Science and technology innovation is an important quality of young talents in the new period requirements and training objectives,as well as hospital's driving force and source of the future sustainable development.Under the innovation-driven strategy,characterized by cultivating young talents of science and technology innovation,with the efforts to develop and improve young talents cultivation way,our hospital aim to accelerate the development of young talent as soon as possible.By setting up a hospital's fund,we gradually formed a stable funding system including the four directions of clinical,nursing,management,education.With the development purposes of young researchers as the main body,adhere to improve the quality of hospital funds is the key,we aim at forming a reasonable fund scale.Through the top level design,strict process management,implementation of opening argument and a series of strong measures to ensure project quality,we promote youth science and technology innovation talents effectively.
2.Expression of microvessel density and apoptosis related factor PCNA and Bax in partial bladder outlet obstruction of rats

Objective To study the changes of detrusor microvessel density and apoptosis related factor PCNA and Bax in different phase of partial bladder outlet obstruction. Methods Forty Wistar rats were divided into control(n =10), sham-operation(n =10) , two-week obstruction( n = 10) and five-week obstruction group(n = 10). The expression of detrusor microvessel density and apoptosis related factors PCNA and Bax were evaluated with immunohistochemistry method. Results The average weight of bladder was (125.5?10. 1) ,(128.5?8.9) ,(380.0?12.4) ,(400.0?12.5) mg in control, sham-operation, two-week obstruction and five-week obstruction group, respectively. The difference between control, sham-operation group and obstruction group was significant (P 0. 05 ) . The expression of microvessel density was 2. 1?1.3,13.3?2.3,36.4?4. 1 and 37.3?5.6 in control, sham-operation, two-week obstruction and five-week obstruction group, respectively. The difference between control, sham-operation group and obstruction group was significant ( P 0. 05 ). The expression of PCNA was (38. 2?17. 2)% , (39. 4?11.4)% , (64. 1?11. 5)% and (46. 2?9. 6)% in control, sham-operation, two-week obstruction and five-week obstruction group, respectively. 3.Application of superparamagnetic nanoparticles for cancer treatment

BACKGROUND: In recent years, nanoparticles has been rapidly developing in tumor hyperthermia, genophore research, and targeted drug therapy, particularly nanoparticle containing drug delivery systems will become another breach in tumor therapy.OBJECTIVE: To summarize the application and mechanism of superparamagnetic nanoparticles for cancer treatment in the medical field.METHODS: A computer-based online search was conducted in Medline for English language publications containing the key words of "superparamagnetic, nanoparticles, targeting" from January 2000 to October 2009. Relevant articles were also searched from CNKI with the same key words in Chinese from January 2005 to October 2009.RESULTS AND CONCLUSION: A total of 123 articles about targeting role of magnetic nanoparticles were included, and there were 24 in Chinese and 108 in English. Articles published earlier, duplicated, and similarly were excluded, and 30 references were finally included. Superparamagnetic nanoparticles characterized by targeting role under external magnetic field, and crystal of ferroso-ferric oxide did not has toxicity to cells. As a gene carrier and drug carrier, superparamagnetic nanoparticles were widely used in medical research and they also provided novel evidences for cancer treatment. By an external magnetic field, how to avoid a comprehensive system of phagocytic endothelial phagocytosis and prevent the course of treatment such as drug-induced thrombus is still inadequate.
4.Radiation enhancement and induced apoptosis in ovarian cancer cells by arsenic t rioxide

Objective To study the effect of arsenic trioxide(As 2O 3) combin ed w ith radiation on the killing of ovarian cancer cells. Methods Using MTT and FCM to detect the cytotoxic and apoptosis at different As 2O 3 concentrations combined with 2 an d 8 ?Gy radiation on ovarian cancer cells(SKOV-3). Radiation survival curves were det ermined by cloning assay with 5?mol/L As 2O 3 combined with 2, 3, 4, 6, 8 and 12?Gy radiation. Curve was used to evaluate the effect of cell killing. Results ⑴ Inhibition of cell proliferation seemed more dependent on the increase of As 2O 3 concentration, ⑵Cell survival rate was lower in the combination of As 2O 3 an d r adiation than As 2O 3 alone, ⑶The apoptosis ratio was increased in 2?Gy and As 2O 3 with increase in As 2O 3 concentration, ⑷D q , D 2 value was decr eased in t he combined As 2O 3 and radiation than radiation only (D q: 1.44 vs 2.78, D 0: 0.85 vs 1.30, SF 2: 0.42 vs 0.87), with radiation enhancement ratio of 1.53 and 2.0 7 according to D 0 value and SF 2. Conclusions Arsenic trioxide is able to enhanc e radiation effect obviously ,especially at lower radiation dose.

6.NUTRITIONAL EVALUATION OF SYNTHETIC OIL——Ⅱ. Effect of Synthetic Oil on Body Weight Gain, Blood Cholesterol, Liver Weight, Liver Fat and Liver Cholesterol Level During the Inducing Period of Hypercholesterolemia

The effects of synthetic oil used as dietary fat source on the body weight, organ weight, liver and serum lipids of rats during the hypercho-lesterolemic inducing period were investigated, and comparison was made with lard and soybean oil diets.Weanling albino rats were fed first with stock diet for 3 weeks. Bloods were collected from tails, and serum cholesterol contents were determined. Based upon the body weight and serum cholesterol level, animals were then divided into 5 groups. Four groups were fed respectively with synthetic oil, synthetic oil plus linoleic acid, lard and soybean oil diets, all were supplemented with 1% cholesterol. The, fifth group served as control, was fed with soybean oil diet without cholesterol. Body weights and serum lipids (cholesterol and triglyceride) were measured at intervals during the experiment. Organ weights and liver lipids (total fat, cholesterol and triglyceride) were determined at the end of the experiment.The results showed that there was marked sex difference between male and female rats, that is, synthetic oil diet caused significant lower body weights and lower serum cholesterol levels to female but not to male rats.Cholesterol inducing diets apparently increased the liver weights of all animals, yet fat accumulation only occured in those livers of rats fed with lard and soybean oil diets. The liver fat contents of animals of both synthetic oil and synthetic oil plus linoleic acid groups were at normal level.This experiment reveals that when dietary cholesterol intake is high, the use of synthetic oil as a dietary fat source for maintaining a normal lipid metabolic condition in the body seems beneficial.
7.The Production and Teaching Application of the Internal Circulation System in the high Simulation of Pediatric Intravenous Transfusion Arm

Objective To compare the difference between traditional model teaching method and the system of internal circulation system with high simulation of the teaching method of the teaching method of the arm of the arm of the intravenous infusion in children.Methods The research objects were taken as the research object,and the model of the arm of the 2013 students in Kunming college was taken as the research object,and the traditional model was the control group.The difference between the two groups were compared between the 100 groups.Results Compared with the control group,the study group's practice skill score (87.8 ± 6.9) points was higher than that in the control group (78.5 ± 7.4) (P <0.05);the teaching cost (2.64 ± 0.4 yuan) was higher than that in the control group (12.89 ± 0.6 yuan) (P <0.05);the success rate of puncture in the study group 82.8% was higher than that in the control group (62.3%) (P <0.05) Conclusion The internal circulation system of high simulation of pediatric intravenous infusion arm model teaching method in basic nursing practice teaching can improve the teaching effect,the students teaching goal,and mobilize the enthusiasm of students learning.
8.Effect of Xianlinggubao Tablet on osteoporosis bone pain

ObjectiveTo identify the effect of Xianlinggubao Tablet on the osteoporosis bone pain and bone density in a population of 50 cases at age of 50 and above. MethodsIn a prospective study, the changes of the clinic bone pain and bone density were observed before and after 4—8 weeks treatment. Results & ConclusionsThe bone pain in 36 of 50 was relieved, but no significant difference was observed in bone density. Therefore, Xianligubao Tablet can relieve the osteoporosis bone pain, which may change the space structure of bone trabecula.
9.Fusion to Mistic improved fatty acid desaturase Fat-1 overexpression in E .coli

Objective To construct a highly efficient expression plasmid of eukaryotic nuclear membrane protein Omega 3 fatty acid desaturase gene Fat‐1 in E .coli .Methods Using molecular cloning technology to construct the recombinant prokaryotic expression plasmid pET32a Fat‐1 and pET32a‐Mistic‐Fat‐1 fused with Membrane proteins expression chaperon mistic ;the two re‐combinant plasmids were transformed into E .coli strain BL21 (DE3) ,the expression of Fat‐1 protein and M110 Fat‐1 protein in‐duced by IPTG were identified by SDS‐PAGE and gray degree analysed the amount of expression ,further identified by Western blot .Results The results of enzyme digestion and sequencing demonstrated that we successfully constructed the prokaryotic ex‐pression vectors pET32a Fat‐1 and pET32a‐Mistic‐Fat‐1;SDS‐PAGE and Western blot showed that Fat‐1 fatty acid desaturase wasn′t significantly induced ,but the overexpression of M110 Fat‐1 fusion protein was obtained in E .coli ,accounting for 15% of the total amount of whole cell proteins .Conclusion The fusion with Mistic proteins to express the Fat‐1 gene has realized the overex‐pression of eukaryotic nuclear membrane integrated protein Omega 3 fatty acid desaturase in prokaryotic host .
10.A new isolation method for peripheral blood circulating solid tumor cells with EpCAM antibody linked nanobeads

Objective To establish an isolation method for solid GTC in peripheral blood using EpCAM antibody-linked nanobeads and evaluate the sensitivity of the method and its application significance. Methods Five, ten, twenty, fifty and one hundred MCF7 (breast cancer), KYSE70 (esophageal cancer), BxPC-3 (pancreatic cancer) and 9811P (stomach cancer) cells were added into 7. 5 ml erythrocyte lysed peripheral blood obtained from healthy volunteers respectively. EpCAM antibodylinked nanobeads were used to enrich cancer cells. The recovery rates of the in vitro added cancer cells were evaluated by fluorescence microscopy. Then, the untreated thirty cases of esophageal cancer (six cases at stage Ⅰ and Ⅱ, twenty-four cases at stage Ⅲ and Ⅳ), thirty-five cases of breast cancer (fifteen cases at stage Ⅰ and Ⅱ , twenty cases at stage Ⅲ and Ⅳ), thirty cases of pancreatic cancer (five cases at stage Ⅰ and Ⅱ , twenty-five cases at stage Ⅲ and Ⅳ), thirty-three gastric cancer (thirteen cases for stage Ⅰ and Ⅱ ,twenty cases at stage Ⅲ and Ⅳ) were enrolled to enrich the peripheral blood CTC. Thirty healthy volunteers and thirty gastritis patients served as two groups of control. Meanwhile the enriched CTC was identified by IF and HE staining. FISH was used to analyze the copy number of chromosome 8 and chromosome 20 in two hundred esophageal cancer, breast cancer, pancreatic caner and gastric cancer CTC. Results After DAPI staining and mixing with 7.5 ml peripheral blood from healthy donors, the average cell recovery rates of KYSE70, MCF7, BxPC-3 and 9811P cells evaluated under fluorescence microscope were 87%, 87%, 86% and 88% (within group), and the recovery rates of 5 gradient dilution levels were 88%, 85%, 87%, 88% and 87% (intergroup). With a high sensitivity, this method was able to isolate one cancer cell in 107 white blood cells of peripheral blood. The positive rates of more than 2 CTC in the peripheral blood detected by this method were 50% (15/30) of esophageal cancer, 63% (22/35) of breast cancer, 70% (21/30) of pancreatic cancer and 61% (20/33) gastric cancer patients respectively,but no CTC was detected in the peripheral blood of healthy volunteers and gastritis patients (P = 0. 000).The aneusomy of chromosome 8 and chromosome 20 were found in 80% esophageal cancer, 75% breast cancer, 65% pancreatic cancer and 59% gastric cancer. Conclusions The CTC isolation technique with EpCAM antibody-linked nanobeads is sensitive and accurate. The aneusomy of chromosome 8 and 20 is frequent in CTC from esophageal cancer, breast cancer, pancreatic cancer and gastric cancer.
