1.Effect of Constraint-induced Movement Therapy for Motor Function of Upper Extremity after Stroke

ObjectiveTo explore the effect of constraint-induced movement therapy (CIMT) for motor function of upper extremity after stroke.Methods28 stroke patients with upper-limb hemiparesis were divided into two groups, 14 cases in each group. The observation group received constraint-induced movement therapy, treated with intensive shaping training with constraint of the unaffected arms. While the control group received routine rehabilitation with no constraint of the unaffected arms. The effect was evaluated by The Fugl-Meyer Asseseement(FMA).ResultsThe scores of FMA is higher after than before treatment in the observation group, (P<0-05). The flexibility of upper extremity of the observation group improved more significantly than that of the control group after treatment (P<0-05).ConclusionCIMT can enhance the effect of rehabilitation in the function of upper extremity and the activities of daily living of hemiplegia patients.
2.Effect of Motor Relearning Programme on Lower Limbs Function of Stroke Patients

ObjectiveTo observe the effect of motor relearning programme(MRP) training on lower Limbs function of stroke patients.MethodsSixty patients were randomly and equally allocated to training group (30 patients) and control group (30 patients). The control group was prescribed Bobath therapy while the training group received the MRP training. The lower limbs function was then assessed by the Fugl-Meyer Assessment Scale.ResultsLower limbs function improved significantly in both groups while the training group was better than the control group after three months(P<0-05). ConclusionMRP training can promote the recovery of lower limbs function of patients with stroke.
3.Effect of Physiotherapy on Knee Osteoarthritis

Objective To observe the effect of physiotherapy on knee osteoarthritis. Methods 40 knee osteoarthritis patients were dividedinto treatment group (n=20) and control group (n=20). The control group accepted conventional therapy, acupuncture and moxibustion,fumigation, massage, etc. The treatment group accepted physiotherapy in addition, including strength training and joint activity training.They were assessed with Lysholm Knee Scoring Scale (LKSS) and Visual Analogous Scale (VAS). Results The scores of LKSS and VASimproved in both groups after treatment, and improved more in the treatment group than in the control group (P<0.01). Conclusion Physiotherapycan further improve the stability of knee and release the pain in knee osteoarthritis patients.
4.Prokaryotic soluble expression, purification and function study of LEDGF/p75 protein.

HIV-1 integrase (IN) is a key enzyme for the viral replication. The protein-protein interaction (PPI) between HIV-1 IN and a cellular cofactor lens epithelium-derived growth factor (LEDGF/p75) is a validated target for anti-HIV drug discovery. In order to build the platform for screening inhibitor against PPI between IN and LEDGF/p75, the vector containing the LEDGF/p75 protein cDNA was constructed and expressed in Escherichia coli and the function of the LEDGF/p75 protein was assayed. The LGDGF/p75 encoding gene optimized according to the preference codon usage of E. coli, was synthesized and cloned into the expression vector pGEX-4T-1 to form a recombined plasmid, then transformed into host cell E. coli BL21 (DE3). The recombined clones were identified and confirmed by BamH I/Sal I digestion and sequencing, the successfully recombined plasmid in the host cell was induced by IPTG and the condition of the expression was optimized. The expressed protein was purified by the Ni2+ affinity chromatography column and SDS-PAGE was used to analyze the molecular weight and specificity. In addition, ELISA assay was used to analyze the function of the recombinant protein. The recombinant LGDGF/p75 was soluble, and expressed highly and stably in E. coli. The protein was proved to enhance HIV-1 IN strand transfer activity in vitro by ELISA. It will be helpful to build the platform of screening inhibitors against PPI between IN and LEDGF/p75.
5.Digital Image Measurement in Rhinoplasty and Its Reliability Confirmation

For developing Digital Image Expert System in rhinoplasty, the accurate and reliable Digital Image Measuring System must be first set up, and the data from the Digital Image Measuring System must be reliable with the data measured by hand. The measuring system mainly consists of a computer with an image processing board, a TV camera and a head fixing device. Ten adults were selected randomly and some indexes of head and face were measured with the measuring system and by hand respectively. The reliability of the measuring system was confirmed by the statistical analysis of data measured. Finally, the results were also discussed.
6.Effects of Ankle Intensive Training on Motor of Lower Limbs in Later Stroke Patients

Objective To observe the effects of ankle intensive training on the recovery of lower limbs motor in stroke patients in later stage of rehabilitation. Methods 30 stroke patients with course ≥ 3 months were divided into the treatment group (n=15) and control group (n=15). The control group received routine treatment, and the treatment group received ankle intensive training in addition. They were assessed with Fugl-Meyer Assessment (FMA) of lower extremity and 10 m walking speed. Results The scores of FMA and 10 m walking speed improved in both groups after treatment (P<0.05), and improved more in the treatment group than in the control group (P<0.05). Conclusion Ankle intensive training can promote the motor recovery of lower limbs in later stroke patients
7.Common problems and their countermeasures in data monitoring of No.1 Military Medical Project

Data monitoring of No.1 Military Medical Project is very important for the accuracy of information system data. It's supposed that the statistics staff monitor the data in the network. That is, the data in daily and monthly medial reports should be checked and the checking software should be applied to the examination of the data on the first page of the case history to verify their logicality and nonvacancy.
8.Isolation, idetification and anti-HIV-1 integrase activity of culturable endophytic fungi from Tibetan medicinal plant Phlomis younghusbandii Mukerjee.

A total of 52 endophytic fungi were isolated from roots and stems of Tibetan medicinal plant Phlomis younghusbandii Mukerjee. These fungal isolates were molecularly identified based on ITS sequnces and 28S sequences distributed to 12 genera, including Phoma, Chaetosphaeronema, Fusarium and Leptosphaeria, etc. Among them, the dominant genus was Phoma. Extracts of all strains were evaluated for anti-HIV-1 integrase activity by using soluable integrase expressed in E. coli BL21 (DE3). The results showed that seven samples from five fungal endophytes PHY-24, PHY-38, PHY-40, PHY-51, PHY-53, which belonged to genus Chaetosphaeronema, inhibited strand transfer reaction catalyzed by HIV-1 integrase with IC50 values, of 6.60, 5.20, 2.86, 7.86, 4.47, 4.56 and 3.23 microg x mL(-1) respectively. In conclusion, the endophytic fungi of Phlomis younghusbandii Mukerjee are valuable for further screening anti-HIV-1 integrase agents.
9.Molecular cloning and characterization of S-adenosyl-L-methionine decarboxylase gene (DoSAMDC1) in Dendrobium officinale.

S-Adenosyl-L-methionine decarboxylase (SAMDC) is a key enzyme in the polyamines biosynthesis, thus is essential for basic physiological and biochemical processes in plant. In the present study, a full length cDNA of DoSAMDC1 gene was obtained from symbiotic germinated seeds of an endangered medicinal orchid species Dendrobium officinale, using the rapid amplification of cDNA ends (RACE)-PCR technique for the first time. The full length cDNA was 1 979 bp, with three open reading frames, i.e. tiny-uORF, small-uORF and main ORF (mORF). The mORF was deduced to encode a 368 amino acid (aa) protein with a molecular mass of 40.7 kD and a theoretical isoelectric point of 5.2. The deduced DoSAMDC1 protein, without signal peptide, had two highly conserved function domains (proenzyme cleavage site and PEST domain) and a 22-aa transmembrane domain (89-110). Multiple sequence alignments and phylogenetic relationship analyses revealed DoSAMDC1 had a higher level of sequence similarity to monocot SAMDCs than those of dicot. Expression patterns using qRT-PCR analyses showed that DoSAMDC1 transcripts were expressed constitutively without significant change in the five tissues (not infected with fungi). While in the symbiotic germinated seeds, the expression level was enhanced by 2.74 fold over that in the none-germinated seeds, indicating possible involvement of the gene in symbiotic seed germination of D. officinale.
10.Absorption of flavonoids from Abelmoschus manihot extract by in situ intestinal perfusion.

To explore the mechanism of the absorption of flavonoids from Abelmoschus manihot flowers, in situ intestinal recirculation was performed to study the effect of the absorption at different concentrations and different intestinal regions. To evaluate the conditions of the absorption of six flavonoids from Abelmoschus manihot flowers, the concentrations of Abelmoschus manihot in the perfusion solution were determined by HPLC at predesigned time. And we have investigated the inhibitory effect of six flavonoids from Abelmoschus manihot flowers on P-glycoprotein (P-gp) drug efflux pump. The results demonstrated that the absorption rates of flavonoids from Abelmoschus manihot flowers are not significantly different (P > 0.05) at various drug concentrations, the absorption of flavonoids from Abelmoschus manihot flowers is a first-order process with the passive diffusion mechanism. The absorption rates of each of flavonoids are significantly different. The absorption rate of flavonoid glycoside was lower than that of aglycone; the flavonoids from Abelmoschus manihot flowers could be absorbed in all of the intestinal segments. The best parts of intestine to absorb hyperoside and myricetin are jejunum and duodenum, separately. Verapamil could enhance the absorption of isoquercitrin, hyperoside, myricetin and quercetin-3'-O-glucoside by inhibiting P-glycoprotein (P-gp) drug efflux pump.
