis of the diffuse reflectance infrared Fourier transform (DRIFT) spectroscopy. When all of the isolated OH-groups on the filler surface disappeared (where the K-M function values was zero), the monomolecular coverage of the gamma-MPS molecules on the filler surface was indicated by the linear regression analysis.
RESULTSThe relationship tallied with negative linear correlation between the K-M function values of the isolated OH-groups on the barium glass surface and the concentrations of gamma-MPS. Each gamma-MPS molecule occupied 0.21 nm(2) when the monomolecular coverage was formed on the barium glass surface.
CONCLUSIONSThe result of this study indicated the optimal amount of silane coupling agent on silanated barium glass filler during the production of resin composite.

8.Surveillance on the dynamics of leptospirosis epidemics in the Chongqing Section of the Three Gorges dam area.

OBJECTIVETo analyze the changes of leptospirosis epidemic characteristics before and after the Phase 2 'reservoir store water project' in Chongqing section of the Three Gorges dam area and to provide prevention, control and intervention measures to prevent the spread of leptospirosis from infectious focus to the Three Gorges dam area and downstream region of Changjiang River.
METHODSChangshou district and Fengdu county were selected as surveillance sites. We monitored the source of infection through examining the serum antibody of patients, healthy groups together with farm cattle measured by micro agglutination test (MAT).
RESULTSporadic cases were reported before and after the storage of water in the reservoir. There was no significant difference found between mouse density before and after the Phase 2 reservoir project (chi2 = 1.00, P > 0.05). The main species of rat were Sewer rat before and Insectivorea after the storage of water. The germ-carrying rate of rats was 1.72% (10/583) and positive carrying rate of rats was 16.51% (18/109) when using PCR. Results showed a significant difference when comparing it to culture method (chi2 = 51.80, P < 0.01). Positive rate of leoptopirosis appeared in the serum of patients was 73.33% (33/45) with the major serum group as the Australia group. The rate of infection among the healthy group was 26.84% (233/868). There was significant difference seen between the serum antibody positive rate of epidemic prophase (23.85%) and epidemic anaphase (29.86%) of the healthy group (chi2 = 3.99, P < 0.05). The GMRT of ox serum antibody of leoptopirosis was 29.97 with Bailen group as the predominant microbial population.
CONCLUSIONThere was no epidemics of leptopirosis occurred in the Three Gorges dam area. There was no significant difference between mouse density before and after the storage of water in the reservoir. However, the major species of rats had a change. The natural infection level of people living in the dam area was low, but there existed potential of leoptopirosis outbreak.

9.Properties of tactile responses of neurons in rat thalamic ventroposterolateral nucleus.

OBJECTIVETo determine whether the convergences of tactile information also occur at thalamic ventroposterolateral nucleus in rats, we investigated the properties of tactile responses of the thalamic ventroposterolateral nucleus in rats.
METHODSUnit responses were recorded extracellularly from thalamic ventroposterolateral nucleus in anesthetized rats.
RESULTSAmong 156 neurons examined, 140 neurons (89.7%) had the single, continual and small receptive fields, and 16 neurons (10.3%) had two discrete receptive fields. Some neurons?exhibited different responses to the same intensity stimulation which delivered to different points in their receptive fields. In addition, 4.5% neurons (n = 7) responded only to locomotive stimulation but?not to a punctiform tactile stimulation.
CONCLUSIONThe majority of neurons in ventroposterolateral nucleus of rats have the spatial, temporal and submodal characteristics of cutaneous receptors, while the minority of neurons exhibit the responses of interaction of different peripheral receptors. Therefore, it is concluded that there are convergences of tactile information at the ventroposterolateral nucleus of rats.

10.Effect of morselized bones on binding of the canine hydroxyapatite-coated femoral stem.

BACKGROUNDNon-cement femoral stems are recognized in clinical use, but there are still some problems. The aim of this research was to make non-cement femoral stems to be press-fit with the medullary cavity.
METHODSTwenty-four healthy adult mongrel dogs were randomly divided into experimental and control groups. In the right hip joint, an artificial femoral bone replacement surgery was conducted. For the experimental group, the replacement surgery of hydroxyapatite (HA)-coated femoral stems was done, while autogeneous morselized bone was implanted into the medullary cavity. For the control group, morselized bone was not implanted. At postoperative 1, 3, 6 months, a test for interfacial shear characteristics was conducted in the MTS810 Tester. The comparison between the two groups' bone-prostheses in shear strength for their interface from shearing destruction was made. A histological observation to check prosthesis-bone interface contact ratios and bone growth was carried out.
RESULTSFor the experimental group, shear strength was 0.317 MPa in 1 month, 1.447 MPa in 3 months, and 1.621 MPa in 6 months. For the control group, shear strength was 0.195 MPa in 1 month, 1.023 MPa in 3 months, and 1.483 MPa in 6 months. The difference was statistically significant. Stereomicroscope-based observation showed that the number of trabecular bones in the experimental group was larger than that of the control group, and bone growth of the former group was better than that of the latter group. Inverted microscopic observation showed that the binding degree between the prosthesis and trabecular bone of the experimental group was higher than that of the control group. Comparatively, the experimental group's trabecular bone had more stromal cells.
CONCLUSIONSThe morselized bones can effectively improve the biological bonding strength and bone-contact ratios in the short term for the HA-coated femoral stem and accelerate the bonding process. The use of morselized autogenous bones played a good role in bone in-growth of the femoral bone stem surface.
