1.Influence of Hiwigene targeting silence in chemotherapy sensitivity of breast cancer MCF-7/ADM cells

Objective:To explore the changes of chemotherapy sensitivity of breast cancer MCF-7/ADM cells after Hiwi gene targeting silence, and to illuminate the role of Hiwi gene in resistant mechanism of breast cancer MCF-7/ADM cells.Methods:The MCF-7/ADM cells were transfected with Hiwi control,siRNA1 Hiwi gene and siRNA2 Hiwi gene as Hiwi control group, Hiwi siRNA1 group and Hiwi siRNA2 group.The expression levels of Hiwi gene mRNA and protein in breast cancer MCF-7/ADM cells were detected by Real-time PCR and Western blotting methods to judge the transfection rates in various groups.After transfection,the breast cancer MCF-7/ADM cells in various groups were treated with the different concentrations(0,0.1,0.5 and 1.0 mg·L-1) of adriamycin, and the cell resistant sensitivities were detected by MTT method.Results:Compared with control group(0 mg·L-1 adriamycin), the expression levels of Hiwi gene mRNA and protein inMCF-7/ADM cells in Hiwi siRNA1 and Hiwi siRNA2 groups were significantly decreased (P<0.01).Compared with control group, the survival rates of Cells in Hiwi siRNA1 and Hiwi siRNA2 groups were significantly decreased after treated with different concentrations of adriamycin,and the survival rates in Hiwi siRNA1 and Hiwi siRNA2 groups were significantly decreased(P<0.01);they were (48.15±6.28)% and (41.73±6.17)% when the concentration of adriamycin was 1 mg·L-1,and the sensitivities to the adriamycinwere obviously enhanced(P<0.01).Conclusion:The target silencing Hiwi genes in MCF-7/ADM cells is efficient.The sensitivity of MCF-7/ADM cells to adriamycin restores after Hiwi gene silencing.
2.Action of corn filament beverage on falling blood fat in hyperlipemia rats and mice

Objective To observe the action of lowering blood fat of corn filament beverage(CFB) on hyperlipemia rats and mice.Methods Sixty rats were randomly divided into control group,alloxan model group,kanglishu capsule 0.70 g?kg-1 group,CFB 7.00,3.50,1.75 mL?kg-1 groups(n=10).Rats in each group were exhibited once a day for 21 d,on the seventh day,fourteenth day,twenty-first day, the contents of serum total cholesterol(TCHO) and triglyceride(TG) were determined,and liver malonaldehyde(MDA) level was measured.Another 60 mice were divided into 6 groups with the method mentioned above,mice in each group were exhibited once a day for 14 d,on the fourteenth day,the serum contents of TCHO and TG were determined,and liver MDA level was determined.Another 72 mice were randomly divided into control group,egg yolk model group,Expellant and Cleanning Fat Capsule 0.4 g?kg1 group,CFB 10.0,5.0,2.5 mL?kg-1 groups(n=12).Mice in each group were exhibited once a day for 10 d.16 h before the last administration,except control group,the mice in other groups were injected with 75% yolk physiological salt solution 0.5mL through the abdominal cavity,and began to starve,1 h after the last administration, the contents of serum TCHO and TG were determined,and liver MDA level was measured.Results Compared with alloxan model group,TCHO and TG in hyperlipemia rats of CFB 7.00 mL?kg-1 all reduced obviously(P
3.The role of end-tidal carbon dioxide partial pressure in fluid responsiveness assessment in septic shock patient

Objective To assess whether end-tidal carbon dioxide partial pressure (PET CO2) can predict the fluid responsiveness in septic shock patients.Methods Septic shock patients under mechanical ventilation without spontaneous breathing and with the need of a fluid challenge test were included in this study.Heart rate,central venous pressure,pulse pressure,PErCO2,and CI before and after the fluid challenge test were conducted in all the patients.Results Of the 48 septic shock patients included,34 had preload responsiveness,14 had no responsiveness.△CI and △PET CO2 after the fluid challenge test involume responders were (0.85 ± 0.47) L · min-1 · m-2 and (3.5 ± 2.5) mmHg respectively,which were higher than those in no volume responders (P < 0.05).The fluid-induced changes in PET CO2 and CI were correlated (r =0.072,P < 0.05).The AUCRoc of fluid challenge-induced △PET CO2 as the predictor for volume responsiveness was 0.943,and its sensitivity was 87.9% and specificity was 93.4% with a critical value of 5%.The AUCRoc of △PP as the predictor for volume responsiveness was 0.801,and its sensitivity was 68.1% and specificity was 73.2% with a critical value of 10%.Conclusion The changes of PETCO2 induced by a fluid challenge test can predict fluid responsiveness with reliability,and have a better sensitivity and specificity than the changes of PP.
4.Stereological study of quantitative relationship between morphological and functional changes of the lungs in blast injury in rats

Objective To observe the effects of low frequency electrical nerve stimulation on sensory and motor functioning in patients with hemiplegia and hemianesthesia caused by acute cerebral infarction.Methods Sixty-one acute cerebral infarction patients with both hemiplegia and hemianesthesia were randomly divided into a treatment group and a control group.Thirty patients in the control group received conventional treatment.Thirty-one patients in the treatment group were treated with low frequency electrical stimulation of the peripheral nerves of the affected extremities in addition to the conventional treatment.Sensory function,motor function and performance in the activities of daily living(ADL)were evaluated before and 14 days after treatment.Results Sensory function,motor function and ADL performance in both groups improved significantly over the 14 days.Furthermore,the difference between the two groups after treatment was significant.Conclusion Low frequency electrical nerve stimulation can improve sensory function,motor function and ADL performance in acute cerebral infarction patients with both hemiplegia and hemianesthesia.

Objective To investigate the sensitization to HLA antigens and relationship between sensitization and allograft function after liver transplantation. Methods The panel reactive antibody (PRA) in serum was determined by using LAT ELISA kit in 15 recipients before and after liver transplantation. Results Moderate sensitization was detected in one of 15 cases before transplantation. The patient experienced one episode of mild rejection, which was reversed by treatment with Methylprednisolone. 7.Analysis of errors of computer-assisted cervical pedicle screw installation

[Objective]To evaluate the accuracy of computer-assisted cervical pedicle screw installation and discuss the reasons of screw malposition.[Method]Totally 144 cervical pedicle screws were installed by computer-assisted navigation in 25 patients with lower cervical spinal disease.All the patients were examined by X-rays and CT after operation.Screw position and direction were measured on the sagittal and transsection image of intraoperative navigation and post-operative CT.The result was analyzed statistically.[Result]Two screws perforated the upper pedicle wall,3 deviated from the lateral pedicle wal1.There was no statistical difference in screw position or direction between navigation and CT image.Navigational images were based on the principle of rigid tissue.[Conclusion]Computer-assisted navigation can improve the accuracy of cervical pedicle screw installation.Screw malposition is responsible for the excursion phenomenon.
8.Hospital Required Infection in Patients with Acute Cerebral Infarction

OBJECTIVE To analyze the bacteria strains isolated from the patients with acute cerebral infarction and to prevent and decrease the occurrence of hospital required infection.METHODS We studied 826 patients with acute cerebral infarction in the department of neurology,investigated the origin of infection and etiological factor,and analyzed the sensitivities to antibiotics of 1127 bacteria strains.RESULTS It was common that hospital required infection occurred in the patients of acute cerebral infarction,respiratory and urinary systems were the most common sites.Klebsiella Pneumoniae was the top one in gram-negative bacteria(18.94%),Esherichia coli was the top one in gram-negative bacteria(8.23%).CONCLUSIONS Standard medical management,effective therapy to the original diseases,proper application of antibiotics play the key roles in preventing and decreasing hospital required infection.
9.Effect of ginseng combined with transplantation of bone marrow mesenchymal stem cells on functional recovery of rats with spinal cord injury

Objective To explore the effect of ginseng combined with transplantation of bone marrow mesenchymal stem cells(BMSCs)on functional recovery of rats with spinal cord injury(SCI). Methods Forty-eight female Wistar rats were randomly assigned to following four groups(n=12 per group):the sham operation group was treated by opening the vertebral lamina and exposing spinal cord without SCI;the SCI model group was reproduced by using improved Allen bump method and afterwards no treatment was given;methyl prednisolone(MP)group was treated by MP pulse treatment after SCI,including intravenous injection of MP 30 mg/kg immediately after SCI and 4 hours later the same injection was repeated,and then the same intravenous injection 2 times daily,3 days in total;The ginseng+BMSCs group was treated by orally taking ginseng ultra-microgranules 300 mg/kg after SCI,twice a day for 20 days and BMSCs 5μL(concentration 1×107 cell/μL)transplantation was carried out in SCI region on the 7th day after SCI. In the above 4 groups,the ethological observation(BBB scores)was done regularly and on the 30th day after operation,silver staining was applied to investigate the changes of spinal cord,and neuro-electrophysiological tests including somatosensory evoked potential(SEP)and motor evoked potential(MEP)were performed. Results In sham operation group,after surgery the movement of both hind limbs became temporarily sluggish and on the 7th day their functions recovered to approximately normal. In SCI model group,after injury paralysis of both hind limbs occurred,while in the MP group and ginseng+BMSCs group,different degrees of functional recovery of the injured limbs developed,and the recovery in ginseng + BMSCs group was more significant. Compared with sham operation group,after surgery the BBB score was reduced markedly at various time points in SCI model group;compared to the SCI model group,the BBB scores in MP and ginseng+BMSCs groups were increased significantly,especially more remarkable in ginseng+BMSCs group(all P<0.05),and beginning from the 12th day after operation,the difference became obvious (5.23±1.22 vs. 3.61±1.03, P<0.05). Histological detection showed that in sham operation group,the structure of spinal cord was complete,neurons distributed evenly in the gray matter and a large number of silver staining positive nerve fibers paralleled to each other and arranged regularly;in SCI model group,fragmented construction was present and the defects of gray and white matters were prominent. Compared with the SCI model group, the extents of tissue necrosis in MP and ginseng + BMSCs groups were ameliorated. The neuro-electrophysiological tests demonstrated that in SCI model group,the loss of normal wave form occurred. Compared with SCI model group,in MP and ginseng+BMSCs groups,after treatment latent periods(ms)were shortened prominently in different degrees of SEP and MEP,and their peak-to-peak values(mV)were increased obviously;the improvement in potential in ginseng+BMSCs group was greater than that in MP group〔SEP:latent period(ms):3.31±0.36 vs. 4.66±0.33, peak-to-peak value(mV):0.10±0.01 vs. 0.05±0.01,MEP:latent period(ms):3.40±0.13 vs. 4.24±0.31, peak-to-peak value(mV):41.12±0.56 vs. 16.46±2.83,all P<0.05〕. Conclusion A combined treatment of ginseng and BMSCs transplantation can effectively promote the recovery of neural function for rats with SCI.
