1.The research on teaching method of full-time medical junior college students

Objective: Strengthen the school education of full-time medical junior college students,and to explore the teaching method of them. Methods:The teachers should adjust the teaching method in view of the teaching difficult points of full-time medical junior college students, to teach students in accordance with their aptitude:1)To go into the inner world of students, in-depth understand the characteristics of students;2)Enrich the teaching method and stimulate students' interest in learning; 3)To use the PBL teaching method properly; 4)To change the traditional examination methods;5)To strengthen the training of Teachers. Results:Self-confidence of the medical college students is strengthened, learning interest is improved, and learning effect is improved obviously through the teaching methods above. Conclusion:It can provide a good reference for teaching of medical junior college students through the methods of strengthen the school education of medical junior college students, and cultivate more qualified excellent medical talents for basic medical and health institutions.
2.Analysis on vision changes and related factors of transnasal transsphenoidal pituitary adenoma resection

Objective To investigate the factors affecting the vision recovery after transnasal transsphenoidal pituitary adenoma resection.Methods The vision of the left eyes and the right eyes were compared respectively.The vision after and before the operation was also compared by using paired t-test,related factors were analyzed by multiple variable logistic regression model.ResultsThe paired t-test indicated that both eyes' visions were improved after transnasal transsphenoidal pituitary adenoma resection( P <0.01 ).The tumor diameter and the vision before operation were two factors affecting the post-operative visions by cumulative logistic regression model.The larger the diameters of the tumors were,the better the vision recoveries after operation( Wald x2 =0.047,OR =2.240,P =0.047).Worse vision lose before operation came up with a better recovery after operation ( Wald x2 =9.684,OR =0.010,P =0.001 ) ; however,age,gender,pathological category,the extent of tumor removal and the time of the vision loss have no significant affects on the vision recovery after operation.ConclusionThe transnasal transsphenoidal pituitary adenoma resection can improve the vision of the patients,and the degree which would to be improved is correlated with the tumor diameter and the vision lose before operation.
3.Application value of susceptibility weighted imaging in brain vascular malformations

Objective To discuss the clinical value of susceptibility weighted imaging (SWI) in the brain vascular malformations. Methods Imaging data of 34 patients with brain vascular malformations proved by digital subtraction angiography (DSA) or pathology obtained on Siemens Sonata 1.5T MR system were studied prospectively, and compared with those of conventional MRI (cMRI) and SWI. Results All 41 lesions of 34 patients with brain vascular malformations showed clearly by SWI. These patients were diagnosed by surgical findings or DSA. These nidus comprised 19 cavernous angiomas, 9 arteriovenous malformations and 6 cerebral venous malformations. Conclusion SWI should be used for clinical diagnosis of brain vascular malformations, and providing more complete and detailed information combining with other sequence.
4.The reverse effect of Gli2 gene silencing on epithelial-mesenchymal transition in hepatocellular carcinoma cell line SMMC-7721

Objective To explore the effect and mechanism of Gli2 on EMT and invasion in the hepatocellular car-cinoma cell line SMMC-7721.Methods shRNA of Gli2 and Negative control (NC) shRNA were constructed and transfected into SMMC-7721 cells.shRNA-Gli2 group,shRNA-NC group and blank group were set up .Transwell chambers assay , adhesion experiments were used to detect the ability of invasion ,homogeneous and heterogeneous cells intercellular adhesion of each group .Meanwhile, the qRT-PCR, Western blot were used to examine Gli2, E-cadherin ,N-cadherin and vimentin mRNA and protein expression .Results In different hepatocellular carcinoma cell lines and be along with the increasing ability of the invasion in hepatocellular carcinoma cell lines , Gli2 expres-sion was higher .Compared with the shRNA-NC group and blank control group ,the interfered group extensive cells invasion ability was inhibited ( P <0.05 ) , homogeneous cells intercellular adhesion increased and heterogeneous cells intercellular was opposite ( P <0.05 ) , meanwhile , the expression of E-cadherin was declined significantly ( P<0.05 ) ,the expression of N-cadherin , vimentin raised significantly ( P<0.05 ) .Conclusions Silencing Gli2 gene can reverse the EMT of the hepatocellular carcinoma cell line SMMC-7721 and inhibit cell invasion ,its mecha-
nism may be related to the up-regulation of E-cadherin and the dow N-regulation of N-cadherin, vimentin.
5.Gene cloning,prokaryotic vector constructing for the major allergen ovomucoid in Hen's egg white and its recombinant expression

Objective:To clone and express the gene of ovomucoid,which is the main allevgen in egg white.Methods:Using total RNA of chicken oviduct as template,the gene of ovomucoid was amplified by RT-PCR.The homology was analyzed by comparision with the sequence in GeneBank.Subsequently,the PCR product of the ovomucoid gene was cloned into prokaryotic expressing vector pET-28a and was expressed by the challenge of IPTG.Results:The whole gene of ovomucoid,one of the main allergens in egg white,was successfully cloned.The cloned ORF sequence contains 633 bp,including stop codon,encods for 210 amino acids.Sequence analysis shows that the ovomucoid gene displays 99% nucleotide identities with the published sequences.The molecule weight of ovomucoid protein obtained was 21 kD.By the challenge of IPTG,SDS-PAGE analysis showed that the ovomucoid gene was overexpressed in E.coli BL21(DE3).Conclusion:The gene of ovomucoid is cloned and overexpressed in E.coli BL21(DE3).This study will be the basis for the further research.
6.Retroperitoneal laparoscopic surgery for huge adrenal benign tumors: report of 9 cases

The clinical data of 9 patients with huge adrenal tumors (≥6 cm) undergoing retroperitoneal laparoscopic adrenalectomy between February 2015 and September 2016 were retrospectively analyzed.There were 2 males and 7 females aged 29-65 years.The tumors were detected due to investigation of paroxysmal hypertension in 4 cases, and during the health check-up in 5 cases.The lesions were located on the right side in 4 cases, on the left side in 4 cases and bilaterally in 1 case.The operations were all successful, there was no case transferring to open surgery.The average operation time was 70 min (50-100 min),mean bleeding volum was 100 ml(50-200 ml).The postoperative diagnosis: 4 cases of pheochromocytoma, 2 cases of cortical adenoma, 2 cases of neurilemmoma and 1 case of neuroinoma.The average follow-up time was 13.5 months (6-24 months) and no recurrence was found.

The effect of vascular endothelial growth factor (VEGF) overexpression on matrix metalloproteinase-2 (MMP-2) in nasopharyngeal carcinoma (NPC) cells in vitro and the possible mechanism involved were investigated, and the correlation between the expression of VEGF and MMP-2 in NPC evaluated. The NPC cells were transfected with PAd-trackVEGF165 plasmid. The expression levels of VEGF and MMP-2 mRNA and protein in NPC cells were detected by semi-quantitative RT-PCR and Western blot respectively. It was found that the expression of VEGF and MMP-2 mRNA and protein was significantly increased in NPC cells after transfection of VEGF165. It was concluded that the expression of VEGF was correlated to the in vitro invasion of NPC cells, and the induction of MMP-2 by VEGF was a key process of NPC cell invasion.

Hippo signaling pathway plays a significant role in the development of colorectal cancer,and acts as an important regulatory pathway which regulates cell proliferation and cell differentiation.Multiple proteins and genes in Hippo signaling pathway,especially the downstream YAP protein,play important roles in the occurrence,metastasis,drug resistance and recurrence of colorectal cancer.YAP and other related genes can be used as targeted markers for predicting drug resistance to colorectal cancer.Hippo signaling pathway gene network can be used as targeted therapy or combination therapy,thus providing new ideas for the treatment of colorectal cancer.
9.Transurethral Plasmakinetic Enucleation of the Prostate Combined With Suprapubic Cystostomy Rotary Resection in the Treatment of Big Benign Prostatic Hyperplasia

Objective To investigate the efficacy and safety of transurethral plasmakinetic enucleation of the prostate (PKEP)combined with suprapubic cystostomy rotary resection in the treatment of large volume(>100 ml)benign prostatic hyperplasia(BPH). Methods From June 2015 to December 2015,28 patients with BPH[volume,(112.6 ±9.8)ml]were given transurethral PKEP.The resected gland was pushed into the bladder and the vesical stoma was extended.Whereafter a 10 mm trocar was introduced via suprapubic cystostomy, and then rotary resection of glands was performed and removed under transurethral observation. Results All the operations were successfully completed.The operating time was(85.8 ±26.5)min,the enucleation time was(38.0 ±11.3)min, and the rotary resection time was(47.5 ±20.2)min.Two patients had much flushing fluid extravasated into retropubic space and were given no special treatment.Two patients suffered the complication of transient stress urine incontinence after removal of catheter,and they were recovered within 4 weeks.Follow-up for 3-6 months found no urethral stricture. The IPSS scores at 3 months after operation were(13.5 ±3.6)points. Conclusion Transurethral PKEP combined with suprapubic cystostomy rotary resection in the treatment of large BPH is safe.
10.Clinical analysis of transurethral plasmakinetic enucleation with shovel electrode in treatment of large-volume benign prostatic hyperplasia

From June 2017 to December 2018, 60 patients with large-volume prostatic hyperplasia underwent transurethral plasmakinetic enucleation of the prostate (PKEP) with shovel electrode in Luhe Hospital. During the procedure the resected glands were pushed into the bladder and the tissues were broken and sucked with STROZ Tissue Morcellator. All operations were performed successfully. The mean total operating time was (63.0±20.5) min, including (46.0±11.3)min for enucleation and (17.0±10.2) min for morcellation time. None of the 60 patients received blood transfusion perioperatively, and there was no transurethral resection prostate (TURP) syndrome. Four patients suffered the complication of transient stress urine incontinence after removed catheter, they were recovered within 4 weeks; no true urinary incontinence or urethral stricture occurred in 6 months of postoperative follow-up. Compared with the traditional technique of sheath+electric ring to enucleate the gland, the shovel electrode enucleation has shorter operation time, lower incidence of transient stress urine incontinence after operation, and reduced medical cost.
